Correlation Between Skin Roughness and Dermal Density of Skin Wrinkle Evaluation (피부 주름 평가에 있어서의 피부 거칠기와 진피 치밀도의 상관성 연구)

  • In this study, we evaluated the crow's fret of 95 Korean female by using mechanical assessments; Skin-Visiometer SV 600 and Dermascan C. Transparency profilometry (Skin Visiometer) were using a very thin skin print which allowed parallel light to pass through and was analysed immediately after production. High-frequency (20 MHz) ultrasonography (Dermascan C) enabled non-invasive evaluation in skin thickness and echodensity. We found a correlation between skin roughness and dermal density. Particularly, we showed a significant correlation between skin roughness (R2) and dermal thickness. Also, we demostrated a significant negative correlation between dermal density and dermal thickness (p < 0.05). Therefore, the ultrasonography system might be considered a very useful method in wrinkle evaluation with the transparency profilometry. Further study will be required.

The Temporal and Spatial Expression of the Cytokeratin in Keratinocytes during Cutaneous Wound Healing on the Amphibian(Bombina orientalis) (양서류 피부 상처회복과정 중 각질화세포 cytokeratin의 분포)

  • The system of wound healing is very complex biological processing that includes inflammatory, reepithelialization, and matrix construction. For identification of the transitional pathway of the keratinocytes, we have employed immunohistochemical analysis using cytokeratin antibody after wounding. Epithelium in skin of the frog(Bombina orientalis) was examined with transmission electron microscopy. Cytokeratin was expressed in normal basal and gland cavity cells. At 3-hour basal layer cells were strong positive, however cells of the upper layer were negative reaction. Day1 and 2 after post-wounding, regenerating epithelial cell layer was positive reaction, especially basal layer cells were strong positive. At day 10 after wounding, the degree of positive reaction to basal cells of regenerating epithelial tissue was equal to day 7 wound tissue. At day of 19th, basal and spinous layer cells were strong positive reaction. Regenerating epithelial cells were positive but some basal cells were strong positive at day 27. From this result, we identified that the migration of the keratinocytes in amphibian skin wounds is initiated from basal layer fells and the keratinocytes migrate into basal and middle of the wound area.

Simulation of Energy Absorption Distribution using of Lead Shielding in the PET/CT (PET/CT 검사에서 납 차폐체 사용에 따른 에너지 흡수 분포에 관한 모의실험)

  • Energy absorption distribution according to lead shielding for 511 keV ${\gamma}$ ray was evaluated using a Monte Carlo simulation in PET/CT. Experimental method was performed about the depth of skin surface(0.07), lens(3) and the depth(10) was conducted by using ICRU Slab phantom. Difference of energy absorption distribution according to lead thickness and effect of air gap according to distance of lead and phantom. As a result, study showed that using a lead shielding makes high energy distribution by backscatter electron. As a distance between lead and phantom increased, energy absorption distribution gradually decreased. 9 cm or more air gap should exist to prevent effect of backscatter electron which reaches skin surface, when 0.25 mmPb shielding is used. Also 1 cm or more air gap was needed to prevent the effect in 0.5 mmPb. If air gap was not concerned, 0.75 mm or more lead thickness was necessary to prevent effect of backscatter electron.

  • 각종 섬유소재 중 흡한 흡수성이 우수한 100% 면과 폴리에스테르 섬유를 이용하여 본 과제의 목표에 부합되는 최적의 제직, 제편 조직을 설정, 생산하여 상품화하였으며, 그 내용은 아래와 같다. (1) 흡한, 속건성 직물개발 : 직물의 구성인자(실의 번수, 밀도, 중량, 두께)에 따른 흡수성, 속건성을 검토하여 용도별 직물의 설계조건을 제시하였다. (2) 고흡한, 속건성의 2층 구조 편성물 개발 : 편성물의 구조를 피부측은 흡수하기 쉽고, 외측은 수분을 외부로 발산하기 쉽도록 2층 구조의 편성물을 제조하였다.

Histologic Changes of Rabbit Skin Induced by Progressive Tissue Expansion (Tissue Expansion이 가토의 피부에 미치는 조직학적인 변화)

  • Soft-tissue expansion is a new surgical technique of providing donor tissue in modern reconstructive surgery. This technique provides a quantity of tissue of similar color, texture, and hair-bearing qualities for reconstruction of adjacent defects. It is known that the expanded skin shows several constant histologic changes including the increase in collagen fibers and vascularity within dermis, and thinning of subcutaneous tissue and dermis. In this study, the author observed serial histologic changes of rabbit skin induced by progressive tissue expansion upto excessive expansion of 6 times. The results are as follows : 1. Changes in the thickness of the epidermis was minimal until 3 times of expansion, but slight thinning was observed at excessive expansion state. 2. The thickness of the dermis was progressively decreased, and collagen fibers in the dermis was rapidly increased in early phase of expansion. 3. The vascularity in the dermis was also progressively increased. 4. The skin appendages showed no structural changes even in excessive expansion. 5. The panniculus carnosus showed no atrophic changes and the thickness was maintained in excessive expansion.

