Analysis on the Components of the Angelica dahurica Root (구릿대(Angelica dahurica) 뿌리의 성분 분석)

  • This study was carried out to analyze the components of the Angelica dahurica root for the studies of the physiological function. General components of A. dahurica root were 71.7% moisture, 10.6% carbohydrate, 9.3% crude protein, 6.6% crude ash and 1.9% crude fat. The content of reducing sugar was 1,850 mg/100 g. The total amount of free sugar was 80 mg/100 g, in which 19.3 mg/100 g fructose, 27.8 mg/100 g glucose, 28.4 mg/100 g sucrose and 4.5 mg/100 g maltose were present. In the results of mineral analysis, the content of K was the highest(2,145.03 mg/100 g) followed by 286.35 mg/100 g Mg and 145.23 mg/100 g Ca. The total amount of hydrolyzed amino acid was 71.68 mg/100 g, in which 20.98 mg/100 g essential amino acid and 50.70 mg/100 g of non-essential amino acid were present. Among them, proline(11.74 mg/100 g) was the highest. Total free amino acids were contained 17.04 mg/100 g, in which 6.67 mg/100 g of essential amino acids and 10.37 mg/100 g of non-essential amino acids were present. Among them alanine(5.96 mg/100 g) was the highest. Total content of amino acid derivatives was 3.37 mg/100 g.

Effect of Essential Amino Acid Deficient Diets in Feeding Response and c-fos Expression in Rats Brain in Response to Methionine Deficiency (필수아미노산 결핍에 의한 섭식반응과 Methionine 결핍이 흰쥐의 뇌내 c-fos 발현에 미치는 영향)

  • This study was conducted to investigate the effect of essential amino acid(EAA) deficient diets on short-term feeding response and the Fos expression in brain area when methionine deficiency diet fed, and thereby to know the mechanism of feed intake regulation. In all trials, experimental diets were formulated with pure amino acid mixture to level of 15% nitrogen. Rats were adapted to a 6-hr single-meal feeding per day(17:00${\sim}$21:00). Feed intake and body weight were monitored every hour after 7-day of feeding of individual EAA deficient diets in Exp. Ⅰ. In Exp. Ⅱ, Fos immuno- histochemistry was determined in various regions of brain to identify the regions that is related to suppressed feed intake following feeding methionine-deficient diet. Fos expression was examined to know the initial sensitive region in the brains of rats at 3h after feeding of the control and methionine deficient diet(-Met). Initial response to EAA deficiency diets was severely depressed in methionine deficiency diet, but the depression was low in threonine deficiency diet. However, the feed intake at 3rd day in rats was depressed in the order of His(71%), Leu(68%), Ile(66%), Thr(63%), Trp(61%), Val(55%), Phe(52%), Met(51%), Lys(44%) and Arg(24%). Fos immunoreaction in neural regions(PPC, amygdala and EPC) of pyrifrom cortex was increased in the -Met group more than in the control diet group, but those in LH, VMH and PVM were similar. Thus, based on these data, the PPC was identified as the initial response area in the -EAA diet.

Analysis of Minerals, Amino Acids,and Vitamin of Lespedeza cuneata (비수리의 미네랄, 이미노산, 비타민 분석)

  • For developing functional biomaterials, chemical analysis of Lespedeza cuneata including minerals, amino acids and vitamin were investigated. Minerals of L. cuneata were found to be calcium, potassium, magnesium, phosphorus, sodium, iron, manganese, zinc and copper. Among the free amino acid, proline was 33.77 mg% that were 67.2% of free amino acids and essential amino acids were 7.49 mg%. Total amino acids were analyzed as 2,817 mg% and the content of glutamic acid (496.00 mg%) was highest. In case of vitamin, the highest components was vitamin E with 33.03 mg%.

  • To study nutritional values of snake head and carp cooking, the content of minerals and amino acid including general components were analyzed. Crude fat in snake head and carp were 3.4%,2.8% respectively and crude protein 1.7%, 2.9%. Calcium in snake head and carp were 127.1mg%. 16.6mg% and iron were 5.6mg%. 5.2mg% respectively. The amino acid composition of snake head and carp showed high content of glycine, glutamic acid, proline and alanine while the content of phenylalanine, valine, histidine, isoleucine, methionine and tyrosine were relatively low. Total amino acid of carp showed higher than that of snake head.

