Germination of pelleted seeds in Rehmannia glutinosa Libosch (피막처리된 지황종자의 발아)

  • Seed coating and pelleting techniques have been used in many crops to improve the germination vigor of tiny and light seeds. Cultivated Chinese-foxglove (Rehmannia glutlnosa) has been infected by various types of virus derived from vagetative propagation of rootstock. Seed characteristics and alginate-coated seed germination rate have been investigated to get basic information for healthy seedling production through seed propagation. Chinese-foxglove showed different numbers of seeds per pod from 61 in Seocheon local to 207 in Jiwhang 1 and 1,000grain weight also varied from 70mg of Seocheon local to 130mg of Jiwhang 1. Seeds of Chinese-foxglove has dormancy because that seed collected last year more stimulated than that of this year at germination test. Optimum alginate concentration for pellected seeds germination was 2%.

Effects of Cryopreservation on the Seed Germination and Growth Properties of Seedlings of Maackia amurensis (초저온 저장이 다릅나무 종자의 발아와 유묘의 생장특성에 미치는 영향)

  • This study was conducted to investigate effects of cryopreservation by vitrification on the seed germination rate and growth and physiological properties of seedlings of Maackia amurensis. Cryopreservation significantly decreased the germination rate of seeds of M. amurensis, but the reduction of germination rate was mitigated by the treatment of cryoprotectant (plant vitrification solution, PVS2) before plugging into liquid nitrogen and fast thawing rate after cryopreservation. Long-term PVS2 exposure decreased seed germination rate, whereas cryopreservation time didn't have influence on seed germination rate. In addition, growth and physiological properties of seedlings were not affected by PVS2 exposing time and cryopreservation time. Therefore cryopreservation could be widely used as a technique of long-term ex situ conservation without any damage and deterioration of cells or tissues of the forest seeds. However, in order to increase the effect of cryopreservation, we have to develope the lower toxic cryoprotectant and suitable techniques to the structural or chemical properties of a variety of seeds.

Effect of Low-Dose Gamma Radiation to Enhance Germination Rate in Bottle Gourd and Pumpkin Seeds (참박과 호박 종자의 발아촉진을 위한 저선량 감마선의 효과)

  • To establish the seed treatment method of the gamma radiation for germination enhancement of bottle gourd(Lagenaria siceraria Standl.) and pumpkin(Cucurbita ficifolia Bouche), seeds were irradiated with the dose of 1$\sim$20 Gy of the gamma radiation. The germination rate in irradiated group was higher than that of the control, depending on seed condition. In the case of bottle gourd, the germination rate of 70$\sim$80% germinative seed was increased at 4$\sim$20 Gy irradiated groups. Especially, it was the highest at 4 Gy irradiated group. The germination rate of 80$\sim$90% germinative seed was the highest at 4 Gy irradiated group. The germination rate of 70$\sim$80% and 80$\sim$90% germinative seed at 4 days after sowing was increased 66% and 26% at 4 Gy irradiated group, respectively. In the case of pumpkin, the germination rate of 70$\sim$80% and 80$\sim$90% germinative seed was increased at 2 Gy and 8 Gy irradiated group, respectively. Effect of the low-dose gamma radiation on the early growth was enhanced depending on their germination enhancement. Especially, it was the highest at 2 Gy irradiated group of 70$\sim$80% germinative seed of pumpkin. Catalase activity of bottle gourd was increased at 4 Gy irradiated group. Catalase and peroxidase activity of pumpkin were increased at 2 Gy and 8 Gy irradiated group. These results suggest that the germination, early growth and physiological activity of bottle gourd and pumpkin seeds could be promoted by the low dose gamma radiation.

Effects of Scarification, Temperature and Sulfuric Acid Treatments on Seed Germination of White Lotus (Nelumbo nucifera) (백련(Nelumbo nucifera) 종자의 발아에 미치는 종피의 파상, 온도 및 황산처리 효과)

  • This study was conducted to determine the effects of scarification temperature, and sulfuric acid treatments on seed germination of white lotus collected from the Muan districts, Jeonnam in Korea. Without scarification, white lotus seeds were not germinated at all at 15 days after seeding. However, seeds sacrificed at basal parts showed 100% germination rate at $25^{\circ}C$ 6 days after seeding. All the seeds floated (0.90 g) and soaked (1.18 g) in the water were completely germinated. Especially, the lotus seeds soaked in the water at $25^{\circ}C$ showed high germination rate. Seeds treated with 80% sulfuric acid for 40-160 hours were germinated completely within 6 days after seeding. No difference in seed germination rate of white lotus stored up to 0, 12 and 24 months after harvest was observed. Overall results would be useful means for propagation and production of white lotus.

