Studies on the Antimicrobial Effect of Extracts of Propolis (프로폴리스 추출물의 항균 활성에 대한 연구)

  • Approved chemical preservatives have been widely used to preserve foods and increase their shelf life. There are increasing demends of the partial or complete removal of chemical preservatives from foods vecause of adverse health effect of chemicals. In this study, the possibility of natural antimicrobial compounds, Korean propolis as food preservatives are investigated. Propolis samples were extracted on various concentration of ethanol. Propolis extracts extracted with 100% ethanol showed the highest inhibitory effect aginst food spoilage microorganisms. The 100% ethanol extracts of propolis were selected and the antimicrobial activites of 100% ethanol extracts of proplis against several food spoilage microorganisms were examined. Bacillus subtilis, Micrococcus luteus, Escherichia coli, Staphylcoccus aureus, Bacillus cereus, Shigella sonnei, Salmonella choleraesuis, Erwinia rhapontici, and Vibrio parahaemolyticus as food poisoning microorganisms were chosen for the examination. The Propolis extracts had antimicrobial activity against food spoilage microorganisms. When the microorganisms were treated with propolis extracts, the population of food spolige microorganisms were decreased by 1~9 log.

Various Factors giving Impacts On the Satisfaction level of spectacle Wearers (안경원 이용자의 만족에 영향을 미치는 요인)

  • This study has been implemented to identify the level of satisfaction of the Glass spectacle wearers on various services being given by optical shop workers and help them to provide services of higher quality to the spectacle wearers. The objectives of this study are the spectacle wearers who visited to the 5 Designated optical shops located in Seoul area. All the data has been collected for the 2 months period (Oct. 1 through Dec. 1. 1999.) By using structured, self-described research form. All the collected data were 250 copies in total but evaluated 222 copies only as we excluded 28 copies which has uncertain reply/contents. Major results are as follows: 1. The proportions of objectives by sex was male 54.1%, female 45.9% and by Academic career was University graduates 35.1%, High school graduates 32.4%. 2. The factors impects on the satisfaction level of spectacle wearers were the Employee's service and the status of frame. 3. the factors impacts on the intention for revisit the optical shops were facility status, the responsiveness on wearer s opinion and the employee s services. The factors impect the wearers to build up intention to recommend the visited optical Shops to the 3rd costomer were employee s services, the status of frames, the facility Status and the price level they paid. Considering the results of this study above mentioned, in order to raise up the satisfaction level of glass spectacle wearers, it s thought that the employee (optician) of optical shops have to well acquainted fluent knowledge on the glass spectacles and detailed to the spectacle wearers in a gentle manner and let them build up the reliabily.

Antifungal activity of Streptomyces costaricanus HR391 against some plant-pathogenic fungi (여러 식물병원성 진균을 억제하는 Streptomyces costaricanus HR391의 항진균능)

  • In this study Streptomyces strains were isolated from soils and their antifungal activities and involved mechanisms were investigated. Among over 400 isolates of actinomycetes, Streptomyces costaricanus HR391 was selected as a potential antagonist to control several plant-pathogenic fungi. S. costaricanus HR391 inhibited mycelial growth of Fusarium oxysporum f. sp. raphani, F. oxysporum f. sp. niveum, F. oxysporum f. sp. lycopersici, and Rhizoctonia solani by 26.5, 26.2, 21.2, and 23.8%, respectively compared to those of uninoculated control after 7-day incubation on PDB medium. S. costaricanus HR391 produced $89{\mu}M$ of siderphore, and showed fungal cell wall-degrading activity including $0.46{\mu}mol/min/mg$ of chitinase and $0.83{\mu}mol/min/mg$ of ${\beta}$-1,3 glucanase. S. costaricanus HR391 secreted 87.49 mg/L of rhamnolipid, and produced 9.49 mg/L and 4.3 mM of lipopeptide, iturin A and surfactin, respectively, all they are membrane-disrupting biosurfactants. It also produced antimicrobial peptide and antibiotics phenazine. In addition to antifungal substances, S. costaricanus HR391 secreted plant growth-promoting phytohormones, zeatin, gibberellins and IAA. These results suggest that S. costaricanus HR391 may be utilized as an environment-friendly biocontrol agent against some important pathogenic fungi.

