Hypopigmentary Effect of Milk Thistle Extract Silymarin (엉겅퀴 추출물 실리마린의 피부 미백효과)

  • We found that silymarin exhibited the inhibitory effect on melanogenesis in a spontaneously immortalized mouse melanocyte cell line, Mel-Ab. Silymarin is a standardized extract obtained from the dried seeds of milk thistle (Silybum marianum Gaertn.). Silymarin significantly prevented melanin production in a dose-dependent manner with an $IC_{50}$ value of 28.2 ${\mu}g/mL$ without effects on cell viability. Also, silymarin inhibited tyrosinase activity in melanocyte, while it did not affect the catalytic activity of cell-free tyrosinase. Furthermore, Western blot analysis indicated that silymarin decreased the expression of tyrosinase protein. Silybin A/B and isosilybin A/B were also able to inhibit melanin production and tyrosinase expression in protein level. Double blind study on the clinical efficacy of a cream containing 2 % silymarin showed that silymarin have a significant skin whitening effect. Therefore, this study suggests that silymarin may be useful as a natural skin whitening agent.

The Inhibitory Effects of Acanthopanax sessiliflorum Seeman on Melanogenesis (오가피추출물의 멜라닌 생성 저해 효과)

  • To develop a new natural whitening agent for cosmetics, we investigated effects of Acanthopanax sessiliflorum Seeman on melanogenesis. We prepared phenolic acid-rich extract including two phenolic acids, chlorogenic acid and caffeic acid, as predominant constituents from Acanthopanax sessiliflorum Seeman. Phenolic acid-rich extract showed ROS scavenging activities in 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical and xanthine/xanthine oxidase system with the $IC_{50}$ values of $3.43{\pm}0.35{\mu}g/mL$ and $158.91{\pm}1.57{\mu}g/mL$, respectively. Phenolic acid-rich fraction reduced melanin contents of B16 melanoma cells dose-dependantly and the decrease was $27.27{\pm}2.66%$ at a concentration of $100{\mu}g/mL$. And the phenolic acid-rich fraction reduced intracellular tyrosinase activity about $53.67{\pm}8.55%$ at a concentration of $100{\mu}g/mL$. Phenolic acid-rich extract inhibited tyrosinase and TRP-2 expression at protein level. These results suggest that phenolic acid-rich fraction reduced melanin formation by the inhibitions of tyrosinase activity and expression in B16 melanoma cells. Therefore, we suggest that phenolic acid-rich extract could be used as a whitening ingredient in cosmetics.

Effect of Vegetal Complex Raw Food on Plasma Glucose and Lipid Composition in High Fat Diet-induced Diabetic Rats (식물류를 이용한 생식용 천연복합식품이 고지방 식이에 의한 당뇨병 유발쥐의 혈청 glucose함량 및 지질구성에 미치는 영향)

  • This experiment was conducted to develop and design a product that is convenient and helpful to both patients and ordinary people who want to maintain their health. In this experimentation, the experimental rat have been given with high fat food to induce Diabetes. Organic brown rice was given as their main energy source and natural complex food, most of materials were freeze-dried, was given to observe its effects on their glucose amount in the plasma. The results are as follows. During 28 days of basal diets, as observing their weight variations, normal group gained weights, but the natural complex food(NCF) groups lost their weights. As comparing the amount of neutral fat levels in the plasma between the diabetes group who just had basal group and one who had NCF, group with NCF groups' level was much lower than the other group who just had basal food. As comparing total cholesterol levels in the plasma, with diabetes group with basal food, $50\%$ and $100\%$ NCF groups' levels were lowered than that group. Regarding HDL-cholesterol level, all NCE groups had around 30mg/dl and there were not much fluctuation among groups. As for LDL-cholesterol level, the $100\%$ NCF group had the lowest level and showed almost the same as the normal group. The amounts of the glucose in the plasma were lowered in all NCF groups. Among those NCE groups, $100\%$ NCF group had 120mg/dl to reach almost the same as the normal group.

