Gomisin A Inhibits Tumor Growth and Metastasis through Suppression of Angiogenesis (Gomisin A의 신혈관형성 저해를 통한 종양 성장 및 전이 억제 효과)

  • Cancer chemotherapy drugs command a large share of the market, and the development of new therapeutics with high efficacy and specificity is an active area of study. Recently, the development of cancer therapeutics from natural products targeting angiogenesis has drawn attention due to conventional chemotherapeutics showing serious side effects and resistance in cancer cells. In this study, we investigated the pharmacological efficacy of Gomisin A, an active ingredient of Schizandra chinensis baillon, on tumor growth and metastasis. Administration of Gomisin A at 10 and 100 ${\mu}g/ml$ reduced tumor growth in vivo by $80.5{\pm}8.1%$ and $96.2{\pm}2%$, respectively, compared with positive tumor controls. Treatment of Gomisin A in normal and various tumor cell lines did not exert significant toxicity. Mice treated with Gomisin A at a concentration of 10 and 100 ${\mu}g$/head showed a significant reduction in tumor-induced angiogenesis of $151{\pm}16.9%$ and $98.5{\pm}29.5%$, respectively. Furthermore, tumor metastasis analysis revealed that the administration of Gomisin A at a concentration of 10 and 100 ${\mu}g$/head inhibited tumor metastasis by $13.5{\pm}8.56%$ and $58.3{\pm}9.12%$, respectively. In addition, Gomisin A significantly decreased cell adhesion of the B16BL6 cells to the extracellular matrix. These results demonstrate that Gomisin A inhibits tumor growth via suppression of angiogenesis and tumor metastasis inhibition, without cellular toxicity. The pharmacological efficacy of Gomisin A suggests that it may be a potential candidate for the development of cancer drugs.

Decreased of Clusterin mRNA Expression of Epididymis Following Exposure to Bisphenol A Diglycidyl Ether during Gestation and Lactation in Sprague-Dawley Rats (임신 및 수유기간 동안 Bisphenol A Diglycidyl Ether 노출에 의한 랏트 부고환 clusterin mRNA 발현량 감소)

  • Bisphenol A diglycidyl ether(BADGE)는 비스페놀 A와 에피클로로하이드린의 축합에 의해 만들어지는 물질로 상업용 액상 에폭시 수지의 주성분이다. 본 연구는 clusterin mRNA 발현이 BADGE의 노출된 생식기계 독성에 연관되어 있는지를 연구하기 위해 수행하였다. BADGE는 SPF Sprague-Dawley 임신 랏트에 임신 6일부터 수유기가지 하루에 한 번 0(대조군)과 375mg/kg/day를 경구 투여하였다. 수컷 새끼는 일반 사항과 몸무게, 일반 발달 지표(예, 항문과 생식기 사이의 거리, 이개개전, 절치붕출, 유두잔류, 안검개열, 고환하강, 포피박리 등)등을 관찰하였다. 대조군과 투여군에서 다섯 마리의 수컷 새끼는 출생 후 3, 6와 9주에 부검하여 부고환의 조직학적 변화 등을 관찰하였다. BADGE 375 mg/kg/day 투여군에서 항문과 생식기 사이의 거리는 대조군보다 길어지는 경향을 보였다. 출생 후 6주와 9주에서 부고환의 상대 무게는 대조군보다 약간 증가하였으나 조직학적인 변화는 관찰되지 않았다. BADGE 투여 군에서 clusterin mRNA 발현량은 대조군에 비해 3주에 56%, 6주에 57% 그리고 9주에 86% 감소하였다. 이런 결과는 랏트의 부고환에서 clusterin은 BADGE에 반응하는 유전자 중 하나일 수 있다는 가능성을 나타낸다.

Assessment of biofilm formation of Lacticaseibacillus rhamnosus LRH020 (프로바이오틱스 Lacticaseibacillus rhamnosus LRH020의 미생물막 형성 평가)

  • Biofilms are complexly structured communities of microorganisms composed of surface-attached microorganisms, where their effects on the host have been controversial. In this study, we investigated the potential biofilm-forming capacity of Lacticaseibacillus rhamnosus LRH020 (DSM25568) by detecting genes known to promote biofilm formation. It was shown that the aggregation substance gene (asa 1) was presented in the LRH020 strain. Therefore, we investigated the phenotypic activities of the gene asa1 via two methods: biofilm formation and auto-aggregation activity. It was shown that the strain LRH020 had significantly less ability to form biofilm compared to the positive control strain Enterococcus faecalis ATCC 19433. Furthermore, LRH020 exhibited biofilm-forming activity comparable to Lacticaseibacillus rhamnosus GG (LGG), widely used probiotics. The auto-aggregation activity of LRH020 was also within the safe range similar to that of LGG. In conclusion, this study shows that both biofilm-forming and auto-aggregation activities of the LRH020 are comparable to one of the most studied probiotics strains, LGG.

