Effect of Addition of Lactic Acid Bacteria on Quality of Rye Silage Harvested at Early Heading Stage (젖산균 첨가가 출수초기 호밀 사일리지의 품질에 미치는 영향)

  • This study was conducted to investigate the effect of addition of lactic acid bacteria inoculants on quality of rye silage harvested at early heading stage. The nutritive values in lactic acid bacteria (LAB) inoculated group showed similar results to control. The pH of rye silage in LAB inoculation significantly decreased as compared to control (p<0.05). In addition, the content of lactic acid in LAB inoculation significantly increased (p<0.05), but the content of acetic acid in LAB treatments decreased. In addition, lactic acid bacterial counts in LAB inoculation significantly increased as compared to control (p<0.05). Therefore, we suggest that rye silage could be improved by LAB inoculation.

  • This study aimed to investigate the impact of enhancing the electrode conductivity and mitigating the production of hydrogen peroxide - a by-product arising from lactate oxidation - on the performance of lactate electrodes. The electrical conductivity of the electrode was improved by modifying the surface of carbon paper with single-walled carbon nanotubes. Catalase was introduced to effectively eliminate the hydrogen peroxide produced during the lactate oxidation reaction. The carbon paper electrode, with simultaneous immobilization of both lactate oxidase and catalase, yielded a current 1.7 times greater than the electrode where only lactate oxidase was immobilized. The electrode in which lactate oxidase and catalase were co-immobilized on the surface of carbon paper modified with single-walled carbon nanotubes, produced a current of 171 µA, which was more than twice as much current as the carbon paper with only lactate oxidase immobilized. The optimized electrode showed a linear response up to lactate concentration of 20 mM, confirming that it can be used as a sensor electrode.

  • This study was conducted to investigate the effect of cocoon silk fibroin hydrolyzate(CSFH) on shelf-life extension of kimchi. CSFH was added in kimchi in the concentration of 0.3%(w/w), 0.6% and 0.9%. Kimchi with CSFH showed higher pH and lower titratable acidity than control and it was supposed that the growth of lactic acid bacteria was inhibited by CSFH. In model system, for complex lactic acid bacteria, CSFH showes antimicrobial activity.

  • $M_4$-LDH isozyme was partially purified from the skeletal muscle of Agkistrodon blomhoffii brevicaudus. The protein was injected into rabbits and the resulting antiserum was tested for reactivity with crude preparations of LDH isozymes of fifteen vertebrate species. Antisera against $M_4$-LDH isozyme of A. blomhoffii brevicaudus reacted very strongly with the LDH isozymes, except the $H_4$-LDH isozyme, of A. saxatilis and A. caliginosus but weakly with those of Rhabdophis tigrinus at fixed conditions. A. caliginosus showed a difference in the immunodiffusion test and was considered to be a species less related to others of genus Agkisrodon. The suggestion that the H and M lactate dehydrogenase subunits are immunclogically distinct has been reaffirmed in the present study.

  • Accumulation of toxic waste byproducts in hybridoma culture can limit cell growth and monoclonal antibody (MAb) productivity, and one of the major toxic metabolites is lactate produced via glycolytic pathway of glucose metabolism. The factors affecting the glucose to lactate conversion rate were investigated. The conditions of high initial glucose concentration and high growth rate stimulated glucose to lactate conversion rate. The glucose-controlled fed-batch culture was investigated, and 19% reduction in lactate formation and 41% enhancement of MAb titer could be achieved by fed-batch culture.

  • TLC condition was developed for its simple separation and quantitative analysis of lactic acid. Rapid and clear separation of lactic acid by silica gel TLC plate was obtained by using nitromethane : 1-propanol : $H_2O$ (2 : 5 : 1.5, v/v/v) and a suitable dipping solution of 40 mg bromocresol purple in 100 mL 5% ethanol (pH 10.0). The lactic acid was shown as a bright yellow spot on a light cinnabar background. The quantitatively detectable concentration range of lactic acid was between 0.5 and 4% with 99.4%, confidence. Quantitative TLC analysis result was confirmed with HPLC and with enzymatic Quantitative analysis methods (by using lactate dehydrogenase).

  • 효모와 젖산균에 의해 발효된 소위 sourdough빵은 천연의 고유 향미와 그리고 건강식품이라는 인식과 더불어 계속 소비가 증가되고 있는데, 이것은 젖산균에 의해 생성된 젖산과 아세트산에 의해 약간의 신맛과 독특한 풍미가 제공되고 저장성이 향상되기 때문이다. 이에 본 연구에서는 냉동 빵의 품질 향상을 목적으로 sourdough의 냉동반죽 특성을 조사하였다. soudough용 wheat brew 제조에는 L.plantarum과 Lbrevis혼합균주(0.1%)의 발효상태가 양호하였으며 wheat brew를 첨가한 경우 farinouam 특성중 반죽의 stability가 감소되어 반죽의 발효시간이 단축된 short time법을 반죽제법에 적용하였다.   • 유포자 젖산균 Bacillus sp의 식품이용을 위한 기초로서 유포자 젖산균의 발육, 포자형성 등에 관하여 연구한 결과는 다음과 같다. 탄수화물 중 dextrin이 좋았고, Mn, Ca ion, biotin 등은 생육, 포자형성, 내열성에 대하여 효과가 좋았다. 배양의 최적조건은 호기조건하에서 $37~45^{\circ}C에서$ 36시간 이상 배양하는 것이었다.

  • Lactobccillus acidophilus와 Saccharomyces cerevisiae를 대두유에서 혼합 배양시켜 최적 젖산발효조건을 검토하였다. 대두유에서의 혼합배양시 20시간 이상 배양했을 때 산생성이 촉진되었고 배양온도는 34$^{\circ}C$ 부근으로 했을 때 가장 좋았다. 대두유에 sucrose와 skim milk가 젖산발효에 미치는 영향을 조사한 결과 sucrose 는 1.5% 첨가시 skim milk는 2.0% 첨가시 첨가하지 않고 배양할 때 보다 2배 정도의 산생성을 보였다.

  • 질염 환자의 질분비물 도말에서 광학현미경상에서 고배율로 관찰한 결과 정상 상피세포에서는 질내 정상 pH를 유지하기 위해 무수한 젖산균의 존재를 확인할수 있었다. 그러나 저배율, 고배율의 광학현미경상에서 진균에 감염된 세포 주변에서 젖산균(lactobacilli)은 관찰되지 않았다. 주사전자 현미경 상에서 Candida albicans는 기능을 탈락한 상피세포의 세포질에 기생하며, biofilm의 성장환경을 만들며, budding을 통해 2차 딸세포를 형성하며, 성장과 증식을 끊임없이 반복함을 확인하였다. 성장과 재생이 주기적으로 일어나지 않은 면역력이 약화된 임산부나, 호르몬의 부족으로 인한 위축성 질염 환자는 상피세포층이 떨어지면서 나오는 글리코겐을 이용해 질내에 존재하는 lactobacilli균이 젖산을 풍부하게 만들 수 없으므로 감염증 발생이 더 높아진 것으로 사료된다.
