This study was performed to examine analytical method of a quinolone compound, oxolinic acid in paddy soil by HPLC coupled with UV detector. Two types of soil texture in different regions were used for this experiment. Oxolinic acid was extracted by a 4 M-KOH : MeOH(1 : 3, v/v) mixtures and acidified followed by liquid-liquid partitioning in dichloromethane. Dichlormethane layer was dehydrated, evaporated and analyzed by HPLC (262 nm). Retention time was 10.2 min. The standard calibration curve of oxolinic acid showed linearity ($r^2>0.999^{**}$, y=378.99x+135.08) in the range of $1{\sim}40$ ng. The mean recoveries, evaluated from fortified soil samples at two concentration levels of 0.2 mg/kg and 1.0 mg/kg, were $90.9{\pm}4.52%$(C.V. 4.97%) and $95.0{\pm}0.23%$(C.V. 0.24%) for soil 1 and $92.2{\pm}1.15%$(C.V. 1.25%) and $93.1{\pm}0.31%$ (C.V. 0.33%) for soil 2, respectively The detection limits of two types of soils were same as 0.05 ppm. Overall, the present analytical method of oxolinic acid by HPLC coupled with UV detector seems to be used reasonably.
The study researched germination of the plants and growth of Fungus according to concentration of aqueous extracts in order to provide basic data for developing natural agricultural resources by using Persicaria longiseta. The seed germination of Amaranthus spinosus was inhibited at 25% P. longiseta extract, while Agrostis stolonifera ssp. palustris was not affected at all concentrations tested. Especially, the seed germination rate and fresh weight of Trifolium incarnatum at 20% P. blumei extract were higher than those of control plot. The early growth of most receptor plant seedlings was promoted at 25% and 50% of P. blumei extracts, but the radicle growth of all receptor plants was significantly inhibited at > 25% of P. longiseta extract. The response of receptor plants to P. longiseta extract was different according to the plant species and the plant parts. The growth of plant pathogenic fungus in PDA medium showed an increasing inhibition tendency with increasing concentrations of P. longiseta extract. Especially, P. longiseta extract showed the greatest antimicrobial activity against Phytophthora infestans, Phythium graminicola, and Pythium venterpoolii. The content of total phenolic compound in P. longiseta was higher in leaves (1082.3 mg/L) but lower in roots (228.6 mg/L) and stems (207.8 mg/L), which is an allelopathic chemical. As these results are summarized, P. longiseta have competitive advantage because they release phenolic compounds with allelopathic effect and affect on germination, growth and fungi growth on underground flora compared to native plants and they have eligibility for natural herbicide and germicide.

We investigated characterization as cosmetic substances of chestnut inner skin extracts with in vitro antioxidant activity. Total phenolics of various extracts from chestnut inner skin were the highest 60% methanol (164.82 mg/g), and ethyl acetate fractions (191.14 mg/g). We found that the both samples from chestnut inner skin dose-dependently increased in vitro antioxidant activities (DPPH radical scavenging activity and inhibition of lipid peroxidation). In addition, the both samples also showed a strong UV absorption in the range of UV-B (290-320 nm). Especially the 60% ethanol extracts presented higher inhibitory effect on elastase (46.40% at $100{\mu}g/mL$) than that of the ethyl acetate fractions, so that it showed in vitro anti-wrinkle activity. Finally, the 60% methanol extracts and ethyl acetate fractions showed anti-bacterial activity against skin pathogenic bacteria. Consequently, these results suggest that the chestnut inner skin can be used for cosmetic industry.

