Mouse Single Oral Dose Toxicity Test of Chongmyung-tang Aqueous Extracts (총명탕(聰明湯) 열수(熱水) 추출물의 마우스 단회 경구투여 독성 실험)

  • Objectives & Methods : The objective of this study was to evaluate the single oral dose toxicity of Chongmyung-tang (CMT) in ICR mice. Korean traditional herbal prescription CMT has traditionally been used as a neuroprotective for treatment of learning disability and memory improvement. CMT, lyophilized aqueous extracts (yield=9.7%) were administered to female and male mice with oral dose of 2,000, 1,000 and 500 mg/kg (body weight) according to the recommendation of Korea Food and Drug Administration (KFDA) Guidelines. Animals were monitored for mortality, changes in body weight, clinical signs and gross observation during 14 days after administration upon necropsy; organ weight and histopathology of 14 principle organs were examined. Results : We could not find any CMT extracts treatment related mortalities, clinical signs, changes in body and organ weight, or gross and histopathological observations against 14 principle organs up to 2,000 mg/kg in both female and male mice, except for some accidental sporadic findings which did not show any obvious dose-relations and most of which also demonstrated in both the female and male vehicle control mice in this experiments. Conclusions : Based on the results of this experiment, the 50% lethal dose ($LD_{50}$) and approximate lethal dose (ALD) of CMT extracts after single oral treatment in female and male mice can be considered to be over 2,000 mg/kg, and is likely to be safe in humans.

CONDITIONS FOR PROCESSING OF MEATY TEXTURED FISH PROTEIN CONCENTRATE FROM ALASKA POLLACK AND MACKEREL (명태 및 고등어의 축육과 유사한 어육조직단백질 농축물의 가공조건)

  • For the effective utilization of the coastal fish resources in Korea, an investigation on the optimum processing conditions and the quality of a textured fish protein concentrate similar to the texture of animal meat has been carried out with the fish meat of Alaska pollack and mackerel. A noodle shaped product was prepared with the fish meat paste after the adjustment of pH and salt content. The product was soaked in $96\%$ ethyl alcohol to produce textured fish protein concentrate and then dried. The processing conditions were estimated with the rehydration capacity of the textured fish protein concentrate(FFC). The quality of the final product was evaluated with chemical composition, sensory test and texture measurement. The optimum pH and salt content of the fish meat for the processing of meaty textured FPC were 7.5 and $1.0\%$ respectively. The most effective soaking conditions were as follows:soaking time, 40 min. ; temperature of alcohol, 5 to $20^{\circ}C$;amount of alcohol, 4 times the weight of tile fish meat paste, number of soaking in alcohol, 4 times. The alcohol remaining in meaty textured FPC could be removed effectively by forced air drying. The yield and the contents of protein and lipid in the meaty textured FPC from Alaska pollack were $19.9\%\;84.3\%\;and\;0.5\%$ and those from mackerel were $29.8\%,\;78.1\%\;and\;3.6\%$ respectively. The content of essential amino acid in the meaty textured FPC from Alaska pollack and mackerel was not inferior to that of beef, textured soybean protein and FAO pattern. Beef meat can be substituted with the meaty textured FPC up to $50\%$ in processing meat balls withoutanysignificantlossinthetaste, ordor and texture.

Growth and Postharvest Freshness of $Tah$ $Tasai$ Chinese Cabbage ($Brassica$ $campestris$ var. $narinosa$) Baby Leaf Vegetable as Affected by Brushing Treatment during Cultivation (재배 시 brushing 처리에 따른 어린잎 채소 다채의 생육과 수확 후 선도 차이)

