Effects of cytokinins, GA, and IBA on in vitro propagation of Vitex negundo var. insica (좀목형 (Vitex negundo var. insica) 신초의 기내증식에 미치는 cytokinin, GA 및 IBA의 영향)

  • To develop an efficient micropropagation technique for Vitex negundo var. insica, which is known as aromatic and medicinal tree, the effects of various plant growth regulators (PGRs) on in vitro shoot proliferation and rooting were evaluated using the newly-developed shoots of a 3-year-old tree. Multiple shoot induction was achieved effectively on WPM (woody plant medium) supplemented with 0.5-2.0 mg/L BA, and the highest shoot number (7.9/explant) was obtained at the concentration of 1.0 mg/L BA. Typically 1 or 2 superior shoots (about 3.4 cm) were induced on hormone-free WPM. Combined treatment of BA 2.0 + GA 0.5 mg/L appeared to effective on shoot proliferation and rooting. Plant growth regulators added in shoot proliferation medium had strong impact on subsequent rooting as well. Overall, shoots induced by BA treatment resulted in high rooting rates while the effect was reduced gradually by ascending BA levels. TDZ of low concentration also revealed a similar tendency as BA, but the rooting ability was strongly inhibited at the concentration of 0.5 mg/L, and rooting was never observed at the concentrations higher than 0.5 mg/L. Combined treatment of BA and IBA had positive influence in both shoot proliferation and rooting. These results suggest that Vitex negundo var. insica could be effectively micropropagated via axillary bud cultures.

  • Mitochondria have multiple functions in cells: providing chemical energy, storing cellular $Ca^{2+}$, generating reactive oxygen species, and regulating apoptosis. Through these functions, mitochondria are also involved in the maintenance, proliferation, and differentiation of stem/progenitor cells. In the brain, the subventricular zone (SVZ) is one of the neurogenic regions that contains neural stem cells (NSCs) throughout a lifetime. However, reports on the role of mitochondria in SVZ NSCs are scarce. Here, we show that rotenone, a complex I inhibitor of mitochondria, inhibits the proliferation and differentiation of SVZ NSCs in different ways. In proliferating NSCs, rotenone decreases mitosis as measured through phosphorylated histone H3 detection; moreover, apoptosis is not induced by rotenone at 50 nM. In differentiating NSCs, rotenone blocks neurogenesis and oligodendrogenesis while glial fibrillary acidic protein-positive astrocytes are not affected. Interestingly, in this study there were more cells in the differentiating NSCs treated with rotenone for 4-6 days than in the vehicle control group which was a different effect from the reduced number of cells in the proliferating NSCs. We examined both apoptosis and mitosis and found that rotenone decreased apoptosis as detected by staining cleaved caspase-3 but did not affect mitosis. Our results suggest that functional mitochondria are necessary in both the proliferation and differentiation of SVZ NSCs. Furthermore, mitochondria might be involved in the mitosis and apoptosis that occur during those processes.

Continuous Production of Phalaenopsis Clones by Basal Shoot Culture (호접란 줄기기저부 절편배양을 통한 조직배양묘의 연속생산)

  • This study was conducted to establish a practical masspropagation system of Phalaenopsis clones from basal shoot segments. The frequency of PLB (protocorm like body) induction was compared with various explants. Basal shoot segments showed the most successful result of 45%, while root tips, stalk node segments, stalk leaves and mature leaves represented low frequency (below 5%). The PLB induction ratio in the culture of basal shoot segments was examined with 11 different Phalaenopsis varieties, and the majority of varieties, including pink flower lines, showed an about 30% rate of PLB formation. Especially, when whole basal shoot parts without cutting were inoculated onto PLB induction medium, giant PLB was induced from explant. This giant PLB was green color and big in size compared with normal PLB. When dissected giant PLB segments inoculated onto PLB multiplication medium, only normal size of PLBs were induced from them. PLBs induced by basal shoot culture were transferred onto proliferation medium and then shooting medium, from which normal plants were formed. Therefore, this culture method is considered as effective and practical protocol for Phalaenopsis mericlone production. In addition, it is suggested that clones of an infinite number can be produced consecutively by this culture system through repeated cycles of PLB induction and proliferation using the basal shoot segment of flask plant.

