항생물질과 RNA 분해효소를 동시에 생산하는 방선균의 한 균주를 토양으로부터 분리하여 이 균주가 생산하는 RNA 분해효소의 물리화학적 성질 및 분해산물에 대해 검토하였다. 효소반응의 최적 pH 및 온도는 명명 pH 5.6과 $50^{\circ}C이었다.$$37^{\circ}C$ 에서 90분간 열처리 시켰을 때, 이 효소의 활성은 비교적 안정하였지마는 $50^{\circ}C$ 에서 90분간 열처리 시켰을 때는 효소활성이 심하게 저하되었다. 이 효소의 활성은 $Ba^{2+}$ 에 의하여 50% 정도의 저해 작용을 나타내었지만 EDTA에 의해서는 저해되지 않았다. 이 효소에 의한 RNA분해로 이 효소가 대사산물로서 guanosine, adenosine과 밝혀지지 않은 두 가지의 핵산관연물질을 생산함을 알 수 있었다. 제2보에서는 ENase의 효소학적 성질을 검토하였으며 이후 항생물질 측면에서 검토할 예정이다.
Probiotics have been isolated from various environments and Bacillus species are advantageous among the probiotic bacteria due to their ability to form endospores that can compensate for the limitation of typical probiotics such as lactic acid bacteria. The aim of this study is to investigate the probiotic potential of Bacillus species from Ulleungdo soil, known as an unpolluted environment in Korea. Soil samples were collected from various areas of Ulleungdo, and Bacillus spp. were isolated, and assessed for antibiotic resistance and enzymatic activity. Six Bacillus spp. were not resistant to all tested antibiotics and subsequently tested for enzyme activity. We found the six Bacillus spp. were all inactive β-glucuronidase enzyme, which can have detrimental effects on human health, and one of Bacillus spp. showed an activity of Leucine arylamidase suggesting its probiotic potential.
The effects of some soil conditions on the degradation rate and decomposing pattern of thiolix were investigated and the obtained results are summarized as follows: Thiolix degraded more rapidly in flood soils than in noon-flooded, and in wet soils than in dry soils under non-flooded soils. The degradation rates in non-flooded soils increased with higher pesticide concentration. Thiolix was more persistent in non-flooded soils under soil sterelization than under non-sterilization and degraded rapidly in glucose application. The metabolites identified from the soils by TLC and GLC include Thiolix alcohol, Thiolix sulfate, Thiolix ether and a unknown metabolite. Soil enzyme, acid phosphatase activity decreased at higher pesticide concentration, lower moisture contents of soil and the activity in glucose application was increased. Soil enzyme, urease and dehydrogenase activity decreased at higher pesticide concentrations.
Wetlands constitute a transitional zone between terrestrial and aquatic ecosystems and have unique characteristics such as frequent inundation, inflow of nutrients from terrestrial ecosystems, presence of plants adapted to grow in water, and soil that is occasionally oxygen deficient due to saturation. These characteristics and the presence of vegetation determine physical and chemical properties that affect decomposition rates of organic matter (OM). Decomposition of OM is associated with activities of various extracellular enzymes (EE) produced by bacteria and fungi. Extracellular enzymes convert macromolecules to simple compounds such as labile organic carbon (C), nitrogen (N), phosphorus (P), and sulfur (S) that can be easily taken up by microbes and plants. Therefore, the enzymatic approach is helpful to understand the decomposition rates of OM and nutrient cycling in wetland soils. This paper reviews the physical and biogeochemical factors that regulate extracellular enzyme activities (EEa) in wetland soils, including those of ${\beta}$-glucosidase, ${\beta}$-N-acetylglucosaminidase, phosphatase, arylsulfatase, and phenol oxidase that decompose organic matter and release C, N, P, and S nutrients for microbial and plant growths. Effects of pH, water table, and particle size of OM on EEa were not significantly different among sites, whereas the influence of temperature on EEa varied depending on microbial acclimation to extreme temperatures. Addition of C, N, or P affected EEa differently depending on the nutrient state, C:N ratio, limiting factors, and types of enzymes of wetland soils. Substrate quality influenced EEa more significantly than did other factors. Also, drainage of wetland and increased temperature due to global climate change can stimulate phenol oxidase activity, and anthropogenic N deposition can enhance the hydrolytic EEa; these effects increase OM decomposition rates and emissions of $CO_2$ and $CH_4$ from wetland systems. The researches on the relationship between microbial structures and EE functions, and environmental factors controlling EEa can be helpful to manipulate wetland ecosystems for treating pollutants and to monitor wetland ecosystem services.

인공적으로 디젤류에 오염시킨 5kg의 토양이 충전된 두 개의 실험 column을 이용해서 외부 온도변화 효과를 최소화하기 위해서 $25^{\circ}C{\pm}2.5$로 유지시켰으며, 유량속도도 10ml/min로 일정하게 유지시켰다. 또한 수분은 활동범위 능력의 $60{\sim}80%$에서 유지시켰다. 따라서 연속식과 간헐 주입식 모드를 운전비용의 감소 및 효율의 향상 측면에서 모드의 평가를 위한 방법으로 이용기간 동안 총세균 수, 유류분해균 수의 변화, 탈수소효소 활성도의 변화 추이 분석을 하였다. 두 venting 모드별로 본 총 종속영양균 수와 유류분해균 수는 모두 대등한 성장률을 보였으며, 반응일 수에 따라 비교된 탈수소효소 활성도 반응 90일 동안 비슷한 활성을 유지 했다.

