Development of Vacuum Nozzle Seeder for Cucuribitaceous Seeds - Optimum Operating Conditions of Vacuum Nozzle Seeder - (박과 종자용 진공노즐식 파종기 개발 -진공노즐식 파종기의 최적 작동조건-)

  • 진공노즐방식의 파종기로 파종작업할 경우 박과채소의 대목으로 사용되고 있는 대립종자들의 최대파종율을 얻기위하여 참박, 특토좌, 흑종호박 종자에 대한 파종시험을 실시하였다. 1) 적정한 노즐구경과 홉착부압은 참박종자가 1.5mm, 7.97kPa, 특토좌종자가 1.5mm, 10.63kPa, 흑종호박종자가 2.0mm, 5.31kPa과 1.5mm, 13.28kPa로 나타났다. 2) 최적작동조건에서 참박종자, 특토좌종자 및 흑종호박 종자의 최대 파종율은 97.6%, 98.8% 및 97.6%로 나타났다. 3) 종자가 커지고 무거워짐에 따라 노즐의 구경이 큰 것이 유리한 것으로 나타났는데, 대목으로 사용되는 종자에서는 1.5mm노즐을 범용으로 사용가능하다는 결론을 얻었다. 4) 노즐개수가 많아지면 부착압력이 낮을 경우에는 파종율에 차이가 있었지만 부착압력이 높아지게 되면 파종율의 차이는 약간 낮아졌다. 5) 참박, 특토좌, 흑종호박 종자의 최대파종율을 나타내는 흡착부압 P = η.4W/$\pi$d$^2$로 나타낼 수 있고 여유계수 η ≒ 10으로 나타났다.

Effect of Low-Dose Gamma Radiation to Enhance Germination Rate in Bottle Gourd and Pumpkin Seeds (참박과 호박 종자의 발아촉진을 위한 저선량 감마선의 효과)

  • To establish the seed treatment method of the gamma radiation for germination enhancement of bottle gourd(Lagenaria siceraria Standl.) and pumpkin(Cucurbita ficifolia Bouche), seeds were irradiated with the dose of 1$\sim$20 Gy of the gamma radiation. The germination rate in irradiated group was higher than that of the control, depending on seed condition. In the case of bottle gourd, the germination rate of 70$\sim$80% germinative seed was increased at 4$\sim$20 Gy irradiated groups. Especially, it was the highest at 4 Gy irradiated group. The germination rate of 80$\sim$90% germinative seed was the highest at 4 Gy irradiated group. The germination rate of 70$\sim$80% and 80$\sim$90% germinative seed at 4 days after sowing was increased 66% and 26% at 4 Gy irradiated group, respectively. In the case of pumpkin, the germination rate of 70$\sim$80% and 80$\sim$90% germinative seed was increased at 2 Gy and 8 Gy irradiated group, respectively. Effect of the low-dose gamma radiation on the early growth was enhanced depending on their germination enhancement. Especially, it was the highest at 2 Gy irradiated group of 70$\sim$80% germinative seed of pumpkin. Catalase activity of bottle gourd was increased at 4 Gy irradiated group. Catalase and peroxidase activity of pumpkin were increased at 2 Gy and 8 Gy irradiated group. These results suggest that the germination, early growth and physiological activity of bottle gourd and pumpkin seeds could be promoted by the low dose gamma radiation.

  • To observe the stimulating effects of low dose ${\gamma}-radiation$ on the seed germination, early growth and physiological activity in the gourd, seeds of three cultivars (Partner, Support, FR Yongja) were irradiated at the dose of $0.5{\sim}30\;Gy$. The hormetic effects of the low dose ${\gamma}-radiation$ on the growth were different from each cultivar. Low dose ${\gamma}-radiation$ had promoting effects on the germination at the optimum dose of 2 Gy and 8 Gy and on the early growth at the optimum dose of 4 Gy, 16 Gy and 20 Gy in Partner and Support cultivar and 1 Gy in FR Yongja cultivar. Irradiation didn't have significant effects on the chlorophyll contents of gourd plantlet. The antioxidant enzyme activity of gourd plantlet and early growth increased in low dose irradiation group.