  • 패모(貝母)는 땅속의 뿌리줄기에 있는 2매의 비늘조각(인편(鱗片))이 마치 조개처럼 합쳐 있어 패모라 불리우며, 백합과로 숙근성 다년생 초본의 절패모(浙貝母; Fritillaria thungergii) 및 동속식물의 비늘줄기를 기원으로 하며, 중국의 강소 절강 안휘 등지에서 대량으로 재배되고 있다. 패모의 줄기는 15~40cm정도로 곧게 자라며, 꽃은 4~5월에 피며, 윗부분의 엽액에 1개씩 밑을 향하여 달린다. 종자는 결실률이 매우 낮아 이용을 할 수 없어 번식은 대부분 비늘줄기의 인편이나 자구를 이용한다. 약용부위는 인경(鱗莖)(비늘줄기)이 사용되는데, 인경(鱗莖)에는 알칼로이드물질인 verticine, fritillin, peimisine, peimine 등의 주요 성분을 함유하여 약리작용으로는 중추신경마비, 혈압강하 및 진해작용이 있다. 한방에서는 진해, 거담, 배농, 폐렴, 기관지염 등으로 이용하는 중요한 약초 중의 하나이나, 값싼 중국산 패모의 수입으로 우리나라에서는 재배가 붕괴되어 현재는 재배면적이 거의 없는 자원고갈 약초로 재배면적 확보가 시급한 실정에 있어 이를 되살리기 위한 노력이 절실한 실정이다. 따라서 패모 자구 증식을 위하여 온도와 $GA_3$, 상토의 조건에 따른 시험을 수행한 결과 패모 자구의 증식 효과는 저장온도 온도가 낮을수록 자구의 증식이 많아 $5^{\circ}C$에서 주당 5.5개로 구수의 증식효과가 높았으며, 상토는 모래와 밭흙에서 각각 3.1개, 2.5개로 많았으나, $GA_3$과 온탕처리에 있어서는 차이가 없었다.

Micropropagation of a Rare Tree Species, Empetrum nigrum var. japonicum K. Koch via Axillary Bud Culture (희귀 수종 시로미의 액아줄기 유도 미세번식)

  • In order to develop an efficient micropropagation system for a rare tree species, Empetrum nigrum var. japonicum K. Koch, the effect of medium salt, cytokinins and auxin at different concentration were evaluated. Shoot induction from axillary bud was better on WPM medium than on MS medium. Although there was no significant differences observed in shoot induction among the salt strengths of WPM medium, whereas healthy shoots were developed on basal WPM medium. In comparison of the cytokinins affecting shoot proliferation, zeatin was better than BA, whereas BA exhibited more effectiveness on shoot elongation. In vitro root formation was better on WPM medium than on 1/2MS medium and achieved the highest rooting rate when 5.0 mg/L IBA treatment. 93% of rooted plantlets were survived on artificial soil mixture after 4 weeks of acclimatization. Above results suggest that a rare tree species, E. nigrum var. japonicum can be micropropagated via axillary bud cultures.

In vitro propagation of a rare and endangered species, Echinosophora koreensis Nakai, by axillary bud culture (희귀 및 멸종위기 수종 개느삼의 액아배양을 통한 기내번식)

  • An efficient micropropagation was established by using axillary bud explants from two-year-old tree(Echinosphorea koreensis Nakai), which has been known as a rare and endangered species. Among various basal media tested, DKW medium was shown to be the best for axillary shoot elongation. The addition of both BA and TDZ to the medium induced 6 to 10 shoots per explant during eight weeks of culture, without showing any abnormal morphology at the shoot proliferation stage. However, high concentration of TDZ(>0.05 mg/L) appeared to cause hyperhydration on either leaf or shoot at the later developmental stage. Approximately 20% of shoots produced roots by the addition of 1.0 mg/L NAA but not by IBA($0.2{\sim}1.0$ mg/L). Ex vitro micro-cuttings were better source for root induction; up to 58.6% of the micro-cuttings rooted when 100 mg/L IBA was applied to the soil(vermiculite). More than 90% of plantlets with roots were successfully acclimatized and grew normally in the field. Therefore, we suggest that this endangered tree species can be effectively micropropagated by axillary bud culture system developed in this study.

Effect of plant growth regulators on micropropagation of a rare and endangered species, Tsuru-rindo (Tripterospermum japonicum) (멸종위기 식물 덩굴용담의 기내번식에 미치는 생장조절제 효과)

  • Various plant growth regulators were tested for shoot proliferation of Tripterospermum japonicum, a rare and endangered species. Among the six different media tested, MS medium was the best for the shoot growth. Whereas BA, upto 3 mg/L, significantly increased shoot proliferation rate, it suppressed the rate at higher levels. Neither kinetin nor TDZ was so effective in proliferating shoots as BA. As for rooting, TDZ strongly inhibited it even at very low concentration though spontaneous rooting was frequently observed from the proliferated shoots during culture of lower concentration BA or kinetin. In contrast, shoot elongation was significantly promoted by $GA_3$. More than 90% of the proliferated plantlets could be transplanted via cuttings into pots containing artificial soil mixture where they rooted and resumed normal growth. Most of the plants bloomed to bear fruits in the following year.

