Effects of Taeumjowetang on Lipid Peroxidation by Free Radicals and Oxidative Damage of Hepatocytes by tert-Butyl Hydroperoxide (태음조위탕(太陰調胃湯)의 항산화(抗酸化) 효능(效能)에 의한 간세포(肝細胞) 보호(保護) 효과(效果)에 관한 연구(硏究))

  • Effects of Taeumjowetang on Lipid Peroxidation by Free Radicals and Oxidative Damage of Hepatocytes by tert-Butyl Hydroperoxide. 1. Purpose The present study was carried out to evaluate the antioxidant effects of Taeumjowetang in vitro. 2. Methods In this study, antioxidant effects of TJT on lipid peroxidation were determined according to the method of TBA. (Abbreviation) TJT : Taeumjowetang, TBA : 2-thiobarbituric acid. 3. Results : 1) TJT inhibited markedly peroxidation of linoleic acid during the autoxidation. 2) TJT inhibited lipid peroxidation induced by hydroxyl radical derived from H2O2-Fe2+ in rat liver homogenate. 3) TJT showed 66% scavenging effect on DPPH radical. 4) TJT exhibited a 25% inhibitory effect on superoxide generation from xanthine-xan thine oxidase system. 5) To investigate the antioxidative effects of TJT on the hepatocytes, cultured normal rat liver cells(Ac2F) were prepared and incubated with or without TJT. After 16~18hr, cells placed in DMEM medium without serum, and then incubated with 1mM t-BHP for 2hr. Viable cells were detected by MTT assay. In this test, TJT protected the cell death induced by t-BHP and significantly increased cell viability in the normal rat liver cell. Beneficial Effect of Taeumjowi-tang on the Cisplatin-Induced Gastrointestinal Dysfunctions in Rats (시스플라틴으로 유발된 랫트의 위장관 운동장애에 대한 태음조위탕의 효과)

  • Objectives This study aimed to observe the effect of Taeumjowi-tang on the cisplatin-induced gastrointestinal dysfunctions in rats. Methods Four groups, each of 8 rats per group, were used in this study. Saline and distilled water treated control rats were intact vehicle control group. Delayed gastrointestinal motility was induced by intraperitoneal treatment of cisplatin 2mg/kg, once a week for 5 weeks(Cisplatin control group). Taeumjowi-tang aqueous extracts(TJ) were orally administered in a volume of 5ml/kg, once a day for 14 days from 4th cisplatin treatment(TJ group). Ondansetron 1mg/kg was subcutaneously treated, in a volume of 1ml/kg, as same as TJ(ondansetron group). We measured the body weights, intestinal charcoal transit ratio, fecal parameters, fundus MDA(malondialdehyde), GSH(glutathione) contents and SOD(superoxide dismutase), CAT(catalase) activities, TPH(tryptophanhydroxylase) and MAO(monoamine oxidase) activities, pyloric gastrin and serotonin contents with their immunoreactive cells, colonic serotonin-immunoreactive cells, the histopathology of pylorus, fundus mucosa and colon. Results 1) The body weight gains, the small intestinal charcoal transfer rates, the fecal parameters(numbers, weights and water contents) were increased in TJ, ondansetron group. 2) The inhibit of fundus antioxidant defense systems by cisplatin were decreased in TJ, ondansetron group. 3) The pyloric TPH activities were increased and the pyloric MAO activities were decreased in TJ group. 4) The pyloric gastric contents and the gastrin-immunoreactive cells were increased and the pyloric serotonin contents and the pyloric and colonic serotonin-immunoreactive cells were decreased in TJ group. 5) The pylorus atrophic changes and the gastric surface erosive damage regions by cisplatin were favorably inhibited by treatment of TJ group. Conclusions The results obtained in this study suggest that TJ favorably retarded the cisplatin related GI(gastrointestinal) dysfunctions and constipation through modulations of GI enterochromaffin cells, serotonin and gastrin-producing cells and antioxidative systems.

