The Kinetics of Protease Production by Bacillus licheniformis (Bacillus licheniformis에 의한 단백질분해효소 생산 Kinetics)

  • Carbon sources and nitrogen sources are known to be very important in protease production by microorganisms. The effects of carbon source and nitrogen source on protease biosynthesis by Bacillus licheniformis were investigated using batch cultures. As initial carbon and nitrogen concentrations of culture medium increased, the specific growth rate of Bacillus licheniformis was increased, while the specific protease production rate was decreased. From the results of batch cultures, a mathematical model which considers the effects of carbon source and cnitrogen source was proposed and the methods to increase the productivity of protease were discussed.

Cultural Characteristics of a Biosurfactant-Producing Microorganism Pseudomonas aeruginosa F722 (Biosurfactant 생산균주 Pseudomonas aeruginosa F722의 배양특성)

  • Productivity of biosurfactant (rhamnolipid) by Pseudomonas aeuginosa F722 was investigated in the several culture conditions and culture composition. Biosurfactant production by P. aeuginosa F722 was amounted to 0.78 g/l as the result of the nitrogen sources and carbon sources without investing of optimum conditions. As for that one was investigated, biosurfactant production by P. aeruginosa F722 was amounted to 1.66 g/l. Biosurfactant production increased twofold because the composition of a modified C-medium was investigated efficiently. $NE_4$Cl or $NaNO_2$ inorganic nitrogens and yeast extract or trypton organic nitrogens were effective, but others inorganic nitrogens and organic nitrogens tested were not efficient far biosurfactant production by P. aeruginosa F722. The optimum concentration of $NH_4$Cl; inorganic nitrogen and yeast extract; organic nitrogen were 0.05% and 0.1%, respectively. In various carbon sources, others with the exception of hydrophobic property substrate (n-alkane) and hydrophilic property substrate (glucose, glycol) were not found to be effective fur biosurfactant production, and 3.0% was better in yield than other concentration of glucose. This yielded C-to-N ratios between 17 and 20. In our experiment, the highest biosurfactant production by P. aeruginosa F722 were observed in 5 days cultivation, containing glucose 3.0%, $NH_4$Cl 0.05%, and yeast extract 0.1% and C-to-N ratio was 20. Optimal pH and temperature for biosurfactant production were 7.0 and $35^{\circ}C$, respectively. Under the optimal culture conditions with glucose, biosurfactant production was amounted to 1.66 g/l. Velocity of biosurfactant production and strain growth increased after nitrogen depletion. The average surface tension of 30 mN/m after the 3 days of incubation under optimal culture condition was measured by ring tensionmeter.

  • Effects of nitrogen souses and C/N ratio were investigated on the production of extracellular microbial surfaclant, sophorolipid, from C. bombiocola. Organic nitrogen sources, such as urea, peptone and yeast extract was found to be more effective for sophorolipid production, than inorganic nitrogen sources. Depending on the nitrogen sources, sophorolipid production pattern varied by increasing C/N ratio. Increased production of sophrolipid could be obtained up to 90g/L by feeding carbon source again 2 days after cultivation.

Optimization of Medium Components for the Production of Crude Biosurfactant by Bacillus subtilis JK-1 (Bacillus subtilis JK-1의 생물계면활성도를 위한 최적 배지 조성)

  • Bacillus subtilis JK-1 showed degradation activity against crude oil, gasoline, kerosene, and light oil, and this strain was used as a crude biosurfactant producing microorganism in this study. To optimize the culture medium for production of crude biosurfactant, the influences of various carbon, nitrogen and mineral sources were assessed. The highest biosurfactant production by B. subtilis JK-1 was observed after 96 h cultivation, containing 1.0% (w/v) soluble starch as a carbon source and 0.5% (w/v) skim milk as a nitrogen source, and carbon to nitrogen concentraion (C/N) ratio was 2.0. For the biosurfactant production 0.1% (w/v) of $KNO_3$ was the most effective mineral source. Comparison of biosurfactant production indicates that B. subtilis JK-1 produces more biosurfactant in the optimum medium established in this study than LB and TSB. Under the optimum medium, the surface tension of culture broth of B. subtilis JK-1 was decreased from 47.3 dyne/cm to 24.0 dyne/cm after cultivation of 48 h.

