Purification and Characterization of Polyphenol Oxidase in Sweet Potato (Ipomoea batatas) (고구마 Polyphenol Oxidase의 정제 및 특성)

  • The present work was undertaken to investigated the purification and characterization of polyphenol oxidase (PPO ; EC 1.10.3.1) in sweet potato, particularly the number of PPO isozymes, and PPO properties such as pH optimum, heat stability, substrate specificity, kinetics, and inhibitor studies. The purification achieved was 23.1 fold from crude extract with a yield of 41.5%. Eight PPO isozymes and twelve PPO isozymes were detected by disc polyacrylamide gel electrophoresis and isoelectric focusing, respectively. The specific activity of each isozyme separated by isoelectric focusing was in the range of $6,000{\sim}46,700U/mg$. This enzyme was sweet below $65^{\circ}C$ and the pH optimum of PPO occurred at 6.0-6.5. The substrate specificity of sweet potato PPO showed the high affinity toward the odiphenolic compounds. Km and Vmax for catechol were found to be 6.7 mM and $20{\triangle}A/min$, me protein, respectively. Inhibitor studies indicated that dithiothreitol was the most potent among the inhibitors used in the present work.

Sorption Characteristics of Uranium on Goethite and Montmorillonite under Biogeochemical Reducing Conditions (생지화학적 환원조건에서 우라늄의 침철석 및 몬모릴로나이트에 대한 수착 특성)

  • Two kinds of uranium species, oxidized uranium(VI) and reduced uranium(IV), were prepared to be interacted with goethite and montmorillonite to identify sorption characteristic of uranium species, which are very sensitive to the redox-reaction. The reduced uranium was prepared by diluting a substantial uranium(IV) that was concomitantly produced during a sulfate reduction via a sulfate-reducing bacterium. The sorption amount of uranium(IV) by the minerals was relatively lower than that of uranium(VI) because the aqueous uranium(IV) had fine colloidal forms to cause its weak adsorption onto the mineral surfaces. We found that the uranium(IV) phase has a nano-colloid character by the transmission electron microscope, suggesting that the uranium species possibly migrating with the flow of groundwater in underground environments can be the colloidal uranium(IV) as well as the ionic uranium(VI).

SIFT based Image Similarity Search using an Edge Image Pyramid and an Interesting Region Detection (윤곽선 이미지 피라미드와 관심영역 검출을 이용한 SIFT 기반 이미지 유사성 검색)

  • SIFT is popularly used in computer vision application such as object recognition, motion tracking, and 3D reconstruction among various shape descriptors. However, it is not easy to apply SIFT into the image similarity search as it is since it uses many high dimensional keypoint vectors. In this paper, we present a SIFT based image similarity search method using an edge image pyramid and an interesting region detection. The proposed method extracts keypoints, which is invariant to contrast, scale, and rotation of image, by using the edge image pyramid and removes many unnecessary keypoints from the image by using the hough transform. The proposed hough transform can detect objects of ellipse type so that it can be used to find interesting regions. Experimental results demonstrate that the retrieval performance of the proposed method is about 20% better than that of traditional SIFT in average recall.

Changes of Rutin and Quercetin in Commercial Gochujang Prepared with Buckwheat Flour during Fermentation (메밀가루 첨가 공장산 고추장의 발효과정 중 Rutin과 Quercetin의 변화)

  • Changes in rutin and Quercetin contents of Gochujang, which was added buckwheat flour, were investigated during steaming and fermentation for 1 month. Not steamed buckwheat flour showed only in presence of rutin. But rutin decreased and quercetin increased after steaming. The koji of buckwheat flour showed maximum contents of rutin and Quercetin at 20 hours during fermentation. The rutin content of Gochujang made with koji of buckwheat flour decreased slightly during fermentation but the Quercetin content increased with decreasing contents of rutin, probably due to degradation of rutin. All rutin degraded completely in Gochujang made with koji of buckwheat flour after 20 days.

Determination of Flavonoid and Limonoid Compounds in Citron (Citrus junos Sieb. et Tanaka) Seeds by HPLC and HPLC/MS (HPLC 및 HPLC/MS를 이용한 유자 씨앗 중 플라보노이드 및 리모노이드 화합물의 분석)

  • Flavonoid and limonoid compounds were determined by HPLC on the methanol and ethanol extracts from citron seeds. The quantities of the compounds in these categories were higher in the ethanol extract than methanol extract. The types of these compounds were detected in larger numbers in the ethanol extract. The content of limonin was the largest in both methanol and ethanol extract among the detectable compounds ; 140.34 mg/100g and 170.98 mg/100g, respectively, and the contents of other compounds, caffeic acid, naringin, lutin, nomilin, were found in large amount in this order. The molecular weights of forty two compounds in ethanol extract were determined with mass spectrums and extracted ion current chromatograms by HPLC/MS.

