Qualities and Early Growth Responses of Paprika Seedlings Grown in High and Low Temperatures (고온 및 저온에서 육묘된 파프리카 묘의 소질과 정식 후 초기 생육 반응)

  • The effect of high and low temperatures on seedling quality while raising of paprika (Capsicum annuum L.) seedlings, and their early development after planting was investigated. The control raising seedling temperature (RST) was $23^{\circ}C$; high temperature, $31^{\circ}C$; and low temperature, $15^{\circ}C$ throughout the raising seedling period. At $15^{\circ}C$ and $30^{\circ}C$, plant height, stem diameter, fresh weight, dry weight, number of leaves, and seedling leaf area were significantly lower than those at $23^{\circ}C$. At 4 weeks after planting, seedling's growth characteristics showed a similar pattern. Compared to $23^{\circ}C$, seedlings raised at $15^{\circ}C$ and $30^{\circ}C$ had an increased dry weight and leaf area per unit time after planting than during the seedling raising period. At 4 weeks after planting, crop growth rate and leaf area index were unaffected by RST, and relative growth rate and net assimilation rate at RSTs of $15^{\circ}C$ and $31^{\circ}C$ were higher than those those at RST of $23^{\circ}C$. At an RST of $15^{\circ}C$, growth speed and net assimilation rates were higher after planting than before planting, according to increased photosynthetic rate. Thus, high and low temperatures during the seedling raising period significantly reduced seedling growth and plant growth after planting. After planting, seedlings raised at $15^{\circ}C$ recovered more quickly than did those raised at $31^{\circ}C$.

  • This experiment was conducted to investigate biological activity of ABA and its analogues on barley shoot growth and peroxidase activity in barley seedlings. The treatments of 3.2 ppm (+)-ABA, 1.2 ppm (S)-(+)-ABA, 0.6 ppm ABA-methyl cinnamate ester compound (AC), and 0.08 ppm ABA-umbelliferone ester compound (AU) inhibited the growth of barley seedlings by more than 80% as compared with untreated control. The increase in barley shoot-inhibit activity of (S)-(+)-ABA became 3-fold than the activity of racemic ABA, and those of AC and AU higher than that of (S)-(+)-ABA by about 2.5 and 16 times, respectively. Peroxidase activities of barley shoots during the early growth stage were kept at a constant levels without ABA treatment, but in the treatment of racemic ABA, (S)-(+)-ABA, AC and AU increased the activities. Furthermore, the peroxidase activities increased as the higher concentration of ABA and its analogues were applied.

Vegetational Changes in the Early Stages after Lumbering of Populus albaglandulosa in Urban Forest (도시림에서 은수원사시나무 벌목 후 초기의 식생 변화)

  • To develop a better restoration technique for altering urban planted forest to more natural forest, the changes of flora, vegetation structure and woody plant growth in the early stage after lumbering only Populus albaglandulosa of the tree layer were studied in Chungdam Park, Chungdam-dong, Kangnam-gu, Seoul for two years (from 1997 to 1998). The results were as follow. First, in the year after lumbering, the species changes occurred not in woody plants but in herbs, which increased by 44%. Three species including Viola verecunda disappeared, and 15 species including ruderals such as Metaplexis japonica and introduced species such as Erechtites hieracifolia appeared newly. Second, lumbering caused the coverages of Quercus aliena, Q. mongolica, Q. acutissima and Sorbus alnifolia in the subtree, or shrub layer, to be increased abruptly. The coverage of Lespedeza bicolor was affected by neighboring plants. Third, in the herb layer the coverage of Artemisia keiskeana was conspicuously increased but those of the others were not. Fourth, by ridding the upper layer canopy, Quercus' growth rates were increased highly. Rates of DBH growth of Q. aliena, Q. mongolica and Q. dentata were increased to 53.0%, 22.9% and 8% in the experimental area, and 23.1%, 8.3% and 6.1% in the control area, respectively, during two growing seasons (from May 11, 1997 to October 27, 1998). The ratios of twig biomasses of the previous year to the next year were 100:565 in Q. aliena, 100:197 in Q. mongolica and 100:644 in Q. dentata in the experimental area. There were also growth ratios of 100:117 in Q. aliena, 100:100 in Q. mongolica and 100:42 in Q. dentata in the control area, respectively. The growth rate increases of Q. aliena and Q. dentata were thus conspicuous in twig rather than in trunk, but that of Q. mongolica was vice versa.

