In Vitro Anti-inflammatory Activities of Herbal Extracts with Eucommia ulimoides and Ulmus davidiana (두충과 느릅 혼합추출물에 의한 염증 억제 효과)

  • This study confirmed the cytotoxicity and anti-inflammatory activities of natural herbal extracts (HE) including Eucommia ulimoides and Ulmus davidiana against human mast cell (HMC-1). HE was extracted with distilled water (at $75^{\circ}C$) and then freeze-dried for 5 days. Finally, the HE was sterilized by gamma radiation with $^{60}Co$ ${\gamma}$ source at room temperature. Cytotoxicity of the HE against HMC-1 cell was measured using cell counting kit-8 (CCK-8) assay. In addition, inflammatory cytokines such as $TNF-{\alpha}$, IL-6 and IL-8 were evaluated by ELISA kit on the HMC-1 cells with calcimycin A23187 and phorbol 12-myristate 13-acetate (PMA). The results showed that HE had no toxicity and reduced $TNF-{\alpha}$, IL-6, IL-8 response on HMC-1 cells.

Modulation of Hyaluronic Acid Properties by Electron Beam Irradiation (전자선 조사를 이용한 히알루론산의 특성 조절)

  • A variety of natural polymers have been used as tissue engineering scaffolds, drug delivery system, and cosmetic materials due to their higher biocompatibility and water uptake. As a major component of extracellular matrix, hyaluronic acid consisting of D-glucuronic acid and N-acetylglucosamine has been popularly used as a hydrogel material. Even though it has good properties to be used in the tissue engineering and cosmetic industry, its higher viscosity has limited a potential use in a variety of applications; only low content should be applied in preparing above products. In the present study, we investigated the effect of electron beam irradiation on the properties of hyaluronic acid. Hyaluronic acid paste containing low contents of water changed to solution after electron beam irradiation ranging from 1 to 10 kGy, which didn't exhibit any alteration of surface properties and morphological change after freeze-drying. However, its viscosity was significantly decreased as absorbed dose increased, which was approximately one by hundred in comparison with the viscosity of original hyaluronic acid solution with same concentration. In addition, it can still interact with positive charged chitosan generating polyelectrolyte complex. Therefore, only viscosity was decreased after electron beam irradiation, whereas other properties of hyaluronic acid maintained. Consequently, these hyaluronic acids with lower viscosities can be used in a variety of applications in tissue engineering, drug delivery, and cosmetic industry.

Evaluation of Quality Properties of Gamma-irradiated Freeze-dried Fruits (감마선 조사에 따른 동결건조과일의 품질 특성 평가)

  • The quality properties of freeze-dried apples, pears, strawberries and pineapples gamma-irradiated at 0, 1, 2, 3, 4, and 5 kGy were evaluated to develop germ-free products for immuno-compromised patients. The initial count of total aerobic bacteria in non-irradiated apples, pears, strawberries and pineapple was 2.5, 3.1, 2.6, and $3.2{\log}\;CFU{\cdot}g^{-1}$, respectively. Microorganisms were not observed in apples after 1 kGy, in pears and strawberries after 4 kGy, and in pineapples at 5 kGy within a detection limit of $10{\log}\;CFU{\cdot}g^{-1}$. In addition, the sterilization of each sample was confirmed at the same dose. The score for the overall acceptance of freeze-dried fruit irradiated at a sterilization dose was 5.5 for apples, 4.1 for pineapples, and 4.0 for the other fruits, whereas that of non-irradiated control sample was 5.6 for apples, 5.2 for pears, and 5.8 for strawberries and pineapples with a 7-point scale. As a result, gamma irradiation of 1 kGy for apples, 4 kGy for strawberries and pears, and 5 kGy for pineapples is sufficient to sterilize each freeze-dried fruit with acceptable sensory properties.

