Isolation and characterization of antifungal violacein producing bacterium Collimonas sp. DEC-B5 (항진균활성 violacein 색소를 생산하는 Collimonas sp. DEC-B5 균주의 분리 및 특성)

  • Forty-nine pigments were extracted from the collections of 106 pigment producing bacteria from the plant rhizosphere soil. Antibacterial activity test was performed in the subjects of the extracted pigments with plant pathogenic bacteria including Xanthomonas axonopodis and Xanthomonas campestris, and with plant pathogenic fungi including Botrytis cinerea, Colletotrichum acutatum, and Fusarium oxysporum. The yellow pigment by Chryseobacterium sp. RBR9 and the red pigment by of Methylobacterium sp. RI13 showed the antibacterial activities against Xanthomonas axonopodis and Xanthomonas campestris. The violet pigment by Collimonas sp. DEC-B5 showed the antibacterial activity as well as the antifungal activities against Botrytis cinerea and Fusarium oxysporum. Especially, the violet pigment inhibited the growth of Botrytis cinerea more than 65% at MIC $20{\mu}M$. Upon the HPLC analysis result for the isolation of pigment with antifungal activity, violacein (91.6%) and deoxyviolacein (8.4%) were isolated for the pigment by Collimonas sp. DEC-B5. The production amount of the pigment was increased more than 10 times higher when D-mannitol 1.5% and yeast extract 0.2% were added as the nitrogen source to SCB medium. This study suggests that produced violacein by Collimonas sp. DEC-B5 will be effective to control strawberry gray-mold rot fungi by its preventive activity.

Cloning and Identification of Differentially Expressed Genes Induced by Fungal Infection from Silkworm, Bombyx mori (누에에서 곰팡이(Aspergillus niger) 감염에 의해 유도 발현되는 유전자의 클로닝과 동정)

  • We tried to identify differentially expressed genes (DEGs) from a silkworm, Bombyx mori, involved in fungal (Aspergillus niger) infection. A total RNA purified from fungal-induced and normal B. mori ($5^{th}$ instar larvae) was used for the cDNA synthesis. Differentially expressed genes were screened by annealing control primer (ACP)-based PCR technique. Comparing the gene expression profiles between fungal infection and control silkworm, we detected 10 genes that were differentially expressed in fungal induction and performed molecular cloning and nucleotide sequencing of the 10 genes. We confirmed the expression patterns of 3 DEGs by RT-PCR. The 3 DEGs over-expressed in fungal infection were identified as lysozyme, enbocin and an unknown gene. They were first identified to be genes induced by fungal infection. Although the detailed functions of 3 genes and their products remain to be determined, the genes will provide insight into the molecular mechanisms of insect-immune systems induced by fungal infection.

Studies on the Antimicrobial Activities of Prunus mume (매실의 항미생물 활성 연구)

  • This study was conducted to investigate the antimicrobial activity of Prunus mume extracts. Prunus mume extracts have growth inhibitory activity to various microorganisms. Gram positive bacteria such as Bacillus cereus, Bacillus subtilis, Micrococcus leteus, Staphylococcus epidrimidis were more easily inhibited than Gram negative bacteria tested Vibrio parahaemolyticus, Salmonella typhimurium, Escherichia coli, Proteus vulgaris and Yeasts by Prunus mume extracts. Among several fractions of methanol extract, EtOAc & BuOH fractions were showed strong antibacterial activities, but those fractions were not showed on fungi.

Cosmetic Effects of Dietary Fiber from Mozuku, Cladosiphon novae-caledoniae Kylin (큰실말(Cladosiphon novae-caledoniae Kylin) 부산물의 화장품소재 특성)

  • To investigate the effect of dietary fiber from mozuku, Cladosiphon novae-caledoniae kylin (C. novae-caledoniae kylin) on the skin care, we measured anti-oxidant activity, anti-microbial activities, tyrosinase activity inhibition and elastic activity. B16F10 melanoma cell (MTT assay) were used to measure cell viability. MC and MI exhibited in vitro antibacterial activity against Staphyloccus aureus (S. aureus) and MRSA without antifungal activity. Mozuku extract (MS) showed excellent tyrosinase inhibition effect compared to arbutin as a positive control (to 49% for tyrosine). The wrinkle-improving effect was relatively low. However, wrinkle-improving effect was relatively low. DPPH free radical scavenging activity was 89% in a concentrations at $500{\mu}g/mL$. These results indicate that the mozuku extracts may be an effective cosmetic ingredient for skin whitening.