Maillard Reaction Products Formed from D-Glucose-Glycine, System and Their Formation Mechanism (D-Glucose-Glycine 계의 Maillard 반응생성물 및 그 생성기구)

  • Equimolar aqueous solutions of D-glucose and glycine were heated at $50^{\circ}C\;and\;95^{\circ}C$ at pH 6.7. The headspace volatiles and the ether extracts from the reaction mixture were analyzed by gas chromatography and gas chromatography-mass spectrometry using a fused silica capillary column. The major components formed were identified as diacetyl, three furfurals, two pyrroles, one furanone, two pyranones and two amides. In order to elucidate the formation mechanisms of the amides formed front amino-carbonyl reaction, two model systems were adopted. N-butylacetamide were formed as major components from diacetyl-butylamine ana glyoxal-butylamine systems, respectively. The results obtained suggest that such ${\alpha}-dicarbonyls$ as 3-deoxy-D-erythro-2,3-hexodiulose and diacetyl generated in the amino-carbonyl reaction react with amino compounds, amides then being formed by cleavage of the C-C bond in the ${\alpha}-dicarbonyls$.

Quality Control on Neutron Activation Analysis for Urban Dust by the Proficiency Test (비교숙련도 시험을 통한 도시대기분진에 대한 중성자방사화분석법의 품질관리)

  • Trace elements in two air filters (V-50, P-50) artificially loaded with urban dust provided from IAEA were determined non-destructively using instrumental neutron activation analysis. Standard reference material (Urban Particulate Matter, SRM 1648) of National Institute of Standard and Technology was used for analytical quality control. About 20 elements in both of loaded filter samples were determined. To evaluate inter-laboratory comparison and proficiency test, analytical data were statistically treated with the results which collected from 49 laboratories, 40 countries participated in this study using neutron activation analysis, particle induced X-ray emission, inductively coupled plasma mass spectroscopy, etc,. From the results of statistical treatment, Z-scores are within ${\pm}2$. Furthermore, accuracy and precision of obtained analytical values are passed according to the criteria of the proficiency test. Consequently, it was proved that analytical quality for air dust samples being performed has been controlled properly.

Three Dimensional Numerical Analysis on Rock Cutting Behavior of Disc Cutter Using Particle Flow Code (3차원 입자결합모델을 이용한 디스크 커터의 암석절삭에 관한 연구)

  • The LCM (Linear Cutting Machine) test is one of the most powerful and reliable methods for designing the disc cutter and for predicting the TBM (Tunnel Boring Machine) performance. It has an advantage to predict the actual load on disc cutter from the laboratory test on the real-size large rock samples, however, it also has a disadvantage to transport and/or prepare the large rock samples and to need an extra cost for experiment. In order to overcome this problem, lots of numerical studies have been performed. In this study, the PFC3D (Particle Flow Code in 3 Dimension) has been adopted for numerical analysis on optimum cutter spacing and failure aspects of Busan Tuff. The optimum cutting condition with s/p ratio of 16 and minimum specific energy of $14MJ/m^3$ was derived from numerical analyses. The cutter spacing for Busan Tuff had the good agreements with those of LCM test and numerical analysis by finite element method.

Changes of the Soil Physic-Chemical Properties and Rice Productions with Methods Applied Organic Materials in Organic Culture (벼 유기재배에 있어서 유기자재 시비방법에 따른 토양 및 수량 특성 변화)

