The Relationship between Some Blood Parameters and Antioxidant Enzyme Activity in Korean Postmenopausal Women (일부 폐경 후 여성에서 영양상태 및 골대사 관련 일부 혈액 지표와 항산화효소 활성과의 관련성 분석)

  • To elucidate the relationship between blood parameters related bone metabolism and antioxidant enzyme activity in postmenopausal period 60 women residing in Iksan area were recruited. Food and nutrient intake of each individual subject were estimated by 24-hour recalls of 3 non-consecutive days. The biochemical markers including total protein, albumin, osteocalcin (intact bone gla protein; BOP), calcium, phosphorus and hemoglobin were measured in fasting blood. In addition, parameters of antioxidative capacity including the activities of superoxide dismutase (SOD), glutathione peroxidase (GPx) , catalase (CAT) and total antioxidant capacity (TA) were monitored in blood, also. The mean age, height, weight, and BM! of subjects were 64.8 years, 151.1 em, 59.5 kg $26.0\;kg/m^2$, respectively. The mean SOD, GPx, and CAT activities were 138.5 U/ml, 1,273.8 U/ml and 314.3 kU/l respectively, and TA was 1.16 mmol/l without significant difference among different age groups. BMI was positively correlated with SOD activity (p < 0.01). SOD activity and CAT activity showed positive correlation with serum albumin (p < 0.05) and hemoglobin (p < 0.01). In conclusion, this study revealed that antioxidant enzyme activity holds a significant relationship with the blood parameters like as serum albumin and hemoglobin in postmenopausal women and further systematic research is needed to investigate the their relation mechanism.

Molecular Phylogenetic Analyses of Scyliorhinus torazame (Carcharhiniformes) Inferred from Cu,Zn Superoxide Dismutase (두툽상어(Scyliorhinus torazame) Cu,Zn-SOD의 분자 계통학적 분석)

  • Copper,zinc superoxide dismutase (Cu,Zn-SOD) plays a key role to the first antioxidant defense system against oxidative stress in diverse aerobic organisms. Due to the housekeeping action of Cu,Zn-SOD, it was reported that the structure and function have been conserved during evolution. In this study Cu,Zn-SOD from cloudy catshark Scyliorhinus torazame was subjected to phylogenetic analyses to know its evolutionary relationship in the vertebrate lineage. Molecular phylogenetic trees inferred by NJ, MP, ML and/or Bayesian analyses showed two shark species, Prionace glauca and S. torazame grouped together with high statistical supports. In general, they placed at the separated position from bony vertebrates. Thereafter, bony vertebrates composed of teleosts and birds/mammals (amniotes) formed a monophyletic group. Each teleost and amniote clade was also supported by relatively high statistical values. These phylogenetic relationships are well congruent with the phylogenetic hypothesis of the ancestral position of cartilaginous fishes to bony vertebrates.

Effects of Regular Physical exercise Habits on the Activities of Erythrocyte Antioxidant Enzyme and Plasma Total Radical-trapping Antioxidant Potential in Health Male Subjects (규칙적인 운동습관이 남자 성인의 적혈구내 항산화효소활성과 혈장 항산화능력(TRAP)에 미치는 영향)

  • In the present work we investigated the effect of regular physical exercise on the activities of erythrocyte antioxidant enzyme, plasma total radical-trapping antioxidant potential(TRAP) and plasma level of lipid peroxidation(malondialdehyde, MDA) in 64 healthy male, aged 34-67 years. The study population were divided in two groups: small amount of exerciser(exercise time less than 10min/d) and moderate amount of exerciser(exercise time more than 20min/d) according to their physical exercise habits measured by a questionnaire. Erythrocyte superoxide dismutase(SOD), glutathione peroxidase(GSH-Px) and catalase(CAT), plasma TRAP, as well as plasma MDA were determined. Erythrocyte GSH-Px and plasma TRAP were higher in moderate amount of exercisers than those in small amount of exercisers by 17% and 26%, respectively. No significant differences were observed in erythrocyte SOD, CAT and plasma MDA between the two groups. Mean exercise time was positively correlated with the erythrocyte GSH-Px activity and plasma TRAP significantly. The results would sugest that regular moderate exercise enhances antioxidant defences against reactive oxygen species and may increase the likelihood of a healthier life span.