발효 콩 추출물의 항돌연변이원성 효과

  • 콩 발효식품은 예로부터 단백질 식품원으로서 뿐만 아니라, 식생활에서 없어서는 안되는 매우 중요한 식품 중의 하나였다. 발효식품에 대한 연구가 부진하였던 과거에는 콩 발효식품은 하나의 식품군으로서의 중요성을 가질 뿐 큰 관심의 대상은 아니었다. 그러나 최근에는 많은 연구자들이 콩 발효시 생성되는 기능성 성분 및 생리활성 효과를 점차 밝혀냄으로서 주목을 받기 시작하였다. 따라서 본 실험에서도 콩 발효에 의한 생리활성 효과를 알아보기 위해 Ames법에 의한 항돌연변이원 효과를 실험하였다. 콩 발효는 국산콩을 이용하여 메주에서 분리한 Bacillus sp. 와 Aspergillus sp.를 복합 발효시켜 동결건조 후, 분쇄하여 실험에 사용하였다. 제조된 발효 콩 분말은 일반분석을 행하였으며, 70% 에탄올로 3회 추출하여 감압농축 후, hexane, chloroform ethyl acetate, butanol 및 aqueous로 분획하여 동결 건조시킨 후, S. typhimurium TA98 및 TA100 균주를 이용한 유전자 복귀 돌연변이 시험을 실시하였다. 그 결과, 70% 에탄을 추출물과 각각의 분획물 자체의 돌연변이원성은 없었다. 또한 항돌연변이원 실험에서는 발암물질로서 직접 돌연변이원인 4NQO와 MNNG, 간접 돌연변이원인 Trp-P-1을 이용하였다. 특히 이들 발암물질 중 MNNG(0.4 $\mu\textrm{g}$/plate)의 경우 TA100 균주에서 ehtyl acetate 분획물에서 다른 분획물보다 높은 86.6%의 억제 효과를 나타내었으며, 대부분의 분획물에서도 70%이상의 억제효과를 나타내었다. 또한 각 분획물에서 농도 의존적으로 억제효과 역시 높았으며, 분획물에 따라 서로 다른 억제효과를 나타내었다.아 저장할 때 대비 저온저장고에서는 111일 동안에 11.7%의 중량감모가 발생하였으나, 신기술투입 저온저장고에서는 5.6%의 중량감모만이 발생하여 약 50%의 중량감모를 줄일 수 있었으며, 배의 색깔이나 경도도 대비구 보다 우수하였다. 4. 배를 비닐로 포장하여 대비 저온저장고에 저장한 경우와 비닐로 포장하지 않고 신기술투입 저온저장고에 저장한 경우를 비교할 때 11월~다음해 1월 까지는 중량감모, 과피색깔 및 경도에 큰 차이가 없었으나, 2월부터는 비닐로 포장하여 대비 저온저장고에 저장한 배의 품질변화가 급격히 증가되어 중량감모, 과피색깔 및 경도가 신기술 투입시 보다 급속하게 나빠졌다.를 저장 25일 경과시까지 유지하였다. 수확 시 높은 품온을 갖고 있는 과일을 산지에서 예냉 처리를 한 후 저온 냉장차를 이용하여 유통한다면 관행 유통 구조보다 고품질의 포도를 유통시킬 수 있는 것으로 사료되며 앞으로는 완숙된 고 당도(12.0~15.0Bx)$^{\circ}$ 포도를 수확 한 즉시 예냉 처리하고 저온 유통한다면 보다 신선한 과일을 소비자에게 전달 할 수 있을 것이다.갈변물질이 생성되었다. 이와 같은 결과로 볼 때, BAAG의 처리는 BAAC의 경우보다 가격은 저렴하면서도 항균력은 우수한 천연 항균복합제재로써 농산물 식품원료에 적용하여 선도유지 기간을 연장할 수 있는 효과를 기대할 수 있었다. 과일 등의 포장제로서 이용할 가능성을 확인하였다.로 [-wh] 겹의문사는 복수 의미를 지닐 수 없 다. 그러면 단수 의미는 어떻게 생성되는가\ulcorner 본 논문에서는 표면적 형태에도 불구하고 [-wh]의미의 겹의문사는 병렬적 관계의 합성어가 아니라 내부구조를 지니지 않은 단순한 단어(minimal $X^{0}$

Antioxidant and anticancer properties of hot water and ethanol extracts from the roots of Smilax china L. (발계(Smilax china L.) 열수 및 에탄올 추출물의 항산화 및 항암활성)

