Distribution of Inorganic N from Fertigated and Broadcast-applied 15N-Urea along Drip Irrigation Domain (점적관수시 관비와 표면시비된 중질소 표지요소의 행동비교)

  • The objectives of this study were to measure the changes in soil moisture regimes and the distribution patterns of inorganic N derived from the fertigated $^{15}N$-labeled urea, and compare them with the results obtained from broadcast-applied soil under the same drip irrigation domain. In fertigated soil, a $^{15}N$-labeled urea solution of $117mg\;N\;L^{-1}$ was applied by surface drip irrigation for 4 weeks. In broadcast-applied soil, no the other hand, 4 g of $^{15}N$-labeled urea(1.87 g N) mixed thoroughly with 5 kg of soil was placed on the surface of packed soil. Soil water status was controlled by drip irrigation scheduled at soil matric potential of -50 kPa. A calibrated time-domain reflectometry probe was installed in the soil vertically 15 cm apart from a drip emitter to control drip irrigation. About 60% of urea-derived inorganic nitrogen was remained in the top zone between 0 and 10 cm depth of fertigated soil, while, most of the inorganic nitrogen (91%) was accumulated in the top zone of broadcast-applied soil. Of inorganic nitrogen derived from urea, the percentage of $NO_3{^-}$ was much higher for fertigation (99%) than for surface application (62%). The relatively lower recovery of urea-derived inorganic nitrogen of broadcast-applied urea-N (51%) than that of fertigated urea-N (89%) was attributable to enhanced $NH_3$ volatilization.

  • To investigate the effect of inorganic nutrient supplement in oyster mushroom (Pleurotus ostreatus), we have conducted some study on cultural and growth characteristics of fruitbody formation and chemical composition of media and fruitbody. When supplemented with slow releasing fertilizer, contamination rate was not different from non-supplemented medium, days for incubation time and first pinhead were faster than non supplemented medium. And fruitbody yield and biological efficiency were increased $10{\sim}28%,\;7{\sim}20%$ respectively, but biological efficiency was decreased when increased supplement ratio. The chemical compositions (total carbon, total nitrogen, ammonia nitrogen, nitrate nitrogen, potassium and phosphate) of slow releasing fertilizer supple mented medium and fruitbody were compared with non-supplemented.

Biological Perchlorate Reduction in Municipal Sewage (도시하수에서 생물학적 퍼클로레이트의 환원)

  • This research was done to evaluate the potential for destruction of perchlorate in municipal sewage. Laboratory experiments were conducted in flasks containing 3 liters of raw sewage. Sewage was mixed with defined amount of perchlorate and various additives. Perchlorate reduction in sewage did occur, but was quite variable, ranging from 0 to 72% over 72 hour. Addition of even a small amount of perchlorate acclimated biomass (167 mg/L SS) significantly reduced the lag and resulted in complete perchlorate removal. Perchlorate reduction in sewage-brine mixtures was inhibited when the dissolved oxygen level was greater than 2 mg/L, and when the mixture salinity was relatively high (conductivity = 14 mS with equivalent TDS = 8 g/L). When nitrate ($NO_3{^-}$) was present with perchlorate in the laboratory flask tests of sewage-brine mixtures, nitrate reduction proceeded first. A significant amount of nitrite ($NO_2{^-}$) accumulated in the sewage-brine mixtures, accounting for about 66% of initial nitrate nitrogen ($NO_3$-N).

Growth of an Obligatory Methanotroph Methylosinus trichosporium OB3b on Methanol (메탄자화균 Methylosinus trichosporium OB3b의 메탄올 기질에서의 성장)

  • An obligatory type II methanotroph Methylosinus trichosporium OB3b was cultivated on methanol as a sole carbon and energy source. The effects of methanol concentration, pH, temperature, nitrogen source and phosphate concentration on cell growth were investigated and the results were compared with the growth on methane, which had been studied previously. When $(NH_4)_2SO_4$ was used as a nitrogen source, the maximal specific growth rate (${\mu}max$) on methanol was $0.20hr^{-1}$ and the carbon conversion efficiency(CCE) was 43%. In comparison, on methane, ${\mu}max$ and CCE were $0.08hr^{-1}$ and 32%, respectively. Ammonia was found to be a better nitrogen source for methanol-growing cells. Cell yield on nitrogen (YX/N) was the same regardless of nitrogen source as 7.14g dry cells/g N, but the yield on methanol(YX/N) was higher with ammonia(0.8g dry cells/g MeOH) than with nitrate(0.64g dry cells/g MeOH). Optimal pH and temperature were 7.0 and $30^{\circ}C$, respectively. Methanol inhibition on cell growth was observed at above 0.5%(v/v). Inhibition by phosphate was observed at above 60mM, although the inhibition on methanol dehydrogenase activity started at a much lower level of 20mM. Based on the experimental findings, the cellular physiology of M. trichosporium OB3b growing on the two closely-related carbon sources were discussed extensively.