Effects of Addition of Inorganic Germanium, GeO2 on the Growth, Germanium and Saponin Contents of Ginseng Adventitious Root in Submerged Culture (무기 게르마늄 GeO2의 첨가가 액체 배양 중 인삼 부정근의 생장과 게르마늄 및 사포닌 함량에 미치는 영향)

  • This study was carried out to determine the optimal submerged culture conditions for production of ginseng root containing germanium using plant tissue culture technology. The ginseng (Panx ginseng C.A. Meyer) .cot induced by plant growth regulators of 0.5 mg/L BAP and 3.0 mg/L NAA was cultured on SH medium and the effects of various $GeO_2$ concentrations, addition time of $GeO_2$ and pH of medium were investigated on fresh weight, saponin production and germanium accumulation in ginseng root. Optimal $GeO_2$ concentrations for fresh weight, saponin and germanium content were 10, 0 and 110ppm, respectively. When $GeO_2$ was added after 2 weeks cultivation of ginseng root, germanium content was higher than that of adding $GeO_2$ at the initial cultivation time, but saponin content and fresh weight were lower. pH 5.5 was found to be the most favorable condition for the growth of ginseng root and germanium accumulation, but saponin production was best at pH 6.0.

Growth and Photosynthetic Rate of Pinus koraiensis and Pinus rigida in Banwol Industrial Region (반월공단지역의 잣나무 및 리기다소나무의 생장과 광합성 특성)

  • This study was performed to investigate the growth and photosynthetic rate of ten-years old Pinus koraiensis and Pinus rigida in Banwol industrial region comparing with those of the control site in Gwangju, Gyeonggi Province. Surface soil of the damaged site in pH 4.2 was more acidic than control site in pH 4.8, assuming the accelerated soil deterioration by acid deposition. The shoot growth, needle survival rate and net photosynthetic rate of P. koraiensis and P. rigida at the damaged site were lower than at the control site. In addition, the quantum yield and chlorophyll a+b and b contents reduced in damaged treatment. These results suggest that an inhibition of photochemical reaction of P. koraiensis and P. rigida at the damaged site of Banwol industrial region could be attributed to combined influence of atmospheric pollution and soil acidification.

  • 미나리 묘를 대량으로 생산하는데 있어서 체세포배를 이용하는 방법이 매우 유리하다. 체세포배는 계절에 영향을 받지 않고 좁은 공간에서 한꺼번에 많은 량의 묘를 생산할 수 있다. 또한 생물반응기 등을 이용하여 대량 생산할 수 있으면 실생묘를 대체할 수 있을 것으로 생각된다. 배양기 내의 공기 환경의 영향은 삼각플라스크를 이용한 소규모 배양에서는 문제가 되지 않았지만 배양기가 커짐에 따라 공기 환경의 변화도 커져 체세포배의 발생과 발아에 영향을 미친다. 체세포배를 배양하는 기간은 배발생과 발아로 나누어, 세포에서 배가 발생하는 처음 2주 동안에는 밀폐 환경, 즉 환기가 거의 되지 않는 환경이 유리하지만 이후의 발아 동안에는 산소를 많이 요구한다. 삼각플라스크를 이용한 소규모 배양에서는 aluminum foil을 플라스크의 뚜껑으로 하여, 배지를 교체할 때를 제외하고는 배발생과 발아가 진행되는 전 배양 기간 동안 이를 유지한다. 배가 발생하는 초기 2주의 경우는 이 밀폐 환경이 발아를 촉진시킨다. 그러나 배가 발아하여 정상 식물체로 발달하는데는 유리하지 않다. 체세포배는 발아할 때 산소를 많이 필요로 하며 에틸렌이 많이 축적되면 발아율이 낮아지며, 적절한 공기 환경이 주어지면 체세포배 발아가 동조화되어 균일한 묘를 얻을 수 있고 수확시기도 예측할 수 있다. 본 실험에서는 배가 발아하는 기간 동안 플라스크 내의 공기 환경을 다르게 하기 위해 플라스크를 막는 뚜껑의 소재를 달리하여 배양한 후 발아율을 측정하였다. 또한 가장 효율적인 환기량을 구명하기 위하여 인위적으로 플라스크에 공기를 넣어 강제 환기시키는 실험을 수행하였다. 미나리 cell은 처음 8일까지는 생장을 하지 않다가 이후 급속히 생장을 시작하여 35일 정도까지는 생장을 하다가 다시 생장이 둔화되었다. 밀폐시킨 삼각플라스크에서 자라는 Cell은 상태도 좋지 않고 전반적인 증식량도 적었다. Cell은 환기정도에 민감한 것으로 판단되며 삼각플라스크에서 약 35일 정도의 생장 주기를 가지는 것으로 사료된다. 배양 3주까지는 플라스틱 뚜껑으로 밀폐시킨 bottle에서 가장 많은 체세포배를 얻었다. Air filter를 달아 2일 마다 신선한 공기를 넣어 주었을 때는 배의 발달이 많이 늦어져 배양 3주째에 다른 처리보다 배의 수가 훨씬 적었다. 체세포배가 발달하는 동안에는 산소를 많이 요구하지 않으나 성숙하는 동안에는 산소를 많이 요구하는 것으로 생각된다.

