The morphology and somatic chromosome numbers of Korean Parasenecio were investigated to estimate their taxonomic values. Some morphological characters, such as the length of whole plants, the numbers of cauline leaves, the shapes of leaf and petioles, the numbers of involucral bracts, floret numbers per head, presence of trichomes on the style, ratio between wide part and narrow part of floret and the size of achene were found to be a good characters for delimiting each taxa. Based upon these characters, six taxa, Parasenecio auriculala var. auriculata, P. auriculata var. kamtschatica, P. hastata subsp. orientalis, P. koraiensis, P. firmus, P. pseudotaimingasa, were recognized about their distribution in Korea, but 2 taxa, P. adenostyloides, P. auriculata var. matsumurana, were needed to be studied their taxonomic position and distribution in Korea. The somatic chromosome numbers of six taxa, P. adenostyloides, P. auriculata, P. auriculata var. kamtschatica, P. hastata subsp. orientalis, P. firmus, P. pseudotaimingasa were 2n=60 and those of P. firmus and P. pseudotaimingasa were reported first in this study.
The chromosome morphology of two Korean Lactuca (L. indica, L. triangulata) is reported herein. The chromosome number and karyotype of a naturalized plant, L. scariola are reported for the first time. The basic chromosome number was x = 9. Polyploid forms were not recorded. The karyotypes of L. indica, L. scariola, and L. triangulata were 2 n = 18 = 2 m+ 7 sm, 2 n = 18 = 1 m + 6 sm+ 2 st, 2 n = 18 = 2 m + 5 sm+ 2 st, respectively. Both L. indica and L. triangulata had satellites at the ends of their short arms. The haploid genome lengths of L. indica, L.scariola, and L. triangulata were $56.3{\mu}m$, $35.3{\mu}m$, and $72.5{\mu}m$ respectively. Each chromosome length of naturalized L. scariola was $2.7-5.2{\mu}m$; the smallest among Korean Lactuca. The chromosome lengths of L. indica and L. triangulata were $4.7-7.6{\mu}m$ and $2.9-7.9{\mu}m$, respectively. The karyotype of L. scariola differed from that of L.indica and L.triangulata both of which belong to sect. Tuberosae. Therefore, L. scariola is thought to belong to sect. Lactuca subsect. Lactuca.
Codonopsis lanceolata ("Deoduck" in Korea) is a perennial herb, and belongs to family, Campanulaceae. Its taproot is used a good source of a wild vegetable as well as an herbaceous medicine. In this study, to develop a bialaphos-resistant transgenic Codonopsis, seed germination mechanism and somatic embryogenesis of the plant were investigated, and Agrobacterium-mediated transformation with bar gene encoding phosphinothricin acetyltransferase (PAT) was performed. Attempt were made to regenerate plant from cells via somatic embryogenesis. When the cotyledons, nodes and leaf disks were cultured on MS medium containing 2,4-D and zeatin, embryogenic calli were induced. Upon transferring the somatic embryos to N6 solid medium without plant growth regulators, they developed into plantlets under continuous illumination. All plants were dead on MS basal medium containing 10 mg/L phosphinothricin (PPT) and Basta, respectively. The explants did not produce calli in the medium containing 200 mg/L kanamycin. The explants were cocultured with Agrobacterium tumefaciens for 2 days, and transformants were selected in MS basal medium containing 1.0 mg/L 2,4-D, 100 mg/L kanamycin and 500 mg/L carbenicillin. After the selection, embryogenic calli were induced and then somatic embryos were produced by subsequent subculturing. The somatic embryos were germiated on N6 basal medium containing 200 mg/L kanamycin and 500 mg/L carbenicillin. PCR analysis showed that nptII and bar genes were introduced in the Deoduck transformants. After the confirmation of bar gene expression in RNA and protein level, the transgenic Deoduck will be used to study the genetics of filial generation with the herbicide control gene, bar.gene, bar.
In order to find optimum conditions for somatic embryogenesis from different individual (2-year-old) in Aralia elata were cultured on MS medium supplemented with 1.0 mg/L 2,4-D, 3% sucrose, and 0.3% gelrite. We also investigated the effect of MS medium salt concentration, BA and ABA on the embryo germination and plant regeneration. While noticeable difference was observed on somatic embryo induction among different individual tree, no apparent difference was seen in both germination and regeneration frequencies. Compared with nonembryogenic calli, embryogenic calli tended to look yellow and/or pale brown in color, slowly growing and soft in their texture. Regardless of BA or ABA treatment, half-strength MS salt medium proved to be better than full strength MS medium in both embryo germination and plant regeneration. Both hypocotyl and cotyledon developments were slightly promoted by adding 0.1 mg/L BA. However, its effect on germination and regeneration seemed inferior to control. ABA treatment on somatic embryos at their torpedo and early cotyledonary stages resulted in poor response in germination and regeneration. Although most regenerated plantlets varied greatly in cotyledon number and shape, they could be developed into normal plants after 4 weeks in culture. More than 95% plantlets were acclimatized in an artificial soil mixture, successfully transplanted to nursery bed and grew normally without any phenotypic abnormalty.

To determine physical and physiological factors for Liriodendron tulipifera L. somatic embryo germination, temporary immersion bioreactor (TIB) system was investigated. It was designed to immerse liquid media with plantlets so that it was able to adjust the immersion time. Immersion of 120 minutes every 4 hours and 60 minutes every 4 hours was found to be effective in germination (91.64%, 85.67%, respectively). However, hyperhydricity of the plantlets was higher in short immersion time (15 minutes every 6 hours) and long immersion time (120 minutes every 4 hours) (51.61%, 34.28%, respectively). Immersion of 60 minutes every 4 hours showed the lowest hyperhydric plantlets, and also it showed the lowest activities of abscisic acid (ABA), superoxide dismutase (SOD), and catalase. The overall results implied that immersion time of media affected germination and growth of somatic embryo, and it was able to make use of germination and growth of L. tulipifera somatic embryos.