  • The biological activities of Smilax china L. rhizome (SCR), hot water (SCRW) and 70% ethanol extract (SCRE) were analyzed. The total phenolic contents of SCRW and SCRE were 51.7 and 100.5 mg/g, respectively. The measured flavonoid content of SCRW ($67.7{\mu}g/g$) was almost double that of SCRE ($31.7{\mu}g/g$). SCRE ($IC_{50}=42.4{\mu}g/mL$) exhibited stronger antioxidant activity in the DPPH system than the positive control ${\alpha}$-tocopherol ($71.3{\mu}g/mL$) or butylated hydroxy anisole ($53.8{\mu}g/mL$) did. SCRE ($IC_{50}=50.3{\mu}g/mL$) also showed stronger ABTS radical scavenging activity, as did ${\alpha}$-tocopherol ($67.1{\mu}g/mL$). The SOD-like activity and Tyrosinase inhibition activity of SCRW and SCRE showed almost the same pattern. The best SOD-like activity and tyrosinase inhibition activity were measured as 24.9% and 20.3% in SCRW at $1,000{\mu}g/mL$, respectively. The cytotoxic effects of the SCR extracts were analyzed via MTT assay on human cancer and normal cells. SCRW and SCRE did not show cytotoxicity up to the concentration of $1,000{\mu}g/mL$ against the normal human cell line HEK293. Against human breast cancer cells (MCF-7), SCRW inhibited MCF-7 growth (by 27.6%) better than the anticancer drug cyclophosphamide (15.5%) at $1,000{\mu}g/mL$. SCRE ($1,000{\mu}g/mL$) inhibited the growth of human lung cancer cells A549 (37.6%) and human stomach cancer cells AGS (53.6%) more effective than did SCRW (21.0% and 35.4%) or CPA (22.2% and 31.7%). These results suggest the potential use of SCRE and SCRW as an excellent antioxidant and antiproliferative substance, respectively.

Development of mixed Th1/Th2-type immune response in mice following immunization with GP63 from Leishmania donovani (내장리슈만편모충 유래 GP63 항원을 마우스에 접종한 후 관찰되는 Th1/Th2-type 복합 면역반응)

  • The $M_r$ 63,000 glycoprotein (GP63) and lipophosphoglycan (LPG) of Leishmania donovani were evaluated as vaccine candidates against visceral leishmaniasis. Mice were immunized with liposomeencapsulated GP63 and/or LPG that were purified from the soluble extract of L. donovani promastigotes, and were challenged with virulent amastigotes. Mice immunized with GP63/LPG in liposomes plus BCG resulted in a 27.4% reduction of amastigotes in the liver compared to the control group (liposomes plus BCG), and mice immunized with liposome-GP63 plus BCG failed to induce a protective immune response against the challenge infection. Immunization of mice with GP63 fused to the Schistosoma japonicum glutathione S-transferase (GP63-GST) plus BCG also failed to elicit protective immunity. To analyze the cause of failure to induce protection, cytokine release from the spleen cells of immunized mice and Leishmania-specific serum antibodies were analyzed. Spleen cells from mice immunized with GP63-GST plus BCG that were exposed to soluble extract of L. donovani in vitro produced 10-fold greater quantities of IFN-gamma and 3-fold greater quantities of IL-5 than cells from mice receiving BCG only or saline. Western blot analysis revealed that sera from these mice had Leishmania-specific antibodies recognizing 1 to 3 antigens of L. donovani with M. W. of 60-65 kDa. Although immunization of mice with GP63-GST induced a strong Th1 response, this study indicated that GP63-GST simultaneously elicited the Th2 response of the CD4+ T-cell, which was known to abrogate the protective immune response conferred by the Th1 effector function.

  • In order to clarify the removal of astringency in persimmon fruits (Diospyros koki L.) and its mechanism, a comparative histology of tannin cells in tile cultivars of astringent persimmon fruits (Sangju Dungsi. Daegu Bansi. Cheongdo Bansi) and a sweet persimmon fruit (Fuyu) was observed. Tannin cells were widespread in all fruits tissue expert for tissue of ovule before full blossom. The epidermal cells of ovary. flower and calyx consist of tannin cell. Arrangement of tannin cells has radiated type toward the upper directions in the calyx. The major part of seed coat consisted of tannin cells. The epidermal cells of persimmon fruits were consisted of small tannin cells, and the inner part of epicarp of the astringent persimmon was consisted of stone cells, but the sweet persimmon was consisted of parenchymatous cells. It was suggested that differantiation of tannin cell In persimmon fruits occured until about the middle of August. Some tannin cells of matured astringent persimmon fruits was coagulated and wall of tannin cell produced protuberance, and most of tannin cells of matured sweet persimmon fruits was coagulated or ruptured.

