Isolation and Optimized Culture Conditions of Fibrinolytic Enzyme Producing Strain Isolated from Korean Traditional Soybean Sauce (간장 유래 혈전분해 효소 생산 균주의 분리 및 배양학적 특성)

  • Bacterial strains exhibiting fibrinolytic activity were screened from traditional Korean soybean sauce. The Fibrinolytic activities of the various isolated microorganism were further examined and the superior strain YJ11-21 was selected for further analyses. Gene sequence analysis of 16S rDNA of the YJ11-21 strain revealed Bacillus licheniformis. Optimal culture conditions were investigated in order to maximize the production of the fibrinolytic enzyme by YJ11-21. Amongst the carbon sources tested, glucose was the most effective for enzyme production and amongst the nitrogen sources tested, yeast extract was seen to be the most effective. A one percent addition of NaCl to the medium resulted in the highest fibrinolytic activity. Interestingly, a 10% addition of NaCl resulted in a high activity together with a high cell growth rate. Therefore, YJ11-21 is speculated of being a halotolerant. The optimum pH and temperature for enzyme production were a pH of 9.0 and $30^{\circ}C$, respectively.

Solubilization of Insoluble Phosphates by Penicillium sp. GL-101 Isolated from Soil (토양에서 분리한 Penicillium sp. GL-101에 의한 난용성 인산염의 가용화)

  • Phosphate solubilizing microorganisms (1,000 bacteria and 200 fungi) were isolated from soil around Kyungnam and Kyungbook regions using potato dextrose agar-calcium phosphate medium. A fungus with the greatest phosphate solubilizing activity was selected and identified to Penicillium sp. GL-101, based on the morphological characteristics of conidiophore and conidia; flask shape of phialide, simple branching type of conidiophore, and columnar shape of conidial head, in malt extract agar and potato dextrose agar media. The optimum temperature and initial pH to solubilize rock phosphate in potato dextrose broth-rock phosphate medium were $25^{\circ}C$ and pH 7.5, respectively. In these optimum conditions, phosphate solubilizing activities of Penicillium sp. GL-101 against four types of insoluble phosphate: tricalcium-phosphate, aluminium phosphate, hydroxyapatite and rock phosphate, were quantitatively determined. As results, this fungus highly discharged free phosphates to the culture broth with the concentrations of 1,152 ppm against tricalcium-phosphate, 565 ppm against rock phosphate, 292 ppm against aluminium phosphate, and 217 ppm against hydroxyapatite, respectively.

Effects of Fruiting Productivity of Grifola frondosa Using Bottle Cultivation according to Different Substrate Composition (잎새버섯에서 배지조성이 병재배 자실체 생산성에 미치는 영향)

  • To determine a favorable substrate formulation for Grifola frondosa, physicochemical conditions, culture properties, and yields according to various substrate formulations were investigated. Based on these analyses, T4 (80:5:15 ratio of oak sawdust to dried bean-curd refuse to corn husk) resulted in a shorter cultivation period and higher yields (weight of fresh mushrooms harvested at maturity) than those of other treatments. The physicochemical properties of T4 were pH 5.4, 2.4% crude fat contents, 54 C/N ratio, 74.3% porosity, and 0.26 g/cm3 bulk density. These results emphasize the importance of optimal substrate development on the production efficiency of G. frondosa mushrooms and have implications for commercial applications.

Correlation between Serum Albumin Level and IgG Level in Minimal Change Nephrotic Syndrome (소아 미세변화형 신증후군에서 혈청 알부민과 IgG의 상관관계)

  • Purpose : Hypogammaglobulinemia has been observed in nephrotic syndrome, but its pathophysiology remains unknown. We evaluated the relationship between the serum IgG and at bumin levels for children with minimal change nephrotic syndrome(MCNS). Methods : The levels of immunoglobulin G(IgG), albumin and total cholesterol of a total of 46 children with MCNS(proteinuria $>40mg/m^2/h$, and serum albumin level <2.5g/dL were analyzed. Results : The mean values of albumin, IgG and total cholesterol in MCNS children were $1.7{\pm}0.3g/dL,\;368{\pm}143mg/dL\;and\;431{\pm}78mg/dL$, respectively. There was an inverse correlation between the albumin values and the total cholesterol values(r=0.68, P=0.0001), whereas there was a direct-proportional correlation between albumin values and the IgG values(r=0.4, P=0.01). Conclusion : The IgG level is associated with albumin level, and it may reflect the severity of urinary protein loss in MCNS. Further studies are needed to evaluate this phenomenon.

