Effect of Eriobotrya folium on Local Fat via Regulation of Lipase Secretion (비파엽추출물의 지방분해효소 조절을 통한 국소 지방분해 효능)

  • Objectives: In this study, the lipolytic effects of Eriobotrya folium extract (EFE) on local fat was investigated in high fat diet (HFD)-induced obesity mouse and 3T3-L1 adipocytes. Methods: C57BL/6J mice (5 weeks) were fed HFD for 6 weeks to induce obesity. EFE (20 mg/ml, $100{\mu}l$) or saline ($100{\mu}l$) as a normal control was injected into left inguinal fat pad region, 3 times per a week for last 2 weeks. After sacrifice, body weight, and histological changes of the inguinal fat pad were evaluated. The expressions of hormone-sensitive lipase (HSL) and adipose triglyceride lipase (ATGL) in inguinal fat pad were analyzed by Western blotting. Also, lipid accumulation and lipases release were determined in 3T3-L1 adipocytes by oil red o staining. Results: EFE significantly reduced the weight of inguinal fat pad and the size of adipocytes in HFD-induced obesity mice compared to control. The treatment of EFE up-regulated the expressions of HSL and ATGL in inguinal fat pads of obesity mice, as well as 3T3-L1 adipocytes. In addition, EFE inhibited the lipid accumulation in 3T3-L1 adipocytes in a dose dependent manner. Conclusions: EFE showed lipolytic effect on local fat of HFD-induced obesity mice by up-regulation of the lipases secretion. This suggests that EFE could be considered as anti-obese substance with lipolytic property on local fat.

Isolation and Identification of Microorganisms Producing the Soy Protein-Hydrolyzing Enzyme from Traditional Mejus (전통메주로부터 대두단백질 가수분해효소 생산성 미생물의 분리 및 동정)

  • In order to develop the enzymatic hydrolysis system concerned with taste and flavor, strains having the high hydrolyzing activity on the soy protein were selected from some traditional Mejus. Two molds and one bacterium producing enzymes which were different in character of hydrolysis were isolated and identified. Leucine and azodye enzyme activities of both M4 and M5 were relatively high among in the isolated molds. And, leucine enzyme activity of B16 was the lowest in the isolated bacteria. These strains were isolated as microorganisms having a dissimilar hydrolysis pattern on the soy protein by enzymatic reactions. Mold M4 on the culture solid media was mycelium colors of white and its sclerotia colors were changed from white to black. According to the result of slide culture, radial conidial head, subclavate vesicle, conidia of subglobose, stipes of uncolored with smooth walls and metula and phialides were existed. Because M4 was taxonomically similar to the characteristics of Aspergillus oryzae (ahlburg) species, M4 was identified and named as Aspergillus oryzae M4.Mold M5 showed white and black mycelium on the MEA medium. Mold M5 colony exhibited grayish-green color and have long(7 mm) sporangiophores at slide culture. Sporangia became brownish-gray and the wall of larger sporangia was broken to form small collars, and smaller sporangia were fomed continually from large basal membrane. Columella is globose and hyaline, and sporangiospores are ellipsoidal of small diameter$(80\;{\mu}m)$. Because M5 was taxonomically similar to the Mucor circinelloides of zygomycetes, M5 was was identified and named as Mucor circinelloides M5. Bacteria B16 colony was opaque white, circular and lobate, and had rod shaped endospore. B16 was found positive in stain, catalase, ${\beta}-glucosidse$ and V-P tests. B16 was found to utilize D-fructose, ${\alpha}-D-glucose$, maltose, D-mannose, D-raffinose, stachyose and sucrose. By the morphological and physiological results, the characteristics of B16 was thought to correspond to that of Bacillus megaterium. However, fatty acid composition was similar to Paenibacillus marcerans, requiring further study for the definite identification. Accordingly, Bacteria B16 was provisionally classified and named as Bacillus megaterium B16.

