Evaluation of the $HApS^{TM}$ Method for the Enumeration of Aerobic Microorganisms and Coliforms in Retailed Meat Samples in Korea

  • This study was designed to compare the effectiveness and applicability of the HApS$^{TM}$ (Hazard Analysis process System; HUKO, Seoul, Korea) based on Petrifilm$^{TM}$ (3M, St. Paul, MN, USA) with the AOAC (the Association of Official Analytical Chemists) standard total aerobic count (TAC) method and coliform count (CC) method for meat products. The comparisons were carried out using 230 meat samples collected from various retailers: 80 pork samples, 80 chicken samples, and 70 beef samples. In the comparison of the correlation coefficient (r) between conventional method and HApS$^{TM}$ method by a linear regression analysis, the correlation coefficients in total microorganism were 0.97767, 0.90712, and 0.95594 in pork, beef, and chicken samples, respectively. The correlation coefficients in coliform count were 0.82062, 0.94833, and 0.96839 in pork, beef and chicken samples, respectively. All the independent t-test on measurement values between conventional method and HApS$^{TM}$ method represented no significant differences in the means between two methods at the 0.05 of significance level($\alpha$=0.05). Based on the high correlation between HApS$^{TM}$ and the AOAC standard methods in the TAC and CC, it might be compatible to employ the HApS$^{TM}$ method to measure the microbial contamination in livestock products. HApS$^{TM}$ method was simpler and less time-consuming in sample preparation and procedures faster than the conventional method. These results suggested that the HApS$^{TM}$ method could be substitute for the conventional methods in the analysis of microbial contamination measurement in meat products.n meat products.

  • Endoglucanase A from Clostridium thermocellum which is resistant to pancreatic proteinase was selected out of numbers cellulases then were expressed in lactobacilli. Recombinant lactobacilli expression vector, pSD1, harboring the endoglucanase gene from C. thermocellum under the control of its own promoter, was constructed. Both L. bulgaricus and L. plantarum were electrotransformed with pSD1. The endoglucanase activities of 0.120 and 0.144 U/ml were found in culture media of L. bulgaricus and L. plantarum containing pSD1, respectively. In vitro survival characteristics of the transformed lactobacilli were tested. Both L. bulgaricus and L. plantarum showed a similar resistance to low pH 3. Moreover, L. plantarum was bile-salt resistant in the presence of 0.3 and 1% oxgall. L. bulgaricus and L. plantarum showed a rather homogenous resistant pattern against the tested antibiotics. Both of the strains were resistant to amikacin, gentamicin, streptomycin, kanamycin, and colistin.

A Molecular Sex Identification Using Duplex PCR Method for SRY and ZFX-ZFY Genes in Red Deer and Elk (붉은사슴과 엘크에서 SRY와 ZFX-ZFY 유전자의 Duplex PCR기법을 이용한 성 판별)

  • This study was focused on discriminating the molecular sexes of red deer and elk by duplex polymerase chain reaction(PCR) using two primer sets. Sex differentiation of mammals is primarily dependent on the presence or absence of sex determining region Y(SRY) gene encoded on Y chromosome which plays a key role for male development. Zinc finger X-Y(ZFX-ZFY) gene, one of X-Y homology gene group was found on X- and Y- chromosomes, respectively. At first, the nucleotide sequences were characterized for the intron 9 flanking region of ZFX-ZFY genes. The intron 9 of ZFX and ZFY is 529-bp and 665-bp in length, respectively. A transposable element sequence similar to bovine SINE element Bov-tA was detected only in ZFY gene of Cervidae. Sexing analysis was conducted by duplex PCR assay for amplification of SRY and ZFX-ZFY genes. Two differentially amplified patterns were found: one for females has a common band amplified only from ZFX as a template, and another for males had three bands(a common ZFX and two male-specific ZFY and SRY). On the separate tests using each gene, the results was identical to those from duplex PCR assay. Moreover, the results from PCR assays provide also identical information to phenotypic investigation of individuals of red deer, elk as well as their hybridized progenies collected from two isolated farms. These results suggest that it may be a rapid and precise method for determining the sexes by duplex PCR amplification using Y-chromosome specific SRY and X- and Y- homologous ZFX-ZFY genes showing sexual dimorphism in red deer and elk without any other controls.

