Infection and Rapid Detection of Perkinsus sp. In Cultured Babyneck Clam, Ruditapes philippinarum from Western Coast of Korea (서해안 양식 바지락에 발생한 Perkinsus sp. 감염증과 신속검출)

  • An apicomplexan parasite, Perkinsus sp. was observed from the cultured baby clams, Ruditapes philippinarum, collected from the coast of Kochang and Taean (South Korea), where it caused seasonal mortality of clams. Several milky-white cysts were observed on the surface of gill and visceral mass of parasitised clams. The trophozoites of parasite had eccentric nucleus and proliferated by schizogony in gill, mantle, hepatopancrease and reproductive tissues, resulting in the formation of granuloma and the intensive infiltration of hemocytes in the tissues. During incubation in FTM, trophozoites increased in size, resulting in prezoosporangia which appeared as round black spheres when colored with Lugols iodine solution. The prevalence of Perkinsus sp. in clams was Kochang, 73.1%; Taean, 94.8% (during 9-mo. survey) and showed size-dependent infection. Hemacolor kit was useful to reduce time for diagnosis of the trophozoite of Perkinsus sp. that has been responsible of massive motalities in the clam.

A report on the 2012 mass summer mortalities of black rockfish, Sebastes schlegeli in the Southeast Sea, Korea (2012년 고수온기에 발생한 남동해권역 조피볼락의 대량폐사)

  • From July to early September 2012, there was mass mortality of fishes, particularly black rockfish, which were being raised in the floating fish cage along the coast of Gyeongsangnam-do. The amount of damage was 1,802,000 fishes and the causes were confirmed to be rapidly rising water temperature and repeated daily changes in water temperature. The water temperature in this area of the sea rose to the maximum $28.4^{\circ}C$ and the daily range of changes in water temperature was maximum $6.5^{\circ}C$. As a result of investigating biological diseases of 194 fishes in 49 fishery areas, major pathogenic organisms such as red seabream iridovirus (RSIV), Vibrio sp. and Vibrio spp. or Microcotyle sp. were detected in rockfish in some fish farms. It is considered that the major causes of the mass mortality were high water temperature accompanied by repeated daily changes in water temperature, it is considered that biological diseases influenced the increase in the perish of fishes.

  • 1. 질병명 : Neosporosis 2. 본 질병의 개요 역사 및 역학 : 1984년 노르웨이에서 부전마비로 폐사한 개를 검사하던 중 뇌척수염과 근염을 확인하고 T. gondii와 다른 새로운 원충성 질병을 최초로 보고한 이래 Dubey등이 미국에서 톡소플라즈마 감염으로 진단되었던 30두의 개조직에 대한 재검토를 실시하여 그 중 10 두는 새로운 원충성 질병으로 확인한 후 Neospora caninum을 명명하였다. 현재까지 본 원충은 개, 소, 염소, 말, 사슴 등에서 자연발생 예가 보고되었다. 특히 소에서는 유산의 한 주요한 원인체임이 한국을 비롯한 여러 나라에서 확인되었다.   • Iridoviridae과에 속하는 lymphocystis disease virus(LDV)는 비교적 큰 20면체 DNA 바이러스이며(Murphy et al., 1995), 림포시스티스병(lymphocystis disease, LD)을 유발하는 원인체로서 140여종 이상의 해수 및 담수 어종에서 보고된 바 있다(Wolf et al., 1966). 우리 나라의 경우, 주요 양식 어종인 넙치에 주로 감염되어 2차 감염과 빈혈 및 합병증 등에 의해 폐사를 유발할 뿐만 아니라 상품성을 저하시킴으로서 경제적 손실을 야기한다.   • About 10% mortality occurred in cultured rainbow trout Oncorhynchus mykiss at a marine farm in Jeju in 2014. Diseased fish showed markedly abdominal distension and distended stomach. Although parasites, bacteria or viruses were not isolated from diseased fish, numerous Candida sp. were isolated from distended stomach. In experimental infection with Candida sp., mortality was not observed in most fish and clinical sign of distended stomach was not observed in the fish. These results suggest that Candida sp. may not be the etiologic agent of stomach distension syndrome. This is the first report of stomach distension syndrome in seawater farmed rainbow trout in Korea.

The Effects of Different Farrowing Space on Lactating Sow Performances and Growth Performances of Piglets (분만 면적의 차이가 분만돈의 생산성과 자돈의 성장능력에 미치는 영향)

  • This study was conducted to investigate the effect of different farrowing space, narrow and wide, on the sow performances and piglet growth performances. The $1.5m^2$ of narrow farrowing space was determined as the size of farrowing crate. The $3.0m^2$ of wide space for farrowing sow was the same of farrowing pen to allow the behavior freedom of sow. Baby piglets in the wide farrowing space was protected from crushing of sow by installation of safety bar. The pregnant sows used in this study were stall-housed during gestation and moved to each farrowing spaces on 8 d before parturition. Feed intake, backfat thickness and body condition score of sow were not affected by both farrowing spaces. However, the changes in backfat thickness and body condition score between farrowing and weaning in wide farrowing space were lower (p<0.05) than in narrow farrowing space. The return to estrus of sow was remarkably decreased in wide farrowing space compared to sows in narrow farrowing space. The lower number of stillbirth and higher mortality of piglets were observed in wide farrowing space. From the results, although wide farrowing space could be practically acceptable in terms of sow performances, possible cause of mortality of piglets should be scrutinized through observation of piglet and sow behaviors.