Fibrinolytic enzyme is thought to be involved in extracellular matrix remodeling during regeneration. We investigated the expression and characterization fibrinolytic enzyme activity during earthworm tail regeneration. Electrophoretic analysis of fibrinolytic enzymes induced during regeneration revealed that at least seven types of fibrinolytic enzymes were expressed, which had molecular weight of 12, 19, 23, 27, 32, 45 and 58 kDa, respectively. These fibrinolytic enzyme activities were dramatically increased within 1 day after amputation. These activities were maintained by 7 days postamputation, followed by decrease to control level from 14 days after amputation. Alltypes of fibrinolytic enzyme activities were inhibited by treatment of PMSF and aprotinin, and were insensitive to EDTA and exogenous Ca$^{2+}$. These results indicate that the fibrinolytic enzymes are serineproteinase. Other characteristics including specificities for extracellular matrix proteins are under investigation. Based on these results, we are trying to find out the relationship among expression of proteinases, extracellular matrix remodeling, and dedifferentiation, which are believed to be essential processes during regeneration.
One hundred-eighty extracts of soil collected from ginseng (Panax ginseng C.A. Meyer) fields were subjected to lettuce germination test, electrolyte leakage, cell viability and antioxidant enzyme activity test. Regardless of various cultivation periods, there was no significant difference in soil pH, the content of organic matter and available phosphate in ginseng fields. Based on lettuce seed germination test, six soil extracts showing inhibition of germination and/or seedling growth were selected for further study. Selected soil extracts markedly inhibited cell viability of ginseng cultured cells but leakage of electrolytes were not affected by the treatment. Enzyme activity of superoxide dimutase in ginseng cultured cells was not affected by the treatment with the soil extracts. However, those of peroxidase and catalase were significantly inhibited by the treatment with soil extracts which showed inhibition of lettuce seed germination and seedling growth.

갈대(Phragmites communis Trin.)란 염분이 있는 곳에서 자라는 염생식물로서 우리나라 전역에 분포하고 있다. 지난 50년 동안 우리나라의 여러 습지에 걸쳐서 갈대는 우점종으로 자라왔고, 육지와 수중서식지에서 갈대의 확산범위는 증가하고 있다. 갈대의 확산은 다른 습지 식물의 서식지를 파괴하고, 갈대가 번식하면 동 식물들의 번식 자체가 어려울 뿐 아니라 갈대숲에 포식자가 늘어나 살아가기 어려운 환경으로 변하기때문에 갈대를 체계적으로 관리할 수 있는 방안이 마련되어야 한다. 본 연구는 우점종이 다른 두 습지에서 갈대군집의 성장률을 관찰하고, 토양의 화학적 분석과 식물의 생리적 분석을 통해 갈대군집 성장에 미치는 영양염류의 영향을 규명하였다. 연구 대상지는 한강하구에 위치한 장항습지와 성동습지로서 동일하게 갈대가 분포하며, 장항습지에는 줄 군락이 성동습지에는 새섬매자기 군락이 우점하고 있다. 분석 항목은 이화학적 항목을 비롯하여 용존유기탄소(DOC, dissolved organic carbon), 체외미생물효소활성도(Extracellular enzyme activities), 암모니아성 질소($NH_4^+$), 질산성 질소($NO_3^-$)을 분석하였다. 실험결과, 두 습지 갈대의 성장은 7월부터 9월에 증가하였고 성동습지의 토양성분이 점토질로 형성되어 높은 수분함량과 유기물함량을 유지하고 있기 때문에 갈대의 밀도가 높고 성장률이 활발한 것으로 나타났다. 또한 미생물활성과 환경인자간 양의 상관관계를 보아 환경인자들이 미생물 활성을 자극하고 미생물들은 식물의 성장을 촉진하여 영향을 주며, 반면 식물 뿌리는 enzyme을 생성하는 미생물에게 C 삼출물을 공급해 enzyme 활성에 영향을 미칠 것으로 사료된다.

A bacterial strain NO. AM-28, showing proteolytic activity against defatted soybean was isolated from domestic soil. The isolated strain was identified as Aeromonas hydrophila by both the biochemical tests using API kit and the analysis of cellular fatty acid profile with MIDI system. The protease production from A. hydrophila AM-28 was highly enhanced when it was cultivated in the medium containing glycerol as a carbon source, tryptone or $(NH_4)_2HPO_4$ as a nitrogen source, and $CaCl_2$ as a mineral source. The optimal pH and temperature for the enzyme was 8.0 and $65^{\circ}C$, respectively. The enzyme was stable up to $55^{\circ}C$ and at pH values ranging from 7.0 to 13.0. The enzyme activity was inhibited by phenylmethylsulfonyl fluoride and EDTA, indicating that serine residue and metal ions be involved in enzyme activity.
The extracellular cholesterol oxidase produced from a soil microorganism HSL613 was purified and partially characterized. Through a series of purification procedures including concentration with CH2 concentrator, DEAE-cellulose column chromatography and gel filtration on Superose12, the purified enzyme was shown to have a specific activity of 108 units/mg protein giving 30.8-fold purification and final yield of 66%. The molecular weight of the enzyme was estimated to be 59,500 daltons by SDS-PAGE. The optimum temperature and pH for this enzyme were $50^{\circ}C$ and 6.0, respectively. The activity of the purified cholesterol oxidase was inhibited by $Ag^{2+}$, $Hg^{2+}$ and SDS.