Plant Growth-Promoting Effects of Antagonistic Bacillus sp. YJ-3 against Fusarium Wilt of Watermelon-Rootstock Gourd (수박 만할병균에 길항하는 Bacillus sp. YJ-3에 의한 대목용 참박 생육촉진효과)

  • Twelve antagonistic strains against Fusarium wilt of watermelon-rootstock gourd were selected from 54 bacterial isolates which were isolated from the rhizosphere of crop plants growing in various locations. They showed strong inhibitory effects on growth of Fusarium osysporum f. sp. niveum, the causal agent of watermelon-rootstock gourd Fusarium wilt. Among these antagonists, the isolate YJ-3 was the most pronounced in growth-promoting ability for watermelon-rootstock gourd. The growth of watermelon-rootstock gourd in bed soil inoculated with YJ-3 was better by 46 and 13% than those in commercial bed soil alone and in bed soil inoculated with commercial microbial inoculant, respectively. The antagonistic plant growth-promoting rhizobacterium, strain No. YJ-3, was identified as Bacillus sp. on MIDI system. Furthermore, Bacillus sp. YJ-3 showed antifungal activity on growth against Alternaria cucumerina, Botrytis cinerea, Colletotrichum orbiculare, Didymella bryoniae, Rhizoctonia solani and Fusarium oxysporum.

Transformation of Gourd through Leaf Explant Regeneration (잎 절편의 재분화에 의한 참박 형질전환)

  • In order to develop a disease-resistant root stock for the growth of watermelon, an efficient regeneration system of the gourd(Lagenaria leucantha Duch.) inbred line GO701-2 via organogenesis was established in this experiment. Using proximal parts of cotyledon explant excised from germinated seedling in vitro, maximum adventitious shoot formation (39%) was achieved on MS medium where cytokinin (BA) and auxin (IAA) were added at a concentration of 3mg/L and 0.1mg/L, respectively. Roots of the elongated shoots were successfully formed on MS medium without adding any plant growth regulators. The cucumber CsGolS1 gene known as a resistance gene against biotic and abiotic stresses, was constructed into the binary vector pBI121 under the control of CaMV 35S promoter. When the gene was introduced into the genome of gourd by Agrobacterium-mediated transformation, putative transgenic plants were obtained with the transformation efficiency of approximately 20 percent.

Effects of Low Dose $\gamma$-Radiation on the Growth, Activities of Enzymes and Photosynthetic Activities of Gourd (Lagenaria siceraria) (저선량 $\gamma$선 조사가 참박의 초기 생육과 효소 활성 및 광합성 능에 미치는 영향)

  • Gourd seeds were irradiated with the doses of 0-20 Gy to investigate the effect of the low dose $\gamma$-radiation on the early growth and physiological activity. The stimulating effects of the low dose y- radiation on the early growth were not noticeably high, but were increased generally at 4-16 Gy irradiation group. The catalase and peroxidase activity of cotyledon from seeds irradiated with $\gamma$- radiation were increased at 8 Gy irradiation group. The photochemical activity of leaf was noticeably high at 4 Gy irradiation group. The photochemical yield of PSII, estimated as Fv/Fm, decreased with increasing illumination time by 50% after 4 hrs in the control and 8 Gy irradiation group, while Fo slightly increased. However, Fv/Em in the 4 Gy irradiation group decreased by 40% of inhibition, indicating that photoinhibition decreased by the low dose $\gamma$- radiation. Changes in the effective quantum yield of PSII, $\varphi_{PSII}$ and 1/Fo- l/Fm, a measure of the rate constant of excitation trapping by the PSII reaction center, showed similar pattern to Fv/Em. NPQ decreased by 70% after photoinhibitory treatment with showing similar pattern between the control and the irradiation group. These results showed the positive effect of low dose $\gamma$- radiation on the seedling growth and the reduction of photoinhibition in the 4 Gy irradiation group.

Effects of Low Dose Gamma Radiation and Seed Moisture Content on Germination and Early Growth of Vegetable Crops (저선량 방사선 조사 시 종자수분함량이 채소류의 발아와 초기생장에 미치는 영향)

  • To investigate the effects of low dose gamma radiation and seed moisture content (SMC) on germination and early growth of vegetable crops, seeds of chinese cabbage (Brassica campestris L.), radish (Raphanus sativus L.), red pepper (Capcicum annuum L.), figleaf gourd (Cucurbita ficifolia Bouche) and bottle gourd (Lagenaria siceraria Standl), with different SMC were irradiated with different doses ($0{\sim}20\;Gy$) of gamma-ray by irradiator ($^{60}Co$, ca.150 TBq of capacity, AECL). Vegetable crops in which low dose gamma radiation was irradiated in seeds with different moisture content showed different response in seed germination and early growth to low dose gamma radiation. The germination rate of chinese cabbage, figleaf ground and bottle gourd irradiated with $2{\sim}8\;Gy$ showed interactive responses against relative SMC. Also, significant interactions occurred for the early growth between those factors. The stimulating effects of gamma radiation were more pronounced for hydrated seeds of chinese cabbage, radish, figleaf gourd and bottle gourd showing prominent responses with $2{\sim}10\;Gy$ irradiation, particularly for chinese cabbage and bottle gourd. These results suggest that radiation may promote germination and early growth of vegetable crops through interaction with SMC.