Production of monoterpenoid flavor compounds by suspension culture of peppermint cells (페파민트 세포의 현탁 배양시 생육 및 정유생성 특성)

  • To investigate the production of monoterpenoids by Mentha pipperita cells in suspension culture, effects of media formulation, plant growth hormones, initial pH of the media, and cold stress on the production of essential oil and menthol were analyzed. Among the media employed, Lin-Staba medium resulted in the best essential oil production. Addition of 100 mg/l of yeast extract to the Lin-Staba medium induced the cells to produce large amount of essential oil and high content of menthol (0.39 g/l and 19.6%, respectively). In the effect of plant growth hormone, auxine were more effective than cytokinins. At initial pH of 4.7, oil production was good but menthol content was low. However at pH 5.7 the trend was reversed. When the culture temperature was lowered from $27^{\circ}$ to $10^{\circ}$ during 6 hour-dark period, growth was not changed much but essential oil production and menthol content was increased and reached to 528 mg/l and 21%, respectively.

Effect of Iron Ion on Cell Division and Microcallus Growth in Mesophyll Protoplast Cultures of Arabidopsis thaliana (철이온이 Arabidopsis thaliana 초기 원형질체배양의 세포분열 및 미세 캘러스 생장에 미치는 효과)

  • This study was performed to investigate the effect of iron ion on the mesophyll protoplast culture of Arabidopsis thaliana. Mesophyll protoplasts were isolated and cultured in a modified IMH medium supplemented with various concentrations of Fe-EDTA. Relatively low concentration of Fe-EDTA (<0.02 mM) induced the low level (4.8%) of cell division. In addition the cell division and microcallus growth were dose-dependently stimulated by 0 to 1 mM of Fe-EDTA. In 0.5 to 1 mM concentration range of Fe-EDTA, microcolonies were readily formed and the plating deficiency (8.5%) also showed maximal rate. However more than 1 mM of Fe-EDTA inhibited the initial growth of protoplase. The overall results suggest that Fe2+ion concentration plays an important role at the early developmental stage of protoplast regeneration.

  • This work was conducted to estimate amounts of irrigating water during the growing periods of pepper after estimating water consumption in the plastic film house in 1997 and 1998. Evapo-transpiration (ET) under conditions of a black and white PE mulch and sandy or clay loam soil which enhanced the growth and yield of red peppers was greater than that at the bare and sand soil. Average ET of pepper grown in pots accounted for 56.5%∼79.7% of total supplying water in 1997 and 1998. Most of ET was proportioned to the transpiration amount (91∼94%), but there was some difference between amounts of ET and transpiration plus evaporation. Although 57 depended on conditions of the soil texture and plant growth, transpiration for pepper growing periods was amounted to 337.7∼774.3 m in the clay soil,910.6 m in the sandy loam soil, and 253.1 mm in the sandy soil.

Growth Characteristics of Angelica gigas Nakai on Transplantation Season (April and Autumn) (정식계절(봄, 가을)에 따른 참당귀(Angelica gigas Nakai)의 생장특성)

  • This study compared the growth characteristics of A. gigas according to the transplanting season, in the cultivation located in Yeongju, Gyeongsangbuk-do, transplanted in the autumn and spring. As a result of that the plant survival rate were observed the highest autumn transplanting (9.4%; autumn-92.2%, spring-82.8%) and bolting rate were observed lowest spring transplanting (7.1%; autumn-37.3%, spring-30.2%). Growth characteristics (height, leaf length and width, stem diameter) were observed the highest autumn transplanting in June and highest spring transplanting in August. The early growth is high growth due to long rooting time in autumn transplanting, but the difference in the ground growth between the two experiments was insignificant when the main growth period was from June to August. Further analysis of the growth characteristics and marker components of roots of A. gigas can be used to determine the optimal planting time and the establishment of high quality cultivation technology.