  • 벼 유기재배에 있어서 녹비작물을 이용하여 화학비료를 대신하고 있으나 녹비를 이용하기 위해서는 월동 전에 파종하고 이듬해 벼 이앙 전에 토양에 환원을 해야 하는 번거로움이 있다. 따라서 벼 재배 직전에 유기자재를 이용하여 화학비료를 대신하고자 했을 경우 유기자재를 전층시비와 표층시비의 차이에 따른 토양중의 이화학적 특성과 벼의 수량특성의 변화를 구명하였다. 벼 유기재배시 토양양분공급용으로 이용되고 있는 유기자재 4종을 공시하여 유기자재의 질소 성분량(7kg/10a)을 기준으로 하여 이앙 20일전에 시비방법별로 전층시비와 표층시비 2처리로 구분하여 전량 기비시비하고 경운한 다음 동진1호를 시험품종으로 하여 2년 연속 시비처리와 벼를 재배하면서 일어나는 토양의 이화학적 특성과 벼 생육 및 특성의 변화를 시기별로 조사하였다. 시험 전 토양의 화학성은 표층시비구의 염류농도, 가리와 석회의 함량이 다소 높아서 염류농도가 전층 시비구 보다 높은 조건의 토양이었다. 유기자재별 무기화 정도는 전층시비보다 표층시비를 할 때 약 20~30일 정도 빨랐다. 토양 중의유기물 잔존함량은 시비방법간의 큰 차이는 없었으나 표층시비를 할 경우 후기로 갈수록 다소 증가되는 경향이었으나, 전질소 잔존함량은 감소되었다. 토양 액상과 공극율은 전층시비>표층시비였으며, 입단 형성력도 같은 경향이었다. 토양 효소활성은 PME의 활성은 유기자재를 전층처리하였을 때 촉진되었으며, $\beta$-Glucosidase의 활성은 전층보다 표층처리시 활성이 높았다. 시비방법에 따른 벼의 수량 특성은 시비방법별로는 표층시비를 할 경우 전층시비보다 4~7%의 높은 특성을 보였으며, 관행대비 1년차에는 3~9%의 낮았으나, 2년 연속처리를 할 경우 대조구와 비슷해 지는 경향이었다.

온도 stress에 따른 ZTO TFT의 특성 변화

  • 최근 연구와 생산에 가속이 붙기 시작한 AMOLED는 모두 LTPS TFT를 사용하고 있다. LTPS TFT는 높은 전자 이동도를 가지고 있기 때문에 현재 각광 받는 AMOLED에 잘 맞는다. 하지만 LTPS TFT는 균일성이 낮고 고비용이라는 문제점이 있으며, 현재 대면적 기술이 부족한 상태이다. 극복방안으로 AMOLED를 타겟으로 하는 Oxide TFT와 a-Si TFT의 기술이 발전되고 있다. Oxide TFT는 AMOLED backplane으로 사용될 수 있는 강력한 후보 중의 하나이다. Oxide TFT는 단결정 산화물과 다결정 복합 산화물 두 가지 범주를 가지고 있다. 본 연구에서는 다결정 Oxide TFT의 하나인 ZTO TFT를 연구함으로서 Engineer의 근본적 이슈인 저비용에 초점을 맞추어 소자특성을 확인해보도록 한다. n-type wafer 에 PE-CVD 장비를 이용하여 SiNx를 120 nm 증착하고, channel layer인 ZTO 용액을 spin-coating을 이용하여 형성하였다. 균일하게 형성된 ZTO의 결정을 위하여 $500^{\circ}C$에서 1시간 동안 공기 중에서 annealing을 하였다. 과정을 거친 ZTO는 약 30 nm 두께로 형성되었다. Thermal evaporator를 이용하여 Source, Drain의 전극을 형성 하고, wafer 뒷면에는 Silver paste를 이용하여 Gate를 형성하였다. 제작된 소자를 dark room temperature 에서 측정 하였다. 측정된 소자는 우수한 전기적 특성과 0.96 cm2/Vs 인 이동도를 얻어냈다. 이러한 소자의 안정성에 따른 전기적 특성을 관측하기 위하여 상온에서 $100^{\circ}C$ 까지의 온도 스트레스를 주었다. Stress에 따른 소자는 상온에서 시작하여 온도가 올라갈수록 이동도가 낮아지고, 문턱전압 증가와 SS이 커짐을 알 수 있었다. 캐리어의 운동 매커니즘에서 온도가 올라가면 격자진동의 영향을 크게 받음으로서 캐리어의 이동도가 낮아져 전기적 특성이 낮아지는 점이 본 연구에도 적용됨을 알 수 있었다. 본 연구를 통하여 화학적 안정성을 지닌 소자라는 점과 더불어 여타 TFT공정에 비하여 현저히 낮은 공정비용을 통하여 AMOLED가 요구하는 수준의 특성에 가까운 소자를 제작할 수 있다는 것을 확인하였으며 앞으로의 추가적인 연구에 따라서 더욱 완성된 공정기술을 기대할 수 있었다.
