Immunolocalization of Wound-Inducible Insoluble Acid Invertases in Pea (Pisum sativum L) (완두콩(Pisum sativum L.) 상처에서 유도되는 불용성 산성 인버타제의 면역조직화)

  • Invertase, that hydrolyzes sucrose into glucose and fructose, plays a great role in carbohydrate reallocation between the photosynthetic source tissue and various sink tissues. Invertase also occurs in a variety of isoforms for various functions in plants. Insoluble invertases were extracted only in buffer solutions containing high concentrations of salt. Within these classes, acid invertase has an optimum activity at acidic pH (pH 4-5). Induction of insoluble acid invertase (INAC-INV) in leaf, stem, and root tissues in response to physical wounding has been investigated. To detect the localization of INAC-INV within the plant, immunolocalization has been performed. In this study, the accumulation of INAC-INV was noticeable to reach maximum levels on 72 hr after mechanical injuries. INAC-INV was induced in wounded leaves 3 times more than control leaves. Immunolocalization results showed that INAC-INV accumulated in wall appositions and intercellular spaces. INAC-INV was also localized at sieve cell walls in phloem tissues close to the site of wounding. Taken together, this study suggested that INAC-INV induction upon wounding injuries can play a role on responses to the high energy demand for wound healing process.

Effects of Hypoxia on the Fertilization and Early Development of Sea Urchin, Strongylocentrotus nudus (둥근성게 (Strongylocentrotus nudus)의 수정과 초기 배발생에 미치는 빈산소의 영향)

  • Dissolved oxygen is one of the most important factors controlling growth in aquatic organisms. Hypoxia is generally defined as dissolved oxygen less than 2.8 mg $O_2/L$ (equivalent to 2 mL $O_2/L$ or 91.4 mM). Therefore, hypoxia zone can cause a serious problem in marine ecosystem. In this study, to investigate embryotoxic (fertilization and embryo development rates) effects of hypoxia on sea urchin Strongylocentrotus nudus were exposed to dissolved oxygen levels of 7.6 mg $O_2/L$ (normoxia) and 1.8 mg $O_2/L$ (hypoxia) for 2 days at $15^{\circ}C$ and 33 ‰. Also, Expression levels of stress related gene (HSP70) and antioxidant related gene (glutathione reductase) in the sea urchins exposed to hypoxia were confirmed by Immunoblotting and RT-PCR analysis. In results, we showed that developmental rates were dramatically reduced in hypoxia condition. Molecular analysis demonstrated that higher HSP70 (5.5 fold) and glutathione reductase gene (2.79 fold) were present in the sea urchin exposed to hypoxia. Our results suggested that hypoxia can cause the abnormal development and elicits a stress and antioxidant response on sea urchin.

Evaluation of Sun Protection Factor (SPF) and Protection Factor of UVA (PFA) of the Sunscreen Containing Microalgal Extracts and MAAs (미세조류 추출물 및 MAAs 함유 자외선차단제의 자외선차단지수(SPF) 및 자외선 A 차단지수(PFA) 평가)

  • For the sunscreen development from natural resources, a possible usage of microalgal extracts or mycosporine-like amino acids (MAAs) was investigated. Sunscreens containing 7% of microalgal extracts or MAAs derived from microalgae, Chlamydomonas hedleyi, were prepared and they were applied to human research. Through this clinical research, the values of Sun Protection Factors, Sun Protector Factor (SPF) and Protection Factor of UVA (PFA), of sunscreen containing microalgal extracts or MAAs were determined: SPF values of microalgal extracts and MAAs indicated 9.07 and 9.42, respectively, while PFA ones did 2.43 and 2.41. Due to more than 2 of PFA value in both sunscreens, they can be classified into [PA+]. Taken together, although sunscreen containing microalgae-derived extracts or MAAs does not effectively protect UV irradiation, its capacity can be satisfied if inorganic UV-protecting compounds are added.

Rapid Detection of Cochlodinium polykrikoides by Sandwich Hybridization Integrated with Invertase Assay (Invertase 분석이 통합된 sandwich hybridization에 의한 Cochlodinium polykrikoides의 신속 검출)

  • Rapid and accurate identification of marine microalgae causing harmful algal blooms (HABs) is a crucial tool for predicting and managing HABs. We previously developed a nuclease protection assay sandwich hybridization (NPA-SH) method for the in situ detection of blooming microalgae Cochlodinium polykrikoides. In this study, we improved the applicability of the NPA-SH method for the detection of C. polykrikoides by simplifying the reaction step. For this purpose, invertase (INV) was conjugated to the signal probe instead of using fluorescence, and sucrose was used as a reactant to induce a color reaction. The INV-signal probe conjugation was confirmed by SDS-PAGE and epifluoromicroscopy. The treatment time and appropriate amounts of the probe and sucrose that optimized the reaction were determined. As a result, the developed INV-SH reduced the treatment time in the field compared with NPA-SH, and also enabled the use of a relatively small volume and low-priced personal glucose meter, as well as an absorbance meter. INV-SH is the first C. polykrikoides species identification technology to which INV has been applied and could be an improved field technique.

