Transepidermal Water Loss, Stratum Corneum Hydration and Transmission Electron Microscope Observation on Acetone Damaged Canine Skin Barrier Model (아세톤에 의해 손상된 개 피부장벽 모델에서의 경표피수분소실도 및 각질층보습도 평가와 전자현미경적 관찰)

  • The purpose of this study is to establish experimental canine skin barrier disruption model in dog. The study was designed to investigate the predictive properties of acetone damage using as effect parameters transepidermal water loss (TEWL) and stratum corneum (SC) hydration. To compare the structures of SC intercellular lipids in normal and acetone damaged skin, TEM observations were performed. Six clinically normal, male Beagles without dermatological problems were chosen for this study. Acetone damage was performed at 48hrs after clipping. Efficacy measurements for TEWL and skin hydration were performed before ($t_1$) and after ($t_2$) damage in a temperature- and humidity-controlled room ($2{0\sim}22^{\circ}C$, 50-60%). TEWL and SC hydration values were decreased in the acetone damaged model compared with non damaged skin. In TEM observation of acetone damaged sample, the intercellular lipid lamellae exhibit abnormal and incomplete structure compared with those of normal skin. It seems that acetone damage would be one of canine skin epidermal barrier disruption model for the study of canine atopic dermatitis (AD) as well as dry skin in veterinary dermatology research.

  • 두가지 방법의 Tension Suture가 개에서 손상을 입은 피부의 접합에 평가되어졌다. 이들 술식은 Vertical Mattress Tension Suture와 'Walking'봉합이다. 7주후에 타원형으로 손상된 피부의 반흔조직을 추적하여 면적을 측정하였다. 흠손된 피부의 반흔조직의 평균 넓이는 V.M.T.S의 $5.29cm^2$에 비해 'Walking'봉합의 경우는 $2.07cm^2$였다. 이는 'Walking'봉합의 경우가 명백히 반흔조직이 좁다는 것이 관찰되었다. 더구나 어떤 손상의 경우더라도 'Walking'봉합은 열개(봉합한 자국)가 발견되지 아니했다.

Visual Imaging of Calcium Ion Distribution in Acetone and Tape Stripping Damaged Canine Epidermis (개에서 피부손상에 의한 표피내 칼슘이온 분포상)

  • The purpose of this study is to establish experimental canine skin barrier disruption model, the study was designed to observe calcium ion in skin frozen tissue of canine skin and also the modulation of calcium ion distribution of normal skin with disrupted skin such as clipping, acetone, tape stripping damages according to time. To compare the changes of calcium ion gradient after damages, the distribution of calcium ion in the canine epidermis was visualized by blotting to gel containing chemical indicator (Calcium Green-1) with fluorescent microscope and the effects of skin barrier damages were examined according to time. Three mins and 1hr after acetone damage, the gradations of epidermis and hair follicle showed more radiant and disappeared after 48 hrs. On the contrary, 3mins and 1hr after tape stripping damage, the gradations showed more radiant than those of acetone damage, and these gradations were stabilized after 48 hrs. The method we presented here could show the visual image of the calcium ions in frozen tissue without further preparation, and it might be useful to investigate the role of calcium ion in the canine epidermal barrier recovery, however, it might be need further methodological improvement to get accurate quantitative information.

The Protective Effect of Mineral Water Against UVB Irradiation (미네랄워터의 피부 장벽 보호 효과 연구)

  • Bicationic minerals such as calcium and magnesium are known to protect the skin barrier. The principal objective of this study was to evaluate the skin barrier protective effects of mineral water, which is composed of calcium, magnesium, manganese, and fluorine. UVB irradiation induces a destruction of tight junction (TJ) components. The TJ permeability barrier was also disrupted by UVB irradiation. We employed a skin equivalent model to assess the efficacy of mineral water in this regard. Mineral water maintained the structure of the skin equivalents following UVB irradiation. The results of the TJ permeability assay showed that mineral water helped to maintain the TJ permeability barrier after UVB irradiation in skin equivalent model. Mineral water supported the structure of TJ components and restored the occludin protein level in differentiated normal human keratinocytes after UVB irradiation. In conclusion, we found out the protective effect of mineral water against UVB irradiation.