Induction and in vitro proliferation of adventitious roots in Dendropanax morbifera Lev. (황칠나무(Dendropanax morbifera Lev.)의 부정근 유도 및 기내증식조건)

  • 황칠나무(Dendropanax morbifera Lev.)는 두릅나무과(Araliaceae)에 속하며 학명에서 뜻하는 바와 같이 목본 (Dendro), 전능약(Panax)이라는 의미가 있고 나무인삼이라 불리기도 하며 줄기에 상처를 내면 노란액이 나온다고 해서 황칠나무(D. morbifera)라는 이름이 붙여졌다. 두릅나무과는 우리나라에서 최고의 약재들로 손꼽히는 인삼(Panax ginseng), 가시오갈피(Eleutherococcus senticosus) 등의 약용식물을 포함하고 있어서 황칠나무는 황칠수지액 이외에 약용식물로서의 무한한 개발 가능성을 내포하고 있다. 따라서 본 실험은 황칠나무의 기내 부정근 유도 및 증식조건의 확립을 목적으로 수행되었다. 우선 황칠나무의 기내 발아체로부터 부위(잎, 줄기, 뿌리)를 달리하여 부정근을 유도한 결과, 잎은 줄기나 뿌리보다 양호한 부정근의 유도를 보였다. 또한 유도된 부정근을 이용하여 옥신의 종류에 따른 부정근 유도율을 조사한 결과 IBA와 NAA는 IAA와 2.4-D보다 높은 유도율을 보였다. IBA의 농도에 따른 유도율과 증식효율은 IBA가 1.0 mg/L 첨가되었을 때 가장 높은 유도 및 증식효율을 보였다. 최적의 액체배지조건을 확인하고자 sucrose의 농도와 염농도를 달리하여 실험한 결과 1/2MS 배지는 MS 배지보다 10%정도 높은 증식율을 보였다. 액체배양 된 황칠나무의 부정근을 각각 1/2MS 배지에 30 g/L sucrose, 3.0 mg/L IBA가 첨가된 5 L volume 생물반응기에 4주 간 배양한 대조구와 2주 후 IBA의 농도를 1.0으로 낮추어 배양한 실험구에서 2주후 IBA의 농도를 낮추어 배양한 실험구에서 대조구보다 약 2배 높은 부정근의 증식량을 보였다. 결국, 황칠나무의 종자발아체를 이용하여 부정근의 유도 및 증식조건에 필요한 기내배양조건을 확립하였고, 플라스크와 생물반응기 배양을 통해 효율적인 실험실 내 증식조건을 확립하였다. 본 실험결과는 향후 황칠나무 천연추출물을 활용한 향장품/식,의약품 소재의 대량확보 차원에서 중요한 가치를 내포하고 있다고 조심스럽게 사료된다.

Shoot Regeneration of Mature Nodal Segments in Poplar (Populus deltoides) (미류나무 성숙 절간조직으로부터 식물체 증식)

  • Mature nodal segments of 2-year-old greenhouse stock plant were cultured on Murashige and Skoog basal medium supplemented with the different kinds and various concentrations of cytokinins to produce multiple shoots in in vitro condition. The most adventitious shoots were produced from excised ends of nodal segments. The highest average number $(24.6\;{\pm}\;4.6)$ of shoots was produced with the combination of BA 1.0mg/L and TDZ 0.1mg/L in MS medium. In addition, several shoots were formed from lenticels of bark cambium with the same treatment. These concentrations promoted high shooting capability upto $94.6\%$ and NAA was the best cytokinin among five different PGR sources.

Influence of Plant Growth Regulators on Shoot Multiplication of Thymus quinquecostatus Celak. (백리향 (Thymus quinquecostatus Celak.)의 기내증식에 미치는 식물생장조절물질의 영향)

  • In vitro culture system was established to induce multiple shoots of Thymus quinquecostatus Celak. by investigating the effects of cytokinins. Stem explants were cultures on MS medium supplemented with either five drfferent plant growth regulators or their combinations under light or dark condition. The most effective cytokininsource was the combination of BA 1.0mg/L and TDZ 0.1mg/L for producing shoots (6.05$\pm$1.51), zeatin 2.0mg/L and TEZ 0.1mg/L for elongating shoots (3.27$\pm$0.66cm) under the light condition. In addition the most effective cytokinin was 2-ip 2.0mg/L for producing shoots(5.20$\pm$1.81), zeatin 2.0mg/L for elongating shoots(5.64$\pm$1.24cm)under the dark condition. Overall, the average percent for in vitro shooting was greater than 89.58%.
