Inhibitory effect of Smilacis Glabrae Rhizoma on nitric oxide production in the macrophage cell line RAW 264.7 (토복령(土茯笭)이 RAW264.7 대식세포주에서 산화질소 억제에 미치는 영향)

  • Objectives : 본 연구의 목적은 제습, 해독, 통리관절등의 효능이 있는 토복령이 RAW264.7 대식세포주에서 lipopolysaccharide(LPS)로 처치하여 생성되는 nitric oxide(NO)와 prostaglandin $E_2$($PGE_2$)에 억제작용이 있는지 관찰하고자 하는 것이다. Methods : 토복령이 RAW264.7 대식세포주에 세포독성이 유무를 관찰하기 위하여 농도별 MTT assay를 수행하여 세포생존율을 측정하였다. 또한 LPS로 염증유발된 RAW264.7 대식세포주에서 토복령의 농도별 처치가 NO 및 $PGE_2$ 생성억제에 미치는 영향을 관찰하고자 NO 및 $PGE_2$ assay를 수행하였다. Results : 토복령의 농도별 처치가 RAW264.7 대식세포주에서 세포독성을 유발하는지 MTT assay로 측정한 결과 세포독성은 관찰되지 않았으며, 토복령은 LPS처치로 인하여 증가된 NO 및 $PGE_2$ 생성을 통계학적으로 유의하게 감소시킴을 관찰하였다. Conclusions : 본 연구를 통하여 토복령은 RAW264.7 대식세포주에서 LPS로 유도된NO 및 $PGE_2$ 생성을 효과적으로 억제시킴으로써 추후 염증질환의 치료제로서 가능성을 확인하였다.

Quality Characteristics of Pork Kalbi Sauce Added with Smilax china L. Extract during Storage (토복령 추출물 첨가로 인한 돼지갈비 소스의 품질 특성)

  • The present study was designed to study the antioxidant activity and physicochemical and microbiological characteristics of Pork Kalbi sauce added with Smilax china L. extract(0%, 0.25%, 0.5% and 0.75%) for 5 weeks. Total microbial counts were slightly higher in control and 0.25%-added group, therefore, more addition of Smilax china L. extract lowered total microbial counts. Alpha-diphenyl-β-picrylhydrazyl(DPPH) radical scavenging activity was significantly higher in 0.75%-added group compared to those in others(p<0.05). Also, thiobarbituric acid reactive substances(TBARS) was significantly lower in 0.5% and 0.75%-added groups compared to others(p<0.05). These results indicated that Smilax china L. extract addition increased the antioxidant activity of Pork Kalbi sauce and could improve the shelf-life during storage.

Antimicrobial activity against Propionibacterium acnes bacteria of Smilacis glabrae Rhizoma extracts (토복령 추출물의 여드름 균에 대한 항균활성)

  • This study was conducted to measure the antimicrobial activity and antiseptic effect of Smilax china root extracts against Propionibacterium acnes and to develop natural antimicrobial extracts as an alternative for synthetic preservatives, which have recently been controversial. Extracts were obtained from dried Korean Smilax china root at room temperature using two solvents, distilled water and 95% EeOH, separately. According to the results of this study, the antimicrobial effect of Propionibacterium acnes 3314 and Propionibacterium acnes 3320 against Propionibacterium acnes was outstanding. In particular, the clear zone of the extract using distilled water showed the highest activity with 16.61 mm. As for the result of the collection of contaminated skin sample, the clear zone was 24.48 mm. This indicates that Smilax china root extracts show a high activity against skin flora as well as a high antimicrobial activity against Propionibacterium acnes. In conclusion, it is confirmed that Smilax china root extracts can be used as raw materials for cosmetics that have antimicrobial activity and antiseptic effect, and it is expected that Smilax china root extracts will be used as basic materials for the development of future natural preservatives.

Antioxidant activity and protective effects on oxidative DNA damage of Smilax china root (토복령의 항산화 및 산화적 DNA 손상억제 활성)

  • Recently, cancer incidence in modern society is increasing sharply. DNA damage is caused by intrinsic or extrinsic factors in the human body, cells protect themselves by defense mechanism against DNA damage. Also, Aberrant DNA and deficient DNA repair are closely associated with various diseases, including aging and cancer. Researchers are interested in search for proper materials to inhibition for DNA damage. As knew the side effects of synthetic antioxidant, some researches have been conducted about cancer prevention materials derived from nature. Root of Smilax china, in Liliaceae, is used detoxification and tumor treatments traditionally. However, studies on the inhibitory effect of DNA damage haven't progressed. In this study, antioxidant activity and protective effects on oxidative DNA damage of S. china root were confirmed, relationship between those activities and contents of phenolic compounds in plants were established. S. china root effectively removed 1,1-diphenyl-2-picryl-hydrazyl radicals and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid radicals. The quantification and identification of phenolic compounds were confirmed by high performance liquid chromatography analysis, its antioxidant activity was associated with some phenolic compounds. In addition, protective effects against hydroxyl radicals and ferrous ion-induced oxidative DNA damage were confirmed in plasmid DNA. In the cellular levels, S. china root suppressed the expression of ${\gamma}$-H2AX and p53 protein in NIH 3T3. Besides, S. china root suppressed H2AX and p53 mRNA levels. In conclusion, S. china root had the effect on DNA protection and antioxidant.