The Effect of Feed Additives Supplement on Prerpartum and Postpartum Feed Intake, Milk Production and Metabolic Disorders of Dairy Cows (전환기 젖소의 사료첨가제 급여가 사료섭취량, 산유량 및 대사성장애 발생에 미치는 영향)

  • The objective of this study was to evaluate a feed additives used mainly in lactating cow diets in transient pregrent cow diets. The study was conducted as a completely randomized design with forty Holstein pregnant cows to determine the effect of feeding Aspergillus oryzae(T2), Saccharomyces cerevisiae mixture(T3) and enzyme(cellulase, xylanase) - releasing chemicals(ERC) (T4) on the dry matter intake, milk yield, milk composition and metabolic disorders. Dry matter intake was similar among treatments for 3 weeks prepartum but cows fed enzyme tended to increased feed intake compared to no additives treatment both in calving day and for 3 weeks postpartum. Cows fed Aspergillus oryzae, Saccharomyces cerevisiae and ERC produced more milk than those fed no additives. However, there is no significant difference among treatments. Concentration of glucose was not significantly different among treatment prepartum but that in plasma of cows fed ERC was higher at calving and 3 weeks postpartum compared to others. Increase in NEFA began at 3 weeks prepartum and accelerate during the final 7 days before calving at all treatments but lower for ERC-treated cows at calving and 3 weeks postpartum. Ca concentration not different among treatment prepartum and postpartum. Corticoid content decreased significantly for cows fed ERC compared to those fed non-additives. Metabolic disorder was not occurred in cows fed ERC. However, ketosis and displased abomasum were happened 1 cow when fed non-additives, metritis 1 cow when fed Aspergillus oryzae and retained placenta 1 cow in all treatments except cows fed ERC.

Crude Protein Requirements for Maintenance of Hanwoo Steers by Protein Balance Trial (단백질 균형시험에 의한 한우 거세우의 유지 조단백질 요구량)

  • This experiment was conducted to determine crude protein requirements for maintenance (CPm) of growing Hanwoo steers. Eighteen Hanwoo steers (173.7kg) were allocated randomly to diets with three levels of crude protein in the concentrate by replacing ground corn with corn gluten meal: 0% (LCP), 7.2% (MCP) and 14.2% (HCP), respectively. Hanwoo steers in each treatment were fed diets containing rice straw (20%) and concentrates (80%). CP intake was 5.5, 7.8 and 10.7g/BW0.75 for LCP, MCP and HCP, respectively. Protein balance was -0.05, 2.36 and 4.07g/BW0.75 for LCP, MCP and HCP, respectively. Intercept of the regression equation between CP intake and retained CP indicated that CPm was 5.56g/BW0.75. The estimate of CPm adopted by Korean Feeding Standard (2002) was almost 20% higher than that adopted by Japanese Feeding Standard for Beef Cattle (2000); this would need to be verified with more feeding trial data set including protein requirements for growth.

  • Present review is to introduce an omasal sampling technique in rumen proteolysis and to consider some information on the omasal sampling technique with particular emphasis on methodological aspects. Use of the omasal sampling technique provides a new opportunity for accurate estimation of rumen metabolism with overcoming limitations of previous in vivo, in vitro and/or in situ methods. The potential advantages of the present technique compared with post-ruminal sampling techniques include following points; 1) only rumen cannulated animals are required, 2) less endogenous nitrogen (N) is contaminated in omasal digesta and 3) omasal digesta are devoid of exposure to acid peptide hydrolysis occurring in the abomasum. Estimates of soluble non-ammonia N (SNAN) in omasal digesta indicate that the assumptions underlying the in situ method that rapidly degradable N fraction can be degraded at an infinite rate and only insoluble dietary N escapes the rumen may be not valid. Quatitatively higher peptide concentration rather than free amino acid and soluble protein in escapable SNAN suggests that hydrolysis of peptide to amino acid may be the rate-limiting step in rumen proteolysis.

An Analysis of Ex-post Evaluation on Korea-EU FTA with respect to the Agricultural Sector (한·EU FTA 농업부문 사후영향평가)

  • As the FTAs' implementations are accelerated, an Ex-post assessment, such as an analysis of the tariff schedule and agricultural trade has been emerging as an important national issue for an agricultural sector Korea-EU FTA, which was to be implemented in the five years from July 2016 as one of the giant FTA. The purpose of this study was to determine the demand of an ex-post assessment on agricultural sector as a trade negotiation procedural law. In addition, by providing policy direction for the agricultural policy part requiring amendments and supplements through ex-post assessment, the conflicting arguments between agricultural and non-agricultural sector can be evaluated more objectively. The current evaluation method on the economic impact ex-post assessment of a FTA is generally compared using the change in trade balance before and after the time of FTA implementation. On the other hand, this comparison cannot be said to be the pure FTA effects and objective, tightening economic impact assessment of the FTA in all combined situations, such as the effects of exchange rates and international macroeconomic changes and climate change & occurrence of pests. Over the last 4 years, however, Korea-EU FTA's total accumulated agricultural GDP loss was measured to be 2,178 billion by these research attempts with dynamic analysis as ex-post assessment methodology. The greatest impact was mainly livestock and pork followed by cereals and vegetables. In addition, this research is expected to contribute to policy evaluations in the future.

