대두 단백질을 효소로 가수분해 하였을 때 생성되는 peptide의 항균활성을 조사하고 천연항균제로서 이용 가능성을 검토하기 위하여 분리 대두 단백질에 5종의 단백질 가수분해 효소를 작용시켜 생성된 가수분해물의 항균력을 측정하고 한외여과하여 분자량별로 분리된 각 fraction의 항균활성과 HPLC로 정제하여 항균성 peptide 의 아미노산 결합순서를 분석하여 다음과 같은 결론을 얻었다. 분리대두 단백질에 5종의 단백질 분해효소를 작용시켜 제조한 가수분해물 중 Asp.saitoi protease로 작용시킨 것이 항균활성이 높았다. Asp. saitoi protease로 작용시킨 대두 단백질의 가수분해물을 membrane filter로 여과한 결과 분자량 1000-3000 fraction에서 항균활성이 가장 높았다. 분자량 1000-3000 범위을 가진 가수분해물의 MIC는 0.5-0.8mg/$m\ell$ 이었으며 그람 양성균과 음성균 모두의 증식을 억제하는 경향을 보였다. 분리 대두 단백으로부터 얻어진 항균성 peptide는 121$^{\circ}C$, 10분간 열처리하여도 안정하였으며 한외여과에 의하여 분자량 1000-3000범위의 가수분해물을 동결건조하여 gel filteration하였을 때 2개의 fraction에서 항균 활성을 나타내었다. HPLC결과 RT 16.02 의 peak에서 항균활성이 확인되었고 질량은 1,633이었으며 아미노산 결합순서는 H$_2$N-G-P-P-G-V-V-A-T-V-V-A-A-R-COOH 이었다.
Antimicrobial activity of Caesalpina sappan L. extract (CS extract) against animal husbandry disease-related bacteria was studied. The CS extracts showed a significant antimicrobial activity against Gram(+) bacteria and this antimicrobial activity was most significant against Staphylococcus epidermidis. Minimum inhibitory concentrations (MIC) of CS was in the range of 0.8∼16 mg/ml and 0.8∼10 mg/ml, in the case of MeOH extracts and EtOH extracts, respectively. In addition, the antimicrobial activity of each solvent fraction was most significant with EtOAc layer. The antimicrobial activities of CS extracts against most microbial strains were stable by either heat treatment or acid treatment, whereas those against S. aureus and Sal. typhimurium were reduced by acid treatment. The inhibitory effect of CS extracts on microbial cell growth was further examined by the addition of 0, 100, 300, and 500 ppm of CS extracts into growth medium. The growth of gram(+) bacteria, S. aureus and S. epidermidis, was inhibited for 72 hours in all ranges of CS extracts added, but the growth of gram(-) bacteria was only inhibited when at least 100 ppm of CS extracts were added. Taken together, the antimicrobial activities of CS extracts were more effective against gram(+) bacteria compared to those against gram(-) bacteria.
The aim of this study was to evaluate the antimicrobial activities of leek and garlic extract juice and powder against Escherichia coli, Salmonella enteritidis, and Staphylococcus aureus in a single strain and a mixed strains system. Garlic juice and powder showed higher antimicrobial activity against Sta. aureus than that of E. coli or S. enteritidis. The antimicrobial activities of the leek and garlic powders decreased with increasing temperature and time, but stabilized at various pHs. The antimicrobial effects of mixtures of various ratios between the leek and garlic extracts increased with an increase in garlic extract content. No synergistic effects by the leek and garlic mixtures were observed. In a mixed strains system, the antimicrobial effects of leek and garlic powder were similar to those in a single strain system.
Methylelaiophylin generated superoxide radicals in Bacillus subtilis and showed antibacterial activity against a broad range of gram positive bacteria. The inhibition of DNA synthesis was more sensitive than one of RNA synthesis. A recombination-deficient mutant strain of B. subtilis was 2-fold more sensitive than a wild strain, and this sensitivity was reduced in the presence of an antioxidant, dithiothreitol. Methylelaiophylin generated superoxide radicals in B. subtilis lysates, and this suggests that the antibacterial activity of methylelaiophylin is related to the generation of active oxygen species in the cells.