Durability of concrete using sulfur-modified polymer (개질유황 폴리머를 사용한 콘크리트의 내구성 평가)

  • Most of the sulfur is obtained from desulfurization of natural gas and crude oil. In Korea, more than 120 tons of sulfur are produced by refinery, and about 50 % of the produced sulfur is used as a raw material for the production of fertilizer and sulfuric acid. Modified sulfur is manufactured from excessive sulfur that could be used to improve concrete properties, and this study evaluated concrete strength and durability that contains modified sulfur. Flexural and compressive strengths of concrete with sulfur modified polymer were comparable to those of OPC concrete with mixing water at similar temperatures, while the strengths increased a little as mixing water temperature increased. It was also confirmed that the resistance to freeze-thaw damage was more dependent on entrained air characteristics obtained by a proper use of air entraining agent than on the use of sulfur modified polymer. When concrete was immersed in 5 % sulfuric acid, the rate of reduction in compressive strength of OPC concrete was less than 1/4 of the strength reduction of concrete with sulfur modified polymer. Also, the resistance of concrete with sulfur modified polymer to scaling due to the use of de-icing salt was evaluated as Class 1, while that of OPC concrete was evaluated as Class 4, as aggregates were exposed. Accordingly, it is believed that sulfur modified polymer could be effectively used for bridge deck concrete since sulfur modified polymer improves the durability of concrete.

Development of Natural Seasoning from Alaska Pollack Skin Gelatin Using Continuous Three-Step Membrane Reactor (연속식 3단계 막 반응기를 이용한 명태피 젤라틴으로부터의 천연조미료 개발)

  • The hydrolysates of three kinds [FSEH(first step enzymatic hydrolysate), SSEH(second step enzymatic hydrolysate), and TSEH(third step enzymatic hydyolysate)] were prepared by continuous hydrolysis of Alaska pollack(Theragra chalcogramma) skin gelatin with three-step membrane enzyme reactor. The molecular weight distributions of FSEH, SSEH, and THSE are 9,500∼4,800Da, 6,600∼3,400Da, and 2,300∼900Da, respectively. The contents of amino acid having sweet taste (glycine, proline, serine, alanine, hydroxyproline, glutamic acid, and aspartic acid) were about 70% of total amino acid being in the three kind hydrolysates. We also tried preparing of natural seasonings (complex seasoning and enzymeatic hydrolysale sauce) using the hydrolysates. From the results of sensory evaluations, complex seasoning containing TSEH was nearly equal to shellfish complex seasoning on the market. The mixture sauce which was made by mixing of 80% enzymatic hydrolysis sauce and 20% fermented soy sauce, was at least similar to the tradition soybean sauce in product quality, too.

  • LNG plant projects tend to be implemented in overseas owing to its characteristics, so their project management scheme is somewhat different from those of general projects. Value chain in a LNG plant project includes exploration/production of gases, physical liquefaction/chemical conversion processes, transportation and storage. Key factors in the chain include liquefaction process (including ultra-low temperature liquefaction) to convert natural gas into liquid materials or fuel, and Front End Engineering Design (FEED) package, as well as Engineering, Procurement and Construction (EPC) technology comprising control, operation and construction. Success of a complex LNG plant project implemented in overseas depends on decision-making process in project management. Accordingly, to develop a decision-making model in of plant construction, the study extracted none factors in project management by EPC stage and assessed importance of each factor. The result showed that items in both project management and project risk management are important. Especially, the study developed a decision-making model in the construction stage of a LNG plant project based on the project management factors and importance assessment. The developed decision-making model would lay groundwork in building a decision-making system in construction stage of project management.