Gamma-ray-induced skin injury in the mini-pig: Effects of irradiation exposure on cyclooxygenase-2 expression in the skin (감마선조사에 의한 돼지 피부장애에 cyclooxygenase-2의 발현변화)

  • The basic concepts of radiation-induced skin damage have been established, the biological mechanism has not been studied. In this study, we have examined the effects of gamma rays on skin injury and cyclooxygenase(COX)-2 expression. Gamma irradiation induced clinicopathological changes in a dose- and time-dependent manner in mini-pig skin. The histological changes were consistent with the changes in gross appearance at 12 weeks after irradiation. After three days' irradiation, apoptotic cells in the basal layer were found more frequently in irradiated skin than in normal skin, with the magnitude of the effect being dose-dependent. The thickness of the epidermis transiently increased 3 days after irradiation, and then gradually decreased, although changes in the epithelial thickness of the irradiated field were not observed with irradiation doses over 50 Gy. In the epithelium, there was an initial degenerative phase, during which the rate of basal cell depletion was dependent on the radiation dose (20-70 Gy). One week after irradiation, COX-2 expression was mostly limited to the basal cell layer and was scattered across these cells. High COX-2 expression was detected throughout the full depth of the skin after irradiation. The COX-2 protein is upregulated after irradiation in mini-pig skin. These histological changes associated with radiation exposure dose cause the increased COX-2 expression in a dose-dependent fashion.

Protective Effect of Glycyrrhiza glabra Extract on UV-induced Skin DNA Damage (감초추출물(Glycyrrhiza glabra Extract)의 피부에서의 DNA 손상 방지효과)

  • Ultraviolet B (UVB) damages DNA residues in skin keratinocytes. In particular, the formation of cyclobutane pyrimidine dimers (CPD), a pyrimidine residue damage in DNA, is considered a representative indicator of skin photoaging. In this study, we confirmed defensive effect of Glycyrrhiza glabra (G. glabra) extract against UVB induced DNA damage. First of all, we confirmed UVB dependent amount of CPD formation in human keratinocyte cell line. UVB induced CPD was decreased by G. glabra extract by dose dependent manner. In addition, it was confirmed that the expression of mRNA of DNA damage recovery factors was increased by G. glabra extract. Consequently, through this study, it was possible to confirm the DNA protection effect of G. glabra extract in skin keratinocytes.

Effects of Pleurotus eryngii extract against inflammation in activated RAW 264.7 cells and UV-induced skin damage in mice (활성 RAW 264.7 세포에서 항염증 및 자외선 유도 마우스 피부손상의 개선에 대한 새송이 추출물의 효과)

  • The aim of this study was to investigate the anti-inflammatory and ultraviolet (UV)-protective effect of Pleurotus eryngii extract (PEE) in activated RAW 264.7 cells and UV-induced mouse skin damage. The results showed that PEE strongly inhibited the production of inflammatory mediators such as nitric oxide, interleukin $(IL)-1{\beta}$, and IL-6 at high concentrations in LPS-stimulated RAW 264.7 macrophages. In addition, PEE treatment suppressed erythema, melanin index, and epidermal thickness to a greater degree than ascorbic acid (AA) treatment in UV-irradiated mice. Finally, PEE treatment inhibited the infiltration of mast cell to a similar degree of AA treatment. Therefore, these results indicate that PEE could improve inflammation and skin damage in immune cells and UV-irradiated mice. This study may provide positive insights into PEE as a functional food and cosmetic ingredient for treatment of inflammation and skin damage.

Effect of Topical Application of Intercellular Lipids on Sodium Lauryl Sulphate-Damaged Skin Barrier Function in Dogs (개에서 sodium lauryl sulphate에 의한 손상 피부에 대한 각질세포간 지질의 국소적용 효과)

  • Ceramide, cholesterol and free fatty acids are the major intercellular lipids, maintaining the integrity of the skin barrier. However, the roles of these lipids in canine skin barrier function are little known. The aim of this study was to evaluate the repairing effects of 2% ceramide (CER), 2% cholesterol (CHO), 2% linoleic acid (LIN) and 2% intercellular lipid mixture (ILM) on damaged canine skin barrier by 1.25% sodium lauryl sulphate (SLS). Transepidermal water loss (TEWL), skin hydration, skin pH and skin thickness were assessed. Histological profiles and transmission electron microscopic (TEM) profiles were assessed on day 12. SLS effectively induced the canine skin barrier damage. TEWL was significantly decreased by topical application of CER and ILM in SLS and vehicle-treated skin on day 8 and 12, respectively (p < 0.05, p < 0.0 I). By end of the experiment all lipids significantly decreased the TEWL as compared with SLS and vehicle control, but CER and ILM more significantly decreased the TEWL than UN and CHO, respectively (p < 0.01). Skin hydration was significantly increased by CER and ILM during experimental periods (p < 0.01). Skin pH was significantly decreased by CER, LIN and ILM. In histological profiles, the thickness of the stratum corneum (SC) was significantly increased by the SC lipids as compared with vehicle and SLS (p < 0.01). Especially, CER and ILM showed more prominent improvement of barrier recovery. In TEM of the SC, SLS induced exfoliations of corneodesmosomes in the SC, and CER and ILM effectively protected exfoliations of corneodesmosomes on SLS-damaged canine skin. These results indicated that topical application of CER and ILM dramatically improved damaged-skin barrier function by SLS. Also, it was considered that the use of CER or ILM was recommended for the management of skin barrier dysfunction by irritant and inflammatory skin disorders such as atopic dermatitis.