In vitro inhibitory effects of Yangguksanhwa-tang and Taeumjowi-tang on CYP450 isozymes (양격산화탕과 태음조위탕 추출물의 in vitro CYP450 효소 활성 억제 효과)

  • Objective : Herb-drug interactions have become an important issue because of the consumption of herbal remedies has increased in the world. Yangguksanhaw-tang (Liang ge san huo-tang) and Taeumjowi-tang (Tai yin tiao wei-tang) are typical herbal formulas on Sasang constitution medicine (four-constitution medicine). This study was aimed at evaluating the effects of Yangguksanhaw-tang and Taeumjowi-tang on drug metabolizing enzymes, cytochrome P450 (CYP450) isozymes. Methods : Vivid$^{(R)}$ CYP450 Screening Kits were used to measure of CYP3A4, CYP2C19, CYP2D6 and CYP2E1 activities. This method is based on the use of fluorescent CYP450 substrates that are efficiently metabolized by specific CYP450 isozymes to yield a product with altered fluorescent properties. The percent inhibitions of CYP450s by herbal formulas were calculated. Results : Yangguksanhaw-tang inhibited CYP2C19 and CYP2E1 activities higher than that other CYP450 isozymes. The $IC_{50}$ values of CYP2C19 and CYP2E1 were 159.83 ${\mu}g/mL$ and 261.40 ${\mu}g/mL$, respectively. The CYP2E1 activity was inhibited ($IC_{50}=215.17{\mu}g/mL$) higher than that other CYP450 isozymes by Taeumjowi-tang. Conclusions : These results suggest that Yangguksanhaw-tang may inhibit the metabolism of co-administered drugs whose primary route of metabolism is via CYP2C19 or CYP2E1. Taeumjowi-tang could inhibit the metabolism of co-administered drugs, which are substrates for CYP2E1. Therefore, co-administration of the herbal formulas and other conventional drugs should be undertaken with care.

Analysis of Adverse Events in Weight Loss Program in Combination with 'Gamitaeeumjowee-Tang' and Low-Calorie Diet (가미 태음조위탕과 저열량 식이를 병행한 체중 감량 프로그램에서 발생한 이상 반응 분석)

  • Objectives: The objective of this study was to assess the safety of 'Gamitaeeumjowee-tang' by analyzing adverse events in weight loss program in combination with 'Gamitaeeumjowee-tang' and low-calorie diet. Methods: A retrospective review of adverse events in weight loss program in combination with 'Gamitaeeumjowee-tang' and low-calorie diet from the electronic medical chart (n=124) between June 2015 and December 2016 was conducted. Three Korean Medicine Doctors (KMDs) reviewed adverse events for two times, during week 2 to 4 and at week 10, after starting weight loss program. Adverse events were evaluated in terms of causality, severity and system-organ classes. Also, agreement among the three KMDs was made through further discussion in case of disagreement after independent review. Results: The overall rate of adverse events was 37.1% during week 2 to 4 and 16.9% at week 10. For causality of adverse events using the World Health Organization-Uppsala Monitoring Centre causality categories, 52.2% were evaluated 'possible' at week 2-4 and 57.1% were evaluated 'unlikely' at week 10. All symptoms were evaluated as 'mild' by LDS scale. Nausea (15, 12.1%) was the most frequent adverse event at week 2-4 and dizziness (6, 4.8%) was the most common at week 10. Conclusions: Adverse events decreased over time. There were no serious adverse events and none of the subjects were dropped due to adverse events. Continuous study is needed to prove the safety of 'Gamitaeeumjowee-tang' for treating obesity.

Effects of Dietary Therapy with Korean Herbal Medicine and Cultivated Wild Ginseng Pharmacopuncture on Change of Body Composition: A Retrospective Study (산삼복합약침과 한약을 병행한 식이요법이 체성분 변화에 미치는 영향: 후향적 연구)

  • The purpose of this study was to investigate the effects of dietary therapy with Korean herbal medicine and cultivated wild ginseng pharmacopuncture on change of body composition retrospectively. We analyzed the medical records of 26 patients, who carried out dietary therapy with Korean herbal medicine and cultivated wild ginseng pharmacopuncture at Moemfit Clinic, Charmjin Oriental Medicine for 8-12 weeks. And exercise and nutrition were recommended self-developed food, Moemfit Shake, Moemfit Hankki and walking exercise for one hour a day. The body composition values (body weight, body mass index, body fat mass, skeletal muscle mass, waist-hip ratio, visceral fat area, percent body fat) decreased after dietary therapy with Korean herbal medicine and cultivated wild ginseng pharmacopuncture significantly. But, percent skeletal muscle increased significantly. Results from this investigation showed that dietary therapy Korean with herbal medicine and cultivated wild ginseng pharmacopuncture has positive effects on changes of body compositions.

A Case report of ascites in Liver cirrhosis treated with Taeumjowetang-gamibang (태음조위탕가미방(太陰調胃湯加味方)으로 복수를 동반한 간경변 환자 치험 1례)

  • Liver cirrhosis is a disease of the liver in which normal cells are replaced by scar tissue. This condition results in the failure of the liver to perform many of its usual functions. Liver cirrhosis include acsites, jaundice, portal hypertension, varices etc. This report is about one case of ascites with liver cirrhosis. In this case, we administrated Taeumjowetang-gamibang was administered and acupuncture was done in treatment of ascites and its symptoms. After administration of Taeumjowetang-gamibang medication, clinical symptoms and prominently improved and ascites dwindled to nothing. This reports suggests Taeumjowetang-gamibang has desirable effected on ascites.

