In vitro Long Term Conservation of Potato Germplasms (감자 유전자원의 기내 장기보존 방법)

  • This study was carried out to establish the in vitro culture system of potato germplasms for minimizing the occurrence of variation and maximizing the culture period. We used osmoticum such as sorbitol or mannitol with sucrose in the absence of plant growth regulators. The growth of potato germplasms in the medium containing osmoticum was increased when the growth temperature was lowered. After six months storage in low temperature, plant heights of tetraploid was somewhat higher than those of diploid with the exception of stn-16 and the difference due to media was not observed. But after twelve months storage, survival rates of plants cultured in LSM 1(sucrose and sorbitol) was higher than those of plants cultured in LSM 2(sucrose and mannitol). The survival rate of stn-16, diploid wild species, was approximately 75% and it was considerably high. In Atlantic, tetraploid cultivated variety, every individual was survived.

Effect of Gibberellin Seed-Spray on Seedling Emergence and Growth in Dry-Seeded Rice (벼 건답직파재배에서 Gibbrellin의 종자분무처리가 출아 및 초장신장에 미치는 영향)

  • Seedling emergence and .stand establishment in dry-seeded rice is unstable at present due to deep seeding, low temperature at seeding time, excessive drought or raining etc. This experiment was conducted to elucidate the effect of gibberellic acid(GA$_3$) on the seedling emergence and growth in dry-seeded rice. GA$_3$, IBA and Metalaxyl were treated by seed-soaking or-spray. Hwaseongbyeo, a japonica rice, was used. The seed-soaking treatment with GA$_3$ significantly increased the seedling emergence and plant height as compared with untreated seed. However, no difference was observed in IBA and Metalaxyl treatment. The optimum concentration of GA$_3$ to promote the seedling emergence and growth in dry-seeded rice was about 100ppm for seed-soaking and about 200ppm for seed-spray treatment. The seed-spray treatment of GA$_3$ with 200ppm promoted earlier and faster seedling emergence and elongated seedling height as compared with untreated seed.

Effect of Application Method and Concentration of Plant Growth Retardants On Plant Quality of Potted Saxifraga rosacea Moench

  • Four different plant growth retardants (PGRs), paclobutrazol, flurprimidol, daminozide, and chlormequat, were applied to potted Saxifraga rosacea 'Kumoma' and 'Kumoma-Gusa' plants for control of the growth and flowering. Paclobutrazol (10, 20, 40, $80mg{\cdot}L^{-1}$), flurprimidol (5, 10, 20, $40mg{\cdot}L^{-1}$), daminozide (500, 1000, 2000, $4000mg{\cdot}L^{-1}$), and chlormequat (50, 100, 200, $400mg{\cdot}L^{-1}$) were applied to the plants by a foliar spray or drenching. In 'Kumoma', application of $40mg{\cdot}L^{-1}$ paclobutrazol by a foliar spray or drenching reduced plant height by 12.5 and 12.6 cm, and flower length by 3.4 and 3.3 cm, respectively. On the other hand, in 'Kumoma-Gusa', drenching of paclobutrazol reduced plant height by 10.7 to 12.6 cm and flower length by 2.0 to 3.9 cm with increasing concentration, but the number of florets almost fell to 20 as compared to 40.5 in the control. 'Kumoma-Gusa' plants drenched with $80mg{\cdot}L^{-1}$ paclobutrazol and sprayed with $40mg{\cdot}L^{-1}$ flurprimidol had the shortest heights of 10.7 and 9.9 cm, and floral length of 2.0 and 1.5 cm, respectively. A flurprimidol drenching at $40mg{\cdot}L^{-1}$ delayed the harvest by 3-13 days as compared to the control and the smallest number of florets, 15.6, was observed in this treatment. In both cultivars, chlormequat and daminozide did not effectively influence the growth and flowering. However, number of florets increased to more than 41 at all concentrations and up to 63, the greatest floret number, with chlormequat drench in 'Kumoma-Gusa'. These results demonstrated that over $40mg{\cdot}L^{-1}$ of paclobutrazol or 5 to $20mg{\cdot}L^{-1}$ of flurprimidol could be used as PGRs to control the growth of floral length and flowering for improving potted plant quality in S. rosacea 'Kumoma' and 'Kumoma-Gusa'.