  • An experiment was conducted to find out the effect of brushing treatment during cultivation on the postharvest quality of the baby leaf vegetable, specifically tah tasai Chinese cabbage (Brassica campestris var. narinosa). The effect of mechanical brushing during cultivation on the postharvest quality was determined in terms of the quality changes in weight loss, gas partial pressure, leaf color, and appearance during storage using a 30-${\mu}m$-thick polypropylene film at $16^{\circ}C$. The brushing treatment included brushing with A4 paper back and forth 50 times a day. The study revealed that the growths on the brushing-treated plant group were less than those on the control group. The structure of the leaf tissue of the brushing-treated plant also tended to be less compact than that of the non-treated plant. The brushing treatment resulted in less growth and denser plant tissues as well as in differences in the gas $O_2$ consumption and $CO_2$ accumulation after packaging. For the gas partial pressure, the $O_2$ consumption and $CO_2$ accumulation of the brushing-treated plant tended to be less than those of the non-treated plant. There were no differences, however, between the brushing-treated plant and control groups in the SPAD value and appearance. The study results also suggested that after packaging, the effects of the brushing treatment during cultivation on the quality of the tah tasai Chinese cabbage baby leaf vegetable was not significant. As such, it is recommended that effective post-harvest methods of improving the product quality of the baby leaf vegetable be further investigated.

Characteristics of EVA-Polymer Modified Mortars Recycling Rapid-chilled Steel Slag Fine Aggregate (급냉 제강슬래그를 재활용한 EVA-폴리머 시멘트 모르타르의 특성)

  • For the recycling of rapid-chilled steel slag, the mechanical strengths and physical properties of EVA-polymer modified mortars with the various replacement ratios of rapid-chilled steel slag were investigated. Twenty five specimens of polymer modified mortars were prepared with the five different amounts of EVA-polymer modifier (0, 5, 10, 15, 20 wt%) and rapid-chilled steel slag (0, 25, 50, 75, 100 wt%). For the investigation of the characteristics of polymer modified mortars, the measurements such as water-cement ratio, unit volume weight, air content for fresh mortar and compressive strength, flexural strength, water absorption, hot water resistance, porosity and SEM investigation for curing specimens were conducted. As a results, with an increase in the replacement ratio of rapid-chilled steel slag, water-cement ratios decreased but unit volume weight increased remarkably. With increasing EVA-polymer modifier and the replacement ratio of rapid-chilled steel slag, percent of water absorption decreased but compressive and flexural strengths increased remarkably. By the hot water resistance test, mechanical strengths decreased but total pore volume and porosity increased remarkably. In the SEM observation, the components of specimen were shown to stick to each other in the form of co-matrix phase before hot water resistance test, but polymer modifier of co-matrix phase was decomposed or deteriorated after hot water resistance test.

Effect of Glutathione-Enriched Saccharomyces cerevisiae FF-8 on Tissues Lipid Peroxidation in Orotic Acid-Induced Fatty Liver Model Rats (지방간의 과산화지질에 미치는 글루타티온 고함유 효모 Saccharomyces cerevisiae FF-8 균주 급여의 영향)

  • Glutathione is a well-known chemotherapeutic agent and a popular nutritional supplement for liver disease and oxidative stress. Our previous studies reported the suppressive effects of the glutathione-enriched Saccharomyces cerevisiae FF-8 (FF-8) strain on carbon tetrachloride- and alcohol-induced oxidative stress in rats. The purpose of the current study was to investigate the effect of the FF-8 strain on lipid peroxidation in tissues of rats with orotic acid (OA)-induced fatty liver. OA treatment showed a significant decrease in body weight gain compared to the normal diet, and simultaneous addition of FF-8 and OA had the same effect. OA treatment produced an increase in liver weight, however, this also increased with simultaneous addition of FF-8 and OA. Liver lipid peroxidation was significantly increased by OA, but was significantly decreased by FF-8 strain treatment. This same tendency was found in the kidney and heart. Concentration levels of hepatic glutathione and zinc are known to be closely associated with the antioxidant system, and OA treatment led to reductions in liver glutathione and zinc concentrations, whereas these were significantly increased by FF-8 strain treatment in OA feeding rats. These results suggest that the glutathione-enriched S. cerevisiae FF-8 strain may positively mediate orotic acid-induced oxidative stress by enhancing glutathione and zinc levels in rat livers.

