Friedewald-Estimated Versus Directly Measured LDL-Cholesterol: KNHANES 2009-2010 (LDL-콜레스테롤의 Friedewald 계산값과 실측값 비교: 국민건강영양조사 2009-2010)

  • Low-density lipoprotein cholesterol (LDL-C) is a major modifiable risk factor for cardio- cerebrovascular disease. In clinical practice, however, it is primarily calculated using the Friedewald formula as a cost-effective method. The aim of this study was to compare Friedewald-estimated and directly measured LDL-C values and assess the concordance in guideline LDL-C risk classification between the two methods. The data were derived from the 2009 and 2010 Korea National Health and Nutrition Survey (KNHANES). Analysis was done for 4,319 subjects with lipid panels-total cholesterol (TC), high-density lipoprotein cholesterol (HDL-C), directly measured LDL-C using an enzymatic homogeneous assay, and triglycerides (TG). For subjects with TG lower than 400 mg/dL, Friedewald-estimated and directly measured LDL-C were highly correlated (r = 0.958, p < 0.001) and overall concordance was 82.7%. As TG increased, overall concordance decreased. Overall concordance was 85.4% at TG lower than 150 mg/dL; 78.2% at TG of 150-199 mg/dL; and 71.4% at TG of 200-399 mg/dL. The Friedewld equation tended to overestimate LDL-C when TG are of < 150 mg/dL; however, underestimate LDL-C when TG are of ${\geq}150mg/dL$. As a result, Friedewald estimation misclassified 382 subjects (9.1%) in a higher category versus 348 subjects (8.3%) in a lower category. Our findings suggest that overestimation of LDL-C by the Friedewald formula can be a great problem as well as underestimation.

Fatty acids of neutral lipids of salted-dried mullet roe during processing and storages (숭어알의 가공과 저장중 중성지질의 지방산함량변화)

  • The salted-dried mullet (Mugil japonicus) roe is a kind of traditional food particulary in the area of Young-am gun, Chunnam province. The manufactruring method was that the fresh roe was salted by clean wasters, drained, shaped a flat piece with 1.2cm thickness by pressing, and spreaded sesame oils on the surface of the salted roe periodically during wind drying for 20 days. The dried roe was blanched in heated water$(80^{\circ}C/3min.)$ and packaged the dried product for the cooling storages. The fractional compositions of free lipid were 40% of neutral lipids, 12% of glycolipids and 9% of phospholipids and those of bound lipids were 13% of neutral lipids, 10% of glycolipids and 13% of phospholipids in wind drying method. The major fatty acids of the neatral lipids were $C_{16:0},\;C_{18:0},\;C_{18:2},\;C_{18:2}\;and\;C_{20:0}$ which was consisted of free and bound lipids. As the major fatty acid amount of neutral lipids was 30.1% in free lipid, and 11.0% in bound lipid, the nsaturated fatty acid degree(TUFA/TSFA)was 1.12-1.14, the polyenoic acid degree (TPEA/TMEA), 0.32-1.20 and the total essential fatty acid (TEFA), 11.33%.

Studies on the Lipid Components in Sweetfish from Korea-2 Comparison of the contents of neutral and phospholipid components (한국산 은어의 지질성분에 관한 연구- 2 중성지질 및 인지질의 성분 비교)

  • The class and fatty acid composition of neutral lipid(NL) and phospholipid(PL) of Korean sweetfish were experimented. The NL was mainly consisted of triglyceride ($94.8{\sim}99.5\%$), and also identified free sterol($0.29{\sim}2.77\%$), sterol ester and diglyceride in less quantity. Triglyceride content of viscera was much higher than those of other tissues. Main components in the PL were phosphatidylcholine(PC, $7.9{\sim}61.6\%$), phosphatidyl ethanolamine(PE, $19.3\%{\sim}39.3\%$) and followed by diphosphatidyl glycerol and sphingomyelin. PC and PE contents were higher in muscle and head tissues. The major fatty acids in NL fractions of sweetfish were 16:0, 18:1n-9, 16:1n-7, 18:2n-6, 18:0 and 14:0. Fatty acid composition of NL was similar to those of total lipid and were not significantly different among the fishes, the large and small sweetfish. In case of PL fractions, the major fatty acids were 16:0, 18:1n-9, 22:6n-3, 18:0 and 18:2n-6.

