  • Out of 50 seed samples of Sesamum indicum L. tested, Alternaria sesami and Corynespora cassiicola were detected in 29 samples and 38 samples, respectively. Heavy infection of A. sesami and C. cassiicola caused seed rot and seedling blight on water agar as well as in pots. In case of light infection the seed germinated showing elongation of the radicle and hypocotyl, but soon light brown or dark brown lesions appeared at certain parts of the shoot and root where sporulation of the fungi was observed after prolonged incubation in moisture. Four seed disinfectants were compared for the control of seed-borne infection of A. sesami and C. cassiicola. Complete control of seed-borne infection of C. cassiicola was obtained by seed treatment with Benlate T and Homai and seed treatment with Busan 30 was found superior to control seed-borne infection of A. sesami.

  • This experiment was conducted to evaluate the effect of early seed harvest on germination of tobacco(Nicotiana tabacum L.) seeds. Seeds of seven burley tobacco were harvested every two days from 8 to 30 days after pollination and tested for germination. The results are; 1. Seeds harvested 12days after pollination germinated but germination rates were low and varied across cultivars. These seeds will provide viable seeds adequate for breeding program. 2. Germination rates of seeds harvested 24-28 days after pollination were high and showed none significant differences among cultivars. 4. Over-ripen seeds showed low germination rates cused by after-harvest or temporary dormancy.

Effect of Priming Treatments on Seed Germination and Seedling Growth of Sorbus alnifolia (Priming 처리가 팥배나무의 종자 발아 및 유묘 생장에 미치는 영향)

  • Seed priming is a useful technique for rapid and uniform seed germination as well as early seedling establishment. This experiment was conducted to find out the optimum condition for Sorbus alnifolia seed priming with four concentrations of four reagents in germination property and seedling growth performance. The results are summarized as follows: Percent germination (PG) varied 2.67% to 24.67%, and S. alnifolia seeds had the highest PG in the treatment that were primed in 100mM $KNO_3$ solution for 2 days. Mean germination time was the shortest in 200 mM $KNO_3$ solution for 2 days. Seed priming with $KNO_3$ solutions increased germination speed (GS) and germination performance index (GPI) compared with non-primed seeds. Especially seed primed with 100 mM $KNO_3$ solution for 2 days showed the highest GS and GPI. The highest relative growth rate (RGR) and seedling vigor index (SVI) was significantly (p<0.05) different from the control and other treatments, respectively. RGR of height (0.0071) and root collar diameter (0.0141) of seedling from primed seeds were the highest in 400 mM NaCl solution for 2 days. The highest SVI (5.43) was observed in the seedlings from seeds primed in 100 mM $KNO_3$ solution for 2 days. Consequently, the optimum reagent and concentration were $KNO_3$ and 100 mM for the effective germination and seedling growth in S. alnifolia

Germination and Emergence of Eclipta prostrata(L.) L. (한련초의 발아(發芽) 및 출현(出現))

  • Several experiments were conducted to investigate the effects of external factors on germination and emergence of Eclipta prostrata (L.)L. The weight of viable achenes doubled as a result of 90 minutes soaking in water. The germination of E. prostrata was significantly improved by alternating temperatures. At a constant temperature of $35^{\circ}C$, only 78% of the achenes germination, whereas at alternating temperatures of 35/$20^{\circ}C$, 96.5% of the achenes germinated. E. prostrata was more sensitive than rice to moisture stress. No germination of E. prostrata achenes occurred in the absence of oxygen. No germination of E. prostrata achenes occurred in the dark or when they were exposed to green, blue, and far-red light. Germination of E. prostrata achenes was influenced by the duration of illumination after absorption of water. Ten hours of illumination was needed for maximum germination and 2 hours for 50% germination. No significant changes in germination of E. prostrata achenes were observed between pH 3 and 10. A high tolerance of E. prostrata achenes to salt was observed. Emergence of E. prostrata achenes was greatly affected by planting depth. In the upland soil, 74.0% of the achenes planted on the soil surface germinated, and no emergence was at planting depths of 0.5 cm or greater.

