The experimental evaluation of exposure conversion formula using the relationship between optical photo-density, exposure dose and the quality of radiation characteristics of radiation energy to X-ray and ${\gamma}-rays$. The film badge dosimeter is analysed by exposure conversion formula which evaluate image fading characteristics for development time and directional characteristics for incident beam angle. In conclusion, exposure conversion formula evaluated of this study is satisfied with quality decision criterion of the film badge dosimeter.
Since provisions on the technical criteria for personnel dosimetry was amended three years ago, several improvements in the technique of monitoring personnel doses by TLD have taken place, but for the photograpfic film as a personnel monitor, additional investigations should be carried out for its accuracy of dose estimates because of its wide use in the radiation involved industries. So, this paper describes the methods to develope dose evaluation algorithm for photographic film using ISO reference radiations by i) empirical formula, ii) degree-of-fit method, and iii) matrix approximation. These methods show a good agreement between irradiated and calculated dose within tolerance level represented in ANSI N13. 11, and can be used for the dose evaluation of X, ${\gamma}$ and/or radiation fields.
An effort was attempted to utilize an under-utilized protein source, rice bran protein, in coating or wrapping food material for the purpose of protection them from oxidation and bacterial infection. However, the utilization of rice bran protein as a food coating material is limited because the rice bran protein coating material itself can be spoiled by a bacterial infection. Therefore, this study was conducted to produce the economical and antibacterial rice bran protein film by utilizing rice bran and bacteriocin-producing microorganism. Bacteriocin produced by Pseudomonas putida 21025 was partially purified after 33h of shaking incubation at 30$^{\circ}C$. The amount of amino-type nitrogen did not increase in the rice bran protein film containing the bacteriocin any more after gradual increase upto the content of 0.22% for 8 days, while that without the bacteriocin increased continuously, implying that application of the bacteriocin to the rice bran had positive effects on prolonging the shelf-life of not only film itself but also the foods wrapped by this film.
The purpose of our study is to compare the adhesion and biofilm formation abilities of isolates from water discharged from dental unit waterlines (DUWLs). Bacteria were isolated from a total of 15 DUWLs. Twelve isolates were selected for the experiment. To confirm the adhesion ability of the isolates, each isolate was attached to a glass coverslip using a 12-well plate. Plates were incubated at $26^{\circ}C$ for 7 days, and the degree of adhesion of each isolate was scored. To verify the biofilm formation ability of each isolate, biofilms were allowed to form on a 96-well polystyrene flat-bottom microtiter plate. The biofilm accumulations of all isolates formed at $26^{\circ}C$ for 7 days were identified and compared. A total of 56 strains were isolated from 15 water samples including 12 genera and 31 species. Of the 56 isolates, 12 isolates were selected according to the genus and used in the experiment. Sphingomonas echinoides, Methylobacterium aquaticum, and Cupriavidus pauculus had the highest adhesion ability scores of +3 among 12 isolates. Among these three isolates, the biofilm accumulation of C. pauculus was the highest and that of S. echinoides was the third-most abundant. The lowest biofilm accumulations were identified in Microbacterium testaceum and M. aquaticum. Most isolates with high adhesion ability also exhibited high biofilm formation ability. Analysis of adhesion and biofilm formation of the isolates from DUWLs can provide useful information to understand the mechanism of DUWL biofilm formation and development.
The water discharged from dental unit waterlines (DUWLs) is heavily contaminated with bacteria. The development of efficient disinfectants is required to maintain good quality DUWL water. The purpose of this study was to establish a DUWL biofilm model using well-plates to confirm the effectiveness of disinfectants in the laboratory. Bacteria were obtained from the water discharged from DUWLs and incubated in R2A liquid medium for 10 days. The bacterial solution cultured for 10 days was made into stock and these stocks were incubated in R2A broth and batch mode for 5 days. Batch-cultured bacterial culture solution and polyurethane tubing sections were incubated in 12-well plates for 4 days. Biofilm accumulation was confirmed through plating on R2A solid medium. In addition, the thickness of the biofilm and the shape and distribution of the constituent bacteria were confirmed using confocal laser microscopy and scanning electron microscopy. The average accumulation of the cultured biofilm over 4 days amounted to $1.15{\times}10^7CFU/cm^2$. The biofilm was widely distributed on the inner surface of the polyurethane tubing and consisted of cocci, short-length rods and medium-length rods. The biofilm thickness ranged from $2{\mu}m$ to $7{\mu}m$. The DUWL biofilm model produced in this study can be used to develop disinfectants and study DUWL biofilm-forming bacteria.