호산성 방선균과 내중성 방선균

  • Waksman과 Heinrich(1943)에 의해 Streptomyces속이 기술되고 actinomycin, streptomycin등의 많은 항생물질이 발견되면서 방선균(actinomycetes)은 산업적으로 중요한 미생물이 되어 왔으며, 근래에는 항생물질 뿐만 아니라 효소, 효소저해제, 면역조절물질 등 소위 생리활성물질(bioactive compound)의 생산균으로서 주목을 받고 있다. 그러나 지금까지 대부분의 스크리닝이나 연구의 대상은 주로 중성 pH에서 잘 자라는 호중성 방선균(neutrophilic actionmycetes)이었지만 산성이나 알칼리성 토양, 고온, 고열, 고압 등과 같은 특수환경에서 분리된 균주나 희귀 방선균(rare actinomyces)에 대한 관심도 점점 높아지고 있다. 산성 토양에서 분리된 방선균에 관한 연구는 Jensen(1928)의 연구로부터 시작하였지만 Lonsdale(1985)이 침엽수림, 산성탄광 등에서 분리된 방선균을 수리분류할 때까지 활발하게 이루어지지 못하였다. 본 고에서는 호산성 방선균위 분포, 생리적 특성, 수리분류학적 연구, 화학적 분류(chemotaxonomy), 산업적 응용가능성 등을 살펴보고자 한다.

Lipid Components of Sea Squirt, Halocynthia roretzi, and Mideuduck, Styela clava (우렁쉥이 및 미더덕의 지방질성분)

  • Total lipid (TL) content of sea squirt (Ureungsweng-ee), Halocynthia roretzi, and Mideuduck, Styela clava, were 2.0%, 2.1%, respectively. Reviewing the composition of each lipid fraction in total lipids of sea squirt and Mideuduck, it was found that contents of neutral lipids (NL) (36.6%, 36.3%) and phospholipids (PL) (46.2%, 44.5%) were high, while that of glycolipids (GL) (17.2%, 19.2%) was low. The NL of sea squirt and Mideuduck were mainly consisted of triglyceride (49.0%, 59.6%) and free sterol (25.8%, 22.0%), and followed by diglyceride (9.4%, 7.7%), monoglyceride (6.0%, 4.2%), free fatty acid (4.6%, 1.9%) and esterified sterol and hydrocarbon (5.2%, 4.4%). And main lipids in PL were phosphatidylcholine (48.6%, 46.7%) and phosphatidylethanolamine (32.4%, 35.0%), and followed by phosphatidylinositol (9.8%, 7.0%), phosphatidylserine (5.7%, 5.8%) and an unknown substance (3.5%, 5.5%). Fatty acid composition was not significantly different among TL, NL, PL and GL contained in sea squirt and Mideuduck. The major fatty acids of TL in sea squirt and Mideuduck were eicosapentaenoic (21.3%, 18.3%), docosahexaenoic (16.3%, 14.2%), palmitic (13.8, 16.3%) and oleic acid (8.5%, 7.0%), respectively. Fatty acid composition of PL and NL were similar to those of TL. In case of GL fraction the major fatty acids were gadoleic (15.7%, 14.7%), palmitic (13.5%, 14.7%), stearic (11.6%. 9.8%) and oleic acid (8.0%, 8.1%).

Studies on Change of Lipids Improvement-Meju during the Fermentation (개량(改良)메주의 숙성과정(熟成過程) 중 지질(脂質)조성의 변화(變化)에 관(關)한 연구(硏究))

  • Changes of lipid composition in the Improvement-Meju inoculated with Aspergillus oryzae were examined. To investigate those changes systematically, silicic acid column chromatography was used for analysis of glycolipid, neutral lipid, phospholipid, and gas chromatography to examine the change of those fatty acid content. Following results were obtained. The lipid fraction obtained from soaked soybean and cooked soybean were mainly composed of about 93~94% neutral lipid, whereas phospholipid and glycolipid was 4.0~5.0%, 2.0~2.1% level, respectively. During meju incuvation period, neutral lipid decreased gradually, but glycolipid and phospholipid increased. Among the nonpolar lipids prepared from cooked soybean and soaked soybean, triglyceride content was mainly composed of 88~89%, and the content of sterol ester, free fatty acid, diglyceride and sterol was higher in soaked soybean than in cooked soybean. During meju incuvation period, triglyceride content decreased remarkablely, whereas content of sterol ester, free fatty acid and diglyceride increased gradually. From the soaked soybean and the cooked soybean, the fatty acids content of crude lipid, neutral lipid, glycolipid and phospholipid were composed of linoleic acid 54~70%, oleic acid 20.0~22.6%, palmitic acid 11.0~12.4%, linolenic acid 6.0~7.8% and stearic acid 3.4~4.3% in turn and myristic acid showed the trace, palmitic acid was a little higher in glycolipid and phospholipid than in crude lipid and neutral lipid. During meju incuvation period, the change of fatty acid content showed linoleic acid and linolenic acid reduction gradually in the neutral lipid, glycolipid and phospholipid. On the other hand, palmitic acid, stearic acid and oleic acid increased gradually, the maximum value was at the 4-days. The change of glycolipid fatty acid and phospholipid fatty acid was examined. 9-kinds including traced 3-kinds was detected. It was supposed that traced 2-kinks was occurred for incuvation, and those are the matter investigating in the future.

