Contents of Bioactive Constituents and Antioxidant Activities of Cultivated and Wild Raspberries (재배종 및 야생 산딸기의 기능성 성분 함량과 항산화 활성)

  • In order to select the raspberry cultivars that have high contents of bioactive constituents and high antioxidant activities, 7 cultivated and 2 wild raspberries which were selected and cultivated in the Bokbunja Institute were evaluated for their physicochemical characteristics, bioactive constituents, and antioxidant activities. The wild raspberry of Asan was the smallest among the sample raspberries but it had the highest sugar and lowest acid contents among the raspberries. Another wild raspberry of Ulleungdo had the highest total phenolic compound and ellagic acid contents, 182.97, $55.25mg{\cdot}100g^{-1}FW$, respectively, although it was small and had low sugar and high acid contents. Among the widely cultivated raspberry cultivars in Kimhae, 'Wangttal' cultivar was a big raspberry with 12.80% sugar content, and another unknown raspberry cultivar was as small as the wild raspberry with 14.60% sugar content. Although 'Wangttal' and the unknown raspberry cultivars cultivated in Kimhae possess lower contents of total phenolic compound (159.62, $165.94mg{\cdot}100g^{-1}$) and ellagic acid (45.7, $52.1mg{\cdot}100g^{-1}$ ) than the wild raspberry of Ulleungdo, the contents of total flavonoids (14.28, $14.90mg{\cdot}100g^{-1}$) and total anthocyanins (28.69, $30.48mg{\cdot}100g^{-1}$) were higher. Also the wild raspberry of Ulleungdo, 'Wangttal', and the unknown raspberry cultivar of Kimhae had higher antioxidant activities measured by FRAP (Ferric reducing antioxidant power), DPPH (2,2-diphenyl-1-picrylhydrazyl), and ABTS (2,2' azinobis-(3-ethylbenzothiazoline-6-sulphonic acid) assays. The present study shows that three raspberry cultivars could be potent resources for raspberry breeding and functional material development.

Quality Properties of Omija Beverage Based on the Storage Container and Storage Temperature (용기 및 저장온도가 오미자음료 품질에 미치는 영향)

  • This study compared the physicochemical quality characteristics of the Omija beverage for 36 weeks (9 months) depending on the storage container and storage temperature. From 3 weeks of storage, the optical density showed a significant difference according to the storage container, and the soluble solid contents showed a significant difference according to the storage container, storage temperature, and storage container${\times}$temperature. The total polyphenol and total acidity decreased slightly in the late storage period at all transparent and brown bottle treatments but the pH increased slightly. Generally, changes in the quality characteristics of Omija beverage at $4^{\circ}C$ storage were less than that at $20^{\circ}C$ and $36^{\circ}C$ storage. Correlation analysis of each factors, optical density, soluble solid content, total polyphenol, pH, and total acidity showed a positive correlation with each item according to the storage period. The storage period, particularly the transparent bottle treatment, appeared to have a significant impact on reducing the optical density. A few quality changes were observed after the brown bottle treatment for 36 weeks. In addition the stability according to the storage temperature was confirmed in each treatment. The brown bottle treatment is effective in maintaining the quality of Omija beverage for 36 weeks.

  • Basic optimal extraction condition and stability data were determined for prediction of usefulness of olive leaf as functional food material. Solid contents of olive leaf extracts increased with increasing extraction temperature and ethanol content, and was the highest (38%) under $85^{\circ}C$, 80% ethanol, and 5 hr treatment conditions, Total phenol contents and electron-donating abilities of olive leaf extracts also increased with Increasing ethanol content, and were the highest under $25^{\circ}C$, 80% ethanol, and 1 hr treatment conditions, then slightly decreased during storage at $25,\;55,\;and\;85^{\circ}C$. Olive leaf extract showed high stability under acidic storage condition, while low under alkalic condition.

Optimization of Ethanol Extraction Conditions from Glasswort (Salicornia herbacea) Using Response Surface Methodology (반응표면분석법을 이용한 퉁퉁마디 에탄올 추출조건의 최적화)