Antibacterial and Whitening Activities of Coffea arabica Ethanol Extract (커피 에탄올 추출물의 항균 및 미백활성)

  • In this study, Coffea arabica ethanol extract (CAE) was tested for possible functional cosmetic agent. Whitening effect was measured by tyrosinase inhibition assay, and anti-oxidant activity was checked by SOD-like activity. SOD-like activity of CAE showed $94.8{\pm}6.2%$ at $500{\mu}g/mL$. The anti-bacterial activities CAE was evaluated against three different gram-positive bacteria and six gram-negative bacteria including MRSA strains. CAE exhibited in vitro broad spectrum antimicrobial activities of gram-negative bacteria without antifungal activity. CAE was strong exhibited against MRSA CCARM3561. The tyrosinase and L-DOPA inhibitory activities of the CAE lower than those positive control arbutin. CAE reduced melanin contents of B16-F10 melanoma cell in a dose dependent manner and decrease about 89.2% at a concentration $100{\mu}g/mL$. These result highlight the potential of coffee extract as a naturally active and non-toxic antibacterial suitable for cosmetic applications.

  • The ethyl acetate (EtOAc) extracts from needles of Pinus densiflora were showed antimicrobial activities against bacteria, yeast and fungi. The antimicrobial active substance of EtOAc extracts were successively purified with solvent fractionation, silica gel adsorption column chromatography and Sephadex LH-20 column chromatography. The purified active substance was isolated as crystals and identified as benzoic acid by $MS,\;^{1}H-NMR\;and\;^{13}C-NMR$. The amount of benzoic acid was 0.608 mg per gram of fresh needle of Pinus densiflora.

  • The methanol extracts of Aralia elata bark showed antimicrobial activities against bacteria, yeast, fungi. The solvent fractionated acidic fraction that showed the activity was then successively purified with silica gel adsorption column chromatography, Sephadex LH-20 column chromatography, silica gel partition column chromatography, HPLC and TLC. The isolated major active substance was identified as 3,4-dihydroxybenzoic acid by MS, GC-MS, IR, $^{1}H-NMR\;and\;^{13}C-NMR$. The content of 3,4-dihydroxybenzoic acid was 0.869㎎/g in dried bark of Aralia elata.

  • The EtOAc extracts from needles of Pinus densiflora showed antimicrobial activities against bacteria, yeast and fungi. The antimicrobial principle was successively purified by solvent fractionation, silica gel adsorption column chromatography and Sephadex LH-20 column chromatography. The active substance was further purified by HPLC using $C_{18}$ column. The active substance was identified as trans-cinnamic acid by MS, $^{1}H-NMR\;and\;^{13}C-NMR$. The amount of cinnamic acid was $9.27\;{\mu}g$ Per gram of fresh needle of Pinus densiflora.

Optimum Conditions for Production of Mevinolin from the Soybean Fermented with Monascus sp. (홍국균(Monascus sp.) 발효콩의 mevinolin 생산 조건)

  • Soybeans were fermented with Monascus sp. to select strain with highest mevinolin production through solidstate fermentation. Monascus pilosus IFO 480 showed highest yield of 2.2 mg mevinolin per g dry weight without citrinin, toxic fungal secondary metabolite, as byproduct. Nutrient broth used for soybean fermentation with Monascus sp. was composed of 3.4 rice powder, 1.1 peptone, 2.6 glycine, 12.9 glucose, initial pH 6.0 (%, w/v). Mevinolin was present in fermentation substrate largely as hydroxy carboxylate form (open lactone, 91.8-96.8%), which is used as hypocholesterolemic agent. Production of mevinolin continued up to 50 days fermentation time at $30^{\circ}C$.
