Group Sequential Tests Using both Type I and Type II Error Spending Rate Functions on Binomial Response (이산형 반응변수에서 오류 분배율 함수를 적용한 집단축차 검정)

  • In interim analysis, group sequential tests are widely used for the ethical, scientific, and economic reasons. In this paper, we propose the group sequential tests using both type I and type II error spending rate functions when the response variable is discrete, especially binomial distribution, in the interim analysis. In addition, we propose new error spending rate function which covers the formerly proposed. Our method has good property that is flexible, fast and easily applicable. A numerical simulations are carried out to evaluate our method and it shows good performance.

A nonparametric sequential test based on observations in groups (집단관측치에 의한 비모수적 축차검정에 관한 연구)

  • A new nonparametric sequential testing procedure is proposed in the paper. Sequential observations are divided into equally sized groups and a nonparametric statistic, which is appropriate for testing the given hypotheses, is obtained from each group. Then Wald's sequential test is applied for the case where the log probability ratio statistic is replaced by the nonparametric statistic. The properties of such test are evaluated approximately by the Wiener process.

  • A group sequential design can end a clinical trial early if a confirmed efficacy or a futility of study medication is found during clinical trials. Adaptation can adjust the design of clinical trials based on accumulated data. The key to this methodology is considered to control the overall type 1 error rate while maintaining the integrity of clinical trials. The estimation would be more complex and the sample size calculation will be more difficult if the clinical trials have repeated measurement data. Lee et al. (2002) suggested a repeated observation case by using the independent increments properties of the interim test statistics and investigated the properties of the proposed confidence interval based on the stage-wise ordering. This study extend Lee et al. (2002) to adaptive group sequential design. We suggest test statistics for the adaptation as redesigning the second stage of clinical trials and induce the stage-wise confidence interval of parameter of interests. The simulation will help to confirm the suggested method.

  • There are many clinical trials where a large portion of the patients are cured of disease. In such a case, one might be more interested in testing the differences in cure rates rather than other types of differences in failure distribution. For ethical and economic reasons, clinical trials must be repeatedly monitored for evidence of treatment benefit or harm. In this article, we examined by simulation the properties of nonparametric group sequential methods for comparing the cure rates between two treatment groups during the trial in a wide range of alternatives, censoring rates and cure rates.

  • For ethical and economic reasons, clinicla trials must be repeatedly monitored for evidence of treatment benefit or harm. Repeated testing at conventional critical values can substantially inflate the overal type I error rate. To maintain acceptable levels, group sequential procedures have been developed for clinical trials. This article gives a brief overview of the group sequential procedures.

Gender Differences of Knee Angle in Landing From a Drop-Jump: A Kinematic Analysis (수직착지시 성에 따른 슬관절의 형상학적(kinematic) 자료 분석)

  • 이 연구의 목적은 수직착지시 성에 따른 슬관절 가동범위의 차이를 알아보는 것이다. 연구 대상자는 20대의 건강한 성인 48명(남자 25명, 여자 23명)이었다. 연구 대상자에게 40 cm 높이에서 한발로 뛰어내리도록 하고, 2차원 동작측정 기구인 CMS-HS를 이용하여 수직착지시 슬관절의 굴곡각도, 최대 슬관절 굴곡각도, 슬관절의 가동 범위, 각속도를 측정한 후, 남 여 두 집단간의 차이를 알아보았다. 이때 사용한 분석방법은 독립적인 두 표본 t-검정이었으며 축차적(stepwise) 다변량회귀분석을 이용하여 체중과 신장을 조절한 상태에서 남여 간에 차이가 있는지를 분석하였다. 그 결과 수직착지시 슬관절의 굴곡각도에서는 유의한 차이를 보이지 않았으나, 최대 슬관절 굴곡각도, 슬관절의 가동 범위, 각속도에는 유의한 차이가 있었다(p<.05). 체중과 신장이 조절된 조건 하에서 여자는 수직착지시 최대 슬관절의 굴곡각도와 슬관절의 가동범위가 남자보다 작았다. 이러한 결과를 통하여 수직착지시 여자가 남자보다 슬관절 손상의 빈도가 높은 이유 중의 하나는 여자가 슬관절을 덜 굴곡시킴으로 인해 바닥의 충격을 더 많이 받기 때문이라는 가능성을 발견하였다.