Effect of Cultural System and Sonic Strength of Nutrient Solution on the Growth of Dendrobium (Dendrobium phalaenopsis ) Seedlings (양액재배 시스템 및 양액농도가 덴파레(Dendrobium phalaenopsis) 유묘의 생장에 미치는 영향)

  • This study was conducted to evaluate the optimum hydroponic system and nutrient solution for shortening the early growth period and quality improvement of dendrobium ( Dendrobium Phalaenopsis) seedlings. Dendrobium seedlings with 3 to 4 leaves were transplanted in the deep flow technique(DFT) system, aeroponic system, and ebb and flow system with different concentration of balanced nutrient solutions recommended by the Japanese Horticultural Experiment Station. Growth characteristics of shoot and root were recorded and evaulated among treatments. For autumn cultivation, plant height was the longest at the DFT system with quarter concentration of nutrient solution, where aeroponic system with half concentration of nutrient solution. Aeroponic system stimulated the root growth but fresh weight was observed in the plots of DFT system. For spring cultivation, pH values increased up 7.5 at the DFT and aeroponic system, where EC values did not fluctuate regardless of cultural system. Ebb and flow system showed the best result in the growth of plant followed by BFT system and aeroponic system. Higher concentration of nutrient solution within this range of treatment was recommended for the growth promotion of leaf length and width in DFT system. In conclusion, growth responses differed depending on the cultural system, concentrations of nutrient solutions and duration of cltivation.

Effects of Timings and Light Intensities of Supplemental Red Light on the Growth Characteristics of Cucumber and Tomato Plug Seedlings (적색광 처리시기 및 광도가 오이 및 토마토 플러그묘의 생장에 미치는 영향)

  • This study was established to control the overgrowth of cucumber and tomato plug seedlings by controlling of irradiation timings light intensities of red (R) light. Compared with the control, end-of-day (EOD) timing of R irradiation significantly reduced the seeding heights by 21.3% and 14.2% in cucumber and tomato, respectively. In addition, both plant seedlings treated with R light at EOD timing had the thickest stem diameter. Tomato seedlings treated with R light at EOD showed th highest chlorophyll content and the smallest leaf size. Dry weights of above ground plant tissue in both plant seedlings were significantly reduced by EOD R light treatment. And both plant seedlings treated with EOD treatment had the lowest T/R ratio, and the highest compactness rates. The increased intensities of R light resulted in the shorter plant heights in cucumber and tomato plant seedlings. It also reduced the length of hypocotyls and internodes and the size of leaves in the both seedlings. Elevated R light intensities tended to increase the stem diameter and chlorophyll contents. Dry weights of above ground plant tissue and roots in both plant seedlings were reduced by the elevated R irradiation. Cucumber seedlings treated with 2 and 8 $mol{\cdot}m^{-2}{\cdot}s^{-1}$ and tomato seedlings treated with 8 ${\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ had significantly lower T/R ratio the other treatments tested.

Effects of Oligosaccharide and Pseudomonas sp. on the Growth of Potted Kalanchoe During Summer Season (천연올리고당 및 Pseudomonas속 길항미생물의 단독 및 혼용처리가 고온기 칼랑코에 생육촉진에 미치는 영향)

  • Most severe problem in production of potted kalanchoe during summer season is retardation of growth caused by high temperature. The aim of this experiment was aimed to investigate the effects of natural products such as algin-oligosacchride and glucosamine oligosaccharide, plant growth promoting rhizovacteria such as Pseudomonas sp. B and Pseudomonas sp. D2, and AG-solution on the growth of potted kalanchoe under the different root zone temperature in the greenhouse. Growth characteristics in terms of plant height, leaf length, leaf width, leaf area, leaf weight, fresh weight of shoot and root and root length were recorded under three root zone temperatures (25$^{\circ}C$, 30$^{\circ}C$, 35$^{\circ}C$). In 25$^{\circ}C$, the mixed treatment of Pseudomonas sp. B and glucosamine oligosaccharide resulted in the best growth in terms of plant height, leaf area and root weight. In 3-$^{\circ}C$, glucosamine oligosaccharide treatment gave fair result in plant height and leaf weight, but the mixed treatemtn of Pseudomonas sp. D2 and algin-oligosaccharide showed better growth on leaf area and root weight. In 35$^{\circ}C$, the mixed treatment of Pseudomonas sp. B and glucosamine oligosaccharide could greatly improve the plant height, leaf area, leaf weight and root weight. These results demonstrated that the mixed treatment of natural products and microorganisms could overcome the detrimental effects caused by high temeprature in the production of kalanchoe.