Characterization of an isolated reovirus from the paradise fish Macropodus opercularis imported from Southeast Asia (동남아시아로부터 수입된 paradise fish Macropodus opercularis로부터 분리한 reovirus의 특성)

  • In 2008, mass mortality was observed in paradise fish Macropodus opercularis which was imported from Indonesia. PCR of these fish found positive for megalocytivirus and Mycobacterium sp., while an unidentified virus was culture-isolated using CHSE-214 cells. In the present study, we investigated characterization of the unidentified virus and its pathogenicity to determine whether the virus was the causative agent of the mass mortality of paradise fish. The unidentified virus induced cytopathic effect (CPE) with syncytia in CHSE-214 and other fish cells, BF-2, GF, SSN-1, FSP and FFN. The virus was resistant against treatments with IUdR, chloroform, acidity at pH 3, basicity at pH 11 and high temperature at $56^{\circ}C$ for 3h. By electron microscopy, the viral particles were spherical having a double capsid structure with approximately 65 nm in external diameter. Viral genome was composed of at least 10-segmented RNA with sizes ranging from 0.7 kb to 3.6 kb. Based on these characters, this virus can be classified into family Reoviridae. This reovirus did not cause any mortality in an artificial experiment conducted by injecting the virus to paradise fish. This indicates that the reovirus is not only responsible for the mass mortality of paradise fish in 2008.

Development of an environment-friendly moving aquatic animal rendering equipment and evaluation of fertilizer value for recycling of fish waste (친환경 이동식 수산생물 폐사체 처리장치 개발 및 재활용을 위한 비료 가치 평가)

  • Although aquaculture production rates grown over the years, aquatic animal diseases occur every year which causes substantial economic losses. When an aquatic animal is infected with an aquatic animal pathogen it is either incinerated or buried according to the aquatic life disease control act. Although these methods prevent the spread of disease, it is not environment friendly. Here, we developed an aquatic animal rendering equipment for disposal of fish waste which is environment-friendly and efficient. Also, fertilizer components of fish waste were evaluated value for recycling. The mobile rendering equipment was designed for field operation and/or high temperature and pressure system, oil and water separator, and shredding drying apparatus. During the experiment (July-2016 to November-2016), a total of 53,824 kg fish waste was collected, and 29,216 kg compost of rendering by-product was made. Also, compost made from viral (Viral hemorrhagic septicemia virus) infected fish did not reflect any detectable pathogen. The concentration of nitrogen, phosphorus, and organic matter in the fish waste compost were 2.17%, 26.98%, and 92.44%, respectively. The results suggest that fish waste used in this study was decomposed efficiently as per the official standard for fertilizer product. This equipment can be useful for efficient inactivation of the aquatic animal pathogenic agents and recycling of the fish waste in an environment-friendly manner.

The Infection Characteristics of Vibrio scophthalmi Isolated from Olive Flounder, Paralichthys olivaceus (양식 넙치, Paralichthys olivaceus에서 분리한 Vibrio scophthalmi의 감염 특성)

  • Recently high mortality of cultured olive flounder, Paralichthys olivaceus occurred frequently at the fish farms in Ulsan, Korea. The diseased fish showed skinny body and swimming behavior around the water surface with liver atrophy and white enteritis as internal signs. The isolated bacteria were identified to V. scophthalmi by biochemical test, nucleotide analysis of 16S rRNA and dnaJ gene sequencing. The pathogen of this study showed strong pathogenicity as 75% mortality to olive flounder by intraperitoneal injection of $1{\times}10^6$ CFU/fish. The pathological sign was not different between the naturally diseased fish and the artificially infected fish. Histopathological changes were shown to liver atrophy, desquamation of the intestinal mucosa and hyaline droplet like as other previous studies.

An Outbreak of Malaria in Penguins at the Farm-land Zoo (자연농원 동물원에서의 펜귄의 말라리아 발생보고)

  • In June of 1983 authors had investigated the occurrence of the avian malaria in the Humboldt penguins imported from Japan to the Farmland Zoo in central district of Korea. The disease was characterized by acute course and high mortality. The peripheral blood smears from the affected penguins demonstrated different developmental stages of Plasmodium sp. in the mature erythrocytes. The predominant gross lesions noticed were pulmonary and epicardial edema and hepato-splenomegaly. Microscopically the lesions were characterized by extensive reticuloendothelial cell hyperplasia with striking feature of exoerythrocytic schizogony affecting a variety of tissues. The report also signifies the first description of a disastrous epizootic of avian malaria in Korea.
