A study investigated the effects of a mixture of Lactobacillus plantarum (L. plantarum) and Bacillus subtilis (B. subtilis) on diarrhea incidence, immune response, and fecal microflora counts in post-weaning piglets. One hundred 28-day-old piglets were randomly assigned to five treatment groups: negative control (NC), free of antibiotics; positive control (PC), 0.03% chlortetracycline; LB 1, a mixture of L. plantarum and B. subtilis (LB) 0.5 kg/ton feed; LB 2, LB 1.0 kg/ton feed; and LB 3, LB 2.0 kg/ton feed. Diarrhea scores for LB 2 and LB 3 from the 2nd week were significantly reduced compared to NC (P<0.05). In terms of the level of IgG and IFN-${\gamma}$, all treatment groups were significantly higher than NC (P<0.05), and the IgG level of LB 3 was significantly higher than that of PC (P<0.05). Furthermore, fecal lactic acid bacteria (LAB) counts for LB 2 and LB 3 were significantly higher than those of NC and PC (P<0.05). In addition, fecal Enterobacteriaceae (ENT) counts for PC, LB 2 and LB 3 were significantly lower than those of NC (P<0.05). Based on the results from this study, it was concluded that a combination of L. plantarum and B. subtilis strains could be used as potential alternatives to antibiotics to prevent diarrhea incidence in piglets.
The growth of V. parahaemolyticus was inhibited by the combination of natural products having antioxidative capacity. The growth of V. parahaemolyticus was controlled effectively by each or the combination of Paeonia japonica, Acanthodanacis cortex, Phellodendri cortex, Mori cortex radicis, Theae folium, Galla rhois, Coptidis rhizomn and Astragali radix. The growth inhibition ratio of the combination of Astragali radix, Phellodendri cortex, Coptidis rhizotna and Paeonia japonica was 95%, and its antioxidative capacity was also 5.4 times higher, and the elimination ratio of hydroxyl radical was more than 80% compared with control. As natural products might activate some parts of human body, they could be utilized as pharmaceuitcal applications, functional foods, antiaging tea, and multifunctional fermentative beverages.
This study was conducted to evaluate effects of mixed microbes addition on physico-chemical, fermentative and microbial parameters of sawdust-based spent mushroom substrates(SMS). The SMS was inoculated with mixed microbes(Enterobacter ludwigii, Bacillus cereus, 2 strains of Bacillus subtillis, Saccharomyces cerevisiae and Lactobacillus plantarum) at 1% level(wet basis) and anaerobically fermented during the different periods(up to 8th week). Compared with the SMS before ensiling, the ensiled one had higher CP, NDF and ADF percentages and lower DM and NFC percentages. However, levels of change were very low. The in situ ruminal disappearance of SMS DM and NDF decreased with the ensiling period prolonged. For fermentative parameters, pH reduced and lactic acid contents increased after ensiling, compared with those after ensiling. At 8th week of ensiling, pH increased and lactic acid contents reduced again, compared with those at 4th week of ensiling; however, the silage still showed favorable fermentation status. Lactic acid bacteria counts did not change throughout 8 weeks of ensiling. Counts of total microbes and yeast reduced after 4th week of ensiling period. Counts of lactic acid bacteria and yeast at 8th week of ensiling were in the levels of 108cfu/g. These results indicate that anaerobic fermentation with microbial addition could be an effective way for the long term(8 weeks) storage of the SMS.
For the investigation of conserved regions in lipase genes, 132 and 24 sequences were obtained from LED (Lipase Engineering Database) and COG (Clusters of Orthologous Groups of proteins), respectively. There was high diversity in lipase genes and peculiar amino acid sequences were conserved for each homologous family of LED. Similar conserved amino acid sequences were detected from COG0657 and Moraxella lipase 1 homologous group of LED. Although many studies have attempted to detect new lipase genes in procaryotes, they have been limited culturable bacteria. The importance of metagenome, including DNA from non-culturable bacteria, is known. Due to the high diversity, we assumed it might be possible to detect new lipase gene from metagenome. Due to the high diversity of nucleotide sequences in lipase genes, 10 primer sets were designed. Designed primer sets were inspected in BLAST of NCBI and they could amplify a part of the lipase gene from 222 to 713 bp. They can amplify 16.7%∼60.0% of each lipase homologous group which was 3.6 fold higher than each sets. They might offer a high probability of detecting new lipase genes, owing to high efficiency and the diversity of lipase genes.

This combination also eliminated more than 80% of hydroxyl radical. These natural products can be utilized as pharmaceuticals, functional foods, antiaging tea, and multifunctional fermentative beverages.