This study aimed to analyze the content and composition of a biological amine, cadaverine, isolated from the venom of worker honeybees (Apis mellifera L.). This biological amine―which has diverse functionality, such as anti-inflammatory and antibacterial effects―has not been previously reported in bee venom. An assay completed in 13 minutes was developed for the cadaverine present in the bee venom using an ultra-performance liquid chromatograph and a Halo C18 column with acetonitrile and water as the mobile phase. The specificity, accuracy, and precision of the assay were verified, and the assay was validated. The linearity for cadaverine in the bee venom was R2=0.99 or above, indicating a moderate level. The limit of detection and limit of quantification were both 0.3 ㎍/ml, and the rate of recovery was 97.6%-99.1%. The relative standard deviation (RSD) of the intra-day precision and inter-day precision for cadaverine was 0.25%-0.44% and 0.25%-1.25%, respectively, with an RSD that fell within 5% indicating excellent precision. Through this novel assay, it was found that the mean content of cadaverine was 1.10±0.05 mg/g. Our results indicated that the linearity, limit of detection, limit of quantification, and rate of recovery of the cadaverine assay were of a satisfactory level, and the cadaverine content of the bee venom was ably determined. This study provides basic data on cadaverine in bee venom, which will prove useful in further studies on the bioactivity of this component.
Among various health functional foods, probiotics constitute the largest market. The interest in probiotics is increasing continuously according to the research results that gut health can control the immune function of the body, prevent diseases, and assist in treatment. In this study, dairy products and dressing sauces were developed using Ficus carica vinegar (FV), and their effects on colon cells were analyzed. When 5% FV was added to regular milk, the satisfaction with the resulting yogurt and ricotta cheese was high. The dairy product was Leuconostoc lactis, and the number of bacteria was more than 1.0×107~1.0×108 CFU/mL. The product satisfied the health food standards as probiotics. An examination of the cell viability of Caco-2 cells, which proliferate similarly to human intestinal epithelial cells, revealed an approximately 19% increase in the proliferation rate when treated with whey at 10%. An antioxidant activity of up to 58% was recorded when the cells were treated with whey at various concentrations. In addition, excellent adhesion was observed for L.latis isolated from whey. This study confirmed that dairy products made using traditionally fermented FV assist intestinal health effectively as the microbiome.

This study was carried out to investigate the feeding effects of dietary supplementation of fermented spent mushroom substrates (F-SMS) from Pleurotus eryngii with Bacillus subtilis CS21 and Saccharomyces cerevisiae on Hanwoo steers. The cellulase and xylanase producing bacteria, designated CS21, was isolated from freshly spent mushroom substrates from Pleurotus eryngii and used as probiotics to fermented spent mushroom substrates. Twenty Hanwoo steers were allocated into two feeding groups and assigned equally to two dietary treatments; Control (TMR) and TMR including 30% F-SMS (30% F-SMS TMR). Total gain and feed intake was significantly greater in the 30% F-SMS TMR than control (p<0.05), but carcass grades were not influenced by the experimental diets. Based on this study, fermented spent mushroom substrates from Pleurotus eryngii with B. subtilis CS21 and Saccharomyces cerevisiae is able to use as an ingredient feed in TMR for Hanwoo steers.

This study was carried out ot investigate the effects of fetal calf serum (FCS) and gonadotropins supplemented to the medium on maturation and fertilization in vitro of porcine follcular oocytes. Ovaries were obtained from gilts at local slaughter-house. Oocyte-cumulus complexes were recovered by puncturing the ovarian follicles(3~5 mm in diameter). The complexes from individual ovaries were pooled in a $0.4m{\ell}$ droplet of medium covered with paraffin oil, then washed twice in fresh droplet and cultured for 36hrs in culture media according to experimental conditions. Boar epididymal spermatozoa were capacitated by preincubation for 4hrs in m-KRB medium and the preincubated spermatozoa were insemenated in the fertilization medium containing the cultured oocytes. The results obtained in this study are summarized as follows: 1. The maturation rates of oocytes cultured in m-KRB and m-KRB supplemented to 10% FCS were 82 and 37%, respectively. When PMSG, hCG. and PMSGt hcG($10Iu/m{\ell}$) were added to the media supplemented to 10% FCS, the maturation rates were 66, 58 and 68%, respectively. 2. Expansion of cumulus cells was not occured in m-KRB and m-KRB supplemented to 10% FCS. However, when PMSG, hCG and PMSG+hCG($10Iu/m{\ell}$) were added to m-KRB supplemented to 10% FCS, the expansion rates of cumulus cell layers were 92, 13 and 91%, respectively. 3. When oocytes were mltured in m-KRB, the rates of penetration and formation of male pronucle: were 93 and 7%, respectively. By adding FCS and gonadotropin to m-KRB, the penetration and formation of male pronuclei were 100 80%, respectively.