Quality Improvement in Perilla III. Varietal Differences of Protein Content and Amino Acid Composition in Perilla (들깨의 성분개량 연구 III. 들깨 품종의 단백질 함량과 아미노산 조성)

  • Korean local collected strains of perilla were analyzed on their protein and amino-acid contents in the seed. The strains analyzed were 80 of which grown in the experimental field of Crop Experiment Station at Suwon in 1991. This experiment was performed to obtain the basic informations on the varietal differences of protein and amino-acid contents and further applied in the breeding project for quality improvement of perilla. The seed protein contents of 80 perilla varieties ranged from 17.9% to 24.6% and the average content was 24.6%. There were no differences in protein contents according to the maturity and 1,000 seed weight, but differed by the seed coat color showing 1.4% higher content in light gray than that of drak brown, which may suggest that the seed coat color could be used as a marker gene for breeding high protein varieties. While the total essential amino-acid contents of collected perilla ranged from 36.41% to 44.85%, the mean of 40.55%, the non-essential amino-acid contents was 47.23%~60.01% with the mean of 55.29%. Moreover, there was high positive correlation between the essential amino-acid contents and seed coat color. The essential amino-acids of perilla has an exceptionally high content of arginine, leucine, but methionine indeed being almost marginal. Of the essential amino-acids, lysine, methionine and valine were contained higher compared with the FAO recommendation level.

  • Potato is the 4th important crop along with rice, wheat and maize. It contains high quality of starch with relatively high content of vitamin C and protein. However, there is a nutritionally limiting factor due to a low level of sulphur-containing essential amino acid including methionine and cysteine. Recently, recombinant DNA technology and metabolic engineering with genes involved in the bio-synthetic pathway have been applied to enhance the level of these essential amino acids. In this report, it has been discussed about the current status and bottleneck on the development of transgenic potato containing high level of sulphur-containing essential amino acids.

Varietal Difference of Protein Content and Amino Acid Composition in Sesame (참깨품종의 단백질함량과 아미노산 조성)

  • To obtain the basic information an seed quality improvement in sesame, protein content of 114 varieties and amino acid composition of 12 varieties was analyzed. Protein content showed the vaietal difference ranged 20.6-30.2% and the mean was 24.72%. The highest variety in protein content was PI158066 (30.2%) originated from U.S.A. Protein content of Korean local varieties were highest among original group analyzed. Seed coat texture and seed coat color affected to protein content so, smooth type was higher than rough type in protein content, and black seeded varieties showed the hight protein content. Amino-acid composition of sesame was uneque in balance and higher than FAO reference. Total amino-acid of variety PI258372 was highest as 25.03%. Essential amino-acid (EAA) /total amino-acid(TAA) ratio of sesame was higher as 42-58.2% than soybean, corn, rice, peanut. Korean local variet 'Samcheck' showed best quality in amino-acid composition as 58.2% in EAA/TAA ratio with high tyrocin and lysine. Total amino acid content was high in order of Korean local '||'&'||'gt; introduced '||'&'||'gt; Korean bred varieties.

Studies on the Amino Acid Components of Korean Hazel Nut (한국산(韓國産) 개암 종실(種實)의 아미노산 조성(組成)에 관한 연구)

  • Free and total amino acids of Korean hazel nut (Corylus heterophylla Fisch. var. Japonica koidz.) were identified by amino acid autoanalyzer (Technicon PNC-1 type). The results indicate that among 16 kinds of all free amino acids contained in hazel nut, arginine was the richest, and then came glutamic acid, proline, alanine, tyrosine and valine in that order. From the total amino acids which were closely related with the nutritional valuation, arginine was in the richest amount and then came with glutamic acid, proline, leucine, aspartic acid and valine.

Amino Acid Composition and Scanning Electron Micrographs of Modified Silkworm Larvae Protein Concentrates (변형번데기 농축단백질 아미노산 조성 및 주사전자현미경에 의한 관찰)

  • This study was designed to determine the amino acid composition and to investigate microstructure by scanning electron micrographs of silkworm larvae protein and modified silkworm larvae protein concentrate. The results were as follows: 1, The protein contents of soybean and silkworm larvae protein concentrate were 70.3% and 84.1%, respectively. 2. In general, the essential amino acid content of silkworm larvae protein concentrate were higher than soybean protein concentrate as well as FAO provisional scoring pattern. Silkworm larvae protein concentrate was especially high in lysine and methionine indicating that it could be a good supplemental source for cereals and beans. Succinylation and acetylation resulted in no difference in most amino acid content. 3. The scanning electron microscopic observations revealed that silkworm larvae protein concentrate had smooth surface topography while defatted silkworm larvae flour showed different shapes and sizes with relatively rough surfaces. Acylated silkworm larvae protein concentrate exhibited less cellularity and denser than protein concentrate. However, succinylated silkworm larvae protein concentrate showed especially good texture indicating that it could increase the functional properties of silkworm larvae protein concentrate.