Morphological Characteristics and Function of Hilum in Safflower Seed Germination (잇꽃 종자의 발아에서 제(Hilum)의 형태적 특성과 기능)

  • This research was carried out to clarify the germination characteristics with reference to hard seed coat in safflower. Morphologically, seed coat surface has hilum and micropyle which were evident during seed development stage. In the flower of developing seeds, the hilum area is connected with placenta of maternal tissue while the micropyle area is connected with the style of pistil. When the seeds imbibed, the hilum surface began to crack and the embryo protrudes through the hilum. To investigate the route for moisture absorption and gas exchange on the seed coat, the hilum and the micropyle were artificially sealed by paraffin. The seeds whose hilum were sealed could not germinate, which indicates that the exchange of moisture and oxygen takes place through hilum in safflower seeds. The germination was tested at $15^{\circ}C$, $20^{\circ}C$, and $25^{\circ}C$ by three substrates with different moisture conditions; top of paper method (hilum submerged in water), between-paper method, and soil seeding. The germination percentages were 31.3% at $15^{\circ}C$, 15.7% at $20^{\circ}C$ and 6.0% at $25^{\circ}C$ in the top of paper method; and 45.5% at $15^{\circ}C$, 30.0% at $20^{\circ}C$ and 14.0% at $25^{\circ}C$ in the between-paper method; and 80.0% at $15^{\circ}C$, 77.0% at $20^{\circ}C$ and 78.0% at $25^{\circ}C$ in the soil seeding, respectively. When the internal structure of hilum was investigated through SEM, it was found out consisting of vascular bundle element. In conclusion, the hilum of safflower seed was closely related with water absorption and gas exchange during initial germination process.

Improvement of Seed Germination in Rosa rugosa (해당화의 종자 발아 연구)

  • Rose seed shows low germination percentages (about 20%) because it has a high amount of substances that inhibit germination in the pericarp. We investigated the effect of orientation of achene, the day after pollination (DAP), and cold storage with or without half-cut in achene for the germination percentage in R. rugosa. Germination percentages of intact or half-cut achenes were investigated in a 16-hour photoperiod at $25^{\circ}C$ room on basal MS medium for two weeks. In germination percentage, maximum 100% was measured within one week when half-cut achenes were cultured on an orientation that the embryos facing to the light. Half-cut achenes at 90 DAP were germinated 100% regardless of cold storage. Various LED lights (red, blue, yellow, green, and white) were illuminated over the half-cut achenes to gain the effect of light color. Germination percentage of R. rugosa seeds under blue LED reached the greatest with 90% within one week of culture and these seedlings were the best with a steady growth rate. It is concluded that half-cut achenes would be an effective method to improve seed germination in R. rugosa without stratification or scarification. This system could be applied to breeding studies in rose cultivars.

The Effect of Temperature of Compost on the Germination Inhibition of Weed Seeds during Livestock Manure Composting (가축 분 퇴비화과정의 온도에 따른 잡초종자 발아억제 효과)

  • This study was carried out to assess the composting temperature effects on germination of curled dock and barnyard millet seeds. After the seeds wrapped in gauze were buried in the compost heap, the seeds were taken out under monitering the temperature for measuring germination rate at 1.5, 3, 10 and 20 days of composting. The germination rate of the seeds was directly related to the temperature of the compost heap regardless of the type of composting method. The seeds that buried in the compost for 1.5 days at a temperature of $60{\sim}70^{\circ}C$ were completely lost their germination ability. However, the germination rate of the seeds retained about 10% even after 20 days when compost temperature was below $51^{\circ}C$. Reulsting data suggests that, it is necessary to keep the compost temperature at least $55^{\circ}C$ for more than 3 days in order to suppress the germination of the seeds. On the other hand, if the temperature of the compost rises by $60{\sim}70^{\circ}C$, the seeds will lose their gemination ability with in 1.5 days. In conclusion, aerobic composting would be more effective in suppressing germination ability of curled dock seed and barnyard millet seeds.

  • Effects of media and low temperature on germination of weeds were examined to get basic information for establishing weed control methods. Soil and agar(0.4%) was good media for germination of weeds at laboratory. Germination was improved by placing weed seeds at 5$^{\circ}C$ low temperature for 10-20 days. Germination was improved by storing weed seeds in water absorbed gauge and in vinylbag at 5$^{\circ}C$ low temperature for 30-40 days. Germination was increased by burial of weed seeds at 10cm of soil depth for 30-60 days.

  • This experiment was conducted to obtain the basic information for the increase ofgermination percent by the seed diameter, specific gravity, different grain filling period and stratification in Bupleurum falcatum seeds. The seed size of B.falcatum ranged from 2.0 to 3.9mm in diameter, The large seed showed the higher germinability, The optimum specific gravity of salt selection for seed screening was 1.05 resulting in screening out seeds with high germinability over 70%.The seeds needed grain filling period for 60 days or longer after flowering to keep high germinability The optimum period of stratification treatment to promote seed germination was about 80 days.

Seed Characteristics and Accelerating Method of Germination in Bupleurum falcatum (시호 종자의 특성 및 발아촉진 방법 연구)

  • The experiment was conducted to determine the seed characteristics and preferable methods to enhance the seed germination rate in Bupleurum falcatum. The optimum temperature for the seed germination of Bupleurum falcatum is 2$0^{\circ}C$. Any significant promoting effects were not found in seed germination with hormone treatments and physical methods. At 15$^{\circ}C$, prechilling combined with 50~200ppm of GA$_3$ treatment raised germination rate by 2 times of control ones. The most positive effect was observed in the treatment of 10$^{-2}$ ~10$^{-3}$ M potassium nitrate only at 15$^{\circ}C$ for 12 and 24 hours. The leachate of Bupleurum falcatum didn't inhibit the germination of Lactuca sativa, showing almost 100% of germination rate, which is suggested that no inhibitors contained in the seeds of Bupleurum falcatum. Observation of embryo conditions under stereoscopic microscope showed that the ratio of seeds with or without embryo is almost 50/50. The results suggested that the lower rate of germination in Bupleurum falcatum was caused by embryolessness of seeds.
