Inhibition of Proliferation and Neurogenesis of Mouse Subventricular Zone Neural Stem Cells by a Mitochondrial Inhibitor Rotenone (미토콘드리아 억제제 rotenone에 의한 쥐의 뇌실 하 영역 신경 줄기 세포의 증식과 신경 세포로의 분화 억제)

  • Mitochondria have multiple functions in cells: providing chemical energy, storing cellular $Ca^{2+}$, generating reactive oxygen species, and regulating apoptosis. Through these functions, mitochondria are also involved in the maintenance, proliferation, and differentiation of stem/progenitor cells. In the brain, the subventricular zone (SVZ) is one of the neurogenic regions that contains neural stem cells (NSCs) throughout a lifetime. However, reports on the role of mitochondria in SVZ NSCs are scarce. Here, we show that rotenone, a complex I inhibitor of mitochondria, inhibits the proliferation and differentiation of SVZ NSCs in different ways. In proliferating NSCs, rotenone decreases mitosis as measured through phosphorylated histone H3 detection; moreover, apoptosis is not induced by rotenone at 50 nM. In differentiating NSCs, rotenone blocks neurogenesis and oligodendrogenesis while glial fibrillary acidic protein-positive astrocytes are not affected. Interestingly, in this study there were more cells in the differentiating NSCs treated with rotenone for 4-6 days than in the vehicle control group which was a different effect from the reduced number of cells in the proliferating NSCs. We examined both apoptosis and mitosis and found that rotenone decreased apoptosis as detected by staining cleaved caspase-3 but did not affect mitosis. Our results suggest that functional mitochondria are necessary in both the proliferation and differentiation of SVZ NSCs. Furthermore, mitochondria might be involved in the mitosis and apoptosis that occur during those processes.

  • To assess the ability of an extravasation defection accessory (EDA) to detect the clinically important extravascular Injection of iodinated contrast material that was delivered with an automated mechanical power injector. The purpose of this study was to assess the ability of this device during clinically important episodes of extravasation. The EDA system was composed of a strain gage, an amplifier and a computer-based system. In the rabbit experimental cases, there were seven true-positive cases (range of the extravasation volumes: $14{\sim}23 ml$). The algorithm results showed seven true-positive cases (range of the extravasation volumes: $7{\sim}16ml$), nineteen true-negative cases, two false-positive cases and no false-negative cases. The EDA system had a sensitivity of 100% and a specificity of 90% for the detection of clinically important extravasation. The EDA system had good sensitivity for the detection of clinically important extravasation and the EDA system has the clinical potential for the early detection of extravasation of the contrast medium that is administered with power injectors.

  • KIST 6MV 가속기는 이온빔 분석 그리고 가속기 질량 분석법(Accelerator mass spectrometry)으로 활용된다. 이온빔 분석으로는 RBS, TOF ERD, PIXE. ${\mu}$-Probe을 할 수 있으며 AMS(Accelerator mass spectrometry)는 액체섬광측정법(LSC)과 비교할때 민감도는 1,000배 정도로 3H, 14C, 26Al,41Ca 을 10-21 ~ 10-18 mole/mg 까지 검출 가능하여. 응용분야로는 BAMS(Biological AMS), 전통과학, 지구과학, 환경과학에 활용되고 있다. 이중 AMS의 생-의학분야(BAMS)의 응용은 최근 매우 중요하게 연구되고 있다. BAMS의 활용 연구에 사용하는 핵종으로는 주로 3H, 14C, 41Ca, 36Cl를 사용하며, 14C 화합물은 쉽게 구할 수 있고, 자연방사선 이하의 낮은 14C labeled drug 사용하기 때문에 1948년 이후 생물학 연구에 혁신적으로 활용되고 있다. 주 활용분야로는 (1) 신약개발은 임상실험 전(Phase 0) 이용되며, 14C로 표지된 bio-molecule을 자연수준의 방사선 농도에서 추적자로 사용하여 질량을 측정하는 방법을 활용. (2) 의과학분야는 인체 내에서의 추적자 연구수행 (3) 항암제 연구는 암조직 중 약물농도와 암효과의 상관성을 연구 (4) 바이오 기술 분야에서는 생약 유효물질의 체내 대사연구 등을 할 수 있어 전세계적으로 활발히 연구가 진행되고 있다. KIST에서는 6MV가속기를 BAMS 연구에 활용하기 위하여 전처리 단계의 Combustion, Gas transfer, Reduction 등을 자체 제작하여 테스트 중에 있으며, BAMS 샘플의 Gas는 호기중의 성분, 대기 성분이 있으며, Liquid는 혈액(혈장,혈청,적혈구), Solid는 DNA, 세포, 장기 뼈, 피부, 식물조직, 사료, Drug 및 그 대사 류가 있다. AMS 측정 결과는 14C/12C 비율로 나타나며 그 결과를 농도로 환산하여 분석하게 된다. 또한 분석 데이터 신뢰를 확보하기 위하여 표준시료 및 품질관리용 시료를 사용하여 BAMS분석법에 대한 검증을 실시하였다.

