A Study on the Maintenance Plan of Trees in Gwanghalluwon Garden for Representing Traditional Space (전통공간 재현을 위한 광한루원의 수목정비방안 연구)

  • This study aims to draw up measures to planting tree and maintain a landscape in traditional space. Preceding comprehensive theoretical consideration of selected species of trees and tree maintenance. And analysis of present condition of planting in cases of Gwanghalluwon Garden, then draw a maintenance plans of planting through species of trees and landscape of planting recorded in literature. The results were as follows. First, Analysis of selected species of trees and tree maintenance that traditional space. A dispute about the selection species of trees in traditional space has been continued until today. Because unconditional reject of foreign trees are limited. In this context, should be sublated that hasty blind faith of records and dichotomous preparation plans such as removal of foreign trees and implicitly planting of native plants. Secondly, Planted trees in Gwanghalluwon garden was investigated and found to the species of trees used in traditional space such as Pine(Pinus densiflora S. et Z.), Sawleaf Zelkova(Zelkova serrata), Ginkgo(Ginkgo biloba), Crape Myrtle(Lagerstroemia indica L.). But, present planting irrelevant to traditional space, except Gwanghallu pavilion area from the spatiality. Thirdly, A look at the records or literature that maintenance of planting through historical research are limited. Because literature was recorded Salix spp., Crape Myrtle, Bambusoideae(Pseudosasa japonica), lotus only among planted trees in Gwanghalluwon garden. Fourth, Gwanghallu zone have nature of history and sense of place. And It was going to restore the appearance on historical. Consequently maintenance plan of planting of Gwanghallu zone should be maintain the current state. Wanwol pavillion zone can be recognized as the original form because they look similar to the Gwangallu zone's buildings. Therefore, it is necessary to secure the sense of place different from Gwanghallu zone by buffer planting for composition of transition space. Wolmaejip zone and lawn zone was marketplace in outside of castle and large forest. Accordingly, this area should be symbolic restoration of the Yulrim(栗林) and representation of the marketplace in outside of castle through aggregation of facilities and administrative facilities in Gwanghalluwon garden. East lawn of the Wanwol pavilion zone is should be maintained the current state that opened place in terms of using thought linked with the Wanwol pavilion zone. Boundary zone of the Gwanghalluwon garden is difficult to associate in terms of historical research and authenticity. Therefore, application of cultural landscape that appeared in literature is be worth.

Effective Herbicide Application on Dry Seeded Paddy Rice (벼 건답휴립직파재배(乾畓畦立直播栽培)에서 효과적(效果的)인 제초제(除草劑) 사용법(使用法))

  • Several rice herbicides were evaluated to determine the herbicidal efficacy on dry seeded paddy rice. The time of herbicide application was within 1 day after seeding(DAS), 18 DAS, and 40 DAS. Under the upland conditions 30 DAS(before permanent flooding) dominant weed species were Echinochloa sp., Digitaria sp., Leptochla sp., weedy rice and other several annual broadleaved weeds belonging to Cruciferae, but after flooding Echinochloa sp., Aeschynomene sp., Eleocharis kuroguwai, Cyperus sorotinus, Ludwigia prostrata, Aneilema japonica were dominat. Early application (1 DAS) of thiobencarb, pyrazolate/butachlor, or chlomethoxyfen/butachlor resulted in very good herbicidal efficacy only during the first 30 days, but not thereafter. Application of propanil+butachlor on 18 DAS did not control the late occurring weeds effectively. Application of bentazone/quinchlorac on 40 DAS successfully suppressed weed growth throughout the remaining rice growing season and showed similar grain yield as hand weeded plot. Weed growth reduced the plant height, panicle number, 1000-grain weight, percent ripened grains, and grain yield of rice by 7, 82, 19, 55 and 87% respectively. There was a positive linear relationship between weed weight and lodging index of rice.