The Effects of Different Type of Triglyceride Supplements on Exercise Performance Time, Energy Substrates, Insulin Hormone and Lipase Activity in the Trained Rats (서로 다른 형태의 지방산 투여가 훈련된 흰쥐의 지구성 운동수행력, 안정시기와 운동스트레스 시기의 에너지 기질, Insulin 호르몬과 Lipase 활성에 미치는 영향)

  • The purpose of this study was to investigate the effects of different type of triglycerides (MCT & LCT) on weight, survival time, energy substrate (FFA, TG, pyruvate, lactate), insulin and lipase in the trained rats. Fifty-four Sprague-Dawley rats were divided into 3 groups: control group (CG, n=18), MCT supplement group (MG, n=18), and LCT supplement group (LG, n=18). They also were divided into 3 periods: trained resting (R, n=6) and trained & exercise load (E, n=6), and survival time test was performed to know the supplemented effects. Body weight of all animals was checked every week, MCT group and LCT group received supplementary MCT and LCT orally and preliminary swimming training for 6 days before the start of main experiment. All animals received 15-minute swimming training 5 times during first week of experiment, and swimming training time was increased 15 minutes every week until it reached 90 minutes at last 9th week. After last swimming training, animals were fasted for 12 hours and blood samples were taken from abdominal aorta in the Department of Animal Medicine at the D university Animal Center. Among the CGE, MGE, and LGE groups, the MGE had the greatest increase in physical performance time. In the FFA levels, there was significant differences(p<.05) in CG, MG and LG groups, and also there was major difference of FFA levels in the MG and LG. In the lipase levels, there was signifi.ant differences (p<.05) in CG, MG and LG groups. MG was the greatest than the other groups. In the insulin hormone levels, there was the great differences (p<.05) in LG compare to CG groups, whereas there was no significant difference in the CG and MG. In conclusion, these results suggest that regular prolonged physical training with MCT supplementation, improves exercise performance time through the increase of energy substrate utilization, lipase activity and FFA levels, irrespective of insulin hormone responses.

  • In order to develop the squid(Todarodes pacificus) sik-hae, the changes of TBA, fatty acids, free amino acids, and the number of microflora fermented at different fermentation temperatures and periods were determined. In addition, pretense from squid sik-hae was partially purified. The number of TBA was the highest after 5-day storage and decreased after that, and lipid oxidation was the highest at $10^{\circ}C$. The amounts of linoleic aid(18:2) and oleic acid (18:1) were about $60\%$ of fatty acid composition of squid sik-hae, and linolenic acid(18:3) and EPA(20:5) significantly decomposed with increasing fermentation periods and temperatures. Pro, His, Aeg, Leu, and Glu were composed mainly of amino acid and the composition ratios of Ser, His, and Arg decreased with increasing fermentation periods whereas, those of Glu, Ala, Val, and Tyr increased. The composition ratios of Glu, Val, and Met increased with increasing fermentation temperatures whereas, those of Ala, Cys, Thr, and Gly decreased. The number of microflora generally increased up to 15-days of storage and decreased after that. The rates of increass and decreass of the microbial number increased in proportion to fermentation temperatures. In addition, the bacteria producing proteases were identified as Bacillus spp. Proteases from $60{\sim}80\%$ ammonium sulfate concentration showed the highest activity and had about 15 binds with molecular weights between 20,000 and 40,000 Dalton by the SDS-PAGE.