Processings of flavoring Substances from Small Kingfish (소형 갈전갱이를 이용한 풍미소재의 개발)

  • To develop natural flavoring substances, optimal hydrolysis conditions for two stage enzyme hydrolysates (TSEH) using small kingfish (Maegari) were investigated. The optimal conditions for TSEH were revealed in temperature at $50^{\circ}C$ for 3 hours digestion with alcalase (Aroase AP-10, pH 8.0) at the 1st stage and 2 hours digestion at $45^{\circ}C$ with neutrase (Pandidase NP-2, pH 6.0) at the 2nd stage. From the results in quality tests of water extracts, autolytic extracts and 4 kinds of enzyme hydrolysates, TSEH processing method was superior to other methods on the aspects of yield, nitrogen contents, taste such as umami intensity and inhibition of off-flavor formation, and transparency of extracts. We may conclude that TSEH from small kingfish was more flavorable compared with the conventional seasoning materials, it could be utilized as the seasoning substances for fisheries processing.

The Anticaries Activity of Hot Water Extracts from Foeniculum vulgare (회향열수 추출물의 치면 세균막 형성 억제 효과)

  • In this research, we screened for glucosyltransferase (GTase) inhibitors that effectively prevent the dental caries from 420 kinds of boiled water extracts of herbs and wild plants and searched for GTase inhibitory activities. Among them, 13 kinds of hot water extracts had high GTase inhibitory activities and especially, we focused on Foeniculum vulgare which showed the highest inhibitory activity on GTase. The boiled water extract of F. vulgare was stable at high temperature and showed as a mixed type of competitive and uncompetitive inhibition kinetic behavior. It did not have antibacterial effect on Streptococcus mutans and had inhibitory activity on GTase. Specially, in the clinical trial, the group treated by boiled water extract of F. vulgare showed more decrease of plague index at 4.8 point than untreated group. These results suggested that boiled water extract of F. vulgare can effectively suppress the plague formation as it inhibits the GTase activity.

  • Hydration process is one of the basic carbonation system. Limestone sludge produced in Pohang Iron & Steel Co., Ltd. We tested for identify of hydration characteristics. The result obtained in this study can be summarized as follows; 1. The classify of limestone sludge is type of ground calcium carbonate(-3mm+325mesh)and the major mineral of calcite, and further more high grade(CaO 51%), fine powder(15~22$\mu\textrm{m}$). 2. Limestone sludge mixed two process sludge, first one is washing process sludge and the other one is wet collect kiln dust. The composition rate is about 8:2. Wet collect kiln dust is major mineral of calcite, too. But the sludge is assumed to one by quick lime, slaked lime and unreacted natural limestone. So, the ideal process is dividing of the washing process sludge and wet collect kiln dust. 3. We manufactured of slaked lime from limestone sludge. To investigate the effect of hydration reactor, the experiments was done with various reactor type as magnetic stirrer, shaking incubator and ultrasonic vibration reactor, respectively. Generally, ultrasonic vibration reactor is excellent hydration for limestone sludge and produced very fine slaked lime powder with ideal distribution. 4. The optimum condition is 10% pulp density, when the manufacture of fine slaked lime powder by ultrasonic vibration reactor. And hydration times to compare the results of the study with ultrasonic vibration reactor of generalized most short time(5~10 min). 5. Finally, the dispersive characteristics of slaked lime powder measured 1~5 $\mu\textrm{m}$ from limestone sludge were compared with those of natural limestone ones(10~20$\mu\textrm{m}$), in order to check applicability of slaked lime with hydration process from limestone sludge.

  • The chemical composition of bottle glass is consisted of Na2O-CaO-SiO2. However the cullet is mornally used in order to decrease the melting tsmperature. This induce the productivity of bottle and decreases the cost. The addition of plate glass decreases the Al2O3 content and in fluence the stone phenomenon and devitification in botle glass. Tus the Feldspar is added in order to increase the Al2O3 content when plate cullet was added in melting. The Tridymite crystal was observed over 7.5% Al2O3 contents, which shown as white crystal in appearance. It is Supposed that the Wollastonite Would be occurred in more over 7.5% Al2O3. This fad id well consised With the Litertctures.
