Streptomyces shows a eukaryotic characteristic that vegetative cell can grow into mycelial form and has morphological and physiological differentiation at a certain period during its life cycle. Streptomyces has been used for the production of many biologically active compounds, such as antibiotics and pronase. Production of second metabolites and differentiation of the vegetative cell share the certain period of its lift cycle. Therefore, second metabolites may affect the differentiation of the vegetative cell. One of the microbial hormone, called A-factor, regulates the production of second metabolites, sporulation and differentiation of the cells. Streptomyces griseus produces streptomycin as well as many different kinds of proteinase. As mentioned, period of proteinases production overlaps with the period of differentiation of the vegetative cells. Protease may play a important role for the differentiation of the cells. In this paper, function of the SGPD gene cloned from S. griseus IFO 13350 tested whether it affects for the differentiation of A-factor mutated S. griseus HH1 and S. griseus IFO13350. pWHM3 and pWHM3-sprD plasmid was transformed into S. griseus HH1 and S. griseus IFO13350. Chymotrypsin activity of the cultured medium of the transformants with pWHM3-sprD plasmid didn't show any change with that of the transformants with plasmid only. The transformants with pWHM3-sprD plasmid didn't show the increase of the production of actinorhodin as well as morphological change in S. griseus IFO 13350 and HH1, as well. The promoter sequences of the SGPA and SGPB gene which encode chymotrypsin-like protease, were compared with that of SGPD gene. Regulatory mechanism of gene expression of proteinase genes will be studied for the development of high production system for protease as well as the function of the proteases.

Methyl tertiary butyl ether (MTBE) is a gasoline additive that boosts the oxygen content in fuel (an oxygenate), resulting in less air-polluting carbon monoxide being released from vehicle exhaust systems. Then, groundwater contamination problems have been developed in areas where the chemical is used. Common sources of water contamination by MTBE include leaking underground gasoline storage tanks and leaks and spills from above ground fuel storage tanks, etc.. Studies on the chemical and these problems are going on abroad vigorously. These studies should be performed in our country as well more actively. This paper reviews on articles on these studies and focuses on the identification of the chemical as a groundwater contamination source.

The effect of biological control of the root-knot nematode, Meloidogyne incognita, on cucumber and lettuce was evaluated with green manure crop species in greenhouse. Nematicidal effect of sudan grass cultivation in cucumber greenhouse was comparable to that of chemical treatment with fosthiazate GR, showing the high activity of 88.6%. Sudan grass cultivation in lettuce greenhouse significantly reduced the number of M. incognita in soil, showing 93.5% of nematiidal activity. In addition, since growth of sudan grass was superior to other green manure crop species, it is considered that cultivation of sudan grass is proper to control M. incognita in greenhouse.

There would be a possibility of uranium contamination around the nuclear power plants and the underground waste disposal sites, where the uranium could further migrate and diffuse to some distant places by groundwater. It is necessary to understand the biogeochemical behaviors of uranium in underground environments to effectively control the migration and diffusion of uranium. In general, various kinds of microbes are living in soils and geological media where the activity of microbes may be closely connected with the redox reaction of nuclides resulting in the changes of their solubility. We investigated the adsorption and precipitation behaviors of dissolved uranium on some solid materials using hydrogen gas as an electron donor instead of organic matters. Although the effect of hydrogen gas did not appear in a batch experiment that used granite as a solid material, there occurred a reduction of uranium concentration by 5~8% due to hydrogen in an experiment using bentonite. This result indicates that some indigenous bacteria in the bentonite that have utilized hydrogen as the electron donor affected the behavior (reduction) of uranium. In addition, the bentonite bacteria have showed their strong tolerance against a given high temperature and radioactivity of a specific waste environment, suggesting that the nuclear-biogeochemical reaction may be one of main mechanisms if the natural bentonite is used as a buffer material for the disposal site in the future.

The crucibles of Baekje Kingdom from the Ssangbukri Site which were used for glass and metal melting had light brown, grayish blue and grayish brown colored bodies. In thin section, the crucibles contained numerous quartz grains and pottery fragments. The surface was covered with fine grained quartz for thermal resistance. Based on decomposition of mica group minerals and formation of mullite detected by X-ray diffraction analysis, it was inferred that all crucibles have been fired over $1,000^{\circ}C$. It was also found that firing temperature has exceeded $1,100^{\circ}C$ in some crucibles because feldspar was not detected. The maximum temperature was assumed at $1,200^{\circ}C$. The magnetic susceptibility values and geochemical characteristics sorted out the crucibles into two groups that differed from the characteristics of the local soils. This reflected geological setting of the site where the alluvium was formed from two kinds of surrounding rock masses, granite gneiss and biotite granite. However, the local soils had similarities with the crucibles in weathering degree and geochemical behavior of major elements. In consequence, it was considered that the raw clay of the crucibles was supplied from the local area of the site.

