Synthesis of Resin Derivatives and Purification of Protein - Synthesis of p-Aminoanilinylsuccinyl-AH-Sepharose 4B and Purification of Protein in Pleurotus cornucopiae - (친화성 고분자 유도체의 합성 및 단백질의 분리정제에 관한 연구 - p-Aminoanilinylsuccinyl-AH-Sepharose 4B의 합성 및 흰느타리버섯 중 단백질의 정제 -)

  • For selective purification of proteins in Pleurotus cornucopiae, affinity chromatography was performed by p-aminoanilinylsuccinyl-AH-Sepharose 4B gel synthesized by treating p-phenylene diamine with succinyl-AH-Sepharose 4B, which was prepared by treating AH-Sepharose 4B with succinic anhydride. The capacity of p-aminoanilinyl ligand group was 6.1 micromole per milliliter of gel. Total apparent molecular weight of the affinity proteins eluted from the synthesized gel was 167 KD, which were a protein complex of 130 KD and 37 KD. The contents of the nonpolar, polar, positively and/or negatively charged amino acids in the affinity protein were 44.57%, 24.75%, 21.25%, and 9.43%, respectively. Total apparent molecular weight of the affinity proteins eluted from the AH-Sepharose 4B gel was 95.2 KD, which were a protein complex of 61 KD, 31 KD and 3.2 KD. The contents of the nonpolar, positively and/or negatively charged amino acids in the affinity protein by AH-Sepharose 4B gel were 44.05%, 29.13%, and 12.91% respectively.

The Effect of 4 Weeks of Treadmill Exercise and Protein Diet on Immunoglobulin and Antioxidant Enzyme in Rats (4주간의 트레드밀 운동과 단백질 식이가 흰쥐의 면역글로불린 및 항산화효소에 미치는 영향)

  • The purpose of this study was to investigate the effects of treadmill exercise and a protein diet on immunoglobulin and antioxidant enzymes in rats. Forty-four male Sprague-Dawley rats, 5 weeks old, were used. Experimental groups were divided into exercise with protein diet group (A, n=11), exercise group (B, n=11), protein diet group (C, n=11), and the control group (D, n=11). Exercise was administered through a treadmill running program (14~18 m/min, $0^{\circ}$ grade, 20 min/day, 5 day/wk) and these rats were given a 40% protein diet for 4 wk. The results of this study are as follows: the protein diet group showed a significant increase in IgG of immunoglobulin compared to the exercise group and control group; the exercise with protein diet group and protein diet group showed a significant increase in SOD activity of antioxidant enzymes compared to the control group; the exercise with protein diet group, exercise group and protein diet group showed a significant increase in GPx activity of antioxidant enzymes compared to the control group; the exercise with protein diet group showed a significant increase in CAT activity of antioxidant enzymes compared to the protein diet group and control group. In conclusion, treadmill exercise and a protein diet were found to help with immunoglobulin and antioxidant enzymes. Further research regarding the effects of exercise and protein diets is required.

Effect of Protein in Diet-induced Hypercholesterolemia Rats (단백질 종류와 급여수준이 흰쥐의 혈중 지질함량에 미치는 영향)

  • This study was conducted to investigate the effect of protein kinds and levels on lipid metabolism in hypercholesterolemia rats. Male Sprague Dawley rats were administrated 1% cholesterol and 0.25% sodium cholate to induce hypercholesterolemia and were fed on diet containing two levels(7, 20%) of casein and soy protein respectively. The rats were sacrified after 5 weeks of feeding periods. Total lipid content in serum was decreased in proportion to level of protein and soy protein was more effective than casein in lipid-lowering effect. Serum triglyceride content was significantly decreased in proportion to level of protein and did not show significance by protein kind. Serum phospholipid content was more significantly decreased in 20% protein groups than that of 7% protein groups. Total cholesterol content in serum was significantly decreased by protein and were decreased in proportion to level of soy protein. Free cholesterol content did not show significance by protein level. The degree of decrease predominated in soy protein groups. Serum HDL-cholesterol content and RL-cholesterol were increased in proportion to level of protein and effect of soy protein was not more significantly than that of casein. Atherogenic index was significantly decreased in proportion to level of protein and soy protein groups were more effective. Serum LDL-, VLDL-cholesterol contents were more significantly decreased in 20% protein groups than that of 7% protein groups. Serum $\beta$-lipoprotein and chylomicrone-cholesterol contents also showed significantly decrease by soy protein.