Effect of Cellular Phospholipase A2 Inhibition on Enhancement of Bt Insecticidal Activity (세포성 인지질분해효소 활성 억제에 따른 비티 살충력 증가 효과)

  • Some bacterial metabolites of Xenorhabdus nematophila (Xn) inhibit phospholipase $A_2$ ($PLA_2$) activity to shutdown eicosanoid biosynthesis in target insects. However, little has been known about the target insect $PLA_2$ of these bacterial metabolites. Eight bacterial metabolites identified in Xn culture broth exhibited significant insecticidal activities against larvae of both lepidopteran species of Plutella xylostella and Spodoptera exigua. Moreover, these bacterial metabolites significantly enhanced insecticidal activities of Bacillus thuringiensis (Bt). To determine target $PLA_2$, we cloned and over-expressed cellular $PLA_2$ ($SecPLA_2$) of S. exigua. Purified $SecPLA_2$ catalyzed phospholipids derived from the fat body and released several polyunsaturated fatty acids. Most Xn metabolites significantly inhibited $SecPLA_2$ activity, but were different in their inhibitory activities. There was a positive correlation between the inhibition of $SecPLA_2$ and the enhancement of Bt insecticidal activity. These results indicate that $SecPLA_2$ is a molecular target inhibited by Xn metabolite.

Production and Characteristics of Protein Hydrolysate from Sharp Toothed Eel (Muraenesox cinereus) (갯장어 효소 가수분해물의 제조 및 특성)

  • Protein hydrolysate was prepared as a natural flavor stock from the sharp toothed eel (Muraenesox cinereus) mince using com-mercially available proteolytic enzymes, Alcalase, Neutrase, Protamex, and Flavourzyme. A 6 hr hydrolysis of mince, to which water of the equal weight of the mince was added, with $2\%$ (w/w, protein weight) Elavourzyme at $50^{\circ}$ yielded a hydrolysate of the highest acceptability. Removing the access lipid in liquified hydrolysate (not dehydrated) after enzyme hydrolysis, five times repetitive extraction using n-hexane (liquified hydrolysate : n-hexane=4 : 1, v/v) was effective, resulting in less than $1\%$ lipid content of the dehydrated-hydrolysate. The amino acid composition of the hydrolysate (prepared with Flavourzyme) was similar to that of the starting material. Hydrolysis led to an increase in concentration of not only total free amino acid but also free amino acid such as serine, glutamic acid, alanine, and methionine responsible for umami taste, especially up to about 40 times for methionine. Major free amino acids in amount were leucine, phenylalanine, valine, alanine, and isoleucine and they comprised about half of the total free amino acids, Moisture adsorption, fat adsorption, emulsifying capacity, and foaming capacity of the hydrolysate were 870.1 $\pm$ $7.9\%$, 352.0$\pm$ $5.3\%$, 50.3 $\pm$ $1.2\%$, and $87.5\pm$ $2.5\%$, respectively, and solubility was 83$\~$$84\%$ at acid pH range of 2$\~$4.

Changes of Some Physicochemical Properties of Yoghurt made from ${\beta}$-Galactosidase-treated Commercial Milks (${\beta}$-Galactosidase 처리 시유로 제조한 요구르트의 이화학적 성분 변화)

  • This study was carried out to hydrolyze lactose in commercial milk by ${\beta}-galactosidase$ from Kluyveromyces fragilis and to compare some physicochemical properties of yoghurts made from control and lactase-treated commercial milks. Quantitative analysis of sugars was performed by gas liquid chromatograph (GLC) on trimethylsilyl (TMS) derivatives. In commercial milk, 94.6% of lactose was hydrolyzed after 2 hours incubation at $40^{\circ}C$ with 6.0 units/ml of ${\beta}-galactosidase$. pH, titratable acidity and viable cell number of yoghurt made from lactase-hydrolyzed (LH) commercial milk were 4.1, 1.04% and $6.5{\times}10^8/ml$ of Str. thermophilus, $8.9{\times}10^8/ml$ of L. bulgaricus after 8 hours incubation at $40^{\circ}C$, respectively, The total contents of amino acid were 2.63% in control and 2.19%. in LH yoghurt. The total contents of free amino acid were 26.95 mg% in control and 17.55mg% in LH yoghurt. Analysis of free fatty acids resulted in that the contents of short chain fatty acids in LH yoghurt were a little higher than those in control. Both in control and LH yoghurt, the palmitic acid content was highest and that was followed by oleic and myristic acid.