  • Gibberellic acid ($CA_3$) is used in many industries and constitutes the primary gibberellins produced by fungi and bacteria. However, there is no information on $CA_3$ production by Methylobacterium oryzae CBMB20, a novel plant growth promoting bacterium. We investigated the favorable carbon (C) and nitrogen (N) sources and ratios and cultural conditions, such as incubation temperature, pH of the culture medium, and incubation period for the maximum production of $CA_3$ by Methylobacterium oryzae CBMB20. Maximum $CA_3$ production was observed in ammonium mineral salt (AMS) broth supplemented with Na-succinate and $NH_4Cl$ as C and N sources, respectively. The maximum $CA_3$ production was found at the C/N ratio of 5:0.4 g $L^{-1}$. The highest $CA_3$ production was obtained when the bacterial culture was incubated at $30^{\circ}C$ for 96 h at pH 7.

Optimum Conditions for the Removal of External Organic Carbon Sources in a Submerged Denitrification Biofilter (탈질 여과조에서 외부 탄소원 제거를 위한 적정 체류 시간과 외부 탄소원 종류 및 질산염 농도에 대한 외부 탄소원의 적정 비율)

  • Denitrification by anaerobic bacteria is one of the most common processes of removing nitrate from recirculating aquaculture systems. This process is affected by many factors such as external carbon sources, hydraulic retention time (HRT), and $COD/NO_3-N$ ratio. Although external organic carbon sources are essential for the denitrification process, these also contribute to increase dissolved organic carbon concentration in recirculating aquaculture systems. So these external organic carbons must be removed from the systems. This study was conducted to find out the optimum operating conditions for the removal of external organic carbons in a submerged denitrification biofilter. Combinations of two external carbon sources (glucose and methanol), two HRT (4- and 8-hour), and four different C:N ratios (3, 4, 5, and 6) were used in this experiment. The removal efficiencies of organic carbon sources at 8-hour HRT were always better than those at 4-hour's (P<0.05). Maximum removal efficiencies were achieved when C:N ratio was 5 in both glucose and methanol. The removal efficiencies of methanol were always better than those of glucose. The maximum removal efficiencies of glucose and methanol were 76.5% and 84.0%, respectively and the removal rates were 223.5 $g/m^2/day$ and 247.1$g/m^2/day$. The maximum removal rates of glucose (290.9 $g/m^2/day$) and methanol (355.6 $g/m^2/day$) were achieved at 4-hour HRT and 5 C:N ratio. But the concentrations of SCOD in the effluent of both glucose ($52.5 mg/\ell$) and methanol ($40.9 mg/\ell$) were too high for rearing fish. Therefore, the optimum operating conditions for the removal of external carbon in a submerged denitrification biofilter were 8-hour HRT and 5 C:N ratio. And methanol showed better efficiency as an external carbon sources.

  • The effects of liquid culture conditions and nutrient sources on the formation of protein-binding polysaccharide (PS) in Cordyceps militaris were examined. The formation amount of PS was increased in proportion to the growth rate of mycelium, in case of higher aeration or lower acidity. The optimum growth temperature of the mycelia was 25$^{\circ}C$ for the formation of PS. The optimum carbon source and nitrogen source were glucose and peptone, respectively. The ratio of C/N was optimal with 3% glucose to 0.5 % peptone. The sugar composition in the PS was greatly changed according to the carbon sources. The mycelium of Cordyceps militaris by liquid culture showed a higher electron donating ability than that by solid culture.