Correlations and Seasonal Variations of Marine Viral Abundances, Bacterial Abundances and Concentration of Chlorophyll-$\alpha$ in Gwangyang Bay (광양만내 해양 바이러스에 대한 엽록소$\alpha$와 박테리아 개체량의 계절적 변이와 상관관계)

  • The marine viral density in the Gwangyang Bay was abundant about 2.0${\times}$10$^{8}$ particles ml$^{-1}$ . For each season, viral abundances were recorded from 9.0${\times}$10$^{8}$ particles ml$^{-1}$ in summer to 0.7${\times}$10$^{6}$ particles ml$^{-1}$ in winter. The spatial distributions of the viral, bacterial and phytoplankton biomass in the Gwangyang Bay were mostly highey in closed estuarine system (Station 2, 5, 10, 12, 16, 20) than open ocean system (Station 28, 38, 42, 46, 51), And the othey closed estuarine system (Station 22, 26, 32, 34) indicated higher viral abundances, lower bacterial and plankton biomass than open oceanic system. In depths of some stations, the bacterial abundances exceeded a hundred fold than viral abundances. Seasonal abundances of marine viruses and their host systems were dynamically changed, and their seasonal variations were closely correlated. In summer, viral and bacterial abundances were increased, and phytoplankton chlorophyll $\alpha$ concentrations were maintained in average values. In winter, viral and bacterial abundances were dramatically decreased, and chlorophyll a concentrations were decreased, but, immediately increased. The viral abundances were peaked in August 2001, and bacteyial abundance, in August 2001 and June 2002, while chlorophyll a concentrations were peaked in April. 2002. In total host and viral abundances, it was seemed that their pools were maintained to steady-states by viral mortality, and viral abundance maintained steady-states. In our assessments, this report is a unique research about marine viral ecology of the Gwangyang Bay in Korea.

Spatial Variation in the Reproductive Effort of Mania Clam Ruditapes philippinarum during Spawning and Effects of the Protozoan Parasite Perkinsus olseni Infection on the Reproductive Effort (여름철 산란기에 있어 바지락 번식량의 공간적 변이와 기생 원생생물 Perkinsus olseni 감염이 바지락 번식에 미치는 영향)

  • Spatial variation in the reproductive effort of Manila clam Ruditapes philippinarum is often closely associated with variation in the seawater temperature and food availability, which determines gonad maturity and the quantity of gamates produced during spawning. Previous studies also have reported that severe infection by the protozoan parasite Perkinsus olseni exerts a negative impact on clam reproduction, retarding gonad maturation or decreasing the reproductive effort. In the present study, we investigated impacts of P. olseni infection on the reproductive condition of Manila clam during a spawning season. Histology revealed that 54% of female clams in Wando off the south coast were in spawning, while only 10% of the female from Gomso and 0% of the female from Seonjaedo in Gyeonggi bay off the west coast were engaged in spawning at the end of May in 2004. Ray's fluid thioglycollate media (RFTM) assay was applied to assess P. olseni infection and indicated that the infection intensity in Wando ($3,608,000{\pm}258,000cells/g$ wet tissue) was significantly higher than the levels in Gomso ($1,305,000{\pm}106,000cells/g$ wet tissue) and Seonjaedo ($1,083,000{\pm}137,000cells/g$ wet tissue, p < 0.001). The size of the ripe female follicle determined from histology was significantly smaller in Wando ($0.032mm^2$) compared to the sizes in Gomso ($0.059mm^2$) and Seonjaedo ($0.052mm^2$, p < 0.05). Accordingly, the number of ripe eggs in the follicle was significantly fewer among clams in Wando (14) compared to the numbers determined in Gomso (23) and Seonjaedo (22). The absolute quantity of egg in ripe clams from Wando (31.01 mg) was also significantly smaller than Seonjaedo (61.79 mg) and Gomso (133.3 mg). Quantity of total protein, carbohydrate, and lipid in the tissue in the Wando samples was significantly smaller than the quantities determined in Gomso and Seonjaedo (p < 0.001). The observed poor reproductive condition and proximate tissue composition of the females in Wando were, in part, explained by the extremely high level of the parasites, sapping the ability to store energy in the host tissues, which is used in tissue growth and the egg production.