Morphogenetic responses were investigated by culturing embryo and leaf explants of Korean wild type tea plant, Camellia sinensis (L.) O. Kuntze. Induction of direct somatic embryogenesis as well as adventitious and/or axillary shoots was obtained from mature zygotic embryo cultures on Murashige and Skoog (MS) basal medium having 5 to $20\mu\textrm{M}$cytokinin a lone. Morphogenetic response was decreased dramatically by the addition of auxins tested. One hundred percent of induced and isolated shoots formed roots after four weeks of culture on half-strength MS or quarter-strength Schenk and Hildebrandt (SH) media supplemented with $10\mu\textrm{M}$indole-3-butyric acid (IBA). Immature zygotic embryos were shown to be a suitable explant for embryogenic callus formation in the presence of 2, 4-dichlorophenoxyacetic acid(2, 4-D) in basal medium. Mature zygotic embryo originated leaves were used to test their ability for mophogenesis by incorporating plant growth regulators such as IBA, naphthyl-1-acetic acid (NAA), and 6-benzylaminopurine (BAP). Apparently, the morphogenetic responses of the cultured explant sources on the types and/or levels of plant growth regulators tested were observed visually.
The process to acquire intron-GUS gene-expressed transformants from somatic embryos (including embryogenic calli) of Rosa hybrida cv. 'Sweet Yellow' using Agrobacterium-meditated transformation method was reported in this study. Somatic embryos including embryogenic calluses were infected with Agrobacterium tumefaciens AGL1 strain (O.D = 0.7~1.6) including intron-GUS gene for 30 min, and were co-cultured for 3 days. After co-cultivation, they were cultured on embryo germination medium (EGM) supplemented with $250\;mg{\cdot}L^{-1}$ cefotaxim at $4^{\circ}C$ for 7 days. Then, transient GUS gene expression was observed. Shoots were regenerated from the shoot primodia induced from the intron-GUS gene-transferred either somatic embryos or embryogenic calli cultured on EGM supplemented with both cefotaxim $250\;mg{\cdot}L^{-1}$ and ppt $2\;mg{\cdot}L^{-1}$. Before induction of rooting from shoots cultured on shoot growing medium supplemented with both cefotaxim $250\;mg{\cdot}L^{-1}$ and ppt $2\;mg{\cdot}L^{-1}$, the shoots were cultured on multi-shoot induction medium supplemented with both cefotaxim $250\;mg{\cdot}L^{-1}$ and ppt $2\;mg{\cdot}L^{-1}$ to induce multi-shoots. When expression of the gene from a part of the multi-shoots was identified by GUS transient assay, the putative transgenic multishoots were transferred to rooting medium supplemented with cefotaxim $250\;mg{\cdot}L^{-1}$. After the formation of healthy roots, transgenic plantlets were transferred to the greenhouse after acclimatization. The expression rate of the intron-GUS gene in the multi-shoots was 100%.
Measurement of compositions and somatic cells in raw milk by chemical methods usually requires a lot of time, skilled labor and expensive analytical equipments. Recently, near-infrared reflectance spectroscopy (NIRS), which is a rapid, cost-effective and non-destructive technique, has been extensively used for safety and quality evaluation in the field of dairy products. However, less study has been performed to evaluate the effect of transmitted light on milk quality during NIRS analysis. Therefore, the objective of this study was to analyze the changes in milk quality using transmitted light. Raw milk samples collected from dairy farm from Siga prefecture in Japan were analyzed for fat, protein, lactose, solids not fat, total solids, milk urea and citric acid using the Milko scan 4000. Somatic cells in raw milk samples were counted by the Fossomatic 5000. Transmittance spectra of 50 ml raw milk samples were obtained by the Lax-Cute lighter in the 400 nm or less, 689 nm, 773 nm, 900 nm and 979 nm. As a result, milk fat as well as somatic cell count was increased by 2.6% and 9.0%, respectively. The other compositions were, however, changed within the relative error of the measurement. Further studies are needed to apply raw milk quality evaluation using the UV band by accumulating more samples and more data.
A photoautotrophic micropropagation methodology in liquid culture medium has a number of advantages for large-scale propagation of plants. This paper describes an improved system for the mass propagation via somatic embryogenesis of the medicinal plant Kalopanax septemlobus Nakai. Somatic embryo-derived young plantlets of K. septemlobus were cultured either under heterotrophic conditions with sucrose on half-strength MS medium with $30gL^{-1}$ sucrose, under heterotrophic conditions without sucrose, or under photoautotrophic conditions (MS liquid medium without sucrose under forced aeration) for four weeks before transferring the plantlets for acclimatization. Plantlets grown under photoautotrophic conditions had more leaves, higher chlorophyll content, a higher net photosynthetic rate (NPR), and a higher survival rate. The results indicate that the photoautotrophic conditions with a forced ventilation system are effective in enhancing the growth of plantlets and the rate of net photosynthesis. The plantlets grown under photoautotrophic conditions had a high survival rate (92%) upon ex vitro transplantation. Our study shows that autotrophically produced plantlets acclimatize better and sooner upon ex vitro transplantation than conventionally cultured plants.