Evaluation of In Vivo Safety of Inhibitory Compounds from Cheongmoknosang Mulberry Leaves against Helicobacter pylori (청목노상 뽕잎으로부터 분리한 Helicobacter pylori 저해물질에 대한 In Vivo 안전성 평가)

  • Biological compounds (caffeic acid, rosemarinic acid, and chlorogenic acid) from mulberry leaf extracts were administered to mice in order to confirm their stability. All male and female mice survived upon a 4,000 mg/g dose in an acute toxicity test, and they also survived after injection of 2,000 mg/kg for 13 weeks repeatedly. Therefore, the level of toxicity was not high. In a comparison of the control and test groups, there were no significant differences upon naked eye inspection, and the weights of stomachs infected by Helicobacter pylori were not significantly different. Regarding the effects on immune cells, NO of macrophages decreased more than that of control when medicine was administered. The spleens of the female mice group proliferated slightly in LPS and Con A within 48 hr, whereas the other test group showed a similar level and the cell toxicity of natural killing cells decreased. Therefore, we concluded that caffeic acid, rosemarinic acid, and chlorogenic acid from mulberry leaf extracts are not harmful for the treatment of infected patients the development as a healthy functional food.

Thirteen-week Repeated Oral Dose Toxicity Study of WK-38 in Rats (랫트에서 WK-38에 대한 13주 반복경구투여 독성에 관한 연구)

  • The subcronic toxicity of WK-38, a herbal preparation for the atherosclerosis, was examined in male and female Sprague-Dawley rats. WK-38 is composed of Rhei Rhizoma, Magonoliae Cortx, Moutan Cortex Radicis. Rats were treated with the test substance at a dose of 5 mg/kg, 50 mg/kg and 500 mg/kg intragastrically for 13 weeks. Control rats were treated with vehicle only. No death and abnormal clinical signs were observed throughout the administration period. Also there was no difference in net body weight gain, food and water consumption, organ weight, gross pathological findings, and urine analysis among the groups of rats treated with different doses of the WK-38. Hematological findings and biochemical examination revealed no evidence of specific toxicity related to WK-38. The results suggested that under the condition employed in this study no observation effect level (NOEL) of WK-38 would be 500 mg/kg/day.

Isolation and Properties of Cytotoxic Polyene Antibiotics Produced by Myxococcus stipitatus JW117. (Myxococcus stipitatus JW117이 생산하는 Polyene계 세포독성 물질의 분리 및 특성)

  • Drug resistance is one of the most significant impediments to successful chemotherapy of cancer. Multidrug-resistance (MDR) is characterized by decreased cellular sensitivity to anticancer agents due to the overexpression of P-glycoprotein. By employing adriamycin-resistance CL02 cancer cells, we undertook the screening for agents which were effective to multidrug-resistant cancer cells. As a result, a myxobacterial strain JW117 was selected for study since the solvent extract of cell mass of the strain was found to exhibit significant activity against the CL02 cancer cells. Cytotoxicity-guided chromatographic fractionation led to the isolation of phenalamides $A_2$ and $A_3$. The producing organism was identified as Myxococcus stipitatus by taxonomic comparison with type strains of Myxococcus sp. as well as its morphological and physiological characteristics. Phenalamides$ A_1$,$ A_2$ and $A_3$ were as active against drug-resistant cancer cells CL02 and CP70 as against the corresponding sensitive cells with $IC_{50}$ values ranging from 0.23~0.57 $\mu\textrm{g}$/ml.

Safety Evaluation of Korean Mistletoe Extract (한국산 겨우살이 추출물의 안전성 평가)

  • Mistlero C was shown to be non-genotoxic in a series of genotoxicity tests, including a bacterial reverse mutation test and a combined in vivo mammalian erythrocyte micronucleus test. In a bacterial reverse mutation assay, no significant increases in the number of revertant colonies, compared to the negative control, was detected in $5,000{\mu}g/plate$ of Mistlero C. In addition, with Mistlero C, no changes were shown in the number of micronucleated polychromatic erythrocytes (MNPCE) among 2,000 polychromatic erythrocytes compared to the negative control. Mistlero C was administered orally in rats to investigate acute toxicity. The $LD_{50}$ values in rats were above 2,000 mg/kg. In a repeated dose, 13-week, oral toxicity study conducted in rats, no compound-related adverse effects were shown at doses of Mistlero C of up to 1,000 mg/kg body weight/day. The results of these studies support the safe use of Mistlero C in food for human consumption.

Research Trends for Soil-Related Algal Toxicity (토양 관련 조류독성 연구동향)

  • Soil ecological risk assessment requires terrestrial toxicity data based on trophic levels including plants, earthworms, nematodes, and springtails. To expand the trophic levels, it is needed to consider primary producer algae, nearly distributed in terrestrial environment, as representative terrestrial test species. In this study, we collected research cases focused on soil-related test species and exposure media from SCI papers, and analyzed exposure media, test species, test chemicals, and other test methods, for reviewing research trends of soil-related algal toxicity. Up to now, in the soil-related algal toxicity, test species were 8 cases (Pseudokirchneriella subcapitata, Chlorella vulgaris, Scenedesmus bijugatus, Chlorococcum infusionum, Scenedesmus subspicatus, Nostoc linckia, Synechococcus elongatus, and Chlorococcum sp.) and endpoints were cell count or photosynthetic pigment content. Also, 5 of exposure media were liquid medium, soil extracts, porewater, agar medium, and soil. Most of papers used algae isolated from natural soils or soil extracts. There were only one case for assessing algal toxicity in soil medium. More researches regarding algal toxicity in soil environments need to be conducted consistently.