  • The lipid compositions of corns produced in Korea were analyzed. Free and bound lipids of the corn kernels were fractionated, quantitated and compared by silicic acid column, thin layer and gas liquid chromatography. Corn kernels contained 5.02% total lipids, which is consisted of 4.09% free lipids and 0.93% bound lipids. Free lipids comprised of 89.61% neutral lipids, 3.75% glycolipids and 6.40% phospholipids, while bound lipids contained 14.26% neutral lipids, 46.06% glycolipids and 37.18% phospholipids. In the neutral lipids of free lipids, triglycerides were predominant (67.68%) and minor components such as esterified sterols, free sterols, free fatty acids, 1,3-diglycerides, 1,2-diglycerides were present. But in the neutral lipids of bound lipids, esterified sterols were not present and the contents of triglycerides were lower (47.68%) and free fatty acids were higher than those of free lipids. Among the phospholipids in free and bound lipids, phosphatidyl choline and phosphatidyl serines and phosphatidyl inositols were also present as minor components. The major fatty acids in the three lipid classes were linoleic, oleic and palmitic acids.

  • Changes in lipid components of powdered Katsuobushi during processing such as boiling, smoke-drying were examined. The total lipid (TL) in raw skipkack was $1.8\%$ and it consisted of $79.2\%$ neutral lipid (NL), $7.8\%$ glycolipid (GL) and $12.5\%$ phospholipid (PL), while powdered Katsuobushi product showed $4.5\%$ TL which consisted of $82.5\%$ NL, $9.2\%$ GL and $8.3\%$ PL. The contents of triglyceride, digalactosyl diglyceride, phosphatidyl choline and phosphatidyl ethanolamine decreased when free fatty acid, diglyceride and monogalactosyl diglyceride increased during processing. Also formation of Iysophosphatidyl choline was identified in PL of product. Total fatty acid contents of raw fishes, boiled sample, 1st smoked sample and powdered Katsuobushi were 721.8mg, 589.8mg, 549.6mg and 473.1mg expressed in $C_{23:0}$ as the equivalents of fatty acid contents. Most fatty acid contents of TL revealed a tendency to decrease during processing, there were about $43\%$ decrease in polyenes, $36\%$ decrease in monoenes, and $26\%$ decrease in saturates quantitatively. The major fatty acids in TL, NL, GL and PL of samples were generally $C_{16:0},\;C_{18:0},\;C_{16:1},\;C_{18:1},\;C_{20:5}$ and $C_{22:6}$.

Gravitropism in the Salt-Stressed Primary Root of Maize (Zea mays) (염분 스트레스에 노출된 옥수수(Zea mays) 뿌리의 굴중성 반응)

  • We investigated gravitropic responses in the primary root of maize (Zea mays) seedlings which were exposed to salt stress. The maize roots salt-stressed with higher than 100 mM NaCl or KCl started to reveal enhanced gravitropic curvature after 2 hours form the gravi-stimulation. Such a promotion was not caused by sodium phosphate, but invoked by potassium phosphate, indicating the active component is $K^{+}$. Because NaCl increased gravitropic curvature, despite that $Na^{+}$ did not played any role, we evaluated the role for $Cl^{-}$ by comparing the effects of $MgCl_2$ and $MgSO_4$. The enhancement of the curvature only with $MgCl_2$ revealed that $Cl^{-}$ played a role in the gravi-response, indicating the involvement of anion channels. These results suggest that both of $K^{+}$ and $Cl^{-}$ play roles in the regulation of osmosis that is required for cell expansion in gravitropism as well as in nyctinasty and stomatal opening.

Root Gravitropic Response of Phytochrome Mutant (phyAB) in Arabidopsis (Arabidopsis 피토크롬 돌연변이체(phyAB)의 뿌리 굴중성 반응)

  • Phytochrome double mutant (PhyAB) showed the delayed root gravitropic response compared to the wild type (WT) in Arabidopsis. After 8 hr of gravistimulation, the gravitropic response of mutant showed 48% of the WT. The delayed response started at 1.5 hr after gravistimulation. And we measured the ethylene production in the root segments of WT and mutant for 12 hr. Ethylene production of mutant decreased about 40% of the WT at 12 hr. This result suggested that the phytochrome might be linked with ethylene production in some way. Generally, ethylene inhibits the growth of plant organs including roots. We measured the root growth rate in the presence of ACC (1-aminocyclopropane-1-carboxylic acid), a precursor of ethylene. And WT showed the inhibition of root growth with ACC, but mutant did not show the inhibition as WT did. To confirm the relationship between the ethylene and gravitropic response, we measured the gravitropic response with ACC. In the presence of $10^{-6}$ M ACC, WT showed the 37.4% inhibition compared to the control (no ACC), whereas mutant showed the only 6.6% inhibition of control (no ACC). This research suggested the relationship between phytochrome and gravitropic response through an ethylene production.