  • The purpose of this research was to investigate the correlation Among Sasang Constitutional Disease and Examination of the pulse. I have gone over literatures of mainly ${\ulcorner}$Dongyi Soose Bowon${\lrcorner}$ and the others Oriental Medical book was studied about the Pulse Diagnosis. And then I came to get some conclusion as follows. 1. Soeumin(소음인) the initial-stage symptoms of wulkwang disease(울광증) ; when the Superficial Pulse and the Superficial+ Moderate Pulse is made a diagnosis, Ceongunggyegitang(천궁계지탕) and Gunggyuhyangsosan(궁귀향소산) can be used. 2. Soeumin(소음인) the initial-stage blood disease symptoms of wulkwang disease(울광증) ; when the Minute+deep Pulse is made a diagnosis, Palmulgnnjatang(팔물군자탕) and Guakhyanggeonggisan(곽향정기산) can be used. 3. Soeumin(소음인) the initial-stage symptoms of mangyang disease(망양증) ; when the Yang region Superficial Pulse and the Yin region Weak Pulse is made a diagnosis, Hwanggigyegitang(황기계지탕), Bojungikgitang(보증익기탕) and Sengyangikgitang(승양익기탕) can be used. 4. Soeumin(소음인) the symptoms of taeum disease(태음증) ; when the Minute Pulse and Deep+Thin Pulse is made a diagnosis, Sasang Prescription can be used. 5. Taeumin(태음인) the Coldness syndrome in esophagus(위완한증) ; when the Superficial+Tight Pulse with weak power on left hand Pulse is made a diagnosis, Taeumjowetang(태음조위탕) can be used. 12. Taeumin(태음인) the Dryness-Heat syndrome(조열증) ; when the Flood+Large Pulse, Long Pulse and Long+Large Pulse is made a diagnosis, Galgeunhaegitang(갈근해기탕) can be used. The Effects of Taeyeumjowee-tang and Taeyeumjoweebaemahwang-tang on Obese rats (태음조위탕(太陰調胃湯)과 태음조위배마황탕(太陰調胃培麻黃湯)이 비만 흰쥐에 미치는 영향)

  • Objective : The aim of this study is to investigate the effects of Taeyeumjoweetang (TJT) and Taeyeumjoweebaemahwangtang (TJBM) medication on body weight change and serum total cholesterol, HDL cholesterol, LDL cholesterol, triglyceride, free fatty acid, total lipid, phospholipid level, and CBC of obese rats. Methods : Obese rats induced by high-fat fed are medicated for 7 weeks. 32 rats are divided into four groups depending on the medication; normal group (general-fat fed and no-medication), control group (high-fat fed and no medication), TJT group (high-fat fed and TJT medication) and TJBM group (high-fat fed and TJBM medication). After 7 weeks, obesity related index and CBC are analysed. Results : There is a significant statistical difference between control group and TJBM group for the body weight change, the total cholesterol, HDL cholesterol, and triglyceride level: total cholesterol values of control and TJBM are $59.25{\pm}9.56 mg/d{\ell}$ and $39.13{\pm}9.08 mg/d{\ell}$, respectively; HDL cholesterol values of control and TJBM are $22.0{\pm}1.69 mg/d{\ell}$ and $13.75{\pm}2.19 mg/d{\ell}$, respectively; triglyceride values of control and TJBM are $12.75{\pm}3.37 mg/d{\ell}$ and TJBM : $8.13{\pm}1.64 mg/d{\ell}$, respectively. There is a significant statistical difference between control group, TJT, and TJBM group for free fatty acid and phospholipid level: free fatty acid values of control, TJT, and TJBM are $936.25{\pm}89.59 {\mu}m/{\ell}$, $738.75{\pm}109.47{\mu}m/{\ell}$, and $720{pm}150.71{\mu}m/{\ell}$, respectively; phospholipid values of Control, TJT, and TJBM are $87.13{\pm}11.86mg/d{\ell}$, $70.5{\pm}12.22 mg/d{\ell}$, and $60.5{\pm}11.1 mg/d{\ell}$, respectively. Conclusion : These results suggest that medication of TJT and TJBM is effective for the treatment of obesity. Especially, TJBM is more effective than TJT.