A Study on Error Compensation for Quadrature Modulator in Frequency Direct Conversion Method (주파수 직접변환방식의 직교변조부 에러보정에 관한 연구)

  • In this study, a method of error compensation for channel gain imbalance, phase imbalance and local oscillator leakage in the modulator of frequency direct conversion is suggested. The compensation of channel imbalance can be carried out by using the received power after transmitting test signal. By applying this method, the phase imbalance conversion with frequency can be easily compensated since this method is rarely affected by the transmission channel. It is confirmed that the algorithm proposed in this study(iteration coefficient=11) converges faster than conventional algorithm(iteration coefficient=43). From the numerical results, the DC-offset, channel gain, phase imbalance compensation coefficient and iteration number converges into($f_1$=0.0199999, $f_2$=-0.050001, $C_{22}$=0.9133, $C_{12}$=-0.0524, N=13) when the local oscillator leakage is not considered. However, it converges into($f_1$=-0.02, $f_2$=-2.2476, $C_{22}$=0.9133, $C_{12}$=-0.0524, N=16) when the local oscillator leakage is considered.

  • 수질오염 저감을 위한 유기물질의 처리를 위해 생물학적 처리에 중점을 둔 하 폐수 처리시설의 증가에 따라 BOD로 대표되는 생분해성 유기물질은 꾸준히 감소하였으나, COD농도의 경우 뚜렷한 감소를 보이지 않고 있다. 유기물질 중 특히 난분해성 물질의 증가는 상수원으로 사용되는 하천의 경우 조류 증식 등을 유발하고 여과공정에 영향을 미쳐 상수처리의 효율을 떨어뜨리는 등의 환경적인 문제를 야기할 수 있다. 기존 BOD, COD로 대표되는 유기물질의 경우 각 유기물질별 상관관계 및 유출특성 등에 대한 연구는 많이 이루어졌으나, 난분해성 물질에 대한 조사는 상대적으로 미진한 실정이다. 특히 동진강수계는 '새만금 제2단계 수질개선종합대책' 등 장기적인 수질개선 대책들이 추진되면서 하천의 BOD5 및 TP는 감소추세를 보이고 있으나, COD는 개선되지 않고 증가하는 추세를 보이고 있다. 동진강의 말단부의 경우 COD/BOD5의 비율이 지속적으로 증가하는 추세를 보이고 있어 상대적으로 난분해성 유기물질에 대한 관리의 중요성이 대두되고 있다. 이러한 COD 및 COD/BOD5의 증가 추세는 국내 4대강에서 전반적으로 나타나고 있는 추세로서, 정부는 2013년부터 하천 및 호소의 생활환경기준에 TOC 항목을 도입하여 2016년부터 호소환경기준의 대표항목을 COD에서 TOC로 대체하였으며, 폐수배출시설에 대해서도 점차적으로 확대 적용할 계획으로, 난분해성물질을 포함하는 총유기물질 관리를 위해서 수질 및 수생태계 환경기준에서 유기물질에 대한 환경기준을 BOD5와 COD에서 TOC로 변경하는 정책을 추진 중에 있다. 본 연구에서는 동진강 유역의 본류 및 지류에서 TOC등 유기물질의 시공간적인 분포와 오염원별 배출특성을 현장조사를 통해 조사 분석하였다. 조사 시기는 건기시, 강우시로 구분하여 수행하였으며 조사항목은 일반수질항목, 유기물질 항목, 유기물질 성상 분석 등에 대하여 수행하였다. 난분해성물질 관리를 위한 기초자료확보를 통하여 새만금호의 수질개선 및 목표수질 달성에 기여 할 것으로 판단된다.

