Evaluation of the Sealing Capacity of the Supercritical CO2 by the Measurement of Its Injection Pressure into the Tuff and the Mudstone in the Janggi Basin (초임계이산화탄소(scCO2) 주입압력 측정에 의한 장기분지 응회암과 이암의 scCO2 차폐능 평가)

  • The laboratory scale experiment was performed to evaluate the sealing capacity of the capping rock such as tuff and mudstone, measuring the intial supercritical $CO_2$ ($scCO_2$) injection pressure and the $scCO_2$-water-rock reaction for 90 days. The drilling cores sampled from 800 m in depth around the Janggi basin, Korea were used for the experiment. The mineralogical changes of mudstone and tuff were measured to evaluate the geochemical stability during the $scCO_2$-water-rock reaction at $CO_2$ storage condition (100 bar and $50^{\circ}C$). The rock core was fixed in the high pressurized stainless steel cell and was saturated with distilled water at 100 bar of pore water pressure. The effluent of the cell was connected to the large tank filled with 3 L of water and 2 L of $scCO_2$ at 100 bar, simulating the subsurface injection condition. The $scCO_2$ injection pressure, which was higher than 100 bar, was controlled at the influent port of the cell until the $scCO_2$ begin to penetrate into the rock and the initial injection pressure (> 100 bar) of $scCO_2$ into the rock was measured for each rock. The mineralogical compositions of mudstones after 90 days reaction were similar to those before the reaction, suggesting that the mudstone in the Janggi basin has remained relatively stable for the $scCO_2$ involved geochemical reaction. The initial $scCO_2$ injection pressure (${\Delta}P$) of a tuff in the Janggi basin was 15 bar and the continuous $scCO_2$ injection into the tuff core occurred at higher than 20 bar of injection pressure. For the mudstone in the Janggi basin, the initial $scCO_2$ injection pressure was higher than 150 bar (10 times higher than that of the tuff). From the results, the mudstone in Janggi basin was more suitable than the tuff to shield the $scCO_2$ leakage from the reservoir rock at subsurface.

  • This paper propose a method that controls facial expression of 3D avatar by having the user select a sequence of facial expressions in the space of facial expressions. And we setup its system. The space of expression is created from about 2400 frames consist of motion captured data of facial expressions. To represent the state of each expression, we use the distance matrix that represents the distances between pairs of feature points on the face. The set of distance matrices is used as the space of expressions. But this space is not such a space where one state can go to another state via the straight trajectory between them. We derive trajectories between two states from the captured set of expressions in an approximate manner. First, two states are regarded adjacent if the distance between their distance matrices is below a given threshold. Any two states are considered to have a trajectory between them If there is a sequence of adjacent states between them. It is assumed . that one states goes to another state via the shortest trajectory between them. The shortest trajectories are found by dynamic programming. The space of facial expressions, as the set of distance matrices, is multidimensional. Facial expression of 3D avatar Is controled in real time as the user navigates the space. To help this process, we visualized the space of expressions in 2D space by using the multidimensional scaling(MDS). To see how effective this system is, we had users control facial expressions of 3D avatar by using the system. As a result of that, users estimate that system is very useful to control facial expression of 3D avatar in real-time.

Characterization of Asian dust using steric mode of sedimentation field-flow fractionation (Sd/StFFF) (Steric 모드의 침강장-흐름 분획법을 이용한 황사의 특성분석)

  • Asian dust particles are known to have sizes ranging from a few nanometers up to about a few micrometers. The environmental and health effects depend on the size of the dust particles. The smaller, the farther they are transported, and the deeper they penetrate into the human respiratory system. Sedimentation field-flow fractionation (SdFFF) provides separation of nano to microparticles using a combination of centrifugal force and parabolic laminar flow in a channel. In this study, the steric mode of SdFFF (Sd/StFFF) was tested for size-based separation and characterization of Asian dust particles. Various SdFFF experimental parameters including flow rate, stop-flow time and field strength of the centrifugal field were optimized for the size analysis of Asian dust. The Sd/StFFF calibration curve showed a good linearity with $R^2$ value of 0.9983, and results showed an excellent capability of Sd/StFFF for a size-based separation of micron-sized particles.The optical microscopy (OM) was also used to study the size and the shape of the dust particles. The size distributions of the samples collected during a thick dust period were shifted towards larger sizes than those of the samples collected during thin dust periods. It was also observed that size distribution of the sample collected during dry period shifts further towards larger sizes than that of the samples collected during raining period, suggesting the sizes of the dust particle decrease during raining periods as the components adsorbed on the surface of the dust particles were removed by the rain water. Results show Sd/StFFFis a useful tool for size characterization of environmental particles such as the Asian dust.