Effect of Heat Sterilization on Milk Nutrition by Hydrodynamic Cavitation - Vitamin A, B2, Calcium, Phosphorus, Magnesium, Zinc, Fat - (수력학적 공동현상을 이용한 가열 살균처리가 우유의 영양성에 미치는 영향 - 비타민 A, B2, 칼슘, 인, 마그네슘, 아연, 지방 -)

  • This study was conducted to investigate the amount of fat, vitamins and minerals in milk can be affected by hydrodynamic cavitation since cavitation is recently focused on as one of the sterilization methods. Vitamins (vitamin A, $B_2$), minerals (Ca, P, Mg, Zn) and fat contents were measured according to Ministry of Food and Drug Safety's Processing Standards and Component Specifications of Animal Product. Vitamin A & $B_2$ contents decreased approximately 23% & 19%, respectively after cavitation. Minerals content showed no change after cavitation. Also, P and Mg contents were reduced after cavitation to 2 mg/100g and 0.1 mg/100g, showing no change. Milk fat content was 3.46% before cavitation and 3.41% after cavitation, displaying no difference. Hydrodynamic cavitation process displays a possibility to replace existing pasteurizing method, as it does not change vitamin or mineral contents in milk. Therefore, a more clear and systematic research on hydrodynamic cavitation pasteurization is needed to distribute excellent quality milk and to emphasize physiochemical properties and quality change of existing heat pasteurization process.

Ecotoxicity Test Using E. agilis Biomonitoring System (Euglena 운동성 측정장치를 이용한 생태독성평가)

  • The toxic responses of flagellate Euglena agilis Carter to 8 heavy metals (Ag, Cd, Cr (VI), Cu, Hg, Ni, Pb, Zn) were measured using E. agilis system (E-Tox), an automated biomonitoring system. The E-Tox measures cell movement parameters, such as velocity, motility, and forms of the cells, as biological endpoints. $EC_{50}$ values from the E. agilis biomonitoring test were compared with the literature data from the tests with Daphnia magna, Vibrio fischeri and Euglena gracilis. Measurement of the E. agilis movement behavior and D. magna acute toxicity test were also conducted for the wastewater samples. E. agilis is less sensitive than D. magna but is comparable to or more sensitive than V. fischeri and E. gracilis for the heavy metals tested in this study. E. agilis shows prompt changes of these parameters for the toxic metal plating wastewater. Major advantages of the E-tox are automatic, easy to handle and fast ecotoxicity monitoring system compared to other biological test systems. These results imply that E. agilis biomonitoring test using E-Tox can be a putative ecotoxicity test as a good early warning tool for the monitoring of toxic wastewater.

Foreign Case Analysis and Implications for Risk Assessment of Industrial Insects (산업곤충 위해성 평가를 위한 국외사례 분석 및 시사점)

  • Industrial insect is defined as the insect utilized in industries that creates added value. Most of the industrial insects used in Korea are exotic species that are introduced through artificial means. Despite the rapid expansion of market for industrial insects, the system for risk assessment of industrial insects is not being adequately conducted. Although Korea carries out a risk assessment for the species designated as disease and insect pest by Animal and Plant Quarantine Agency, far too little consideration is being given to overall ecosystem, as the control system is covered in the Plant Quarantine Law. To solve this problem, we analyzed the Korean risk assessment system and looked at systems in other countries. The results show that it is essential for stakeholders to reach an agreement to set up fundamental directions for the system. Unless the integration system of taxonomical and ecological information is prepared, the ecological risk assessment should be conservative to protect ecosystems and should also follow the precautionary principle. It also requires cooperation among the ministries. In addition, the results indicated that a differentiation between risk assessment and screening is urgent. Several solutions such as setting up clear objectives in both assessment and screening stages, target species, steering organization and assessment criteria assessment systems from were proposed as practical institutional strategies. Among many foreign countries the assessment system from Ireland equally considers various factors such as economical, ecological safety and management aspects, It is also based on precautionary principle to fulfil its original purpose. It was suggested that the Ireland system would be the best reference that can be modified and applied into the Korean system by considering distinct characteristics of the industrial insects.