Effects of Feral Haw (Crataegus pinnatifida Bunge) on Hepatic Functional Enzyme Activity in Dyslipidemic Rats (야생 산사(Crataegus pinnatifida Bunge)가 이상지질혈증 흰쥐의 간 기능 대사효소의 활성 변화에 미치는 영향)

  • Haw (Crataegus pinnatifida Bunge) has been used in traditional medicine for treating various ailments such as obesity and digestive trouble in Far East countries, including Korea. The present study was carried out to investigate the effects of feral haw on hepatic functional enzymes in dyslipidemic rats. Four groups of male rats (Sprague Dawley strain) were fed different diets for 5 weeks: NND (normal-nondyslipidemic diet) group, NNDH (normal-nondyslipidemic diet + haw extract) group, CDD (control-dyslipidemic diet) group and DDH (dyslipidemic diet + haw extract) group. ALP (alkaline phosphatase), LDH (lactate dehydrogenase), AST (aspartate aminotransferase) and ALT (alanine aminotransferase) activities were significantly higher in the CDD group than the NND group. However, haw extract supplement significantly reduced hepatic functional enzyme activities compared to the CDD group. Lipid deposition of the DDH group decreased compared to the CDD group. The size of adipose tissue decreased significantly in the DDH group compared to the CDD group. These results suggest that feral haw could be used as a food resource and functional food material.

Effects of ethanol extract of Polygonatum sibiricum rhizome on obesity-related genes (황정 에탄올 추출물의 비만 조절 유전자에 대한 효과)

  • In previous studies, we confirmed that the ethanol extract of Polygonatum sibiricum (ID1216) has anti-obesity effects on high-fat diet-fed mice. To identify the obesity-related genes affected by ID1216, we studied its effects both in vivo and in vitro. In mice, single administration of ID1216 increased the expression of obesity-related genes including sirtuin1 (SIRT1), peroxisome proliferator-activated receptor ${\gamma}$ coactivator $1{\alpha}$ ($PGC1{\alpha}$) and peroxisome proliferator-activated receptor ${\alpha}$ ($PPAR{\alpha}$) compared to that in mice administered the vehicle; their downstream genes (uncoupling proteins, acyl-CoA oxidase, adipocyte protein 2, and hormone-sensitive lipase) were also increased by ID1216. In fully differentiated 3T3-L1 adipocytes, ID1216 showed the same effects on anti-obesity genes as those in the animal model. Based on these results, we propose that ID1216 has anti-obesity effects by regulating the $SIRT1-PGC1{\alpha}-PPAR{\alpha}$ pathway and their downstream genes, thereby controlling energy and lipid metabolisms.

Characterization of Symbiotic Bacteria from Entomopathogenic Nematode (곤충병원성 선충로부터 분리된 공생박테리아의 종별 특성)

  • Symbiotic bacteria with highly effective insecticidal activities were isolated and compared with their physiological characteristics from seven species of entomopathogenic nematodes belong to Steinernamatidae and Heterorhabditidae sp., and three of them were identified as Xenorhabdus nematophilus. Culture characteristics, insecticidal activities, pretense activities and fatty acid contents of various symbiotic bactierial isolates were also examined. In the case of cell growth and insecticidal activity, XR-PC and XR-MK were superior to other species when cultured in vitro. The insecticidal activity were highest at the early exponential growth phase, and gradually decreased with time. The protease activity of XR-DR was remarkable compared to other species. In the case of HE-HY, however the pretense activity increased in parallel with cell growth. Interestingly, the fatty acid patterns of Xenorhabdus nematophilus isolated from different emtomopathogenic nematode, showed remarkable differences in their contents of 12:0, 14:0, 16:1 cia 5 and 17:0 cyclo and hydroxy and branch factty acids were varied from 2% to 15% among total fatty acid contents.