Electroporation Conditions for DNA Transfer into Somatic Embryogenic Cells of Zoysia japonica (들잔디 체세포 배발생 세포로의 DNA 전입을 위한 Electroporation 조건 구명)

  • We have reported previously that intact embryogenic cells can be used instead of protoplasts for electroporation-mediated transformation of zoysiagrass and rice. In this study, conditions of the tissue electroporation were examined to optimize the procedures. Embryogenic cell suspensions were established in liquid MS medium containing 2 mg/L of 2,4-D with embryogenic calluses induced from mature embryos of Z. japonica. The suspension-cultured cell clumps were electroporated with 35S-gusA expression vector DNA, and degrees of DNA introduction into the cells were determined by histological expression rates of the gusA marker gene. DNA transfer into the cell clumps occurred in wide range of voltage (100-400 V) and capacitance (10-1980 $\mu\textrm{F}$), but more in the ranges of 200-300 V and 330-800 $\mu\textrm{F}$ DNA concentrations higher than 6 $\mu\textrm{g}$/mL were adequate for GUS expression of the electroporated cells. DNA transfers were confirmed in all three embryogenic cell lines but only in one out of eleven non-embryogenic lines. Positive GUS expressions occurred with DNAs added even 20-40 h after pulse treatments. As a promoter of gusA, Act1 and Ubi1 were effective 7 and 5 times than 35S respectively in number of GUS expression units on electroporated cell clumps. Embryogenic cell clumps survived and regenerated into plantlets after pulse treatments of wide range of conditions.

Interfacial and Mechanical Properties of Glass Fiber Reinforced Epoxy Composites with Different Crosslinking Density after Saline Water Aging (기지재의 가교밀도에 따른 유리섬유 복합재료의 염수노화 후 계면 및 기계적 물성)

  • Condition and properties of composites with different chemical structure of epoxy matrix were observed after saline solution treatment. Epoxy was used as matrix and the flexibility was controlled by using 2 typed-epoxies and 3 types hardeners (amine, acid anhydride and amide). Saline water treatment was conducted with 6 wt% NaCl solution at $60^{\circ}C$ for 0, 15, and 30 days. Cross section was observed and interfacial and mechanical and properties was evaluated. Amine type exhibited the highest crosslinking density and mechanical and interfacial properties whereas water absorbance was lowest. It is because that the water molecules can be hardly penetrate into the epoxy matrix or the interface between epoxy and glass fiber and it leads to saline water resistance of composites.

Retardation Effect on the Breach of the Earth Filled Embankment Using the Stiffener During Overtopping (흙댐 제체의 보강재 설치에 따른 월류붕괴 지연효과)

  • Most embankment of the reservoirs (99.1 %) have been constructed in the earth filled type in Korea because the construction of this type is less expensive and simpler than others such as concrete one. However, it has to be reinforced the slope to prevent the breach due to overtopping or piping under unexpected flood conditions. This study has been analyzed the retardation effect using three types (L, T, $L^*$ shape) of stiffener in order to reinforce embankment when they are collapsed by overtopping flow. Experimental results showed that L-type stiffener is the most effective in delaying the breaching of embankment and reducing the soil erosion when compared with others. The reinforced embankment breaching showed that time delay was occurred about 1.73 to 2.29 times and the peak flowrate was reduced compared to non-stiffener embankments due to energy dissipation by collision and less soil erosion. The embankment breaching mostly leads to major damages because of the lack of repair time. Thus, since these stiffeners can resist the rapid breach, it would be possible to earn the time to emergency repair and lifesaving, as well as reduction of damages of embankment in downward region with decreasing peak flowrate. Results from this study would be used for the basis when establishing the emergency action plan for the reservoirs on the verge of hazard.