  • Response surface methodology (RSM) was used to monitor the characteristics of ethanol extracts from glasswort (Salicornia herbacea). A central composite design was used to investigate the effects of the independent variables of sample ratio, extraction temperature, and ethanol concentration on the dependent variables color, sugar, salinity, yield, electron donating ability, and total polyphenol content of extracts. The maximum $^{\circ}Brix$ (8.46) was obtained under specific extraction conditions, with a sample ratio of 7.04 g/100 mL, an extraction temperature of $89.01^{\circ}C$, and an ethanol concentration of 34.29% v/v. At a sample ratio, extraction temperature, and ethanol concentration of 7.00 g/100 mL, $89.15^{\circ}C$, and 34.14% v/v, respectively, the salinity was 7.35%. When the sample ratio, extraction temperature, and ethanol concentration were 5.56 g/100 mL, $68.61^{\circ}C$, and 99.14% v/v, respectively, the maximum electron donating ability was 86.10%. A maximized total polyphenol content of 1,140.15 mg/100 g was found with the following conditions: sample ratio of 8.6 g/100 mL, extraction temperature of $64.19^{\circ}C$, and ethanol concentration of 71.74% v/v. Overall, the optimal ranges of extraction conditions for effective components of glasswort were 3.38.5.33 g/100 mL sample ratio, $55.87-76.96^{\circ}C$, and 25.00.67.31% v/v ethanol.

Composition of Resveratrol and Other Bioactive Compounds, and Antioxidant Activities in Different Mulberry Cultivars (품종을 달리한 오디 추출물의 Resveratrol 및 기능성 성분과 항산화 활성)

  • To develop high value added healthy functional resource from Korean mulberry, the bioactive compounds and antioxidant activities of three different mulberry cultivars were investigated and compared with blueberry and strawberry. Total phenolics, total flavonoids, and trans-resveratrol of 'Daesungppong' were the highest than the 'Suwonppong', whereas those of the strawberries were the lowest. In case of total anthocyanins, 'Daesungppong' was also the highest and followed by blueberry. The antioxidant activities of 'Daesungppong' using DPPH (2,2-diphenyl-1-picrylhydrazyl), ABTS (2,2'-azinobis-(3-ethylbenzothiazoline-6-sulphonic acid), and FRAP (Ferric reducing antioxidant power) assays were the highest followed by 'Suwonppong'. Meanwhile, there were no significant differences in the antioxidant activities between 'Cheongilppong' and blueberry, and the activities of strawberry were the lowest. Furthermore, there was a strong correlation (r = 0.764-0.897) between DPPH, ABTS, and FRAP assays with the bioactive compounds such as total phenolics, total flavonoids, and total anthocyanins. Therefore, Korean mulberry, especially 'Daesungppong' demonstrated interesting biological properties that suggest its use as a potential source and high value added of natural antioxidant compounds and antioxidant activity.

Antioxidative Capacity and Total Phenolic Compounds of Methanol Extract from Zizyphus jujuba (대추 메탄올 추출물의 페놀성 화합물 함량과 항산화 효능)

  • The antioxidative capacity of methanol extracts prepared from jujubes (Zizyphus jujuba) were investigated by measuring total phenolic compounds, electron donating ability and induction period on lard. The extracts of large, medium and small jujubes of $80\%$ methanol extracts (methanol extracts) and soaking in the water before $80\%$ methanol extracts (water-methanol extracts) were Iyophilized. Total phenolic compounds of small, medium and large jujube of methanol extracts were 326.46 mg$\%,223.13\\;mg\%,\;158.06\;mg\%$ and that of water-methanol extracts were $113.23\;mg\%,\;81.45\;mg\%,\;72.78\;mg\%$, respectively. The Electron donating ability for the water-methanol extracts added samples of large (1.5$\~$3.0$\%$), medium and small jujube $(1.0\~3.0\%)$ were superior to the $0.02\%$ BHT added sample. On the induction period, all samples with jujube extracts were exhibited higher Antioxidant Index (AI: Induction period of lard with jujube extracts divided by induction period of lard) than that of the control and the water-methanol extracts were more effective than the methanol extracts on the antioxidative capacity for the lard. This result indicated that two lyophilized extracts of jujubes showed excellent free radical scavenging ability and long induction period on lard.

Antioxidant Activity of Chinese Mung Bean (중국산 녹두의 항산화 활성)

  • The aim of this study was to investigate the applicability of Chinese mung bean as a natural antioxidant agent. This study evaluated the phenolic compounds content and antioxidative activity of methanol extract from Chinese mung bean. Antioxidative activities were measured by in vitro models such as 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, and 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid(ABTS) radical scavenging activity. The contents of total phenolics and total flavonoids of Chinese mung bean extract were $174.83{\pm}2.90GAE\;mg/g$ and $68.87{\pm}2.84QE\;mg/g$, respectively. The antioxidative activities of Chinese mung bean extract were significantly increased in a dose dependent manner on DPPH radical scavenging and ABTS radical scavenging(p<0.05). The concentration of Chinese mung bean extract that reduces the free radical ABTS about 50%($IC_{50}$) was 2.85 mg/mL. These results suggest that Chinese mung bean may have great potential as a natural antioxidant source linked with health benefits.