Changes in Hematological Responses and Antioxidative Enzyme Activities of Japanese Eel Anguilla japonica Exposed to Elevated Ambient Nitrite (아질산에 노출된 뱀장어(Anguilla japonica)의 혈액학적 반응과 항산화효소의 활성 변화)

  • The study was performed to investigate changes in hematological responses and antioxidative enzyme activities (superoxide dismutase, SOD; catalase, CAT) of Japanese eel Anguilla japonica following exposure to 0 (control), 2.33, 4.60, 6,64 and 8.78 mM nitrite-N in fresh water for 48 h. Hematological parameters such as plasma nitrite, electrolytes, cortisol, glucose, glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT), hemoglobin (Hb), methemoglobin (metHb) and NADH-methemoglobin reductase (NMR) were measured. Plasma nitrite, cortisol, metHb and NMR increased directly with increasing ambient nitrite concentration, while Hb content showed a progressive decline. Levels of plasma potassium, GOT and GPT of the eel exposed to 6.64 mM ambient nitrite were significantly higher than the control fish. The activity of SOD and CAT in plasma, gill and liver of the eel following exposure to nitrite were augmented by increasing ambient nitrite. Levels of plasma nitrite, metHb, NMR, cortisol, glucose and antioxidative enzyme activities of the eel exposed to 2.33 mM ambient nitrite were significantly higher than the control fish. This study suggested that the eel acutely exposed to elevated ambient nitrite causes nitrite-induced stress responses, changes in antioxidative enzyme activities and hematological parameters.

Effects of Total Dietary Restriction on the Contents of Thiobarbituric Acid-Reactive Substance and Antioxidant Enzymes in the Liver and Kidney of Rats (절식이 흰쥐의 간과 신장의 Thiobarbituric Acid-Reactive Substance량 및 항산화효소 활성도에 미치는 영향)

  • The effects of total dietary restriction(100% restriction of energy intake) on thiobarbituric acid reactive substance(TBARS) contents and intracellular antioxidant enzymes activities in the liver and kidney of young male Sprague Dawley rats were studied. The TBARS contents were reduced in both liver and kidney, up to 77% and 79% of the control rats, fed ad libitum, respectively at 7 days after dietary restriction . Superoxide dismutase(SOD) activities in the liver and kidney of rats were increased significantly by total dietary restriction. However, the activity of catalase in kidney was decreased 27% at 6 days after dietary restriction, but this enzyme activity did not change in liver. The changes of glutathione peroxidae(GSHPx) and catalase activities in the liver and kidney of rats with dietary restriction were not significant. These result suggested that dietary restriction reduce the free radical induced by tissue damage, as determined by TBARS content, in both the liver and kidney but the changes of activities of antioxidant enzymes may not be a contributory factor in reducing oxidative damage to tissue.

Effects of Hominis placenta pharmacopuncture on the blood picture and antioxidative activity in rats (자하거(紫河車)약침이 흰쥐의 혈액성상과 항산화효소의 활성에 미치는 영향)

  • Objectives: To investigate the effects of Hominis placenta pharmacopuncture on the blood picture and antioxidative activity in rats. Methods: Sprague-Dawley rats were divided into 3 groups; normal control (n=5), pharmacopuncture at CV12 (CV12 group, n=5), and pharmacopuncture at ST36 (ST36 group, n=5) once every other day for 4 weeks. Blood cell counting was performed and liver superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) activities were analyzed. Results: Values of red blood cell and plasma cell volume were significantly higher in the ST36 group than the normal control. Values of hematocrit, total protein, and albumin were not significantly different among groups. White blood cell count and the percentage of neutrophils, lymphocytes, and eosinophils were not significantly different among groups. However, monocytes and basophils were significantly increased in the ST36, and CV12 groups, respectively. SOD and CAT in the CV12 and ST36 groups were significantly activated than in the normal control group, while the activity of GSH-Px showed no significant difference among groups. Conclusions: Based on the findings of this study, Hominis placenta pharmacopuncture may have positive impact on antioxidative capacity, thus activate various functions of the body.

Protective Effects of Sasa Borealis Leaves Extract on High Glucose-Induced Oxidative Stress in Human Umbilical Vein Endothelial Cells (혈관내피세포에서 고농도 포도당으로 유도된 산화스트레스에 대한 조릿대잎 추출물의 보호효과)

  • This study was designed to investigate the protective effects of Sasa borealis leaves on high glucose-induced oxidative stress in human umbilical vein endothelial cells (HUVECs). Freeze-dried Sasa borealis leaves were extracted with 70% methanol and followed by a sequential fractionation with dicholoromethan, ethyl acetate, butanol and water. The ethyl acetate fraction from Sasa borealis leaves extract (ESLE) was used in this study because it possessed the strongest antioxidant activity among the various solvent fractions. Exposure of HUVECs to 30 mM high glucose for 48 hr resulted in a significant (p<0.05) decrease in cell viability, glutathion (GSH) concentration, activities of antioxidant enzymes including superoxide dimutase (SOD), glutathion peroxidase (GSH-px) and catalase, and a significant (p<0.05) increase in intracellular ROS and lipid peroxidation formation in comparison to the cells treated with 5.5 mM glucose. ESLE treatment decreased intracellular ROS and lipid peroxidation formation and increased cell viability, GSH concentration and expressions of SOD and catalase in HUVECs. These results suggest that ESLE may be able to protect HUVECs from high glucose-induced oxidative stress, partially through the antioxidative defense systems.

