Characteristic of Carotenoid Component from Halophilic Bacteria, Haloarcular sp. EH-1 (호염세균 Haloarcular sp. EH-1으로부터 추출한 카로테노이드 색소의 특성)

  • In order to identification of carotenoid pigments of Haloarcular sp. EH-1 as a food for fish were analyzed. The content of carotenoids cultured in 3 L and 5 L bioreactor were 83.1 and 82.7 mg%, respectively. Identification of each carotenoid was achieved by means of co-TLC and co-HPLC with authentic specimens, spectroscopic and instrumental analyses, and chemical treatments as usual. The main components identified were ${\beta}$-carotene(8.1%), 3-hydroxyechinenone(42.0%) and astaxanthin(25.0%).

Application of Response Surface Methodology for Optimization of Nature Dye Extraction Process (천연색소 추출공정 최적화를 위한 반응표면분석법의 적용)

  • As the use of environmentally friendly and non-disease natural pigments grows, various methods for extracting natural pigments have been studied. The natural color was extracted from parsley, a vegetable ingredient containing natural dyes. Target color codes of green series of natural dyes extracted as variables #50932C (L = 55.0, a = -40.0, b = 46.0) were set with the pH and temperature of extracted natural color coordinates (of the extracted), and the quantitative intensities of natural dyes were analyzed. During the colorimetric analysis predicted by the reaction surface analysis method, a color coordinate analysis was conducted under the optimal conditions of pH 8.0 and extraction temperature of $60.9^{\circ}C$. Under these conditions, predicted figures of L, a, and b were 55.0, -36.3, and 36.8, respectively, while actual experimental ones confirmed were 69.0, -35.9, and 31.4, respectively. In these results, the theory accuracy and actual error rate were confirmed to be 73.0 and 13.8%, respectively. The theoretical optimization condition of the color difference (${\Delta}E$) was at the pH of 9.2 and extraction temperature of $55.2^{\circ}C$. Under these conditions the predicted ${\Delta}E$ figure was 12.4 while the experimental one was 13.0. The difference in color analysis showed 97.5% of the theoretical accuracy and 4.5% of the actual error rate. However, the combination of color coordinates did not represent a desired target color, but rather close to the targeted color by means of an arithmetic mean. Therefore, it can be said that when the reaction surface analysis method was applied to the natural dye extraction process, the use of color coordinates as a response value can be a better method for optimizing the dye extraction process.

Antioxidant activity of hot-water extracts and floral waters from natural plant pigments (천연색소 함유 식물추출물과 플로럴 워터의 항산화 활성)

  • The antioxidant potentials of ten kinds of medicinal plants used as natural dyeing agents were tested. Among the plant extracts and floral waters that were examined in this study, the antioxidant activities of the DPPH and ABTS radical scavengers increased with increasing amounts of the extracts. The hot-water extracts from Aphis chinensis, Caesalpinia sappan L., Rumex crispus L., and Areca catechu effectively inhibited the DPPH and ABTS radicals at concentration below 0.1 mg/mL. The floral water obtained through steam distillation from Areca catechu, Rubia akane Nakai, Coptis chinensis, and Phellodendron amurense showed relatively valid antioxidant activities. In particular, the natural colorants extracted from Rumex crispus L., Areca catechu, and Aphis chinensis effectively suppressed the photogenerated singlet oxygen induced by a photosensitizer in in-vitro assay systems. The concentrations ($IC_{50}$) of the hot-water Aphis chinensis extract required to exert a 50% reduction effect on DPPH, ABTS, and singlet oxygen were found to be 8.5, 8.0, and $210{\mu}g/mL$, respectively. The study results suggest that extracts derived from naturally occurring colorants as well as floral waters could be used as natural antioxidants in the food and health-care industries.

