Relationship of Somatic Cell Score and Udder Type Traits of Holstein Cattle (체세포점수와 홀스타인 유방형질간의 관계)

  • Data were taken from the dairy herd improve- ment program from the year 2000, composed of 10,929 first lactation cows consisting of 290,144 test-day records and 37,723 udder type records. The objective of the study was to estimate genetic and phenotypic correlation between fore udder attachment, rear udder height, rear udder width, udder cleft, udder depth, and somatic cell score (SCS) and to calculate heritability of udder depth, front teat length and SCS in Holstein cattle in Korea. The variance component estima- tion using test day model was determined by a derivative-free algorithm-restricted maximum likeli- hood(DF-REML) analysis method. Generally phenotypic correlations were very low between udder traits and lactation SCS which varied from -0.03 to -0.06. Heritability of all type traits and SCS was smaller than 0.12. The results of this study would be applicable to SCS using linear genetic evaluation for future studies.

Somatic Embryogenesis and Plant Regeneration with Embryogenic Tissue Lines in Pinus densiflora (소나무 배발생조직 라인 별 체세포배 유도 및 식물체 재분화)

  • This experiment was conducted to evaluate growth rate among 15 embryogenic tissue lines (ETLs), comparison of maturation on somatic embryos (SEs) with 13 ETLs and efficiency with various concentrations of gelrite on SEs germination in Japanese red pine (Pinus densiflora). In comparison of ETLs growth rate (folds) with 15 lines, the 05-4 line (5.3 folds) showed the highest rate, on the other hand, the lowest one was recorded in the line of 05-37 (1.4 folds). The 13 ETLs were tested for the extent of SEs production. The best production was recorded in the line of 05-4 (39.8/90 mg F.W.). However, most of ETLs, except 5 lines (05-4, 12, 21, 29 and 37), did not produce SEs at all, therefore, big differences in the ability of SEs production existed among the ETLs tested. Effects of various gelrite concentrations for SEs germination with 3 ETLs were also compared. The highest result was obtained from 0.2% gelrite concentration with 05-4 line (47.3%), there was a inclination that the rate of germination was gradually declined over 0.2% gelrite concentration with the 05-4 and 29 lines. respectively. In contrast, in the line of 05-37, no SEs germination occurred on medium with 0.1 or 0.2% gelrite. In conclusion, the growth rate, SEs production and germination frequency were appeared to deeply depended on the ETLs.

Effect of Abscisic Acid, Kinds and Concentrations of Osmoticum on Somatic Embryo Induction, Germination and Plantlet Regeneration in Larix kaempferi (ABA 및 삼투압제 종류 및 농도에 따른 낙엽송 (Larix kaempferi) 체세포배 유도, 발아 및 식물체 재분화 효과)

  • This study was conducted to examine effects of concentrations of abscisic acid (ABA) or /kinds of osmotica on induction of somatic embryos (SEs), germination and plantlet regeneration in Japanese larch (Larix kaempferi). In comparison of duration of culture, concentrations of ABA and osmoticum, the highest induction number (191/g tissue) of the SE was showed in $60{\mu}M$ ABA+0.2 M sucrose for 4 weeks culture. However, the lowest number (3.5~23.5) of SEs was induced from $4{\mu}M$ ABA+0.1 M sucrose, regardless of culture duration for SEs induction. In comparison of germination efficiency of SEs, the highest induction frequencies of cotyledon (90.9%), hypocotyl (95.8%) and root (96.5%), respectively, were obtained from the SEs that cultured from the treatment of $60{\mu}M$ ABA+0.2 M sucrose with 5 weeks culture. In contrast, the lowest germination response was showed in SEs that induced from the treatment of $4{\mu}M$ ABA+0.1 M sucrose. In comparison of effect of different kinds/concentrations of osmotica for germination and plantlet regeneration, the best response was obtained from the treatment of 0.2 M sucrose with induction of cotyledon (98.3%), hypocotyl (78.4%), root (57.5%) and plantlet regeneration (54.8%), respectively.

Effects of Ascorbate on Somatic Embryogenesis in Carrot Cell Cultures (당근 세포배양으로부터 체세포배 발생에 미치는 아스콜빈산의 효과)

  • This study was conducted to elucidate the effects of ascorbic acid and dehydroascorbic acid on somatic embryogenesis from the cultured cells of carrot. Ascorbic acid in culture medium merely stimulated the proliferation of non-embryogenic cells but dehydroascorbic acid in medium induced embryogenic cells from non-embryogenic cells accompanying the inhibition of cell proliferation. Ascorbic acid in medium inhibited somatic embryogenesis from embryogenic cells while dehydroascorbic acid in medium enhanced somatic embryogenesis from the cells as well as non-embryogenic cells. This enhancement was limited to globular embryos and the maturation to cotyledonary embryos was inhibited by dehydroascorbic acid treatment. From the above results it is suggested that carrot callus cultures on medium containing dehydroascorbic acid could quickly induce embryogenic cells. In addition after brief culture of embryogenic cells on development medium containing dehydroascorbic there by acid the subculture of the cells to MS basal medium resulted in the high frequency production of somatic embryos.

Study on the Somatic Chromosome Numbers of Korean Aster L. and Its Allied Taxa (한국산 개미취속 및 근연 분류군의 체세포염색체수에 관한 연구)

  • The somatic chromosome numbers among the various taxonomic characters about 17 taxa in Korean Aster and its allied taxa were investigated to review accurate scientific name and taxonomic rank. The somatic chromosome numbers of the treated taxa were invariable in same taxa, but variable among different taxa. The treated taxa were divided into two types by basic chromosome numbers, one type was x=9, the other x=8 and x=9 type was subdivided by polyploidy. The somatic chromosome numbers of Aster altaicus var. uchiymae, A. hyatae, Kalimeris chejuensis were reported firstly in this study, and based upon somatic chromosome numbers and leaf morphology, the plants, idenified as Aster pinnatifidus in Korea was considered variant of Kalimeris incisa.

  • 고양이 체세포 복제산자의 생산은 Shin등(2002, Nature 415: 859) 1마리의 "Cc"를 성공한 이래 재현 생산한 보도가 없다. 그리하여 본 실험에서는 체세포 복제산자 생산을 위한 기초 실험으로서 난자의 체외성숙, 체세포핵이식 수정란의 체외발달율을 검토하고자 수행하였다. 성성숙한 고양이에서 난소적출수술로 난소를 적출 한 후 4시간 내에 실험실로 운반하여 면도칼로 난소를 절재한 후 미성숙난자를 채취하여 TCM199 + 0.4% BSA + 1000 IU/ml bST에서 24시간 체외성숙하였다. 체외성숙한 난자는 난구세포를 제거 후 핵이식에 공시하였다.   • 병풀(Centella asiatica)의 엽병 조직절편에서 유도된 배발생 캘러스로부터 체세포배 발생에 의하여 소식물체의 재분화를 이루었다. 엽병 조직절편을 1mg/L 2,4-D와 1mg/L kinetin이 조합 처리된 MS 기본배지에 치상하여 85%의 효율로 배발생 캘러스를 유도할 수 있었으며 이와 같은 배발생 캘러스를 5mg/L NAA와 1mg/L kinetin이 첨가된 배지로 옮겼을 때 체세포배의 형성은 87%까지 이루어졌다. 체세포배는 기본배지의 농도를 절반으로 줄이고 0.2 mg/L NAA와 0.2 mg/L kinetin이 첨가된 배지조건에서 기관분화를 거쳐 소식물체로 재분화되었다.