The application of the Life Cycle Assessment (LCA) methodology to analyze the environmental impact to different swine waste treatment systems was investigated. The first part of LCA is to organize an inventory of parameters and emissions released due to the system under investigation. In the following step of the Life Cycle Impact Assessment, the inventory data were analyzed and aggregated in order to finally get one index representing the total environmental burden. For the Life Cycle Impact Assessment (LCIA) the Eco-indicator 95 method has been chosen because this is well documented and regularly applied impact method. Two different swine waste treatment systems such as aerobic and anaerobic digestion systems were chosen as an example for the life cycle impact analysis. For establishing the parameters to be assessed the agricultural environmental effects to above swine waste treatment systems, it has been observed that there was high at T-P emission in anaerobic digestion system and $CO_2$ emission in aerobic digestion system. For Eco-indicator values per environmental effect for swine waste treatment systems related to one tonne of swine waste, it was shown that there was a negative index for global warm potential and soil acidification in aerobic digestion system, but relatively high positive index for eutrophication in anaerobic digestion system.
Creeping bentgrass (Agrostis stolonifera) is one of the most popular turfgrasses for high-quality playing surface such as putting green on golf courses and athletic fields. Continues damage such as divot injury on creeping bentgrass is major issue to maintain golf course properly. Although plentiful researches to maximize divot resistance have been reported, minimal research has focused on relation between nitrogen (N) sources and divot resistance. The study was conducted to determine the effect of N source for turfgrass divot recovery and overall tee performance. Eleven fertilizer treatments as N sources were applied to creeping bentgrass 'Penncross'. Before the first application, divot injuries were simulated by removing a core of soil and turfgrass from established plots and backfilling with native soil. Data collection included turfgrass color and quality. N release speed did not influenced divot recovery. Frequency of urea application had no effects on divot recovery. Urea with split application had no difference with no treatment for divot recovery. Polyon product especially polyon mini (41-0-0) had the best performance for divot recovery and for maintaining better turfgrass quality. Overall, small particle size of slow-release N form would influence creeping bentgrasss to recover divot damage.
An antagonistic bacterial isolate, that inhibits the growth of plant pathogens, was selected and identified from 5,000 isolates screened from the rhizosphere of various crop plants. An isolate Bacillus sp. N32, tested against Colletotrichum gloeosporioides causing anthracnose disease in hot pepper, produced both a heat resistant antifungal protein and a heat sensitive antifungal protein. The heat resistant protein was partially purified by Ammonium sulfate fractionation and gel filtration chromatography. The bioautography showed that the proteins possessed high antifungal activity. The biosynthetic gene cluster responsible for the heat resistant antifungal protein was cloned from cosmid library using DNA probe obtained from PCR product with the primers targeting the conserved nucleotide sequence of the synthetic genes reported earlier, Most of the clones obtained showed higher homology to fengycin antibiotic synthetic gene family reported earlier. On the other hand, the heat sensitive protein was isolated from SDS-PAGE and electroblotting to determine the N-terminal amino acid sequences. The heat sensitive antifungal protein gene was cloned from the ${\lambda}-ZAP$ libraries using a DNA probe based on the N-terminal amino acid sequences of the heat sensitive protein. We are contemplating to clone and sequence the whole gene cluster encoding the heat sensitive protein for further analysis.
Bacillus thuringiensis CAB530 was isolated from dead Anomata albopilosa (Rutelidae: Coleoptera) and soil of green tea field, and confirmed its insecticidal activities. CAB530 isolate showed a high insecticidal activity against the beet armyworm among the many lepidopteran insects that are difficult to control. $LC_{50}$ value of CAB530 isolate against the second larva of Spodoptera exigua was $1.49{times}10^4$ spore concentration (cfu/$m{\ell}$). SDS-PAGE result of insecticidal toxin protein of CAB530 isolate showed a band at 130 kDa that is similar pattern with B. thuringiensis subsp. kurstaki that took insecticidal activity against S. exigua. Otherwise, the crystal protein of the CAB530 isolate was conformed at 65 kDa level after 30 minute of incubation in S. exigua midgut juice. Six crystal genes (cry1Aa, cry1Ab, cry1C, cry1D, cry1F and cry1I) were identified by PCR. It different from genes of B. thuringiensis subsp. kurstaki. Crystal shape and pattern of toxin protein was similar with B. thuringiensis subsp. kurstaki, however, insecticidal activity and PCR result of CAB530 isolate was similar with B. thuringiensis subsp. aizawai.
In this study Streptomyces strains were isolated from soils and their antifungal activities and involved mechanisms were investigated. Among over 400 isolates of actinomycetes, Streptomyces costaricanus HR391 was selected as a potential antagonist to control several plant-pathogenic fungi. S. costaricanus HR391 inhibited mycelial growth of Fusarium oxysporum f. sp. raphani, F. oxysporum f. sp. niveum, F. oxysporum f. sp. lycopersici, and Rhizoctonia solani by 26.5, 26.2, 21.2, and 23.8%, respectively compared to those of uninoculated control after 7-day incubation on PDB medium. S. costaricanus HR391 produced $89{\mu}M$ of siderphore, and showed fungal cell wall-degrading activity including $0.46{\mu}mol/min/mg$ of chitinase and $0.83{\mu}mol/min/mg$ of ${\beta}$-1,3 glucanase. S. costaricanus HR391 secreted 87.49 mg/L of rhamnolipid, and produced 9.49 mg/L and 4.3 mM of lipopeptide, iturin A and surfactin, respectively, all they are membrane-disrupting biosurfactants. It also produced antimicrobial peptide and antibiotics phenazine. In addition to antifungal substances, S. costaricanus HR391 secreted plant growth-promoting phytohormones, zeatin, gibberellins and IAA. These results suggest that S. costaricanus HR391 may be utilized as an environment-friendly biocontrol agent against some important pathogenic fungi.