Individual Differences of Ozone Resistance for Seed Germination and Seedling Development of Pinus thunbergii (해송의 종자 발아 및 유묘 생장에 대한 오존저항성의 개체간 차이)

  • Individual differences of ozone ($O_3$) resistance for seed production, seed germination and seedling development were examined in this study. Five in each healthy and damaged trees of Pinus thunbergii growing in air polluted area for 12 years were chosen based on visible foliar injury and growth. The cones of P. thunbergii, which were collected from healthy and damaged trees, were analyzed for physical characteristics and seeds from the cones were used to test germination percentage under $O_3$ treatment. The germinated seeds were continuously exposed to $O_3$ treatment and the lipid peroxidation and activities of antioxidative enzymes were determined for both seeds and seedlings. The $O_3$ treatment for seed germination and seedling development were conducted at three conditions: control, 150 ppb and 300 ppb of $O_3$. The non-treated seeds from the damaged trees showed 21.6% lower germination than those from the healthy ones. On the $O_3$ treatment of 300 ppb, seed germination decreased approximately 10% for the healthy trees and 19% for the damaged trees compared to that on the control. The seeds from the healthy trees showed significantly higher activities of superoxide dismutase (SOD), glutathione reductase (GR), and catalase (CAT) than those from the damaged trees. The activities of GR, ascorbate peroxidase (APX), and CAT decreased along with the increasing $O_3$ concentration in two tree grades. Malondialdehyde (MDA) content of seeds was not influenced by $O_3$ treatment for two tree grades. In seedling development, there were no significant differences for length and biomass of needle and root of two tree grades at both the control and 150 ppb of $O_3$. At 300 ppb of $O_3$ treatment, however, the length and biomass of needle and stem decreased for two tree grades but no significant differences was detected in root. The seedlings from the damaged trees were more sensitive to the $O_3$ treatment, showing higher activities of SOD, APX, and CAT and content of MDA compared to those from the healthy tree seedlings. Our results indicate that seed germination and seedling development are vulnerable to increasing $O_3$ concentrations and that attention must be paid to the individual selection of tree species for reforestation.

Effect of Pant Growth Regulators on the Germination and Seedling growth of Wasabia japonica Matsum Seeds (식물생장조절물질(植物生長調節物質)이 고추냉이의 발아(發芽)와 유묘생장(幼苗生長)에 미치는 영향(影響))

  • This study was carried out to investigate the effect of some plant growth regulators on the germination and seedling growth of Wasabia japonica Matsum seeds. The seeds were soaked in different concentration of indolacetic acid(LAA), indolbutyric acid(IBA), $GA_3\;GA_4$, Benzyladenine(BA) and kinetin, and the germination percent, the average germination day and the growth of seedlings were measured. The seeds were released from dormancy and the germination percent was considerably increased when the seedcoat was removed. $GA_3,\;GA_4,$ BA and kinetin were effective in breaking the dormancy, but LAA and IBA were not effective. $GA_4$, BA and kinetin were more effective in accelerating germination of dormancy seed than $GA_3$. The applications of BA combined with $GA_3$ l00ppm showed higher germination percent than that of BA alone within the range 50ppm and 100ppm. Root, hypocotyl and petiol of seedling overgrew by the application of $GA_3\;and\;GA_4$. But effects of BA and kinetin were good on seedling growth compared with those of $GA_3\;and\;GA_4$. Therefore, BA and kinetin could be use for dormancy breaking of seed instead of $GA_3$.

