The object of this study was to investigate nitrogen fixation activity of rhizobium inoculated at seed coat when drought condition was applied in flowering period of soybean c.v. Samnamkong. The rhizobia used in this experiment were indigenous rhizobium, R2l4, RJl-29, USDA110 and USDA122. The experiment was done with 1/2000 Wagner pots in laboratory and greenhouse and was tested in completely randomized design with four replications. Nitrogen fixation activity in coventional culture was the highest in R2l4 and indigenous rhizobium among the five rhizobia strains. As given drought condition from flowering to maturity, nitrogen fixation activity was higher in R2l4 and RJl-29 than indigenous rhizobium. Leaf area and relative index (drought/convention) of pod weight were higher in USDA122, RJl-29 and R2l4 than indigenous rhizobium as given drought condition from flowering to maturity. High positive correlation was observed between nitrogen fixation activity of rhizobium and relative index(drought/convention) of pod weight. High negative correlation was observed between respiration of plant and relative index (drought/convention) of leaf area.

To investigate the effect of changes in microbial communities on arsenic release in soil, experiments were conducted on arsenic-contaminated soils (F1, G7, and G10). The experiments involved three groups of the experimental sets; ① BAC: sterilized soil + Bacillus fungorum, ② IND: indigenous bacteria, and ③ MIX: indigenous bacteria + B. fungorum, and incubated them for seven weeks using lactate as a carbon source under anaerobic conditions. The experimental results showed that higher concentrations of arsenic were released from the IND and MIX soils, where indigenous bacterial communities existed, compared to BAC. Significantly higher levels of arsenic were released from the G10 soil, which showed higher pH, compared to the F1 and G7 soils. In the G10 soil, unlike other soils, the proportion of As(III) among the released arsenic was also low. These results may be attributed to differences in microbial community composition that vary depending on the soil. By the seventh week, the diversity of microbial species in the IND and MIX soils had significantly decreased, with dominant orders such as Eubacteriales and Bacillales thriving. Bacteroidales in the seventh week of the MIX in the F1 soil, Rummeliibacillus in the seventh week of the IND and MIX of the G7 soil, and Enterobacterales in the IND and MIX of the G10 soil were dominant. At present, it is not known which mechanisms of microbial community changes affect the geochemical behavior of arsenic; however, these results indicate that microbiome in the soil may function as one of the factors regulating arsenic release.
This experiment was conducted to find out the some physiological characteristics and nitrogen fixation activities under free-living conditions of indigenous rhizobia isolated from soybean-cultivated (30 fields) and un-cultivated (30 fields) soil. The results were summerized as follows: 1. In free-living condition, only 12.8% and 6.4% of the indigenous rhizobia isolated from soybean cultivated (133 strains) and un-cultivated (125 strains) soils were nitrogenase positive as more than 4 n mole $C_2H_4$ per tube per hour by acetylene reduction assay. 2. The acid-producing rhizobia on litmus milk test was observed to be 20% of the total 160 strains isolated from soybean cultivated soil but about 34% of 166 strains isolated from un-cultivated soil. And the serum zone positive strains were higher in the soybean un-cultivated soil than cultivated soil. 3. The population ratio of fast-to slow-growing indigenous rhizobia based on growth pattern of AMA medium was 35.6% to 64.4% of the total 346 strains. 4. The population of indigenous Rhizobium japonicum counted by MPN method was ranged from $9.2{\times}10^2$ cells per gram of soil in soybean un-cultivated soil to $2.3{\times}10^4$ cells per gram of soil soybean cultivated soil. The number of indigenous R. japonicum in 0-10cm depth of surface layer was higher than low layer.
An antifungal substance was purified from culture broth of Pseudomonas flulorescens 2112 that showed a broad-spectrum antagonistic activity against various phytopathogenic fungi including capsici. The substance was identified as 2,4-diacetylphloro-glucinol basd on NMR analysis. The 2,4-diacetylphloroglcinol showed antibiotic activity in broad acidic range from pH 1.0 to pH 9.0. About 83% of initial activity was remained after incubation for 30min ar $60^{\circ}C$, however, the activity was dropped up to 50% after 30 min incubation in $80^{\circ}C$. When the nucleotides of P. capsici treated with 2,4-diacetylphloroglucinol were labeled with[$^{3}$ H]-Adenin, the newly synthesized and radioactive-labeled RNA was significantly reduced than those of untreated P. capsici. indicating that the 2,4-diacetylphloroglucinol inhibits RNA synthesis.
Acid mine drainage (AMD) producing mine tailings can be beneficially recycled to generate electricity by applying fuel cell technology. Pyrite-containing mine tailings and indigenous bacteria from abandoned mine areas were used to construct fuel cells to investigate the effect of pyrite contents and the presence of iron-oxidizing bacteria. The results showed an enhanced electrical performance with a higher content of pyrite in mine tailings. The inoculation of the indigenous bacteria also enhanced the current density by about three times, and the power density by about 10 times. Overall, this study shows that the combined use of the ecological function of indigenous bacteria from mine areas and mine-tailings in fuel cells does not only contribute to reducing harmful effects of mine tailings but also generate electricity.