The Anti-Obesity Effect of Smilax china Extract (토복령 추출물의 항비만 활성)

  • In this study, the anti-obesity activity of Smilax china methanol extract (SCME) was evaluated using a pancreatic lipase enzyme inhibition assay, and a cell culture model system. Results indicated that, SCME effectively inhibited pancreatic lipase enzyme activity in a dose-dependent manner. Furthermore, SCME significantly suppressed insulin, dexamethasone, 3-isobutyl-1-methylxanthine-induced adipocyte differentiation, lipid accumulation, and triglyceride contents on 3T3-L1 preadipocytes, in a dose-dependent manner. The anti-adipogenic effect was modulated by cytidine-cytidine-adenosine-adenosine-thymidine (CCAAT)/enhancer binding proteins (C/EBP) ${\alpha}$, $C/EBP{\beta}$, and the peroxisome proliferator-activated receptor ${\gamma}$ gene and protein expressions. Moreover, SCME triggered lipolysis effects dose-dependently on adipocyte. Taken together, these results provide an important new insight into SCME, indicating that it possesses anti-obesity activity through pancreatic lipase inhibition, anti-adipogenic and lipolysis effects. SCME may therefore be utilized as a promising source in the field of nutraceuticals. The identification of active compounds that confer the anti-obesity activities of SCME may be a logical next step.

The Effects of Traditional Drug Extracts on Acetaminophen-induced Hepatotoxicit (Acetaminophen의 간독성에 미치는 수종 생약추출물의 효과)

  • Acetaminophen(이하 AAP)은 간의 mixed function oxidase system에 의해 toxic metabolite로 대사되어 간괴사를 일으킨다. 최근에는 지금까지 임상적으로 사용되어온 한약이나 생약의 cytochrome P450의 활성과 간의 glutathion 합성에 미치는 영향에 대한 연구가 활발하게 진행되고 있다. 본 연구에서는 AAP의 간독성을 억제하는 물질을 규명하고자 민간이나 한방에서 보간 또는 해독제로 많이 사용되고 있는 생약을 screening하였다. 그 결과 간의 약물대사 효소계에 엉힝을 미치는 것으로 보고된 강활(Angelica koreana)과 민간이나 한방에서 간질환치료의 목적이나 해독제로 쓰여온 시호(Buplerum falcatum), 토복령(Smilax china) 및 금은화(Lonicera japonica)가 AAP로 유발된 간독성에 미치는 효과를 살펴보았다. 방법: 예비실험을 통해 강활, 시호, 토복령 및 금은화의 MeOH ext.가 AAP로 유도된 간독성에 방어효과가 있음을 확인하고 이를 hexane, ether, ethyl acetate 및 butanol로 계통분획하여 랫트에 전처치한 후 고농도의 AAP를 투여하여 간 손상을 일으키고 혈청중의 효소활성과 지질ㆍcholesterolㆍbilirubin함량 등을 측정하였다.

Anti-Inflammatory Effects and Cytoprotective Effects of Smilacis Chinae Radix (토복령의 항염증 및 세포보호 효과에 미치는 영향)

  • Smilacis Chinae Radix has been used as an anti-inflammatory agent. This study was performed to anti-inflammatory and MAP kinase signaling pathway in vitro. Experimental studies were obtained by measuring the Cytotoxicity, production of NO, PGE2, TNF-$\alpha$ and protein level of catalase, SOD, MAP kinase, The results were summarized as follows: Smilacis Chinae Radix was not cytotoxic effects against Raw264.7 and HEK293 cells. Concentration of $100{\mu}g/m{\ell}$ Smilacis Chinae Radix inhibited the production of NO in the Raw264.7 cell stimulated with LPS. All concentrations of Smilacis Chinae Radix not significantly inhibited the production of PGE2 in the Raw264.7 cell stimulated with LPS. All concentrations of Smilacis Chinae Radix did not inhibit the production of TNF-$\alpha$ in the Raw264.7 cell stimulated with LPS. Smilacis Chinae Radix has not effect of blocking NF-${\kappa}B$ into nucleus in LPS-induced macrophage Raw264.7 cell. Smilacis Chinae Radix has the effect of Cytoprotection through activation of ERK and inhibition of p38 and JNK. Accordingly the results show Smilacis Chinae Radix could induce anti-inflammation and Cytoprotection effects against In vitro, but it needs more research on the precise mechanism of such effects.