Alterations in Mitochondrial DNA Copy Numbers and Mitochondrial Oxidative Phosphorylation (OXPHOS) Protein Levels in Gastric Cancer Tissues and Cell Lines (위암 조직과 세포주에서 mDNA와 OXPHOS 단백질 분석)

  • Alterations in mitochondrial DNA (mtDNA) copy numbers have been reported in patients with stomach cancer and suggested to play a role in gastric carcinogenesis or gastric cancer progression. However, changes in the levels of mitochondrial proteins or mtDNA-encoded oxidative phosphorylation (OXPHOS) proteins in gastric cancer remain unclear. In this study, we investigated mtDNA contents, mitochondrial protein levels, and mtDNA-encoded OXPHOS protein levels in gastric cancer tissues and cell lines. We correlated mtDNA copy numbers with clinicopathologic features of the gastric cancer samples used in this study and used quantitative PCR to analyze the mtDNA copy numbers of the gastric cancer tissues and cell lines. Western blot analysis was used for assessing the amounts of mitochondrial proteins and mtDNA-encoded OXPHOS proteins. Among the 27 gastric cancer samples, 22 showed a reduction in mtDNA copy numbers. The mtDNA content was increased in the other five samples relative to that in normal matched gastric tissues. Mitochondrial protein and OXPHOS protein levels were reduced in some gastric cancer tissues. However, mitochondrial protein and OXPHOS protein levels in gastric cancer cell lines were not always in line with their mtDNA contents. The mtDNA copy numbers were reduced in five gastric cancer cell lines tested in this study. In summary, this study reports a common reduction in mtDNA contents in gastric carcinoma tissues and cell lines, pointing to the possible involvement of mtDNA content alterations in tumorigenesis of the stomach.

Effects of Heavy Metals on Mortality and Metabolite Changes in Mosquitoes, Culex pipiens pallens (Diptera: Culicidae) (중금속이 빨간집모기(Culex pipiens)의 치사율과 대사물질의 변화에 미치는 영향)

  • To investigate the effect of heavy metals (Cu, Cd, Hg, Pb, Zn) on mosquito Culex pipiens, 50% lethal concentration ($LC_{50}$) concentration, total lipid content, lipid content composition, and total protein content were measured. The results showed that the Hg $LC_{50}$ was $0.45mg\;kg^{-1}$ and the Hg toxicity was higher than the other metals. The results also showed that cadmium (Cd) significantly retarded the growth of mosquito larvae among the tested heavy metals. Six types of lipid bands were isolated from mosquito samples exposed to heavy metals, and five of them were identified (phospholipid, cholesterol, fatty acid, triglyceride, and cholesterol ester). The total lipid content of all treatments decreased compared to the control. In addition, the protein content of the control group ($0.51mg\;ind.^{-1}$) was higher than that of the treated group, and the protein content of the larvae treated with Hg and Cd was very low. In conclusion, heavy metals not only lower the survival rate of mosquitoes, but also affect the protein content and lipids in the mosquito's body, thereby causing growth inhibition. These results indicate that heavy metal contamination will affect the size of mosquito population by inhibiting the survival and growth of mosquitoes, and it is expected that it can be used to measure the degree of contamination using mosquito through subsequent studies.

Morphology and Protein Pattern During Microspore-derived Embryogenesis of Brassica napus (유채 소포자 유래 배의 발달과정에 따른 형태와 단백질 양상)

  • 유채(Brassica napus L. cv. Topas) 소포자 배양에 의한 기내 배발생 과정을 관찰하고 발생과정에 따른 총단백질의 변화 양상을 이차원 전기영동을 통하여 분석하였다. 배양에 적합한 소포자의 발달 단계는 4,6-diamidino-2-phenylindole (DAPI) 형광염색으로 핵상관찰을 통하여 결정하였다. 최초의 소포자 생존율은 63.9%였으며 이러한 생존율의 차이로 배양초기에 소포자의 이형성이 나타났다. 배 발달 경로에 따른 최초의 세포분열은 화분벽 안에서 균등분열로 시작되어 배병의 발달이 선행된 후 배 발생이 일어났으며, 구형, 심장형, 어뢰형으로의 배 발달은 여러 조직의 분화와 더불어 빠르게 진행되었다. 소포자배 발달과정을 치상단계의 소포자, 배양 3일째의 초기 분열세포, 구형 및 심장형배, 어뢰형배, 성숙한 자엽단계의 배 등의 5단계로 나누어 각각 2차원 전기영동을 수행한 결과, 최초 소포자 단계에서 나타났던 23개의 단백질들은 배 달달 경로로 진행됨에 따라 사라지고, 배양 3일째에는 8개의 단백질이 특이적 또는 지속적으로 발현되었다. 배의 발달과 더불어 20∼50 kD 사이에서 총 42개의 단백질이 급격히 나타나거나 또는 후기 배로 진행하면서 점차적으로 발현되었다.