  • Ninty three species of domestic weeds were collected and screened for antimicrobial, antitumor, antioxidant and herbicidal activities. Among them, few showed antifungal activities. Cuscuta japonica showed inhibitory activity against Alternaria mali, Ambrosia artemisiifolia and Geranium sibiricum against Phytophthora capsici, Aster yomema and Aster pilosus against Phytophthora parasitica. Ambrosia artemisiifolia, Artemisia princeps, Artemisia capillaris, Ludwigia prostrata, Chrysanthemum zawadskii, Bidens frondosa, and Geranium sibiricum showed broad antibacterial activities. Carex chordorhiza, Artemisia capillaris, Persicaria nodosa, Senecio koreanus, Pariicum bisulcatum, Geranium sibiricum showed antiblebbing activity on human chronic leukemia K562 cell, among them, Persicaria nodosa was the strongist. Angelica decursiva, Equisetum arvense, Cimicifuga heracleifolia, Persicaria nodosa, Geranium sibiricum, Oenothera odorata, Cyperus sanguinolentus showed antioxidant activities. Ludwigia prostrata and Peucedanum terebinthaceum showed strong herbicidal activities.

Effect of Brassinolide and Auxin Type Plant Growth Regulators on Radish Petiole Curvature (Brassinolide와 Auxin류(類)의 식물(植物) 생장조절제(生長調節劑)가 무우 자엽병(子葉柄)의 굴절(屈折)에 미치는 영향(影響))

  • This study was conducted to determine the combining effects of a synthetic brassinolide such as homobrassinolide (HBR) with auxin types on the curvature of radish petiole. HBR has no direct effect on the petiole curvature of radish, regardless of concentrations used. Among the natural known plant growth regulators, NAA at 100ppm showed the highest ourvature at 12 hours after treatment. The synthetic auxins such as 2,4-D and MCP resulted in about three times greater effect on curvature than the untreated control, and their effects lasted till 36 horns after treatment, showing much better effect on curvature than that of the natural auxin types of plant growth regulators. The mixture of MCP at 3 ppm with HBR at 100ppm increased petiole curvature of radish seedlings as much as three times greater than that of a single application of HBR. However, the mixture of HBR at 100 ppm plus 2, 4-D at 100ppm increased the petiole curvature of radish seedlings about eight times greater than that of the HBR single application, showing existance of strong synergistic reaction and the most effective combination, and suggesting possibility of a prectical implementation in agriculture.

A Study on the Planting Density and Planting Species Characteristic Analysis of Park and Green Space in Wonju, Gangwon Province (강원도 원주시 공원녹지의 식재 수종 및 식재밀도 특성 연구)

  • In this study, the analysis carried out on characteristic of planting species and planting density of parks and green spaces in downtown city of Haengjeong-dong in Wonju, Gangwon Province. We found that 8,050 coniferous trees (19 species), 6,037 deciduous trees (61 species) and 77,609 shrubs 103 species were planted in the parks and green space in Haengjeong-dong. However, a conifer Pinus strobus is highly concentrated in all four kinds of places which are neighborhood park, children's park, green buffer zone and landscape green space regardless of their period of construction and usage of surrounding land and other trees were planted in order of Pinus koraiensis, Prunus yedoensis, Acer palmatum, Zelkova serrata, Pinus densiflora. Average planting density of parks and green spaces is $0.03tree/m^2$ for a tall tree and $0.18tree/m^2$ for a shrub, which shows low planting density in most parks and green spaces of Wonju. Also, understory vegetation was not planted enough. Since area of lawn is wide in green spaces, planting density is needed to be increased and tall trees should be planted more. Therefore, we suggest planting more trees in an area where trees are not planted, multi-layered vegetation and increasing in planting density through shrub planting etc., by identifying spaces for trees to be planted within parks and green spaces.