  • Objectives : The objective of this study was to investigate the relationship between the status and necessity of dental prosthesis and mastication function in the Korean elderly. Methods : This study investigated 4,557 elderly people aged over 65 in the 5th Korea National Health and Nutrition Examination Survey(2010-2012). Frequency analysis, ${\chi}^2$ test, and logistic regression analysis were used. Results : Those aged over 75 complained of greater discomfort in mastication function than those from 65 to 74, and it was statistically significant(p<0.001). Mastication discomfort was not so significant in those with higher education and higher income(p<0.001). Those who did not use prosthesis tended to have better mastication function(p<0.001). Denture users complained of mastication discomfort when they used it(p<0.001). Conclusions : This study verified the relationship between the discomfort in mastication and related factors, and will provide the basic data for the access to dental clinic services.

Association between psychological factors and temporomandibular disorders in Korean adults : The fouth Korean national health and nutritional examination survey(2009) (한국 성인의 심리적 요인과 측두하악관절 장애와의 관련성 : 제4기 3차년도(2009) 국민건강영양조사)

  • Objectives : This study aimed to investigate relationship between the psychological factors, the prevalence, and perception of temporomandibular disorder in Korean adults. Methods : A total of 7203 data were derived from Korean national health and nutritional examination survey. All data were analyzed using R-COMMANDER(12.2 SDI) program. Results : The prevalence of temporomandibular disorder in Korean adults was 3.1% and 10.5% of adults with temporomandibular disorder (TMD) had more than one TMD related symptom. Women had higher prevalence rate of TMD related symptom than men. Prevalence rate in TMD was low in the younger age, the higher education level, higher income, and the professionals. Psychological factors including cognition of stress and depression was shown to be closely related with suicidal ideation and TMD prevalence. Conclusions : To prevent TMD, it is necessary to provide the systematic oral health education and to develop the combined psychological counseling with treatment program.

Reconsideration on the Importation Pathway of Ancient Korean Rice(Oryzar sativar L.) (고대 한반도에서 재배된 벼의 전래 경로에 대한 고찰)

  • Importation pathway of rice cultivar into Korea was re-established with considering ancient geo-ecological characteristics of the Sororibyeo excavated from Cheongwon. It is assumed that Sororibyeo settled down in Korea by the importation pathway along the southern seashore of China through old downstream of Geumgang by a southern Korean human race when China and Korea were not yet separated each other by the Yellow Sea. This importation pathway was designated as "Old Geumgang-Sorori Rice Road", in this study. It is further inferred that Korean Peninsula was geographically isolated by ocean after the Ice Age. In consequence, Gawajibyeo, an ancient rice with little genetic variation, was evolved from Sororibyeo, which is estimated to evolve into rice cultivar in Korean Peninsula.

Characteristics in Oxidation of Korean Corn Starch with Sodium Hypochlorite (Hypochlorite에 의한 한국산 옥수수 전분의 산화특성)

  • Effects of sodium hypochlorite (NaOCl) concentration, temperature and pH on oxidation mechanism of corn starch were investigated. The rate of oxidation was dependent on the concentration of hypochlorite, pH and temperature of oxidation. The reaction was either first or second order depending on the concentration of NaOCl. At oxidant concentration of $0.75{\sim}3.0%$ active Cl/g starch, the reaction was first-order and it was second-order at $3.75{\sim}4.5%$ active Cl/g starch. The first-order rate constants were increased with increasing oxidant concentration. The rate of oxidation of starch was highest at pH 7 and decreased with increasing acidity or alkalinity of the medium. As the reaction temperature increased, the rate of oxidation was increased.

  • The effects of cure kinetics and mechanism of DGEBA(diglycidyl ether of bisphenol A)/MDA(4,4'-methylene dianiline) with SN(succinonitrile) and HQ(hydroquinone) as an additive and accelerator were investigated. Cure kinetics was evaluated by Kissinger equation and fractional-life method through DSC analysis. The activation energy has hydroxyl group as an accelerator, the activation energy and the starting cure-temperature were lower than those of DGEBA/MDA/SN system. Cure mechanism of those systems was investigated through FT-IR according to the various SN contents. The ratio was SN : HQ = 4 : 1. It has been known that the cure reactions of an epoxy-diamine system are composed of primary amine-epoxy reaction, secondary amino-epoxy reaction and epoxy-hydroxyl group reaction. But in DGEBA/MDA/SN system, primary amino-CN group reaction and CN group-hydroxyl group reaction were added to the above mentioned reactions. These reactions attributed to the long main chain and the low crossliking density. And in DGEBA/MDA/SN/HQ system, hydroxyl group of HQ formed a transition state with epoxide group and amime group and also opened the ring of the epoxide group rapidly, then amino-epoxy reaction took place easily.