The antifungal activities of 153 extracts from 51 species of Korean mushroom on six phytopathogenic fungi were investigated. The powder of fruit-body of each mushroom was extracted with petroleum ether, 80% ethanol and distilled water. The water extracts of four mushrooms including Amanita virosa showed antifungal activities on Alternaria alternata, Colletotrichum graminicola, Fusarium solani, Idriella bolleyie. The 80% ethanol extracts of seven mushrooms including Boletus auripes showed antibiotic activities against A. alternata, C. gramincola, Cylindrocarpon destructans, F. solani, F. oxysporum cucumerinum, and I. bolleyie. The petroleum ether extracts of six mushrooms including Amanita citrina showed antibiotic activities against A. alternata, C. destructans, and C. graminicola. The 102 extracts of 34 mushrooms including Agaricus arvensis didn't show antibiotic activities.
Various separation techniques of antimicrobial compounds from food materials have proven to be ineffective, involving liquid-liquid extraction and chromatography. We established efficient separation procedure for partially purifying antimicrobial compounds from Korean leek. Separation procedures consisted of ultrafiltration, liquid-liquid extraction, TLC and HPLC. Molecular weight of antimicrobial compounds were less than 5,000 dalton. Major compounds were propenyl methyl disulfide, S-methyl methylthiosulphonate and dimethyl disulfide.
The antimicrobial activity of Orixa japonica Thunb. leaf extract towards 13 microorganism strains was evaluated. Both methanol (MEex) and 70% ethanol extracts showed antimicrobial activity towards Streptococcus mutans, Bacillus cereus, Staphylococcus aureus, and Pseudomonas aeruginosa. MEex showed a higher antimicrobial activity than the 70% ethanol extract. In addition, the dichloromethane fraction (DCMfr) of the MEex also had an antimicrobial effect against the microorganisms examined. The minimum inhibitory concentrations (MICs) towards S. mutans, B. cereus, S. aureus, and P. aeruginosa were 49.22, 24.61, 49.22, and 49.22 mg/mL, respectively. In contrast, the MICs of the DCMfr tpwards S. mutans, B. cereus, S. aureus, and P. aeruginosa were 3.31, 0.21, 1.7, and 1.7 mg/mL, respectively. The MEex antimicrobial activity was not affected by a 3 h exposures to pH in the range of 3-11 or by temperatures were maintained between $80^{\circ}C-100^{\circ}C$ for 6 h. However, the MEex antimicrobial activity decreased at a heat treatment of $121^{\circ}C$ 1 h.
A lactic aci bacteria producing antibacterial substance was isolated from pig feces. This strain was identified as a genus Lactobacillus, through its morphological, cultural and physiological characteristics. Lactobacillus sp. KJ-5 isolated showed the strong inhibitory effect on the growth of Salmonella paratyphi. The production of antibacterial substance was growth associated form during the batch culture of Lactobacillus sp. KJ-5 and the maximum production was obtained at the culture temperature of $37^{\circ}C$ as well as optimum temperature of cell growth. The antibacterial activity of the filtrate of culture broth was decreased by adjusting the pH 6.2 and was not affected by catalase treatment. The antibacterial substance was partially purified by methanol and acetone extraction, whtch exhibited three spots in the thin-layer chromatography and one of them showed an antibacterial activity, This substance also showed the maximum absorption of UV at 270nm and an antibacterial activity was completely inactivated by the treatment of proteolytic enzymes.
This study was carried out to get basic resources for the industrial use of Lactobacillus bulgaricus SP5. The antibacterial activity of the supernatant obtained from Lactobacillus bulgaricus SP5 was tested against the pathogenic bacteria such as Escherichia coli KCCM 11234, Salmonella enteritidis KCCM 3313, Salmonella enteritidis KCCM 12021, and Salmonella typhimurium KCCM 40253. The supernatant of L. bulgaricus SP5 showed antibacterial activity against tested pathogenic bacteria. The antibacterial activity was examined after adjusting pH and heat treatment of supernatant. Heat treatment of supernatant had antibacterial activity against pathogenic bacteria at all temperature. However, pH changes showed no antibacterial activity. Antibacterial activity of the supernatant was confirmed to be due to organic acids (lactic, acetic, phosphoric, succinic, pyroglutamic, citric, malic, and formic acid).
Antimicrobial activity of Korean leek was effective on Staphylococcus aureus, Bacillus cereus, Bacillus subtilis, E. coli O157:H7, Pseudomonas fluorescens, Pichia membranaefaciens. Extract from raw Korean leek and that from frozen one showed antimicrobial activity; in contrast, extract stored frozen condition or extract from blanched one lost antimicrobial activity. Filtration, centrifugation, and vacuum concentration did not affect antimicrobial activity of the leek, nor did the changes in storage pHs. Addition of ground leek to soy milk, soy curd, and broth for cold noodle resulted in the prolonged shelf life by significantly reducing the number of viable cells.