Occurrence of Weed Species on Turf Sod Production Areas in Jangsung-gun, Jeonnam Province (전남 장성지역 한국잔디 재배지 잡초발생 현황)

  • This study was conducted to provide basic information for weed control by surveying the occurrence of weed species in turf sod production areas. Surveys of weed species occurred in turf sod production areas were conducted in Jangsung-gun, Jeonnam province from September 2014 to May 2016. Total 50 sites of turf sod production areas in two soil conditions were investigated. On the upland soil condition in turf sod production areas, 66 weed species in 27 families were identified and classified to 49 annuals and 17 perennials and on the paddy soil condition, 69 weed species in 22 families were identified and classified to 53 annuals and 16 perennials. Based on the importance values, the most dominant weed species on the upland soil condition in the first survey (September 2014) was Digitaria ciliaris (8.49%), followed by Erigeron annuus (7.94%) and Rorippa indica (6.56%). In the second survey (May 2016) was Oxalis corniculata (7.26%), followed by Capsella bursa-pastoris (6.21%) and Conyza canadensis (6.21%). Whereas the most dominant weed species on the paddy soil condition in the first survey (September 2014) was Erigeron annuus (9.52%), followed by Mazus pumilus (7.41%) and Cyperus iria (6.82%). In the second survey (May 2016) was Commelina communis (5.08%), followed by Alopecurus aequalis (5.08%) and Erigeron annuus (4.79%). This information could be useful for estimation of future weed occurrence and effective weed control methods in turf sod production areas in Jangsung-gun, Jeonnam Province.

Distribution Patterns of Macrobenthos during Recent Summer Seasons at the Bongam Sand Tidal Flat of Masan Bay, Korea (마산만 봉암갯벌에 서식하는 대형저서동물의 하계 분포양상)

  • In this study, a series of survey were conducted to identify the distribution patterns of macrobenthos at the Bongam sand tidal flat in Masan Bay. We collected macrobenthos at 9 sampling sites twice in June and September of every year from 2012 to 2017 using a box core sampler (collecting area, $0.025m^2$). There was a total of 50 species with a community density of $6,388ind.m^{-2}$ and a biomass of $313.9g\;wet\;m^{-2}$ during the study period. Polychaetes had the highest number of species and density among the macrofauna, but the mollusks had the largest biomass. The number of species ranged from 10 to 25 during study period but increased to over 20 species in 2014. The density which ranged from $1,508ind.m^{-2}$ to $12,008ind.m^{-2}$ rapidly increased in 2015. The dominant species were all polychaetes such as Heteromastus filiformis, Prionospio japonicus, Hediste diadroma, and Neanthes succinea. The mean diversity index ranged from 1.2 to 1.9, richness index from 1.2 to 2.4, and evenness index from 0.5 to 0.9. From the cluster analysis results, there was a spatial difference in the similarity of faunal composition of macrobenthos and this pattern was maintained throughout the study period, that is, the temporal similarities were higher than the spatial similarities. There was a change in community composition from June 2014 to June 2015 in most of the sampling sites. During this time, the dominant species also changed from H. filiformis and N. succinea to H. filiformis and H. diadroma. The density of opportunistic species such as Capitella capitata and Polydora ligni decreased compared to the early 2000s while the population of H. diadroma increased from 2015. There was little ecological information on H. diadroma such as when and where this species occurred.

The taxonomic entity and distribution of Korean Sedum formosanum (Crassulaceae) revealed in 133 years (133년만에 밝혀진 한국산 주걱잎갯비름(돌나물과)의 분류학적 실체 및 분포)

  • Korean Sedum formosanum N. E. Br. (Ju-geog-nip-gaet-bi-reum in Korean) was first recorded in 1887, 133 years ago. Since then, the species has not been collected and its current state has remained unclear. However, these plants were collected in 2020 and the corresponding taxonomic entity and distribution status were revealed. It is known to be distributed only in the southern region of Japan, the northern islands of the Philippines, and in Taiwan, with Taiwan being the collection site of the type specimen. However, on the basis of the findings of the fourth national natural environment survey, it has recently been established that this plant also grows in the crevices of rocks along the seashores of the Korean islands of Hataedo Island and Sangtaedo Island, Sinan-gun, Jeollanam-do. S. formosanum inhabiting Korea is a large succulent biennial that can attain a height of up to 65 cm and differs from its congeneric species in having erect follicles during the fruiting period. Notably, among the Korean Sedum species, S. formosanum is most similar to S. tosaense, although it can be distinguished from this species with respect to its monomorphic leaves that have rounded apices, and it also bears separate flowering and sterile stems. In this paper, we present a description and photographs of the Korean S. formosanum, indicate the differences between this and related species, and provide a key to related taxa.