  • The effects of temperature. pH, carbon and nitrogen sources on the growth of Rhizoctonia solani were studied by using five hyphal anastomosis groups(four cultural types, 7 isolates) of the fungus. The ranges of optimum temperature were $20^{\circ}C$ in the AG 2-1, AG 2-2 and AG 4, and $25^{\circ}C$ in the AG 1-IA, AG 1-IB, AG 3, AG 5. The optimum pH for the mycelial growth was 6-7 in the fungus. Glucose in the AG 1-lA, AG 1-IB, AG 2-2, AG 3 and AG 5, sucrose in the AG 2-1 and fructose in the AG 4 were the most effective for the mycelial growth, but glycerine, cellulose and lactose were not effectively utilized as nutrients. $Ca(NO_3)_2$ in the AG 1-IA, AG 1-IB and AG 4, asparagine in the AG 2-1, $KNO_3$ in the AG 2-2 and $NaNO_3$ in the AG 5 were the best nitrogen sources for the mycelial growth, but $NH_4NO_3$ was not easily utilized by the fungus. Nitrate and organic nitrogens for the fungal growth were utilized better than ammonium nitrogen.

  • 도시화, 산업화로 인해 하수처리장 유입하수 내 질소 농도가 증가하면서 그에 따른 부영양화 발생, 수생태계에 독성을 미치는 등의 악영향 또한 증가하게 되었다. 하수 내 고농도 질소를 처리하기 위해 1990년 초 연구가 시작되어 현재 보편적으로 사용되고 있는 생물학적 질소 제거 공정은 산소공급과 외부탄소원 보충 과정에서 상당한 비용이 소요된다. 이와 같은 문제점이 대두됨에 따라 고도의 질소 제거 공정이 요구되면서, 경제적으로 개선이 이루어져 기존의 질산화·탈질 공정보다 효율적인 혐기성 암모늄 산화 공정(ANaerobic AMMonium OXidation, ANAMMOX)이 제안되었다. ANAMMOX 공정은 혐기성 조건 아래 전자공여체와 전자수용체로써 암모니아성 질소와 아질산성 질소를 이용해 질소가스 형태로 질소를 제거하는 공정이다. 질산화·탈질 공정과 비교했을 때, 폭기과정에서의 산소요구량 감소, 외부탄소원 불필요, 질소 제거 과정 단축 등의 장점을 가진다. 본 연구는 수처리공정에서의 ANAMMOX 공정의 적용 가능성을 확인하고, 암모니아성 질소대비 아질산성 질소 비율에 따른 Mainstream ANAMMOX 공정의 효율 비교를 통해 공정의 안정성과 높은 제거효율을 확보할 수 있는 NH4+ 대비 NO2- 비율을 도출하는데 목적이 있다. 실험실 규모의 Mainstream ANAMMOX 반응조에 적용한 비율은 선행연구를 비롯한 화학양론식에서 제시된 비율을 바탕으로 산정하였다. 1.00부터 1.30의 전체적인 비율을 Initial과 Advanced 2개의 구간으로 나누어 운전한 결과, 각 구간의 NH4+ 제거효율은 각각 58~86%, 94~99%였다. NH4+ 대비 NO2- 비율이 증가함에 따라 공정의 안정성이 확보되고, NH4+ 및 총질소(TN) 제거효율이 증가하는 경향이 나타났다. 본 연구의 결과는 수처리공정에서의 안정적인 ANAMMOX 공정 적용을 유도하고, ANAMMOX 공정의 성능개선을 도모하는 연구의 기초로 활용될 수 있다.

Effects of Carbon and Nitrogen Sources on the Essential Oil Production and Its Composition in Callus Culture of Mentha piperita L. (탄소와 질소원이 박하정유와 정유성분에 미치는 영향)

  • The effects of carbohydrate sources(sucrose, glucose) and ratio of ammonia to nitrate on essential oil production and its com- positions in the callus culture of Mentha piperita L. were studied. An ammonia : nitrate ratio of 1 :2 was more effective for essential oil production regardless of the media used ; Lin-Staba(LS) and modify Murashige-Skoog(MS) medium. Menthol biosynthesis was enhanced when ratio of ammonia to nitrate was 1 :3 in the LS medium while the ratio was 1 :2 in the MS medium. Lower sucrose concentration(20g /1 ) was much better than higher sucrose concentration(30g /1) for both oil and menthol biosynthesis in the LS medium but higher sucrose concentration(30g /1) was more effective for those in the MS medium. When sucrose was replaced with glucose, menthol biosynthesis was sharply decreased or absent regardless of media used.