Nitrate Uptake by Soil Microorganism, Bacillus sp. GS2 (토양미생물 Bacillus sp. GS2에 의한 질산이온 흡수)

  • Over-application of nitrogen fertilizer keeps increasing the salinity in the soils of greenhouse in domestic agriculture. In order to remove the excess amounts of soil nitrate, soil microorganisms which have high capacity of nitrate uptake were isolated from the upland soils and their nitrate uptake activities were measured. Strain GS2 was able to remove 50 mM nitrate within 12 h. After sequence comparison analysis of 16S rRNA gene, the strain was identified and named as Bacillus sp. GS2. When the growth and nitrate uptake activities were measured, maximal values were obtained at $30-40^{\circ}C$ and $37^{\circ}C$, respectively; however, both were optimal at pH 6-8. In the media containing 50 mM nitrate, Bacillus sp. GS2 removed 43 mM nitrate which is corresponding to 86% removal. Similar amounts of nitrate removal were observed at the nitrate concentrations up to 300 mM, showing a saturation in nitrate uptake at concentrations above 50 mM. These results imply that Bacillus sp. GS2 can be a good candidate for the microbial remediation of accumulated environmental nitrate because of its excellent growth and nitrate uptake activity.

Nitrogen and Phosphorus Removal in Membrane Bio-Reactor (MBR) Using Simultaneous Nitrification and Denitrification (SND) (동시 질산화-탈질(SND) 반응을 적용한 MBR 반응조에서 질소 및 인 제거 특성)

  • Simultaneous nitrification and denitrification (SND) occurs concurrently in the same reactor under micro dissolved oxygen (DO) conditions. Anaerobic zone was applied for phosphorus release prior to an aerated membrane bio-reactor (MBR), and anoxic zone was installed by placing a baffle in the MBR for enhancing denitrification even in high DO concentration in the MBR. Phosphorus removal was tested by alum coagulation in the anaerobic reactor preceding to MBR. DO concentration were 2.0, 1.5, 1.0, 0.75 mg/L in the MBR at different operating stages for finding optimum DO concentration in MBR for nitrogen removal by SND. pH was maintained at 7.0~8.0 without addition of alkaline solution even with alum addition due to high alkalinity in the raw sewage. Both TCODcr and $NH_4^+$-N removal efficiency were over 90% at all DO concentration. TN removal efficiencies were 50, 51, 54, 66% at DO concentration of 2.0, 1.5, 1.0, 0.75 mg/L, respectively. At DO concentration of 0.75 mg/L with addition of alum, TN removal efficiency decreased to 54%. TP removal efficiency increased from 29% to 95% by adding alum to anaerobic reactor. The period of chemical backwashing of the membrane module increased from 15~20 days to 40~50 days after addition of alum.

Factors Regulating the Nitrogen Fixation Activity and Growth of Anabaena variabilis ATCC 29413 (Anabaena variabilis ATCC 29413 의 생장과 질소고정활성의 조절요인)