The Anti-oxidative Activities of Fermented Hibiscus syriacus Extract in High Fat Diet-induced obese C57BL/6 Mice (고지방식이로 유도된 비만 C57BL/6 쥐에서 발효 무궁화 추출물의 항산화 활성)

  • The purpose of this study was to evaluate the anti-oxidative effects of fermented Hibiscus syriacus extract on obese mice. The antioxidant experiments were conducted both in vivo and in vitro. The in vitro experiment was measured through the use of nitric oxide (NO) free radical scavenging activity. The results showed that the scavenging activity of the NO free radical in fermented Hibiscus syriacus extract had higher activity than unfermented Hibiscus syriacus extract. The in vivo experiments were divided into a normal dietary group, a high fat dietary group (CON), a high fat dietary + fermented Hibiscus syriacus extract group (FH), and a high fat dietary + unfermented Hibiscus syriacus extract group. These four groups received intraperitoneal administrations for four weeks. Next, the levels of superoxide dismutase (SOD), catalase (CAT), and malondialdehyde (MDA) in a liver homogenate were measured. The results showed that the levels of SOD and CAT in FH were higher than the CON and MDA levels, which were significantly decreased in FH. Consequently, fermented Hibiscus syriacus extract had an inhibitive effect on oxidative stress and a heightening effect on antioxidant enzyme activity in obese mice, suggesting that fermented Hibiscus syriacus extract could be utilized as a useful, natural antioxidant.

Inhibitory Effect of Aged Black Platycodi Radix Extract on Expression and Activation of Matrix Metalloproteinases in Oxidative-stressed Melanoma Cells (쥐 흑색종 세포에서 산화적 스트레스에 의한 MMPs의 발현과 활성에 대한 흑도라지 추출물의 억제 효과)

  • The root of Playtcodon grandiflorum, called Platycodi radix, has been a favorite edible plant in Asia and contains a large amount of saponins. Melanoma cells (B16F10) were used to investigate the inhibitory effect of aged black Platycodi radix extract (ABPRE) on oxidative stress and matrix metalloproteinases (MMPs). Platycodon radix has been known to have a variety of medicinal effects such as prevention of gastric ulcers, antiallergenic activities, histamine release inhibition, and antioxidant effects. However, the mechanism of its action remains unclear in humans. ABPRE was prepared using ethanol extraction of aged black Platycodi radix. In an antioxidant effect study of ABPRE, it was observed that ABPRE specifically exhibited the scavenging activity of DPPH radical, but did not inhibit the production of malondialdehyde from lipid peroxidation. DNA oxidation was also blocked in the presence of ABPRE. In addition, ABPRE decreased the expression and activation of MMP-2 stimulated by phenazine methosulfate. Furthermore, ABPRE revealed the inhibitory effect on melanin production induced by L-dopa via antioxidant effect and the reduction of tyrosinase expression. Especially, the expression of antioxidant enzymes such as SOD-1 and SOD-2 regulated by Nrf2 was increased in the presence of ABPRE. Therefore, it appears that ABPRE may be a possible chemopreventive agent for the prevention of metastasis related to oxidative stress.

Photosynthetic Efficiency in Transgenic Tobacco Plants Expressing both CuZnSOD and APX in Chloroplasts against Oxidative Stress Caused by Highlight and Chilling (CuZnSOD와 APX를 엽록체에 발현시킨 담배식물체의 Highlight와 Chilling 스트레스에 대한 광합성 효율)

  • In order to understand the protection effects of antioxidant enzymes against oxidative stress caused by various environmental stresses, transgenic tobacco (Nicotiana tabacum cv, Xanthi) plants expressing both copper/zinc superoxide dismutase (CuZnSOD) and ascorbate peroxidase (APX) in chloroplasts (referred to as CA plants) were subjected to highlight (1,100$\mu$mol m$^{-2}$ sec$^{-1}$) and chilling at 4$^{\circ}C$. The protection effects of CA plants using leaf discs were compared with those of transgenic plants expressing either CuZnSOD or APX in chloroplasts (SOD plants or APX plants, respectively) and non-transgenic (NT) plants. CA plants showed about 15% protection in the photosynthetic efficiency (Fv/Fm) of photosystem II relative to NT plants 1 hr after treatment of both highlight and chilling, whereas they showed about 23% protection in the redox state of P700 in photosystem I at 3 hr after treatment. SOD plants or APX plants showed an intermediate protection effect between CA plants and NT plants. These results demonstrated that the coexpression of CuZnSOD and APX in chloroplasts importantly involves in the protection effects against oxidative stress caused by various environmental stresses.