Effects of Persimmon (Diospyros kaki Thunb) Syrup Extracted from Persimmon and Persimmon By-products on Blood Glucose Level (감(Diospyros kaki Thunb) 또는 감부산물에서 추출한 감시럽의 혈당수치에 미치는 영향)

  • The purpose of this study was to measure the blood glucose level and glycemic index (GI) in response to persimmon (Diospyros kaki Thunb) syrup extracted from persimmon and extract of persimmon by-products. Major component analyses of persimmon syrup I (PSI, 95:5 mixture of purified persimmon syrup and non-purified persimmon syrup) and persimmon syrup II (PSII, 50:50 mixture ratio of purified persimmon syrup and non-purified persimmon syrup) were $0.3{\pm}0.1$ and $0.6{\pm}0.2$ mg/g for total polyphenolic compounds and $70.6{\pm}0.6$ and $66.6{\pm}1.6%$ for total carbohydrates, respectively. Blood glucose responses of PSI and PSII were determined using both normal ICR mice and streptozotocin (STZ)-induced diabetic male Sprague-Dawley (SD) rats. Further, oral glucose tolerance test (OGTT) was performed on diabetic rats to assess the effects of the experimental diets. Blood glucose response and OGTT showed that blood glucose levels were significantly lower in mice and diabetic rats fed PSI and PSII compared to those fed diets of sugar, maple syrup, or honey. The GIs of healthy volunteers in response to PSI and PSII were calculated to be 51.9 and 35.7, respectively. On the contrary, the GIs of healthy volunteers fed diets including sugar, maple syrup, or honey were 52.6, 20.0, and 93.0, respectively. These results suggest that persimmon syrup can be used for both the treatment of diabetics and healthy people due to its beneficial effects on blood glucose level.

Nutrients and Antioxidative Activities of Metasequoia glyptostroboides (메타세콰이어나무 열매와 잎의 영양성분 및 항산화 활성)

  • The nutrients and antioxidative activities of fruits and leaves from Metasequoia glyptostroboides were investigated to provide basic data for the future development of functional foods. The nitrogen-free extract contents of fruit and leaves were 69.04% and 50.47%, respectively. Total phenolic content was higher in leaves (21.75 mg/GAE g) compared to the fruit (19.95 mg/GAE g). The mineral components of fruit and leaves mainly consisted of calcium (2,136.08 and 304.85 mg/100 g, respectively), potassium (1,355.53 and 1,144.04 mg/100 g, respectively), and phosphorous (426.30 and 350.50 mg/100 g, respectively). In terms of amino acid composition, aspartic acid, glutamic acid, proline, and leucine were relatively high, but methionine and cystine were low. The hot water extract from leaves was a more potent free radical-scavenger and had higher reducing activities than extracts from fruit. Thus, phenolics of M. glyptostroboides leaves can be utilized as an effective functional food substance for its natural antioxidant properties.

Antioxidant Compounds and Activities of Methanolic Extracts from Steam-Dried Allium hookeri Root (증포 삼채뿌리 메탄올추출물의 항산화 성분 및 항산화 활성)

  • This study was carried out to investigate the effects of steam-drying on antioxidant compounds and antioxidant activity of Alliun hookeri root (AHR). The yield of methanolic extracts, total phenolic content (TPC), browning intensity (280 nm and 420 nm), and organosulfur compound contents (alliin and cycloalliin) in raw and steam-dried AHRs were 10.71~37.40%, $15.53{\sim}36.36{\mu}g/mg$, 0.48~2.09, 0.01~0.25, $1.46{\sim}700.61{\mu}g/mg$, and $1,173.95{\sim}2,182.60{\mu}g/mg$, respectively. Yield, TPC, and browning intensity of AHRs increased by steam-drying, whereas organosulfur compound contents decreased. Of all methanolic extracts from raw and steam-dried AHRs, four-time steam-drying showed the lowest $EC_{50}$ values (0.43, 7.53, 0.34, and 0.48 mg/mL, respectively) for DPPH radical scavenging activity, ABTS radical scavenging activity, SOD-like activity, and reducing power, whereas four-time steam-drying resulted in the highest TPC ($36.36{\mu}g/mg$) and browning intensity (2.09 and 0.25 at 280 and 420 nm, respectively). The antioxidant activities of methanolic extracts from raw and steam-dried AHRs were closely correlated with their TPC, browning intensity, and organosulfur compound content, showing correlation determination coefficient ($R^2$) values higher than 0.82. As a result, four-time steam-drying of AHRs could be useful as potential antioxidant sources.