Identification of Korean Poaceae Weeds Based on DNA Sequences (DNA 염기서열에 기초한 벼과 잡초의 분자생물학적 동정)

  • Korean Poaceae includes approximately 80 species of the agricultural weeds. Precise species identification is the first step for more effective weed management in the agricultural fields. However, the identification of species in Poaceae is not easy without the assistance of taxonomists or identification experts although they are relatively easy to distinguish from the plants of the other family by the unique characteristics of caryopsis. Thus, DNA barcode was suggested as an alternative powerful technique for species identification by using short sections of DNA from a specific region of the genome. Two standard barcode markers of vascular plants, chloroplast rbcL and matK, and a supplementary nuclear ribosomal Internal Transcribed Spacer (ITS) region were used for barcode of major Korean Poaceae weeds, 403 individuals of 84 taxa. All the barcode markers revealed a good level of sequencing success with the lowest 73.7% for matK and the highest 88.8% for rbcL. The barcode sequences were deposited to the National Center for Biotechnology Information (NCBI) database for public use. Combined matK and ITS showed very high resolving power with 92.9%. Besides the identification of weeds for weed managment, the generated DNA barcode data could be used for many other applications such as rapid biodiversity assessment and conservation prioritization.

Effect of Seed Priming on the Enhancement of Seed Germination in Cool Season Turfgrass (Priming 처리가 한지형 잔디류의 발아 증진에 미치는 영향)

  • This study was carried out to identify the optimum condition for priming, as a series of experiments for the enhancement of germination percentage and germination synchronization in cool-season turfgrass seeds. The optimum priming conditions to maximize the germinability in both Kentucky bluegrass and Tall fescue was a 1 day treatment of 100 mM $K_3PO_4$ at $25^{\circ}C$. The seeds treated with the optimum priming condition enhanced the germinability compared to control, and shortened the time for germination with reduced $T_{50}$ and mean daily germination (MDG). However, as the concentration and treatment period of NaOH or KOH increased, significantly decrease in germination percentage was observed. Germination percentage in Creeping bentgrass and Bentgrass was below 10%, regardless of priming treatment. However, the priming treatment with 200 mM $KNO_3$ for three days improved the germination up to 6%, which was not a statistically significant level. The most effective priming period for Kentucky bluegrass and Tall fescue was one day treatment, but the germinability was suppressed as the treatment period was extended until six days. For priming chemical treatment, germinability was improved with the concentration of 100 mM, while it was reduced at higher concentrations of 200 mM and 300 mM.

Evaluation of horizontal gene transfer from genetically modified zoysiagrass to the indigenous microorganisms in isolated GMO field (GMO 격리포장에서의 유전자변형 들잔디로부터 토착미생물로의 수평유전자전달 평가)

  • The release of genetically modified organisms ($GMO_{s}$) into the environment has the potential risks regarding the possibility of gene transfer from $GMO_{s}$ to natural organisms and this needs to be evaluated. This study was conducted to monitor the possible horizontal gene transfer from herbicide-resistant zoysiagrass (Zoysia japonica Steud.) to indigenous microorganisms. We have first examined the effect of field-released GM zoysiagrass on the microbial flora in the gut of locust (Locusts mlgratoria). The microbial flora was analyzed through determining the 165 rDHA sequences of microorganisms. The comparison of the microbial flora in the gut of locusts that were captured at the field of GM zoysiagrass and of wild-type revealed that there is no noticeable difference between these two groups. This result indicates that the GM zoysiagrass does not have negative impact on microbial flora in the gut of locust. We then investigated whether the horizontal gene transfer occurred from GM zoysiagrass to microbes in soil, rhizosphere and faecal pellets from locusts by utilizing molecular tools such as Southern hybridization and polymerase chain reaction (PCR). When the total DNAs isolated from microbes in GM zoysiagrass and in wild-type zoysiagrass fields were hybridized with probes for bar or hpt gene, no hybridization signal was detected from both field isolates, while the probes were hybridized with DNA from the positive control. Absence of these genes in the FNAs of soil microorganisms as well as microbes in the gut of locust was further confirmed by PCR. Taken together, our data showed that horizontal gene transfer did not occur in this system. These results further indicate that frequencies of transfer of engineered plant DNA to bacteria are likely to be negligible.