Effects of Mulberry Leaf Tea Fermented by Monascus pilosus on Body Weight and Hepatic Antioxidant Enzyme Activities in Mouse Fed High-Fat Diet (Monascus pilosus 발효 뽕잎차가 고지방 식이 마우스의 체중과 간 조직 항산화계 효소 활성에 미치는 영향)

  • In this study, we investigated the preventive effects of the mulberry leaf tea fermented by Monascus pilosus on high fat-induced obesity, hyperlipidemia, and fatty liver in mice. Non-fermented mulberry leaf tea powder (UM) and fermented mulberry leaf tea powder (FM) were supplemented with high-fat diet at 2% (wt/wt) dosage for 8 weeks. Both UM and FM lowered body weight gain, feed efficiency ratio, epididymal fat, serum triglyceride, total cholesterol and LDL-cholesterol increased markedly with high fat diet (HC) in mice. FM showed more significant effects when it was compared with UM. In addition, Hepatic lipid peroxides and xanthin oxidase activities of the UM and FM were significantly lower than those of HC, despite the lack of a big difference in the amount of hepatic GSH. Activities of ROS scavenging enzymes and serum alanine aminotransferase activity were also examined as a parameter of hepatic damage. The UM and FM groups showed a recovery to NC group from significant changes induced by HC. Finally, histopathological analyses of liver samples revealed a decrease of lipid accumulation in hepatocytes in the UM and FM groups. These results suggest that UM and especially FM can reduce the development of obesity, hyperlipidemia and fatty liver.

Effect of Dietary Vitamin A or $\beta$-Carotene on Lipid Metabolism of Rats Induced Acute Ethanol Administration (비타민 A와 $\beta$-Carotene의 급여가 에탄올을 급성 투여한 흰쥐의 지질대사에 미치는 영향)

  • 본 연구는 retinyl acetate와 $\beta$-carotene보충식이가 에탄올에 의한 산화적 손상에 미치는 영향을 조사하고자 흰쥐에게 비타민 A와 $\beta$-carotene을 보충한 식이를 5주간 급여한 후 에탄올을 급성으로 투여하여 혈청과 간 조직에서 생성되는 지질과 지질과산화물 함량 그리고 glucose-6-phosphatase 활성도를 측정하였다. 본 실험에서 얻은 결과를 요약하면 체중 증가량은 전 실험군에서 유의적인 차이없이 실험 기간 동안 증가되었으며 사료섭취량과 식이효율을 그리고 간 중량 역시 각 군 모두 유의적인 차이를 나타내지 않았다. 혈청 중성지질 함량은 대조군과 retinyl acetate 보충군에서 높은 함량을 보였으나 통계적인 유의성은 없었다. 총 콜레스테를 함량은 대조군과 $\beta$-carotene 보충군은 비슷한 수준을 나타낸 반면 retinyl acetate 보충군에서는 유의적으로 감소되었다. HDL-콜레스테롤 함량은 대조군에서 가장 높았으며 LDL-콜레스테롤 함량은 유의적이지는 않았으나 retinyl acetate 보충군에서 가장 낮은 함량을 보였다. 동맥경화지수는 전 실험군에서 유의적인 차이는 없었으나 retinyl acetate 보충군에서 가장 낮았다. 간 마이크로솜 내에서 지질과산화물 함량은 재도군에 비하여 $\beta$-carotene 보충군에서 유의적으로 감소되었다. Retinyl acetate와 $\beta$-carotene 보충군 사이에서는 유의적인 차이가 없었으나 급성적인 에탄올 투여시에 $\beta$-carotene의 보충 급여가 지질과산화반응에 방어효과가 있는 것으로 나타났다. 간 조직중의 glucose-6-phosphatase 활성도는 비타민을 정상적으로 공급시킨 대조군이 retinyl acetate나 B-carotene 보충군에 비하여 유의적으로 높았으며 retinyl acetat와 B-carotene 보충군 사이에는 유의적인 차이가 없었다.