A Twenty-Eight Days Inhalation Toxicity Study of N-decane in Sprague Dawley Rats (SD흰쥐를 이용한 n-decane의 28일 반복흡입독성연구)

  • N-decane은 지방족탄화수소로 다른 탄환수소들과 같이 혼합된 형태로 존재하며 페인트 제거제나 드라이 크리닝 제품에 사용된다. 최근의 본 연구팀이 실시한 전자산업계의 MSDS 신뢰성조사 결과에 따르면 세정제의 사용 경향은 과거의 방향족 탄화수소나 CFC. HCFC에서 $C_{10}$이상의 지방족탄화수소 물질로 변화되고 있는 경향을 보여주었다. Stoddard solvent나 나프타 같은 탄화수소 혼합물에 대한 작업 환경노출기준은 설정되어있지만 n-decane에 대해서는 제한적인 독성자료 밖에 없으며 작업환경노출기준은 설정되어 있지 않다. 따라서 작업환경에 대한 적절한 관리기준제시와 독성학적 자료를 제공하기 위해 n-decane을 28일 반복 흡입독성시험을 실시하였다. 6주령 흰쥐로 체중이 229$\pm$10g되는 숫컷과 165$\pm$7g되는 암컷 흰쥐를 4개 용량군 즉 대조군, 저농도군(50ppm), 중농도군(200ppm), 고농도군(800ppm) (각군당 10마리)으로 설정하여 하루 6시간, 주5일로 4주간 흡입쳄버에서 노출시켰다. 28일간 노출 후 n-decane의 노출용량에 따른 암수의 체중에는 유의한 변화가 없었으며 유의한 혈액학적 생화학적 변화도 발견되지 않았다. 고농도로 노출된 수컷 몇 마리에서 고환 세정관에서의 공포화(vacuolization)가 발견되었으나, 간신장, 비장, 폐, 부신, 심장, 뇌 등 다른 장기에 대한 조직병리학적 검사에서는 뚜렷한 조직병리학적인 변화를 발견할 수 없었다.

흰목이버섯 유래 고순도 다당체의 광노화 개선 Global 화장품 신소재 개발

  • 최근 길어진 여름 및 이상고온 현상이 지속됨에 따라 심화되는 광노화 피부의 특징으로는 건조, 굵고 깊은 주름, 탄력저하 및 불균일한 색소침착 등이 나타나게 됨. 화장품 소재는 기존 광노화 관련 화학물질인 Retinol 등을 대체하기 위해 자연 유래 성분을 적용한 신소재 연구를 진행하고 있음. 흰목이버섯(Tremella fuciformis)은 흰목이목에 속하는 버섯류로 자실체는 한천질로서, 주름이 되어 갈라져 있거나 또는 귓불 모양을 이루고 있으며, 크기는 10 cm 정도이다. 중국에서는 보양식의 주재료로 쓰일 만큼 탁월한 항노화 효능이 알려져 있다. 본 연구에서는 흰목이버섯의 자실체에서 추출하여 정제한 β-Glucan의 성분 확인, in vitro 수준의 피부 항노화 효과, 동물대체 독성 시험을 통한 피부독성 확인 및 인체 피부유효성 평가를 통한 항노화 효과를 확인하였다. 흰목이에서 추출, 정제 후 Bio-LC를 통한 유리당 분석 결과 Mannose, Fucose, Glucose를 확인하였으며, Human Keratinocyte에 UVB를 조사하여 광노화를 유발한 피부세포에 피부 자극 및 탄력저하 인자인 IL-6, TNFa 및 MMP-1을 평가한 결과 농도 의존적으로 현저히 개선됨을 확인하였다. 또한 보습 및 피부장벽 개선 인자인 Filaggrin과 Involucrin 생성효능을 평가한 결과 매우 높이 생성됨을 확인하였다. 본 연구결과를 토대로 광독성, 피부감작성 및 안점막 동물대체 독성시험을 실시한 결과 무독성임을 확인하여 피부에 안전하면서 효능이 우수한 것을 in vitro 수준에서 확인하였고, 피부 홍반완화, 주름개선, 탄력개선 및 보습증가 등 광노화 예방효과를 인체를 대상으로 평가한 결과 유의적인 홍반완화, 주름개선, 탄력 및 보습증가효과를 확인하였다. 본 연구결과를 종합하여 볼 때 흰목이버섯에서 추출, 정제한 β-Glucan은 in vitro 수준에서 자외선으로 인한 피부 트러블 완화, 탄력 및 보습개선을 확인하였고 독성시험을 통해 무자극임을 판정하였으며, 인체유효성 평가를 통해 광노화 예방효과를 확인하였으며 본 결과를 통해 아시아 및 글로벌 시장으로 천연유래 항노화 소재로 확장하고자 한다.