Experimental Study on Engineering Performance Evaluation and Field Performance of Environmentally Friendly Functional Concrete (친환경 기능성 콘크리트의 공학적 성능평가 및 현장적용성능에 관한 실험적 연구)

  • In this study, the physical, mechanical, structural, and environmental performances based on field measured data were evaluated to check the suitability of concrete for ecological preservation and cultivation of a hydrophilic environment. More specifically, the study is focused on developing an environmentally friendly functional concrete with river ecology restoration and natural river early formation capabilities. The mechanical performance evaluation results showed that the increase in mix rate of the PVA (Poly Vinyl Alcohol) reinforcement fibers and silica fume caused an increase in the strength. The optimal mix rate was found to be 0.05 volume % PVA fiber and approximately 10% silica fume. The frost resistance evaluation showed that superior performance was gained when 0.05 volume % PVA fiber and 15% silica fume was mixed simultaneously. In the structural performance evaluation, the bending strength was improved by 47.7% compared to plain concrete when mixed with 0.05 volume % PVA fiber. The flexural toughness also saw significant improvement. The environmental monitoring of field performance showed that grasses germinated most rapidly, but the growth of red poppies, a plant that germinates in the spring, was most active with passing of time. Coverage measurements in all of the monitoring locations found favorable coverage of over 95% after 12 weeks. The study results showed that the environmentally friendly functional concrete had outstanding environmental performance.

  • aA series of experiments were conducted to investigate the effect of plant growth regulator treatments on the growth and lateral root formation in soybean sprouts in order to establish the effective method of producing root-less or short-rooted soybean sprouts with larger diameter in the hypocotyl. Major results can be summarized as follows. 1. Soybean sprouts showed fairly uniform elongation rate from 3 to g days after imbibition with daily increase of 3.8cm. The speed of elongation of hypocotyl was reduced whereas that of root accelerated 7 days after imbibition. Lateral roots began to emerge fairly evenly from 5 to 9 days after imbibition with a daily increase of 4.4. 2. Auxins(IAA, IBA, NAA, 2,4-D) inhibited hypocotyl elongation and formation of lateral roots and increased hypocotyl diameter without influencing root length and hook diameter at higher concentrations. The dry weight of cotyledon was increased significantly as compared to that of hypocotyl and root. Among the tested auxins, 2, 4-D was the most effective. 3. BA and 4PU-30 significantly reduced elongation of hypocotyl and root and resulted in the biggest diameter of hypocotyl when treated at higher concentrations. The lowest effective concentration of BA to prevent the formation of larval gal roots was 12.5ppm. The formation of lateral roots could be completely prevented by BA and 4PU-30 treatment but kinetin, zeatin, zeatin riboside resulted in many lateral roots and increased thickness of soybean sprouts with little influence. Cotyledon deformation was found in soybean sprouts treated by 4PU-30. 4. 2, 4-D was the most effective for increasing the hypocotyl diameter while 4PU-30 was the most effective for reducing no. of lateral roots. 5. It can be concluded that among the plant growth regulators tested, BA was effective in reducing root length and increasing hypocotyl diameter. BA 12.5 ppm or 15 ppm may thus be the more practical for production of soybean sprouts. 6. ABA showed no significant effect of growth parameter, however ABA 25 ppm inhibited only no of lateral roots with little influence on the growth of seedling. 7. Ethephon inhibited the elongation of hypocotyl and root and increased hypocotyl diameter at higher concentrations. 8. The combined effect of cytokinins and ethephon was very similar to result of BA treatment alone. As the ethephon concentration increased, hypocotyl diameter and dry weight of cotyledon tended to increase.