Radioisotope Labelling Method of Pine Gall Midge (Thecodiplosis japonensis UCHIDA et INOUYE) (솔잎혹파리에 대한 방사선동위원소 표식방법에 관한 연구)

  • This experiment was carried out to develope a technique for labelling pine gall midge with ·radioactive phosphorus and calcium. This technique evolved would help to obtain basic information on the appropriate concentration of $^{32}P\; and\; ^{45}Ca$ in tagging and determination of adult flight ranges which considered as one of the factors in dissemination. 1. Labelling at the larval stage of pine gall midge by injection of $^{45}Ca$ into pine tree trunk appeared not to be effective for tagging the adults because radioactivity was gradually decreased to background by emergence. 2. The radioactivities of larvae in the galls gradually increased in September but from October 1 the activity was gradually decreased for 10 days then equilibrium was maintained thereafter. The results seem to indicate that larval diet termination occured early October. 3. A maximum level of radiation was detected at 3 weeks after $^{32}P$ injection and thereafter it was gradually reduced. This result implies that $^{32}P$ is not appropriate radioisotope in labelling pine gall midge through trunk injection method. 4. After washing in running water for 10 minutes, the optimum$^{32}P$ concentration for treated overwintered larvae and pupae was found to be 0.5 micro curie per ml. for 30 minutes dipping.

흑염소의 체내수정란 생산에 관한 연구

  • 면양과 염소가 최근 수십년동안 세계여러 나라에서 번식생리의 연구를 위한 모델로 사용되어 왔는데, 체내수정란의 생산에 관한 영역도 유럽을 중심으로 활발하게 연구되어왔다. 수정란생산을 위한 발정동기화방법, 과배란처리 및 수정란회수방법 기술은 현재 상당히 많은 기술진척이 이루어진 상태이나, 우리나라 고유의 재래유전자원인 흑염소에는 이를 위한 기술이 미진한 실정이므로 본 실험에서는 흑염소의 체내수정란생산기술을 확립하여 재래가축 유전자원보존을 위한 기초기술을 마련하고자 한다. 축산기술연구소 남원지소에서 사육하고 있는 체중 20kg 이상의 건강한 흑염소를 이용하여 발정동기화를 위해 controlled intravaginal drug release(CIDR)를 질내에 14일 동안 삽입하고, 과배란처리는 FSH를 CIDR 삽입 12, 13, 14일째에 12시간 간격으로 점감법으로 총20mg을 투여하였으며, PGF$_2$a를 13일째 FSH와 함께 투여하였다. CIDR는 14일째의 아침에 제거하였다. 수컷과의 교미는 CIDR제거 24시간후에 GnRH를 투여와 동시에 실시하였으며, 채란은 교미후 3일째에 외과적인 방법으로 실시하였다. CIDR처리경과에 따른 progesterone농도는 CIDR 주입시 바로 수치가 상승하여 제거전까지 6~12ng/m1의 농도를 유지하였으며, 제거즉시 2ng/ml 이하로 떨어졌다. 채란시 평균 배란점은 16.5개, 미배란난포 9.8개였으며, 회수수정란은 6.0개를 나타내어 채란율은 36.4%를 나타내었다. 회수된 수정란의 발달단계는 4-cell 78.9%, 2-cell 5.3%, fragmentation 15.8%를 나타내었다. 이와 같은 체내수정란생산방법을 기반으로 하여 이후 수정란의 동결 및 수정란이식기법에 관한 연구를 수행한다면 우리나라의 재래가축인 흑염소의 유전자원 장기보존과 생산성향상에 기여할 것으로 사료된다.배양액에 30 embryos/50ul 소적으로하여 38.8$^{\circ}C$, 5% $CO_2$의 탄산가스 배양기에서 각각 7일간 배양을 실시하였다. 조사된 결과는 SAS/STAT를 이용하여 통계분석을 실시하였다. 체외수정 12시간 후에 난자 급속 염색법으로 염색을 실시한 결과, 모든 처리구에서 핵성숙률(76.4~95.2%), 정자침투율(51.1~66.9%), 웅성전핵형성률(95.2~100%), 다정자침입률(18.2~25.6%) 및 평균침입정자수(1.2~l.4개)에서 유의적인 차이가 인정되지 않았다. 체외배양 48시간 난할률을 조사한 결과, 처리구별 차이(53.9~67.9%)는 인정되지 않았으나, 배양 7일째 배반포형성률은 각각 14.5, 25.4, 17.3 및 12.4%로서 25uM의 $\beta$-ME처리구가 유의적(P<0.05)으로 높은 배발달률을 나타내었고, 총세포수에 있어서는 대조구와 처리구간 유의적인 차이가 인정되지 않았다. 따라서 돼지 난포란을 성숙배양할 때, 25uM $\beta$-ME를 첨가배양하는 것이 양질의 돼지체외수정란을 생산하는 하나의 방법으로 조사되었다.다.natural objects and was popular at the time of Yukjo Dynasty, and there are some documents of that period left both in Japan and Korea.   • Structural battery has been researched extensively to combine the functions of the battery and structure without gravimetric or volumetric increments compared to their individual components. The main idea is to employ carbon fabric as the reinforcement and electrode, glass fabric as the separator, and solid-state electrolyte which can transfer load. However, state-of-the-art solid-state electrolytes do not have sufficient load carrying functionality and exhibiting appropriate ion conductivity simultaneously. Therefore, in this research, a system which has both battery and load carrying capabilities using glass fabric separator and liquid electrolyte was devised and tested to investigate the potential and feasibility of this structural battery system and observe electric properties. It was observed that elongating separator decreased electrical behavior stability. A possible cause of this phenomenon was the elongated glass fabric separator inadequately preventing the penetration of small particles of the cathode material into the anode. This problem was verified additionally by using a commercial separator. The characteristic of the glass fabric and the interface between the electrode and glass fabric needed to be further studied for the realization of such a load carrying structural battery system.