A study on the ulcerous inhibitory effects of Taeumchowetang (태음조위탕(太陰調胃湯)의 유탕억제효능(遺瘍抑制效能)에 관(關)한 연구(硏究))

  • The experimental study used in this paper was done to examine the clinical effects of Taeumchowetang for the gastric ulcer and the function of gastrointestinal tract using rats and mice which were administered orally the water extraction from Taeumchowetang. Then, the counter-action of the water extraction on the isolated ileum and gastric fundus, the inhibitory effects of pylorus-ligated ulcer and indomethacin-induced ulcer, the associations with gastric juice secretion, total acidity, pepsin output, the transportability in the small and large intestine, were studied with administering acetylcholine chloride and barium chloride. In addition, it was investigated whether the central nervous system related to pain control and sleeping time was influenced by Taeumchowetang. The following results have been obtained; 1. As resulting from injection of acetylcholine chloride and barium chloride into the isolated ileum of rats and mice, Taeumchowetang led to have an inhibitory effect on the muscle contraction of the ileum. Then, acetylcholine chloride was measured as lower effect than barium chloride 2. For the inhibitory effect on contraction for the gastric fundus strip in rats by either acetylcholine chloride or barium chloride, the one showed low inhibitory effect, on the other hand the other showed density-dependent effect. 3. The water extraction was given on the pylorus ligated ulcer with using two different administration groups of 1,300mg/kg and 2,600mg/kg, each result was measured as 22.9% and 36.5% for an ulcerous inhibitory effect (p<0.05). 4. According to the two administration groups, the preventive effect was tabulated 18.1% and 29.3% for indomethacin-induced ulcer (p<0.05, P<0.01). 5. For associations with gastric juice secretion, total acidity, pepsin output in the administration group 2,600mg/kg, Taeumchowetang was recognized as having an inhibitory effects related to suppressive actions involving gastric juice secretion(p<0.05), and free acidity(p<0.01), but there was no significant association with total acidity and pepsin output. 6. To know the transportability in the intestine, BaSO4 solution was used. The transportability of the small intestine in the administration group 2,600mg/kg was 22.2% which was statistically significant compared with the large intestine's transportability(P<0.01). 7. In the administration group 1,300mg/kg and 2,600mg/kg, analgesic effect with against acetic acid was measured as being 16.8% and 24.4% which was shown as statistically siginificant(p<0.01). 8. No statistically significant association between Taeumchowetang and sleeping time was found in both 1,300mg/kg and 2,600mg/kg by administering phenobarbital-Na. According to the results of this study, Taeumchowetang has agreed with the effects of literature review. Further more in this study, Taeumchowetang also has preventive effects on pylorus-ligated ulcer. Hence, Teaumchowetang can be significant effect such as both anti ulcer agent and increasing gastric activity for the patients who suffer from gastric ulcer.

Anti-obesity and Anti-hyperlipidemic Effects of Taeyeumjowee-tang and its modified prescription (태음조위탕(太陰調胃湯)과 그 가감방(加減方)의 항비만 및 항고지혈증 효과에 관한 연구)

  • Objective : The purpose of this study was to investigate the anti-obesity and anti-hyperlipidemic Effects of Taeyeumjowee-tang and its modified prescription on the animal model of obesity and hyperlipidemia induced high-fat diet. Method : 1) The extracts of Taeyeumjowee-tang (TJT) and its modified prescription, Taeyeumjoweetang gagam-bang (TJGB) were evaluated for its inhibitory effects on obesity. 2) The body weight and feed weight were determined in the pre-treated and post-treated mice and the lipid profiles in the serum were analyzed in order to evaluate the anti-hyperlipidemia action of the extracts. 3) The effect of each extract was investigated for the influences on monoamine oxidase activity and HMG-CoA reductase activity. Results 1. TJT and TJGB extracts dose-dependently reduced the body weight and feed intake in normal mice. The effect of TJGB extract was better than that of TJT extract. 2. TJGB extract diminished the body weight increase and reduced the feed intake in the pre-treatment or post-treatment of the extract 3. TJGB extract decreased the amount of total cholesterol slightly and triglyceride potently after the pre-treatment or post-treatment, but HDL cholesterol exhibited no remarkable change compared with control. 4. TJGB extract weakly potentiated the monoamine oxidase activity, but its effect was better than that of TJT extract. 5. TJGB extract weakly inhibited the HMG-CoA reductase activity, but its effect was better than that of TJT extract. Conclusion : Taeyeumjowee-tang and its modified prescription can clinically be useful as anti-obesity drug and also for the improvement of hyperlipidemia.