Comparison of Germination Characteristics and Various Pre-treatment Methods for Enhancing Germination on Zoysiagrass (한국잔디 종자의 발아특성과 발아율 향상을 위한 다양한 전처리 방법의 비교)

  • Zoysiagrass seeds had low germination or lack of germination uniformity under natural condition. To improve the final germination percentage of zoysiagrass seeds, we investigated the methods of proper pre-treatment to enhance germination rate of zoysiagrass. Seeds were subjected to the testa scarification and four types of seed priming methods, namely, hydro priming (distilled water), osmotic priming (polyethylene glycol -0.5 MPa, -1.0MPa, -2.0 MPa), halo priming (Potassium nitrate 100 mM, 200 mM, 400 mM), and solid matrix priming (seed : microcel-E : distilled water = 4:1:8) with and without 25% KOH treatment in the testa. The scarification improved the germination rate and reduced the germination time. Osmotic priming (PEG -0.5 MPa) with scarification were significantly increased the seed germination rate and decreased MGT, and $T_{50}$. Seed germination rate obtained by osmotic priming (PEG -0.5MPa) with scarification increased above 60% in comparison with natural condition. Conclusively, scarified and primed zoysiagrass seed germinated more rapidly and uniformly in the study. The present results suggested that PEG priming treatment with KOH treatment prior to sowing could improve germination percentage, and this method makes us establish the early zoysiagrasses in the field.

Germination Responses of Echinochloa crus - galli Seeds to Temperature (온도에 대한 돌피(Echinochloa crus-galli) 종자의 발아반응)

  • The germination responses of Echinochloa crus-galli (L.) Beauv. seeds to temperature were examined under the various thermal conditions. While almost all the seeds tested did not germinate immediately after collection, almost of the seeds which were stored for 7, 10, and 17 months showed very high germination percentages (85-95%) at their own constant temperatures between $16^{\circ}C\;and\;40^{\circ}C$. The total thermal time which was required for germination(10-70%) of Echinochloa crus-galli seeds ranged from 539Kh(degree Kelvin X hour) to 1,279Kh in accordance with the distribution function of thermal time, $F({\Theta})=1-[3D^{-3}({\Theta}-m+D)+1]^{-1/2}$, where m is 935Kh and D is 555Kh. Moist chilling treatment at $2^{\circ}C$ for 20 days increased the final germination percentage as well as the germination rate. In the increasing temperature(IT1 regime, E. crus-galli seeds started to germinate at $12^{\circ}C$. and showed greater germination rate with increasing temperatures, with the final germination percentage of 80%. On the other hand, in the decreasing temperature(DT1 regime, the seeds began to germinate at $12^{\circ}C$(10% germination) with the final germination percentage of 20%. An induced dormancy occurred at $4^{\circ}C$ in the DT regime.

Characteristics of Seed Germination and Promotion of Germination Rate in Pollia japonica Thunb. (나도생강의 종자 발아 특성과 발아율 향상)

  • This study was conducted to clarify the characteristics of seed germination and to promote the germination rate in Pollia japonica Thunb. that was pointed to protect by the Office of Forestry in 1997. It was better the germination rate and the growth of in the light than those of in the darkness. The seed germination of Pollia japonica showed the highest germination rate at $20^{\circ}C$ any other treated temperature. To increase the germination rate of Pollia japonica, it was conducted priming treatments and storage treatments at $4^{\circ}C$. Priming treatments using 1% NaOH, 1% KOH for 30min were effective compared to control, but soaking of 100mg/L GA3 for 24 hours, 1% NaOCl for 30min were not useful. It was higher in wet storage than in dry storage. Wet storage of seeds showed a germination rate with 95.3% in 90-day treatment, which improved 55.3% than dry storage in 90-day.

  • To search germination inhibitors in higher plants, first of all, some experiments containing selection of test weed seeds and effects of solvents and surfactants for bioassay establishment were conducted. Then MeOH-extracts of 45 plants were assayed for germination inhibition activities against Echinochloa crus-galli (L.) Beauv, Digitaria sanguinalis (L.) Scop., Cyperus iria L., Portulaca oleracea L. and Oenothera lamarckiana Ser. seeds. Among them extracts from Rhathiolepis ovata Briat and Picea abies (L.) Karst showed strong inhibitory effect (60-90% inhibition) on the germination of tested weed seeds at 5000 ppm. On the other hand, the extract from Youngia sonchifolia Max stimulated the germination and growth of tested weeds.