To develop a selective medium for the isolation and the detection of leuconostocs from the various samples including fermented vegetables, ten strains of leuconostocs and seven strains of lactobacilli were tested for their sensitivity to various antibiotics. The basal-medium containing 5 ${\mu}g/ml$ of novobiocin inhibited the growth of lactobacilli completely, but not that of leuconostocs. On the basis of this result, a new selective medium was developed and to be named NLS medium. This medium contains 1% Tryptone (Difco), 0.1% Yeast Extract (Difco), 2% sucrose, 0.1% Beef Extract (BBL), 0.5% sodium acetate, 0.2% ammonium sulfate, 0.01% magnesium sulfate, 0.2% dipotassium phosphate, 0.05% sorbic acid, 75 ppm sodium azide (Sigma), 0.1% (vol/vol) Tween 80, 30 ${\mu}g/ml$ of Vancomycin (Sigma), 5${\mu}g/ml$ of Novobiocin (Sigma), 0.5${\mu}g/ml$ of cysteine HCI, and 1.5% Agar (Difco). All of the eighty six isolates obtained from some foodstuffs were identified as members of the genus Leuconostoc. Comparative counts with the MRS, PES, LUSM, and NLS medium indicated that the recovery percent was lower than other selective media. Therefore, this result suggested that NLS medium was suitable for the isolation of leuconostocs, but not for counting or enumerating.
Sensitive and specific monoclonal antibody (MAb) was produced from hybridoma (1H11-5) obtained by fusion of myeloma cell (V653) and spleen cell isolated from mouse immunized sulfamthazine (SMZ)-HG-KLH. Direct competitive ELISA was developed for rapid detection of SMZ in milk samples using MAb against SMZ with optimized conditions between MAb and SMZ-HG-HRP conjugate, and applicable conditions for analysis of milk samples were established. Detection range of immunoassay was 0.1 to 100 ppb. Recoveries from spiked raw milk and processed milk samples averaged 82.1-120.7 and 82.1-97.1%, respectively.
In this study, a total of more than 1,000 bacteria, including 739 species of aerobic bacteria, 80 species of urease producing bacteria and 303 species of photosynthetic bacteria, were isolated from red-pepper field soils located in the Gyeongsan Province of the Republic of Korea. Amongst these, 158 species of aerobic bacteria, 70 species of urease producing bacteria and 228 species of photosynthetic bacteria were found to be auxin producing soil bacteria through quantification analysis with the Salkowski test. The latter groupings were then tested for antifungal activities to ${\beta}$-Glucanase and siderophore using CMC congo red agar and CAS blue agar media. In addition, the selected strains were examined for antifungal activity against various phytopathogenic fungi on PDN agar media. Six strains; BCB14, BCB17, C10, HA46, HA143, and HJ5, were noted for their ability to both produce auxin and act as antifungal substances. 16S rDNA sequence comparison analyses of these six strains identified them as Bacillus subtilis BCB14, B. methylotrophicus BCB17, B. methylotrophicus C10, B. sonorensis HA46, B. subtilis HA143, and B. safensis HJ5.
The aim of this study was to evaluate the effect of green tea extract on probiotic potential, physico-chemical and functional properties of yogurt fermented with Lactobacillus acidophilus D11 or Lactobacillus fermentum D37 strains isolated from Doenjang. Probiotic activities such as the resistance to artificial digestive juices and the ability to adhere to epithelial cells were slightly higher in yogurt supplemented with green tea extract than in plain yogurt, which may be attributed to the increase in the number of lactic acid bacteria (LAB) by green tea extract supplementation. Furthermore, the microbiological and physico-chemical properties such as the number of LAB, organic acid production and viscosity were significantly (P<0.05) increased in yogurt added green tea extract compared to plain yogurt fermented with L. acidophilus D11. However, the green tea extract did not significantly (P>0.05) affect these properties of yogurt fermented with L. fermentum D37 strain. Meanwhile, the antibacterial activities against Escherichia coli O157 ATCC 43889, Salmonella enteritidis ATCC 13076, and Salmonella typhimurium KCTC 2514 and antioxidant activities including total phenol content, radical scavenging ability, and ferric-reducing antioxidant power were significantly higher in plain yogurt fermented with L. fermentum D37 than with L. acidophilus D11. The antibacterial and antioxidant activities of the yogurt were significantly (P<0.05) increased in proportion to the concentration of green tea extract added to plain yogurt. Consequently, green tea yogurt fermented with L. acidophilus D11 or L. fermentum D37 was considered to be a useful functional food that can inhibit the growth of pathogenic bacteria and scavenge the free radicals from the body cells.
Nematode-trapping fungi develop trap and consume nematodes are an important part of the subsoil ecosystem and they share a special predator-prey relationship. Four nematode-trapping species, there with adhesive network, Arthrobotrys oligospora, A. sinensis, A. thaumasia and one with constricting ring, Drechslerella brochopaga were collected from soils in Korea and tested their predacity against 12 different nematode species. They were three feeding groups, plant-parasitic (Meloidogyne incognita and Pratylenchus penetrans), fungivorous (Aphelenchus avenae), bacteriovorous (Betlerius sp. and Diplogasteritus sp. in diplogasterid, Panagrolaimus labiatus, P. multidentatus in panagrolaimid, Mesorhabditis irregularis, Pelodera strongyloides and Rhabditis sp., in rhabditid, and Acrobeloides sp. in cephalobid). Results showed that nematode-trapping fungi successfully captured most of nematodes in Petri dish in the group of plant-parasitic nematodes and rhabditids, moderately and variably in other nematodes in 15 days. But it didn't captured A. avenae and Acrobeloides sp. both belongs to c-p group 2. Numbers of Acrobeloides sp. and A. avenae even increased during the test period. The results of this study indicated that nematode-trapping fungi may have specificity among nematode species.