Three experiments were conducted to determine utilization of Song-Gang(equation omitted) stone as the dietary additive for growth and immune stimulant in juvenile olive flounder. In the feeding trial, four diets were formulated to contain 0, 0.5, 1.0 and 2.0% Song-Gang(equation omitted) stone per kg diet (SGS$_{0}$ , SGS$_{0.5}$, SGS$_{1.0}$ , SGS$_{2.0}$ ). Fish averaging 5.0$\pm$0.04 g (mean$\pm$SD) were fed one of four experimental diets in triplicate groups for 8 weeks. There were no significant differences in weight gain, feed efficiency, protein efficiency ratio, hematosomatic index, condition factor and survival among fish fed all the diets. In chanlenge test, fish were infected by intraperitoneal injection of 0.1 $m\ell$ bacterial suspension with Edwardsielia tarda per fish after the feeding trial. Fish fed SGS$_{0.5}$ diet have a significantly higher cumulative survival rate than did fish fed the other diets (P<0.05). In the anti-mold test, Asprrgiilus niger, Penicillium pinophiltfm, Chaeromium globosum were inoculated with Song-Gang(equation omitted) stone using ASTM G-21 method. The amount of Α. niger, Ρ. pinuphiium. C. globosum didn't increase in Song-Gang(equation omitted) stone for 4 weeks for the test period. Therefore, these results indicate that 0.5% Song-Gang(equation omitted) stone per kg diet could increase immune resistance in juvenile olive flounder and Song-Gang(equation omitted) stone could be used as the anti-mold additive in fish feed.
The study was conducted to obtain some genetic information for breeding of mice. Average performance, heterosis and genetic variance were estimated with 362 progenies from a full diallel cross of four lines of mice (BALB/c, CBA, C3H and C57BL). The progenies were reared at the Experimental Animal Farm. College of Agriculture, Kyungpook National University from November, 1984 to February, 1985. Data for litter size. sex ratio, body weight and weaning rate were analyzed into heterosis effects, and genetic variance with Hayman's model. The results obtained are summarized as follows : 1. Average performance was 7.54 in litter size, 53.20 % in sex ratio, 1.55 g in birth weight, 10.45 g in weaning weight and 94.13 % in weaning rate. 2. The estimated heterois was 6.97% in litter size, 7.26 % in sex ratio, 6.08% in birth weight, 3.54% in weaning weight and 2.05% in weaning rate, respectively. 3. Additive gene effects were not observed in litter size, sex ratio, birth weight, weaning weight and weaning rate. In litter size and weaning weight, dominance effects were observed, which were shown due to individual crosses in litter size and were shown mean dominance effects of parental line in weaning weight. Mat-Maternal effects appeared in birth weight and weaning weight, and reciprocal effects were observed in weaning weight.
In order to investigate histopathologically the nature of varietal resistance and infection process of the rice bast fungus, Pyricularia oryzae. this experiment was undertaken by using the resistant cultivar Tongil and susceptible cultivars Norin No.6 and Jinheung in 1973 to 1974. 1) It was found that appressorium of the fungus forms not only at 4 hours after incubation under the favorable conditions but also peak of appressorial formation is at 48 hours treatment. Physical stimulus known to be a definite factor for appressorial formation. The optimum temperature range for appressorial infection was at $24^{\circ}C$ to $28^{\circ}C$, and pH was between 4.8 to 8.0 with 6.8 as the optimum. 2) Although percent of appressorial formation on the leaves of resistant Tongil and susceptiible Norin No.6 were only slightly different, there was a remarkable difference between resistant and susceptible cultivars with regard to percent of hyphal infection and index for hyphal extension. Index of hyphal extension was 1.6-2.7 in Tonyil. while in susceptible cultivar was 3.4-6.6. The rate of discoloration of infected cells, a indication of hypersensitivity, was greater in the resistant than in susceptible cultvar. 3) Therefore, it could be concluded that resistance of Tongil cultivar may be attributable to a higher degree of mechanical barriers as well as a higher level of antifungal substance accumulation.