  • Anabaena variabilis A TCC 29413. a photoautotrophic and nitrogen fixing cyanobacteria. was investigated on the environmental factors regulating the growth and nitrogen lixation activity. A good growth of cyanobacteria] cells was observed due to nitrogen t1xation by the heterocyst differentiation in nitrogen free Allen and Arnon (]/8) medium. The nitrogenase activity was appeared to be in proportion to the cell growth lor 6 days then drastically decreased in the later growth period when the nitraTe was accumulated to high level in the culture to cause the inhibition. The optima] conditions lilr the cell growth and nitrogenase activity of A. varillbili.l were anaerobic. IO.OO0 lux. $30^{\circ}C$ and pH 8 with the nitrogen Cree minimal medium. The activity was significantly inhihited by the low concentrations of ammonium and nitrate. but was stimulated b) the ]ow Ieve] of phosphate and carbonate sources. The treatments of several toxic heavy metals showed strong inhibition of the cell growth and nitrogenase activity by O.3~10 ppm in the order of $Hg^{2+}$ > $Cd^{2+}$ > $Co^{2+}$ > $Zn^{2+}$ > $Ph^{2+}$, and the concentrations for 50% inhibition of the maximum activity were 0.41. 0.47. 0.5 L 0.66 and 8.1 ppm. respectively. The addition of carbohydrates (0.5~ 1.0%) in the dark condition stimulated the growth and activity in the order of sucrose > fructose > glucose.

  • 강변여과방식의 시험 취수가 진행 중에 있는 범람원지역의 산화-환원 반응을 밝히기 위해 10개의 간이 관측정을 설치하여 수질 모니터링을 수행하였다. 분석 결과 하천수에 인접한 지역에서 강한 환원환경이 생성되는 것을 확인하였으며, 이는 하천수 내 용존 유기 탄소가 인근 범람원 지역에 퇴적 산화되어 나타난 결과였다. 즉 퇴적된 용존 유기 탄소가 감소하면서 용존 산소와 질산성 질소는 감소시키지만, 이러한 환경변화가 수처리 시 문제가 될 수 있는 용존 망간의 농도를 증가시키게 되며, 따라서 향후 이 지역에 대한 추가적인 분석과 함께 집수정으로 통해 유출되는 용존 망간에 대한 대책이 필요한 것으로 조사되었다.

The Effect of Anaerobic Fermentation Treatment of Rice or Wheat bran on the Physical and Chemical property of Plastic Film House Soil (쌀겨와 밀기울의 토양 혐기발효 처리가 시설 재배지 토양의 물리 화학성에 미치는 영향)

  • This study was done to assess the physical and chemical properties after anaerobic fermentation treatment which use rice bran or wheat bran in plastic film house soil. The results which investigates the change of soil physical property after treatment 150 days showed a dramatic difference. The physical properties of control soil were the bulk density $1.46Mg\;m^{-3}$, hardness $2.30Kg\;cm^{-3}$, hydraulic conductivity $4.8cm\;hr^{-1}$, water stable aggregate(>0.5mm) 6.7%. Of the soil which treatment the rice bran in comparison to control soil, bulk density and hardness was diminished 12% and 58%, respectively. hydraulic conductivity and water stable aggregate(>0.5mm) were increased 4.5 and 5.2 fold, respectively. And, in the soil which treatment the wheat bran, bulk density and hardness was diminished 14% and 67%, respectively. Hydraulic conductivity and water stable aggregate(>0.5mm) were increased 6.3 and 6.5 fold, respectively. $NO_3-N$ contents of the soil which treated the rice bran or wheat bran after treatment 20 days were diminished 98% in comparison to control soil. The decrease of $NO_3-N$ contents in the soil was investigated with the fact that it is caused by with increase of the soil-microbial biomass. EC of the soil which treated the rice bran were $1.48dS\;m^{-1}$ which was diminished 58% in comparison to control soil. That of soil which treated the wheat bran was increased $3.65dS\;m^{-1}$ in the early stage because of acetic and butyric acid. But it was reduced as under $2.0dS\;m^{-1}$ after treatment 30 days. As the conclusion the anaerobic fermentation treatment with rice or wheat bran was effective to the improvement of soil physical and salt accumulation of the plastic film house soil.

  • These studies were carried out to investigate the effects of rice straw on microflora in relation to nitrogen metabolism in submerged soil. Rice plants were cultured in submerged soil to which rice straw was applied. In the submerged soil applied with rice straw the value of Eh lowered. pH was higher in the upper layer than in the lower. The content of iron(II) in submerged soil increased, while that of ammonium nitrogen decreased when rice straw was applied and nitrate-nitrogen was hardly detected during the rice cultivation period Under application of rice straw the number of denitrifying bacteria observed to increase at the early growing stage of rice plant and to decrease thereafter, and that of nitrate reducing bacteria increased at the late growing stage. The number of ammonium oxidizing bacteria and that of nitrite oxidizing bacteria decreased continually but the latter were rather sharply decreased.