Skin Hydration Effect of Jeju Lava Sea Water (제주용암해수의 피부 보습 효과 연구)

  • Many minerals and nutrient salts are abundant in Jeju lava sea water. The objective of this study was to evaluate the skin hydration effects of Jeju lava sea water. The skin barrier serves as a protective barrier that prevents the loss of moisture. The water holding capacity and water transport of the epidermis have been proposed to be important determinants of skin hydration. Jeju lava sea water increased the mRNA expression of filaggrin and caspase-14 which is related to natural moisturizing factor (NMF) formation. Aquaporins 3 (AQP3) are proteins that facilitate the transport of water across cell membranes. Jeju lava sea water increased the mRNA expression and protein expression of AQP3. We employed a skin equivalent model to assess the efficacy of Jeju lava sea water. In a skin equivalent model, Jeju lava sea water increased the CD44 (hyaluronic acid receptor) which is related to skin hydration. From these results, we found out Jeju lava sea water maybe help to skin hydration.

Morphological Changes in the Skin of Hairless Mouse Fed Various Kimchi Diet (김치종류별 식이가 Hairless Mouse 피부의 형태학적 변화에 미치는 영향)

  • Skin is the most frequently exposed tissues to oxidative stress from exogenous and endogenous sources. Dietary antioxidants, which suppress oxidative stress including reactive oxygen metabolites, play an important role in protecting skin from deleterious reactive oxygen species. Kimchi containing lots of antioxidative compounds shows anti-aging effect on skin. Therefore the morphologic changes on the skin of hairless mice fed diets containing Korean cabbage, mustard leaf, and buchu kimchi for 16 weeks were determined. Although epidermal thickness was decreased with age, kimchi prevented this thinning of epidermis compared to control group. In kimchi groups, the staining area of cytokeratin was smaller and stratum corneum was thinner than control group. It suggests that various kinds of kimchi diets prevent the increase of keratinization in epidermis with aging. Type Ⅳ collagen, a major structural protein of basement membrane supporting matrices, existed greater amount in kimchi groups than control group, especially in mustard leaf kimchi group. Rough endoplasmic reticulum (RER) of fibroblast was well developed in dermis of Korean cabbage and mustard leaf kimchi groups, which means collagen synthesis at dermis increased in those kimchi groups. This morphological changes of skin suggest that kimchi consumption can retard skin aging due to the presence of antioxidant and anti-aging compounds, especially some components of mustard leaf kimchi may largely affect on the skin rejuvenescence.

A Study on the Design of Stearic Acid-Based Solid Lipid Nanoparticles for the Improvement of Artificial Skin Tissue Transmittance of Serine (Serine 의 인공피부조직 투과 개선을 위한 Stearic Acid 기반 고형지질나노입자의 설계 연구)

  • Stratum corneum known as a skin barrier, which maintains water in skin, is the outer layer of the skin. Natural moisturizing factors (NMF) are one of the constituents in stratum corneum and amino acids are the highest components among NMF. In this study, we designed stearic acid-based solid lipid nanoparticles (SLNs) for improved skin penetration of serine (Ser). Ser-capsulated SLN was manufactured by double-melting emulsification method. The mean particle size and zeta potential of SLNs were 256.30 ~ 416.93 nm and -17.60 ~ -35.27 mV, respectively. The higher the degree of hydrophobicity or hydrophilicity of emulsifiers, the smaller the particle size and the higher the stability and capsulation rate. In addition, skin penetration was conducted using SkinEthicTM RHE which is one of the reconstructed human epidermis models. The results of Ser penetration demonstrated that all SLNs enhanced than serine solution. The amount of enhanced Ser penetration from SLNs were approximately 4.1 ~ 6.2 times higher than that from Ser solution. Therefore, Ser-loaded SLN might be a promising drug delivery system for moisturizing formulation in cosmeceutical.

  • Skin mucous cells in the three regions of the body in Pseudobagrus fulvidaraco and Leiocassius nitidus were investigated using three histochemical methods (PAS, AB-PAS and HID). In the two species, components of mucous cell were not distinguished in each region, but presented a differences in position and type, size, and count in all regions. Mucous cells of P. fulvidraco were located in two layers, a superficial epithelial cell and the space within alarm substance cells (ASCs). Mucous cells of L. nitidus were situated only in the superficial epithelial cell layer. The size of mucous cells in the ASC layer were larger than those of the superficial epithelial cells for all measured values: diameter, length, width, and area. In the superficial epithelial cell layer, the average number of mucous cells within the three regions did not differ significantly between species by unpaired T-test, but the average number of those within two cell layers of P. fulvidraco had a similarity at each region by paired T-test. The dismilarity in dorsal and ventral regions in average number of ASCs between P. fulvidraco and L. niditus was confirmed by unpaired T-test.

  • Ultrastructures on the integumentary epidermis of the marbled sole, Limanda yokahamae, were examined by means of the light and transmission electron microscope. Epidermal layer consists of supporting cells, unicellular glands and accessory cells. The supporting cells were classified into superficial cell, intermediated cell and basal cell. The cytoplasm of supporting cells is divided into cortex and medullar part. In the cortex and medullar part, microfilaments and cell organelles are well developed, respectively. Gland cells are present in the superficial and middle epidermis. The cytoplasm of mucous cell reacted to blue in AB-PAS (pH 2.5). Club cell has a roundish central vacuole and well-developed microfilaments in the cytoplasm. Granular cells are occurs in the middle and basal epidermis , and the cytoplasm is occupied with membrane-bounded granules of electron dense. Chloride cells are present in the superficial epidermis , and the cytoplasm is occupied with tubular mitochondria. Three types of pigment cells can be distinguished by electron density of cytoplasmic inclusions.