Effect of Si Content on the Microstructure of Cast M42 Tool Steel

  • 공구강은 C, Cr, V, Mo, W, Co 및 Si 과 같은 첨가원소를 함유한 복잡한 철계 합금으로 주요 특성인 경도, 부식저항성, 열 연화저항성 그리고 인성의 요구수준에 따라 화학성분이 결정된다. 본 연구에서는 1.0C, 0.2Mn, 3.8Cr, 1.5W, 8.5Co, 9.2Mo, 1.0V 조성의 M42 공구강의 미세조직과 열간가공성에 미치는 Si 함량의 효과를 체계적으로 조사하였다. 진공유도용해를 이용하여 Si 함량을 중량비로 0.33 에서 1.7% 까지 변화시켜 $140{\times}140{\times}330\;mm^{3}$ 크기의 잉곳으로 제조하였다. 이렇게 제조한 잉곳들은 $1150^{\circ}C$에서 1.5시간 동안 용체화처리한 후 노냉하였고, 이어 $1180^{\circ}C$에서 15mm 두께의 판재로 열간압연하였다. 공정 탄화물의 형상 및 분해 거동을 중심으로 미세조직을 관찰한 결과 Si 함량이 증가함에 따라 분해속도가 느려지는 것을 알 수 있었고 이로 인해 열간성형성이 급격히 저하되는 것으로 나타났다.

Effect of Granite Porphyry Supplementation on Growth Performance and Meat Sensory Quality in Broiler Chickens (맥반석의 첨가 급여가 육계에서 성장성적 및 관능적 특성에 미치는 영향)

  • This experiment was conducted to investigate the influence of dietary supplementation of granite porphyry (GP) on growth performances and meat sensory quality in commercial broiler chicks. A total of four hundred-fifty 5-day-old male broiler chicks were divided into 15 pens and fed one of five experimental diets for 5 weeks; 0% GP with antibiotics (Control), 1% GP with or without antibiotics, and 2% GP with or without antibiotics. Final body weight and daily weight gain of all GP supplemented groups were slightly higher than those of control. Feed conversion rate was improved in GP 1 % supplemented groups, although there was no significant difference. Feeding antibiotics in addition to GP did not influence the growth parameters. Relative weights of liver and abdominal fat tended to be reduced in broiler chickens fed GP supplemented diets. Feeding of 2% GP diet resulted in a significant improvement in meat sensory quality in terms of taste and tenderness(P < 0.05), but the effect of the 1% GP on meat sensory quality was not significant. The results of this study indicate that GP could be used as a favorable feed additive for production of sensory-enhanced broiler meats.

Preparation Technique of Thermostable Foam-Floater for High Temperature Engine Oil (고온 엔진오일용 내열성 발포부표 제조 기술)

  • When a domestically manufactured floater is used in a general gasoline or LPG car, or a flowmeter is floated long time in the engine oil under above $150^{\circ}C$, the floater will be swelled or its organization will be slackened because weight and volume will be changed due to the osmosis of fuel. In this study, we conducted a research on a manufacturing technique of a foam-floater with the small changse in weight and volume, oil-resistance, and thermal resistance in the high temperature engine oil. When the prepared floater TROF II-3, where Nitrile Butadiene Rubber (NBR) as basic material of the floater was superseded by Hydrogenated Nitrile Butadiene Rubber (HNBR), was floated for 100 h at the engine oil of high temperature ($150^{\circ}C$), the change rates of the weight and the volume were 2.90%, and 2.56%, respectively. These were less than the NBR (TROF I-3) case, where the change rates of the weight and the volume were 10.81% and 3.08%, respectively, Therefore, TROF II-3 was determined to be suitable as an engine-oil floater in high temperature because the change of weight and volume were small, the appearance, and the specific gravity of floater were maintained uniformly in the high temperature.