Effects of the indigenous Vesicular-arbuscular mycurrhizal fungi(VAMF) on early growth response of greenhouse grown crops were experimented. This study was done to evaluate the benefit of indigenous VAMF inoculation on the early growth and the subsequent growth after transplanting of some crops such as cucumber, tomato, hot pepper, eggplant, and melon. Leaf area, shoot dry weight, and plant length of mycorrhizal greenhouse crops showed the tendency of significant or no significant increase over control plants receiving no inoculation. The levels of VA mycorrhizal colonization were increased with plant growth, and infection rates of horticultural crop except hot pepper around one week after transplanting were decreased, while that of 8 weeks after emergence of mycorrhizal seedlings were increased again and infected by around 50% at harvesting time. In spore densities in the rhizosphere soil of craps experimented, the number of spore ranged from $72.7{\pm}26.3$ to $100{\pm}10.3g^1$ on dried soil basis and high density showed in both cucumber and tomato. Total nitrogen contents in shoots were lower in the mycorrhizal plants than non-mycorrhizal one, whereas P uptake in mycorrhizal hot pepper and tomato were highly ramarkable. The K contents in the shoots of mycorrhizal cucumber and eggplent were highly enhanced. Inoculation of the indigenous VAMF enhanced shoot Ca and Mg in both tomoto and melon. The contents of Fe, Zn, Mn and Cu in shoots of mycorrhizal crops were higher than non-mycorrhizal plants and vice versa in case of eggplent. Inoculation of the indigenous VAMF to horticultural crops were effective for alleviation of transplanting shock, and pretransplanting infection improved subsequent growth by reducing the time required for establishment of a functional mycorrhizal symbiosis following transplanting.
Sheath rot and dry rot disease caused by Pseudomonas marginalis and Fusarium oxysporum were serious problems in garlic farmland. In this study, total of 160 indigenous antagonistic bacteria were isolated from 16 farmlands in Yeongcheon, Korea. Among these, 15 strains were able to inhibited P. marginalis and F. oxysporum. The 16s rDNA genes of the selected 15 strains were amplified and sequenced. The strains has strong antagonistic ability against garlic pathogens was achieved Bacillus subtilis YC82, B. vallismortis YC84, B. amyloliquefaciens YC240. The selected 3 strains tested for investigation of antifungal mechanisms further analyses; 3 strains of these validated for production of siderophore, ${\beta}$-glucanase and chitinase using CAS (chrome azurol S) blue agar, CMC-congo red agar and DNS method. The 3 strains were able to utilized insoluble phosphate as dertermined by vanado-molybdate method. The 3 strains verified for production of auxin and gibberellic acid using Salkowski test and holdbrook test. Also, 3 strains showed stimulation germination, stem growth promoting activity on the in vivo test. The 3 strains were able to effectively suppress P. marginalis and F. oxysporum causing sheath rot and dry rot diseases on the in vivo pot test.
In order to improve effectiveness of rhizobia-legume symbiotic nitrogen fixation, ecological and physiological characteristics of indigenous rhizobia distributed in Korea soil, that is, the effects of soybean cultivation, physico-chemical properties and climate on the population of indigenous rhizobia and other soil microbes were investigated. The results were summarized as follows: The population of indigenous rhizobia were ranged from $5.1{\times}10^4$ cells to $196.8{\times}10^4$ cells per gram of soil in soybean cultivated soils but from $1.6{\times}10^4$ cells to $78.6{\times}10^4$ cells per gram of soil in soybean un-cultivated soils sampled from 9 different agro-climate zone. The highest population was observed in a soybean cultivated loamy soil from southern part of Korea. The content of available phosphate, exchangeable Ca, Mg, Cu, and B in soil were positively correlated but active Fe, exchangeable Al, Na, and $SO_4$ were inversely correlated to the population of indigenous rhizobia. The inverse relationship was observed between the number of indigenous rhizobia and actinomycetes.

Eightyseven strains of indigenous Rhizobia were isolated from the nodule of soybean cultivar, Danyup, cultivated in four different soils sampled from continuously soybean cultivated and newly reclaimed fields. The strains were grouped into Bradyrhizobium japanicum (slow-grower:55 strains) and Rhizobium fredii (fast-grower: 32 strains). The both groups could be divided into two sub-groups according to the denitrification characteristics, that is, denitrifying fast-grower (F-I), nitrate respiring fast-grower (F-II), denitrifying slow-grower (S-I). and nitrate respiring slow-grower (S-II). Among the 87 isolates, F-I, F-II, S-I and S-II sub-groups were 10, 22, 48 and 7 strains, respectively. The one-and two-dimensional polyacrylamide gel electrophoretic pattern of the four sub-groups were compared and discernible difference was observed between fast and slow-grower, but the difference was not discernible between subgroups within the same growth rate group.