천연 카로티노이드의 생물활성과 이용전망

  • 카로티노이드는 동식물계에 널리 분포하고 있는 황, 등, 적 혹은 자색의 대표적인 천연색소군으로서 자연계에서 약 1억톤/년 생산되고 있다. 이와 같이 막대한 량의 카로틴의 저장고는 식품의 잎, 동물플랑크톤, 조류 등이며 따라서 카로틴의 산업적 이용은 대단히 중요하다. 카로티노이드의 종류는 현재 약 600여종 정도 밝혀져 있으며 세계적으로 널리 이용되고 있는 것은 천연과 합성품을 포함하여 $\beta$-카로틴, $\beta$-아포카로테날, 칸타잔신, 아스타잔신 등과 그외 아나토추출물, 인삼유, 옥수수유, 파프리카, 사프란, 토마토추출물, 조류(algae) 등을 들 수 있으며 이들 카로티노이드 소재는 의약품, 식품, 사료등 다양한 용도로 사용되고 있다. 한편 $\beta$-카로틴, 아스타잔신 및 일부 카로티노이드 등은 30년간의 역학적 조사와 최근의 실증실험을 통해 암발생을 억제하는 물질로 규명되고 있으며 강력한 항산화성을 나타내고 있을 뿐만 아니라 동물의 번식 및 성장률 개선, 질병발생 억제, 어육의 색상개선 등 고차기능성을 나타내는 것으로 알려지고 있어 향후 다양한 분야에 이용가능성이 높아지고 있다. 본고에서는 최근 주목을 받고 있는 카로티노이드를 중심으로 생물 기능성 및 향후 이용전망에 대해 다루고자 하였다.

Dyeing of Silk Fabric with Aqueous Sorghum Extract (수수 추출물에 의한 견직물의 염색)

  • 본 연구에서는 대량으로 손쉽게 구 할 수 있는 식물염료를 확보하여 새로운 천연색소자원을 탐색하기 위해서 진행되었으며, 시료의 최적 추출 조건을 확립하고 추출된 색소의 실크에 대한 염색 특성을 살펴 최적 염색조건을 설정하고, 염색견뢰도를 평가하여 수수 등겨 추출물의 천연색소로의 활용에 대한 기초 자료를 마련하고자 하였다. 수수 색소 추출 시 생산성과 편리성을 고려하여 추출은 pH 4, 추출온도 $60^{\circ}C$, 추출시간 24시간이 적정조건으로 제시되었으며, 수수 추출물에 함유되어 있는 탄닌의 최대흡수파장은 282nm로 나타났다. 견직물의 pH에 따른 염색 특성은 pH가 증가할수록 K/S값이 증가하여 염착량이 증가하였으며, 염색온도가 높을수록 염색시간이 길어질수록 염착량이 증가하는 경향을 보여주고 있다, 염색온도가 높을수록 염색시간이 경과될수록 색상은 $Y{\rightarrow}YR$로 변화되는 것을 알 수 있었으며 염색온도가 높을수록 채도가 증가하였다. 욕비가 클수록 염착량은 감소하였으나, 채도는 약간 증가하는 것을 알 수 있었다. 매염제 처리에 의한 색상변화에서는 Al, Ni 매염은 색상변화는 적었지만, Cu와 Fe 매염은 명도와 채도를 크게 감소시키고 아울러 색상도 Cu는 적갈색으로 Fe는 진회색으로 변화되었다. Sn과 Ti 매염에 의해서 명도는 약간 감소하고 채도는 약간 상승하여 연한 주황색으로 색상이 변화하였다. 설정된 조건에서 염색된 염색포의 일광견뢰도는 2~3급, 세탁견뢰도 변퇴색은 2~3급, 오염은 4~5급으로 나타났다. Fe로 매염된 염색포의 일광견뢰도는 증가하여 3~4급으로 나타났으며, 매염제 처리에 의한 세탁 견뢰도는 같거나 약간 증가되는 것으로 나타났다.

  • 팥에 함유되어있는 색소와 관련된 연구 중에서는, 검정팥의 색소성분을 분리하여 delphinidin-3-glucoside(D-3-G)로 보고한 Sasanuma의 연구와 검정팥 껍질속에 포함된 안토시아닌의 함량을 평가한 Yoshida의 연구 결과 등이 있다. 본 연구에서는 검정팥 종피에서 색소를 추출하여, 적정 염색조건을 설정하였으며, 매염제별 색상변화 및 견뢰도를 측정하는 과정을 통해 염색 특성을 파악하여 새로운 천연염재로서의 활용 가능성을 진단하였을 뿐만 아니라 응용 연구를 위한 기초자료의 확보에 주안점을 두었다. pH에 따른 염색성에서는 견직물의 경우 pH 4에서 가장 염착량이 많았고, 면직물의 경우에는 주어진 범위내에서는 pH가 높을수록 염착량이 많았다. 염색온도와 시간에 따른 염색성은, 견직물의 경우, 염색온도 $80^{\circ}C$에서는 시간이 경과할수록 K/S 값이 크게 나타났으며, 면직물의 경우도 견직물과 비슷한 결과로 온도가 높아질수록 염색시간이 경과될수록 K/S 값이 크게 나타났다. 견직물에 대한 매염제 종류별 표면색의 변화에서는 Fe 매염포만 $YR{\rightarrow}Y$ 로 변화되었을 뿐 다른 매염제에서는 매염 후에도 색상 변화는 크지 않았다. 매염처리 전 견직물의 일광 견뢰도는 무매염이 4~6등급, Al 처리포는 4~5등급, Cu와 Sn은 3~4등급으로 나타났고 Fe는 2~3등급으로 가장 낮게 나타났으며, 세탁견뢰도에서는 무매염 2등급, 매염제 처리의 경우 2~3등급으로 나타났다. 무매염 면직물의 일광견뢰도는 1~2 등급, Fe 매염은 2~3등급, Cu 2등급 Al과 Sn은 1~2등급으로 매염처리에 의해 전혀 개선되지 않았으며, 세탁견뢰도의 경우 Cu매염이 4등급으로 양호하게 나타났으며 무매염, Al, Sn과 Fe는 3등급으로 나타났다.