  • 임상적으로 건강하고 휴식상태에 있는 한국 경주마의 혈청단백질치를 조사하였다. 총단백질농도는 6.5$\pm$0.3g/100ml(평균$\pm$표준편차)이였다. 알부민 농도는 3.4$\pm$0.2, $\alpha$-글로부린 농도는 0.8$\pm$0.1, $\beta$-글로부린 농도는 1.2$\pm$0.2, ${\gamma}$-글로부린 농도는 1.1$\pm$0.3g/100ml이었으며, A/G 비는 1.15$\pm$0.21이었다. 성, 연령, 계절에 따르는 혈청단백질치의 차이를 통계적으로 분석하여 보고하였다.

  • As a one of the new methods for soybean protein food processings, an attempt was made to decompose steamed soybean protein to amino acids, peptides, and other water soluble forms by the use of protease produced by Aspergillus sojae, and extract them to use as additives of low protein foods or weaning-foods of children. In this paper, as a one part of this subject, the optimum condition, such as optimum pH, temperature, digestion period, raw material's mixing ratio with wheat meal, amount of water added, and effect of sodium chloride addition, to decompose and extract soybean protein were studied. They were obtained when mixing raw materials with wheat meal in the ratio of 10:4 and adding six folds of water at $40-45^{\circ}C$, pH 5-8 for 18 hrs. digestion. Although sodium chloride addition exerted an unfavorable influence upon the decomposition of the aforementioned materials, it was effective to prolongation of digestion period. Under the abovementioned optimum conditions, the maximum extractability of soybean protein as forms of amino acids, peptides, and other water soluble forms, was nearly 70 percent against protein content of raw materials.

Proteolysis of Defatted Rice Bran Using Commercial Proteases and Characterization of Its Hydrolysates (탈지미강 단백질의 가수분해 및 분해물의 특성 연구)

  • The defatted rice bran (DRB) was enzymatically hydrolyzed using eight commercial proteases for 4hr at optimum pH and temperature. Proteolytic hydrolysates were examined in supernatant and precipitate using lowry, semimicro kjeldahl and gravimetric method using weight difference before and after enzymatic hydrolysis. In lowry and kjeldahl protein assay method, two proteases (Alcalase and Protease N) were found to be the most effective enzymes. In gravimetric method, 60.6~118.3 mg protein/g DRB was hydrolyzed after eight commercial proteases treatments. Similar to lowry and kjeldahl method, 118.3 and 107.1 mg protein/g DRB were hydrolyzed after Alcalase and Protease N treatments, respectively. When two or three effective proteases (Protamex, Alcalase and Protease N) were applied at one time to obtain synergistic effect, significant increase (P<0.05) was observed when three proteases were applied at one time (63.4 mg protein/g DRB in lowry method and 204.5 mg protein/g DRB in gravimetric method). This result suggests that Alcalase and Protease N were the most effective enzymes for proteolysis of DRB and three commercial enzymes (Protamex, Alcalase and Protease N) showed the synergistic effect on the hydrolysis of DRB.

Protein Composition of Domestic and Glyphosate-Tolerant Soybean (국내산 및 제초제 내성 콩의 단백질 조성 분석)

  • In order to elucidate the differences of protein profiles among soybean cultivars, the protein composition of three conventional domestic soybean cultivars and two imported ones including glyphosate-tolerant HS2906 was analyzed by total nitrogen measurement, amino acid analysis and PAGE/densitometry. There were no statistically significant differences in the levels of any amino acid, including aromatic amino acids, between glyphosale-tolerant soybean and the conventional soybean WS82. In the extraction of protein, the SDS/buffer system was more efficient than the defatting/water system. The SDS-PAGE/densitometry analysis showed that there was a similar profile of proteins among cultivars, although the amount of total protein ranged from 380.2 mg/g to 423.9 mg/g. In addition, there was no discernable difference of protein profile between glyphosate- tolerant soybean (total protein amount, 380.2 mg/g) and the conventional soybean WS82 (390.2 mg/g), although the amount of ${\beta}$-conglycinin (55 kDa) was lower in glyphosate-tolerant soybean. Meanwhile, the amount of 25 kDa protein was greater in domestic soybean cultivars than imported ones. Thus, normal PAGE/ densitometry method would be useful to analyze the difference in protein profiles of soybean proteins, and furthermore Evaluate the protein profile of proteins between GMO and conventional soybean.