  • This study was carried out investigate optimal additive content of spent medium from oyster mushroom in Volvariella volvacea cultivation. The formation of spent mushroom medium from oyster mushroom consisted of 50% pine sawdust, 30% beet pulp and 20% cotton seed hull pellet. The higher amount of spent medium from oyster mushroom resulted in higher total nitrogen content and C/N ratio of medium decreased according to increasing mixing ratio of spent medium from oyster mushroom. The fruit body yield and biological efficiency of supplemented with 50% spent medium from oyster mushroom was same level of non additive medium.

신 품종 느타리버섯 "청풍, 명월" 육성

  • 느타리 버섯의 다양한 품종과 재배형태가 급변히 요구되는 시기에 충남농업기술원 버섯연구팀의 5년 (1996~2,000)에 걸쳐 국,내외 수집 20 균주를 공시하여 개발 육성한 청풍과 명월 신품종 느타리버섯을 현재 재배되고 있는 기존 느타리품종과 동일여부를 위해 PCR-RAPD분석과 배양적, 재배적 특성을 인공재배 시험한 결과를 요약하면 다음과 같다. 1. 청풍, 명월느타리버섯 균사배 양 온도는 청 풍은 20~$25^{\circ}C$,명 월은 25~3$0^{\circ}C$, 이고 배지는 PDA배지가 양호하고 균사생육은 청풍, 명월 모두 비슷하였으며 배지 pH는 6.5~7.5 였다. 2. 버섯재배가능은 청풍은, 5~26$^{\circ}C$ 명 월은, 7~28$^{\circ}C$로 온도범위가 넓어 재배가 용이하나 적정 생육온도는 청풍, 명월 공히 15~19$^{\circ}C$가 양호 하였다 3. 청풍과 명월의 배양온도 23$^{\circ}C$에서 균사배양기간 23~24일 (850cc/pp),폐면 균상 재배에서 균배양 기간은 청풍, 명월 공히 11~12일, 초발이 소요일수는 2~3일 소요 되었다. 4.초발이시 청풍과 명월 갓 색택은 진청색이고, 생육후기 청풍은 회갈색, 명월은 진회색으로 조직이 치밀하고 저장성이 양호하여 경제성이 기대된다. 5. 다발수는 춘추2호 (13개/다발) 대 비 청 풍11개, 명월12개로 약간 적었으나 개체중은 춘추2호(14g/개) 대비 청풍 16g/개, 명월 14g/개로 비슷한 경향 이었다. 6. 수량성은 균상 폐면재배시 춘추2호 (40kg/3.3$m^2$)대비 청풍 느타리버섯은 (46kg/3.3$m^2$) 15%증수되고, 명월느타리버섯은(41kg/3.3$m^2$)로 3% 증수되었으며 병재배 겸용도 가능하였다. 7. PDA대치배양과 PCR-RAPD시험에서 청풍, 명월 신품종느타리 버섯은 기존 재배 되고 있는 느타리품종과 다른 품종으로 확인 되었다.

Comparison in Cultural Characteristics on Different Nutritions in Bag Cultivation of Lentinula edodes (표고 봉지재배에 있어서 영양원에 따른 재배적 특성 비교)

  • This study was carried out to select the suitable substrates for bag cultivation of Lentinula edodes. We investigated the optimal additive materials and its mixing ratio in bag cultivation of L. edodes, Sanjo 701 ho. The suitable substrates for L. edodes bag cultivation were oak sawdust as new material plus deffatted corn flour, and corn husk as an additive at the ratio 8:1:1(v/v), as the result of shorter mycelial growth, higher biological efficient, and a higher yield than any other substrates.