  • In plants, color is a powerful tool to attract insects and herbivores which act as pollinators and vehicles of seed dispersion. In particular, flower color has held key post for human with aesthetic value. Horticultural industry has developed methods to produce new and attractive color varieties in flowering plants. Carotenoids are one of the main pigments being responsible for red, orange, and yellow colors. Their biosynthetic pathway has been considered as a major target of metabolic engineering for color modification of flowers and fruits. Here, we review the diverse efforts to modify pigment phenotype by the control of carotenogenic gene expression and enzyme levels. Recent reports about regulating degradation and storage of carotenoids will be also summarized to help the creation of engineered flower with novel color phenotype which is not existed in nature.

Dyeing of Treditional Fabrics with Natural Dyeing (전통직물의 천연염료 염색에 관한 연구)

  • The dyeing experiments were done by using tannin pigments and herbs for natural fabrics such as ramie, hemp and silk. The anti-bacterial ability and color fastness were examined for dyed fabrics. Dye materials containing tannin pigment such as oak tree leaf showed rich dyeing on ramie, hemp, and silk fabrics when the pH of dye solution is lower as well as the dyeing temperature is higher. In crimson dyeing, the colour of fabric showed red-purple when the crimson pigment was extracted at 40$^{\circ}C$, while yellow-red when extracted above 70$^{\circ}C$. The colour fastness of dye materials containing tannin pigment was excellent showing 3~4 grade or above, while most of those derived from chinese medicine showed poor colour fastness of below 3 grade. The anti-bacterial ability of dye materials derived from chinese medicine was high, since the sappan wood fabric showed low bacteria reduction rate.

Antimicrobial activities of Monascus koji extracts (식품유해균에 대한 홍국 추출물의 항균활성)

  • Currently, natural food colorants and preservatives are being used for their general health benefits. Monascus koji, the product of certain fungi that grow on rice grains, has been added to many foods for coloring and preservation. In this study, the antimicrobial activities of Monascus koji ethanol extracts were investigated. Six Monascus strains (M. araneosus KFRI 00371, M. kaoliang ATCC 46597, M. pilosus IFO 4520, M. purpureus IFO 4482, M. ruber IFO 32318 and M. sp. ATCC 16437) were selected based on their relative intensity of red pigment. Two Monascus extracts, M. kaoliang ATCC 46597 and M. purpureus IFO 4482, displayed antimicrobial activities against Bacillus subtilis, B. cereus, Micrococcus luteus, Staphylococcus aureus and Salmonella typhimurium in concentration-dependent manners. The two extracts showed their strongest antimicrobial activity against S. typhimurium, a cause of food poisoning. Therefore, these results suggest that Monascus koji could be used as a natural food colorant and preservative.

Skin lightening effect of fermented Panax ginseng extract (자연삼 발효 추출물의 미백 활성에 대한 연구)

  • Panax ginseng is known for various pharmacological activities mainly due to saponins. Since minor saponins, generated by the decomposition of major saponins, generally exert higher activities than major saponins, the fermentation may increase the minor saponin contents in ginseng products. In this study, we tested fermented ginseng extract whether or not provide a safe cosmetic ingredient for whitening purpose. In this regard, fermented Ginseng extract was prepared and evaluated the inhibitory activity toward tyrosinase and the melanin synthesis suppression. The safety was tested via cell viability and toxicity test. The skin lightening effect was also evaluated by clinical study. The fermented Ginseng extract exerted higher activities in tyrosine inhibition and in suppressing melanin synthesis compared to Kojic acid and arbutin. In the clinical test, skin lightening effecte of the sample was clearly higher than vehicle or Vitamin C. We thus concluded that the fermented Ginseng extract may provide a safe cosmetic ingredient for skin lightening purpose.