Skin Permeability Study of Flavonoids Derived from Smilax china: Utilizing the Franz Diffusion Cell Assay

  • Smilax china is known for its excellent antimicrobial, antioxidant, and anti-inflammatory properties. As a foundational study for applying the functionality of Smilax china extracts to cosmetics, it is necessory to investigate the concentration-dependent skin permation characteristics of the flavonoids in the extract, namely quercetin, catechin, and naringenin. Therefore, it serves as a crucial method for conducting this basic research on the functional aspects fo Smilax china extracts for cosmetic applications. This investigation focused on examining the percutaneous permeability characteristics of flavonoids originating from Smilax china. Applying Marzulli's definition, the Kp value of quercetin was categorized as "fast" at 0.1 mg/mL and "moderate" at 0.2 and 0.4 mg/mL. Notably, the permeation rate exhibited a decline with increasing concentration. For naringenin, Flux values were 0.69, 1.07, and 1.42 ㎍/hr/cm2 at concentrations of 0.1, 0.2, and 0.4 mg/mL, respectively, with corresponding Kp values of 6.95, 5.34, and 3.56. Naringenin's Kp value fell into the "moderate" category across all concentrations, and as observed with quercetin, the permeation rate decreased with higher concentrations. Likewise, for catechin, Flux values were 0.75, 1.09, and 1.66 ㎍/hr/cm2, and corresponding Kp values were 7.55, 5.46, and 4.16. Catechin's Kp value was consistently classified as "moderate" across all concentrations. The efficacy of quercetin, catechin, and naringenin, active ingredients in high-performance and anti-inflammatory Smilax china extracts, was found to exhibit skin penetration properties above the average. This confirms their suitability as excellent natural materials for use in functional cosmetics, given their outstanding capabilities in preventing acne and reducing inflammation.

Inhibition of Cell Growth by Anoikis in Various Human Cancer Cell Lines Treated with an Extract of Smilax china L. (토복령 추출물이 처리된 여러 종류의 사람 암세포주에서 아노이키스 세포 사멸에 의한 세포 성장의 억제)

  • The present study examined the cytotoxic effects of a Smilax china L. extract (SCLE) in human cancer (A-549, MCF-7, MDA-MB-231, U87-MG, AGS, MKN-74, and SNU-601) and normal MRC-5 fibroblasts, as well as in mesenchymal stem cells derived from dental tissue (DSC). The 50% inhibitory concentration (IC50) values for SCLE were significantly (p<0.05) lower in the cancer cell lines (A-549, MCF-7, MDA-MB-231, U87-MG, AGS, MKN-74 and SNU-601) than in the MRC-5 and DSC cells. Cell growth was significantly (p<0.05) more inhibited in the cancer cell lines treated with 200 ㎍/ml SCLE than in the normal MRC-5 and DSC, and anoikis-like floating cell morphology was observed in the SCLE-treated cancer cells. The cells detached by SCLE treatment were retrieved daily and assayed for viability and telomerase activity. Cells retrieved at 4 days showed significantly decreased viability and telomerase activity (p<0.05), as well as apoptosis-like abnormal morphology, when compared to cells retrieved in the previous 3 days. The ratio of apoptosis and cells in the G1 phase was significantly (p<0.05) increased in the A-549, AGS, and MCF-7 cancer cells treated with SCLE for 4 days compared to untreated controls. However, after SCLE treatment, cell adhesion was not increased by application of an inhibitor of the associated protein kinase (ROCK) that mainly contributes to the increase in cell attachment. This suggests that the cellular detachment by SCLE is probably controlled by a Rho-independent mechanism(s). These observations indicate that SCLE readily induces anoikis in cancer cells and could serve as a potent agent for cancer chemotherapy.

Evaluation of skin improvement efficacy of herbal medicine extracts on skin keratinocytes stimulated with fine dust PM10 (미세먼지 PM10으로 손상을 유도한 피부각질형성세포에서 한약재 추출물의 피부 개선 효능 평가)

  • Due to the increase in fine dust caused by environmental pollution, oxidative damage and aging of the skin are accelerated. In this study, the antioxidant, hyaluronic acid, filaggrin, MMP-1, and ROS level of selected herbal extracts were evaluated to confirm the protective efficacy of keratinocytes treated PM10. As a result, the antioxidant capacity of 1,1-diphenyl-2-picrylhydrazyl(DPPH), 2,2'-azinobis (3-ethylbenzothiazoline-6-sulfonic acid(ABTS), and FRAP assay increased in a concentration-dependent manner. Keratinocytes the group treated with 300 ㎍/ml of PM10, hyaluronic acid and filaggrin decreased by more than 50%, and increased in the group treated with extracts of Alpinia officinarum, Ulmus macrocarpa, and Ulmus macrocarpa but decreased when the extract was treated, which is evaluated as inhibiting the degradation of collagen and elastin. In addition, in the case of ROS measurement using zebrafish embryos, it was confirmed that the extract was reduced when the extract was treated 25 ㎍/ml, the intensity of fluorescence similar to the negative control was shown, confirming that the production of ROS was significantly reduced. Through this study, the selected oriental medicinal materials, Alpinia officinarum, Ulmus macrocarpa, and Ulmus macrocarpa, protect the skin from fine dust. It is thought that it can be used as an anti-aging product for skin improvement as a material that can be improved or improved.