The adaptability of dry rehydratable film for the qualitative evaluation of coliform bacteria and Escherichia coli was tested. In general, culture methods that employ lactose broth or desoxycholate lactose agar are used for qualitative tests of coliform bacteria. Culture using lactose broth showed a high detection yield and low selectivity when compared to the dry rehydratable film. However, culture methods that employ lactose broth required a long time(48 hrs) for qualitative tests of coliform bacteria and complicated procedures were required to prepare the medium. The detection of coliforms using desoxycholate lactose agar had a slightly higher selectivity than the dry rehydratable film method, but this difference was not statistically significant. However, the preparation of the desoxycholate lactose agar was complicated. EC broth for the detection of E. coli showed the highest detection yield and lowest selectivity; however, this method required complicated procedures for preparation of the medium as well. Overall, the dry rehydratable film had a slightly lower detection yield than the other methods. The detection yield of dry rehydratable film method was over 37.1% at a concentration of 1 cfu/$m{\ell}$. Additionally, the dry rehydratable film method showed high selectivity and did not require preparation. However, because the selectivity of the dry rehydratable film was high, it took a long time(36 hrs) to detect E. coli. Overall, these findings indicate that dry rehydratable film can be used for qualitative detection of coliforms and E. coli.
Water supplied through dental unit waterlines (DUWLs) has been shown to contain high number of bacteria. To reduce the contamination of DUWLs, it is essential to develop effective disinfectants. It is, however, difficulty to obtain proper DUWL samples for studies. The purpose of this study was to establish a simple laboratory model for reproducing DUWL biofilms. The bacteria obtained from DUWLs were cultured in R2A liquid medium for 10 days, and then stored at $-70^{\circ}C$. This stock was inoculated into R2A liquid medium and incubated in batch mode. After 5 days of culturing, it was inoculated into the biofilm formation model developed in this study. Our biofilm formation model comprised of a beaker containing R2A liquid medium and five glass rods attached to DUWL polyurethane tubing. Biofilm was allowed to form on the stir plate and the medium was replaced every 2 days. After 4 days of biofilm formation in the laboratory model, biofilm thickness, morphological characteristics and distribution of the composing bacteria were examined by confocal laser microscopy and scanning electron microscopy. The mean of biofilm accumulation was $4.68{\times}10^4$ colony forming unit/$cm^2$ and its thickness was $10{\sim}14{\mu}m$. In our laboratory model, thick bacterial lumps were observed in some parts of the tubing. To test the suitability of this biofilm model system, the effectiveness of disinfectants such as sodium hypochlorite, hydrogen peroxide, and chlorhexidine, was examined by their application to the biofilm formed in our model. Lower concentrations of disinfectants were less effective in reducing the count of bacteria constituting the biofilm. These results showed that our DUWL biofilm laboratory model was appropriate for comparison of disinfectant effects. Our laboratory model is expected to be useful for various other purposes in further studies.
This paper described an improved technical method required for proper evaluation of personnel exposures by means of the photographic dosimeter developed by KAERI in lower gamma or X-ray energy regions, with which response of the dosimeter varies significantly. With calibration of the dosimeter in the energy range from 30 to 300 keV, the beam spectrum was carefully selected and specified it adequately. The absorber combinations and absorber thickness used to obtain the specified X-ray spectra from a constant potential X-ray machine were determined theoretically and also experimentally. A correlation between the density and exposure for the four separate energies, such as $49\;keV_{eff},\;154\;keV_{eff}\;250\;keV_{eff}\;and\;662\;keV$, is experimentally determined. As a result, it can be directly evaluated the exposure from the measured response of dosimeter.
Purpose: We have developed new film dosimetry algorithm for personal dosimetry and examined its characteristics. Materials and methods: Agfagaevart personal monitoring 2/10 films are used. Films which are in the film badges filtered with Cu 0.3mm, plastic 1.5mm, Aluminum 0.6mm and tin 0.8mm, were exposed by standard dosimetry laboratory. Irradiated energy categories are ANSI N13.1l Category III, and IV. Manual type film precessor and X-rite film densitometor was used. Filtered densities to energy relations and does to transformed densities relations can be obtained ofter transformation of H&D curves to linear shape by polynomal fitting. Reults : Personal dose be determined within 25% error for category m and 15% for category IV. And we are able to evaluate the exposed energy. Conclusion : New algorithm developed in this study is good for personal dosimetry within 30% error range for catergory III and IV. It is expectd to be complete personal dosimetry algorithm with further study for categrory, I, Dand II V.