Studies on the development of mushroom media for bottle culture in new Pleurotus ostreatus 'Miso' (신품종 '미소' 느타리버섯 병재배 배지개발)

  • This study was carried out to develop the best medium of new oyster mushroom for bottle culture. The new oyster mushroom cv. Miso is a Pleurotus ostreatus developed at the Chungnam Agricultural Research & Extension Services. For the bottle culture media, poplar sawdust+beet pulp+cottonseed meal (5:3:2), poplar sawdust+wheat husk meal (8:2), poplar sawdust+beet pulp+cottonseed meal (6:2:2), poplar sawdust+beet pulp+cottonseed meal (4:4:2), and poplar sawdust+beet pulp+wheat husk meal (7:1:2) were used in 850cc PP bottle. The pH was 5~6 and the C/N ratio 19.7~28.3 in bottle culture media. The time of pinhead formation was 5 to 6 days. For the fruiting body formation after inoculation took 29~31 days. The yield of fruiting body of poplar sawdust+beet pulp+wheat husk meal (7:1:2) medium was the highest at 110.4g/bottle compared to other media. Therefore, such cultivation medium would be appropriate for the commercial production of bottle culture in the new oyster mushroom 'Miso'.

Physical and Chemical Characteristics of Cotton Waste Substrate According to Fermentation Conditions for Oyster Mushroom Bed Cultivation (느타리버섯 폐면배지의 발효조건별 이화학적 특성)

  • We have surveyed the variation of physical and chemical characteristics of aerobic and anaerobic outdoor fermentation of cotton wastes using for oyster mushroom cultivation. The inner temperature of cotton wastes fermented aerobically covered with thin cloth and setting pallet at bottom was higher than that of anaerobic fermented cotton wastes covered with P.E vinyl and the maximum temperature was $75^{\circ}C$ at 5th day after fermentation. pH of cotton wastes fermented aerobically was increased up to 8.9 after fermentation of $9{\sim}12$ days, but that of anaerobically fermented was decreased up to 5.0. Total carbon content was decreased but total nitrogen content was increased when fermentation was in progress. Oxygen concentration of cotton wastes fermented aerobically was decreased until 6 days after fermentation but increased after 9 days of fermentation. Ammonia concentration of cotton wastes fermented aerobically and anaerobically was below 10 ppm and $20{\sim}85\;ppm$ respectively. In anaerobic condition the cotton wastes was contaminated with mold ($15{\sim}50%$), where no contamination was found in aerobic condition during spawn running stage. Yields of mushroom grown on cotton wastes aerobically fermented for $6{\sim}9$ days was $23.0{\sim}23.6\;kg$ per $3.3\;m^2$ area.

Production of Poly-3-hydroxybutyrate from Xylose by Bacillus megaterium J-65 (Bacillus megaterium J-65에 의한 xylose로부터 poly-3-hydroxybutyrate 생산)

  • A microorganism capable of producing high level of poly-3-hydoxybutyrate (PHB) from xylose was isolated from soil. The isolated strain J-65 was identified as Bacillus megaterium based on the morphological, biochemical and molecular biological characteristics. The optimum temperature and pH for the growth of B. megaterium J-65 were $37^{\circ}C$ and 8.0, respectively. The optimum medium composition for the cell growth was 2% xylose, 0.25% $(NH_4)_2SO_4$, 0.3% $Na_2HPO_4{\cdot}12H_2O$, and 0.1% $KH_2PO_4$. The optimum condition for PHB accumulation was same to the optimum condition for cell growth. Copolymer of ${\beta}$-hydroxybutyric and ${\beta}$-hydroxyvaleric acid was produced when propionic acid was added to shake flasks containing 20 g/l of xylose. Fermenter culture was carried out to produce the high concentration of PHB. In batch culture, cell mass was 9.82 g/l and PHB content was 35% of dry cell weight. PHB produced by B. megaterium J-65 was identified as homopolymer of 3-hydoxybutyric acid by GC and NMR.