Forage Quality Evaluation of Mutant Lines derived from Gamma-ray Treatments in Rubus fructicosus L. (감마선 조사 유래 블랙베리(Rubus fructicosus L.) 돌연변이 계통의 농업부산물 사료가치 분석)

  • We examined the forage quality to provide the basic information for the utilization of blackberry by-products (leaf and stem) among thirty-five mutant lines derived from gamma-ray. The leaves had higher crude protein (CP) contents than stems. In addition, the leaves showed broad ranges of variation in crude fat, crude fiber and crude ash. The overall forage quality of the stems were similar to those of the original variety. The mean of neutral detergent fiber (NDF) content of leaves and stems in mutant lines were 25.88% and 58.98%, respectively. Also, the mean of acid detergent fiber (ADF) content of leaves and stems in mutant lines were 15.43% and 49.27%, respectively. Forage quality grades of all blackberry leaf were special quality class. However, the blackberry stem ranked $1^{st}$ to $5^{th}$ in quality class. The stem diameter was negatively ($P{\leq}0.05$) correlated with the moisture and crude protein. Also, the fruit length was highly negatively ($P{\leq}0.01$) correlated with the RFV (relative feed value) in blackberry leaf. The one hundred fruit weight was highly negatively ($P{\leq}0.01$) correlated with the crude ash. Based on these results, the by-products of blackberry could provide high quality forage for feeding of live stock.

Detection of Gamma-irradiated Imported Foods (Perilla seeds, Almonds, and Soybeans) by Using PSL, TL, and ESR (PSL, TL 및 ESR 분석을 통한 수입산 들깨, 아몬드 및 대두의 방사선 조사 여부 판별 특성)

  • This study examined radiation detection properties of imported perilla seeds, almonds, and soybeans. Food samples were irradiated at doses of 0.5, 1, 2, and 4 kGy and analyzed by photo-stimulated luminescence (PSL), thermoluminescence (TL), and electron spin resonance (ESR). The results of PSL showed that photon counts of irradiated perilla seeds and soybeans were $32,959counts{\cdot}60sec^{-1}$ and $7,234counts{\cdot}60sec^{-1}$, respectively, at an irradiation dose of 0.5 kGy, and the photon count of irradiated almonds was $5,581counts{\cdot}60sec^{-1}$ at an irradiation dose of 1 kGy. The results of TL showed that this technique is useful for detecting irradiated samples; the TL ratios ($TL_1/TL_2$) measured for the food samples at an irradiation dose of 0.5 kGy were 0.2301 for perilla seeds, 0.4595 for almonds, and 0.4827 for soy beans. Lastly, results of ESR spectroscopy for only soybeans revealed specific signals derived from free radicals captured in the cellulose. In conclusion, PSL, TL, and ESR methods can be used for the detection of gamma-irradiated soybean samples, whereas gamma-irradiated perilla seeds and almonds can be confirmed by a serial detection with a TL method after PSL.

  • A simplified model to assess the performance of engineered barrier for the retardation of radionuclide release in a low- and intermediate-level radioactive waste repository was developed. The model is based on the repository design concept being suggested in Korea, and considers two types of release scenario ; a design-bas release for the design of engineered barrier and a realistic release for the performance assessment. For the numerical illustration, the sample calculations were performed for five radionuclides with different chemical characteristics, and the results were analyzed.

일본의 휴먼팩터연구 개요 - 조직적$\cdot$체계적으로 추진

  • 일본의 전력중앙연구소는 최근 휴먼팩터연구센터의 연구개요를 정리발표 하였다. 동센터는 동 연구소내에 7월 1일부로 설치된 것으로써 민간인 총력을 집결하여 휴먼팩터(인간요인)에 기인하는 원자력발전소 사고의 방지책을 체계적으로 연구하는 곳이다. 5개년 계획으로 약10억엔의 연구개발자금을 투입, 그 성과는 일본 전력회사의 휴먼에러방지책에 직접, 반영시켜진다. 소원은 현재 14명이지만, 전력회사에서의 파견으로 확충하고, 피크시에는 20명으로 하고 있다. 동 센터의 연구개요는 다음과 같다.