Growth Responses of Various Ornamental Foliage Plants According to the Amount of Nitrogen Application (질소처리에 따른 여러 관엽식물의 생육반응)

  • This study was carried out to investigate the plant growth of various foliage plants affected by the amount of nitrogen application. There was not an accurate criterion for fertilization to each foliage plant. Moreover, the foliage plants grow slowly during the early stage and this period must be shortened for commercial production. Ficus benjamina, Hedera helix, Philodendron tatei, Rhapis excelsa, and Spathiphyllum spp. were used in this experiment. Nutrient nitrogen concentrations were 120, 150, 180, and $210mg{\cdot}L^{-1}$ (N1, N2, N3, N4) and they were based on the Sonneveld solution. Plant height and width, leaf number, leaf area, fresh and dry weights of shoots were measured for 8 weeks to compare the responses to the different treatments. The nitrogen contents of various plant parts were also analyzed. The growth of Ficus and Hedera was improved at the N1 treatment and that of Philodendron, Rhapis and Spathiphyllum at N4 treatment. The required amounts for nitrogen nutrients were different among the various foliage plants. The nitrogen treatments had no effects on SPAD values and there were no correlations between nitrogen treatments and nitrogen contents of plant parts. Therefore, the various foliage plants should supply with each proper nitrogen treatments to grow faster with better quality. In this case, the plant growth played a more important role than nitrogen levels of plant parts in deciding the proper nitrogen levels for each foliage plant.

Seasonal Growth Inhibition of Perennial Ryegrass (Lolium perenne L.) after Applying Two Plant Growth Regulators (생장조정제 처리 후 생육 시기별 퍼레니얼 라이그래스의 억제 효과)

  • This study was conducted to evaluate the effects of trinexapac-ethyl (TE) and prohexadione-calcium (PC) on the growth and turf quality of perennial ryegrass (Lolium perenne L.). Treatments were designed as follows; control (non-treatment), TE (TE 0.01 a.i. g·m-2·100mL-1), and PC (PC 0.01 a.i. g·m-2·100mL-1). Compared with control, turf color index and chlorophyll content of TE and PC treatments were not significantly different. As applied plant growth regulators (PGRs), shoot length in the TE and PC was decreased by 8.9-12.4% and 6.7-13.6%, respectively, and clipping yield by 44.4-45.8% and 40.6-40.9%. When evaluated with the growth of shoot length after applying PGRs, residual days of TE and PC were 43.6 and 37.9 days after treatment of PGRs (DAT) in the 1st experiment (July 29-August 26), respectively, and 38.3 and 39.5 DATs in the 2nd experiment (September 30-October 28). These results indicated that an application of TE or PC decreased shoot length and clipping yield in the perennial ryegrass, and their residual time was about 40±3 DATs. Perennial ryegrass applied with PGRs like TE and PC was not significant seasonal difference, and might be not phytotoxic.

Growth Modeling of Perilla frutescens (L.) Britt. Using Expolinear Function in a Closed-type Plant Factory System (완전제어형 식물공장에서 선형지수함수를 이용한 들깨의 생육 모델링)

  • Growth modeling in plant factories can not only control stable production and yield, but also control environmental conditions by considering the relationship between environmental factors and plant growth rate. In this study, using the expolinear function, we modeled perilla [Perilla frutescens (L.) Britt.] cultivated in a plant factory. Perilla growth was investigated 12 times until flower bud differentiation occurred after planting under light intensity, photoperiod, and the ratio of mixed light conditions of 130 μmol·m-2·s-1, 12/12 h, red:green:blue (7:1:2), respectively. Additionally, modeling was performed to predict dry and fresh weights using the expolinear function. Fresh and dry weights were strongly positively correlated (r = 0.996). Except for dry weight, fresh weight showed a high positive correlation with leaf area, followed by plant height, number of leaves, number of nodes, leaf length, and leaf width. When the number of days after transplanting, leaf area, and plant height were used as independent variables for growth prediction, leaf area was found to be an appropriate independent variable for growth prediction. However, additional destructive or non-destructive methods for predicting growth should be considered. In this study, we created a growth model formula to predict perilla growth in plant factories.

Effects of Aeration Level to Root-zone on the Growth and Fruit Yield of the Hydroponically Grown Cucumber Plants in Spring Season (춘계재배시 양액재배 베드내 통기수준 차이가 오이의 생육 및 과실수량에 미치는 영향)

  • 초장과 엽수에 있어서는 별도의 배수판을 설치한 Bio-2처리구에서 높게 나타났고 fan을 이용한 강제환기를 시킨 Fan처리구에서는 낮게 나타났다. 경경과 엽면적에 있어서는 무통기처리구인 대조구에서 높게 나타난 반면 경경에 있어서는 Fan처리구와 배수구 통기구인 Bio-1처리구는 생육후기에 있어서 크게 차이가 나지는 않았으나 본 실험에서는 낮게 나타났다. 엽면적은 Fan처리구가 가장 낮게 나타났다. fan을 설치하여 강제환기를 시킨 Fan처리구가 과실수량과 과중, 과장에 있어서는 다른 처리구에 비해 높게 나타났고 무통기구인 대조구가 가장 낮았다. 전반적인 생육이나 과실수량 및 과실품질에 있어서는 근권의 수분함량비율이 비교적 안정적인 Bio-2처리구가 가장 좋은 것으로 나타났다.