IN VITRO STUDY ON APICAL SEAL AND SEM MORPHOLOGY IN CANALS FILLED WITH RESIN-BASED ROOT CANAL SEALER AND SELF-ETCHING PRIMER (SELF-ETCHING PRIMER와 레진계 근관 SEALER 사용시 근관벽의 형태와 치근단 근관밀폐에 대한 실험적 연구)

  • Gutta-percha와 근관sealer를 사용한 근관 충전은 가장 많이 이용되고 성공률이 높은 방법이지만, 현재 사용되는 모든 근관sealer는 미세누출을 나타낸다. 따라서 더 효과적인 근관 폐쇄를 이루기 위한 재료 및 방법들이 연구되어왔다. 이중 상아질 접착제는 근관 충전재와 함께 사용되어 미세누출을 감소시키지만, 근관 내에 사용하기에 술식이 복잡하고 기술이 요구된다. 본 연구의 목적은 gutta-percha와 레진계 근관 sealer로 충전한 근관에서 self-etching primer를 미리 도포한 경우와 도포하지 않은 경우의 치근단 미세누출을 비교하고, self-etching primer를 도포함에 있어 근관 내에 적용하는 방법에 따른 미세누출을 비교하는 것이다. 또한 근관sealer와 상아질 계면을 주사전자현미경으로 관찰하여, 이 결과를 미세누출과 관련시켜보고자 하였다. 36개의 발거된 사람의 단근치에서 치관부를 절단, 제거하고 ProFile로 근관 형성한 후, 무작위로 선택하여 4개의 군으로 분류하였다. 1군에서는 주사기와 30게이지 주사 바늘로 self-etching primer를 근관 내에 적용하였고, 2군에서는 self-etching primer를 paper point에 적셔 근관 내에 적용하였다. 3군에서는 self-etching primer를 적용하지 않았다. 1, 2, 3군의 치아를 gutta-percha와 AH26 sealer를 사용하여 continuous wave 충전법으로 충전한 후 치근단공 주위 3mm를 제외한 치근변에 nail polish를 2회 도포하였고, 4군(음성 대조군)은 치근면 전체에 도포하였다. 1군과 2군에서 각각 2개의 치아는 주사전자현미경적 관찰을 위해 준비하였다. 모든 치아를 Methylene Blue 수용액에 48시간동안 침적, 수세한 후 치아 장축에 평행하게 양분하여 10배의 실물확대현미경 하에서 치근단 색소 침투를 관찰하였다. Self-etching primer를 도포한 군과 도포하지 않은 군 사이에는 평균 미세누출량에 유의한 차이가 없었다. 주사 바늘로 적용한 군과 paper point로 적용한 군 사이에도 평균 미세누출량에 유의한 차이가 없었다. 상아질과 근관 sealer계면의 주사전자현미경 관찰 결과 일부분에서 긴밀한 접착 관계를 나타내었고, 다른 부분에서는 간극을 나타내었다.
