Morphology and Histology of the Olfactory Organ in the Korean Endemic Species, Pseudobagrus koreanus (Cypriniformes, Cyprinidae) (한국 고유종 눈동자개 Pseudobagrus koreanus 후각기관의 형태 및 조직학적 특성 연구)

  • The olfactory organ of a Korean endemic species, Pseudobagrus koreanus, was investigated to describe its morphology and histology and analyze correlation between habitat environment and its ecological habits and characters, using a stereo microscope, a light microscope, and a scanning electron microscope. Its external morphology showed tubular anterior nostril at the tip of the snout and slit posterior nostril at the base of the nasal barbel. The olfactory chamber showed the rosette structure consisting of numerous lamellae radial and parallel to the medium raphe. The olfactory lamella has the sensory epithelium with olfactory receptor nuerons, supporting cells, basal cells, lymphatic cells, plasma cells and the non-sensory epithelium with stratified epithelial cells, mucous cells, lymphatic cells. In particular, 1) 41~43 (n=20) lamellae, 2) lymphatic cells and plasma cells, 3) mucous cells including neutral polysaccharid may be olfactory characteristics relevant to a stagnant or a standing habitat environment, nocturnal habit dependent on olfaction, and taxonomic characters compared at least to other catfish species.

Differential Potential of Stem Cells Following Their Origin - Subacromial Bursa, Bone Marrow, Umbilical Cord Blood - (줄기세포의 분화능의 기원에 따른 비교 - 견봉하 점액낭, 골수, 탯줄 혈액 -)

  • Purpose: To evaluate the differentiation potential of stem cells and their immunophenotype from 3 different sources. Methods: Our study involved three stem cell sources-subacromial bursal tissue, bone marrow, and umbilical cord blood. We obtained the subacromial bursal tissue and bone marrow from the patients undergoing shoulder surgery. After collecting the sample, we applied specific induction media for neurogenic, adipogenic and osteogenic differentiation. Also, flow-cytometry analysis was done to reveal the cell surface antigens. Results: We obtained 100% (8 cases) neural and adipogenic differentiation, but 62.5% (5 of 8 cases) osseous differentiation among the subacromial bursal tissue group. Bone marrow derived cells showed 100% neural (6 cases) and adipogenic (5 cases) differentiation, but 80% (4 of 5 cases) osseous differentiation. Umbilical cord blood derived cells revealed 97% (65 of 67 cases) neural, 53.7% (29 of 54 cases) adipogenic and 68.4% (39 of 57 cases) osseous differentiation. Immunophenotype analysis revealed that surface markers of bone marrow, subacromial bursal cell and umbilical cord blood derived mesenchymal stem cells are different from each other. Conclusions: Mesenchymal stem cells are potential agents in regenerative medicine and are characterized by expression of surface markers and by their differentiation potential. Our study with stem cells from subacromial bursal tissue, bone marrow and umbilical cord discovered that each stem cell has unique differentiation potential and function based on its origin. Various stem cells show multi-lineage differentiations in vitro which can be correlated to in vivo conditions.

  • 본 논문은 간의 비침습성 점액성 낭성종양의 자동화된 진단을 위한 정량적인 형태학적 추출방법에 대하여 설명한다. 본 논문에서 제안된 형태학적 추출방법은 간의 비침습성 점액성 낭성종양 진단을 위한 병리적인 특징들에 기반을 둔 상피세포 형태학적 특징(원주형과 입방형)과 악성 직전에 나타나는 배상세포 출연을 판단하는 특징, 그리고 주변상피 조직에서 나타나는 기질 여부를 조사하기 위한 핵-비율의 측정방법을 소개한다.

Histochemical Study of the Intestinal Mucosubstances in Sebastes schlegeli, Halichoeres poecilopterus, Bryzoichthys lysimus and Takifugu pardalis (조피볼락, 용치놀래기, 송곳니베도라치 및 졸복 장관 점액질에 대한 조직화학적 연구)

  • The conventional histochemical staining were used to study mucosubstances properties of intestinal striated border and goblet cells in four teleostean species, i. e., Sebastes schlegeli, Halichoeres poecilopterus, Bryzoichthys lysimus, and Takifugu pardalis. The following methods were used; PAS, AB pH 2.5, AB pH 1.0, AB pH 2.5-PAS, AF pH 1.7-AB pH 2.5 and HID-AB pH 2.5 stains. The mucosubstances of striated border in the proximal intestine and rectum of Sebastes schlegeli contained with neutral mucin, middle and distal intestine contained with neutral mucin and acid mucin. The striated border of all the intestines of Halichoeres poecilopterus contained with neutral mucin and acid mucin, and those of Bryzoichthys lysimus and Tnkifugu pardalis contained with neutral mucin only. The amounts of neutral mucin were moderate to considerable in Sebastes schlegeli and Halichoeres poecilopterus, minimal to small in Bryzoichthys lysimus and Tnkifugu pardalis. The amounts and properties in mucosubstances of intestinal goblet cells showed differences in species and regions. The intestinal goblet cells of Bryzoichthys lysimus, and Tnkifugu pardalis contained neutral mucin only while Sehastes schlegeli and Halichoeres poecilopterus contained mixture of neutral mucin, sulfomucin and sialomucin. The amounts of neutral mucin were considerable to large in distal intestine and rectum of Tnkifugu pardalis, while moderate to considerable in all intestines of Sehastes schlegeli, all the intestines except for middle intestine of Bryzoichthys lysimus, and proximal and middle intestine of Tnkifugu pardalis. Also it was minimal to small in middle intestine of Halichoeres poecilopterus. The intestinal goblet cells of Sehastes schlegeli contained mixture of minimal amounts of strong sulfmucin, weak sulfomucin and minimal to small amounts of sialomucin, and those of Halichoeres poecilopterus except for rectum contained mixture of minimal to small amounts of strong sulfomucin and sialomucin.

  • Morphology of the gill in Lateolabrax japonicus was investigated after staining the gill, as a result, we found the gill is composed of gill raker, gill arch, gill filament and gill lamellae. The number of gill raker was 7~10 in the upper and 13~18 in the lower. Ultrastructure of the gill in Lateolabrax japonicus were examined by means of the light and transmission electron microscopes. The gill have primary filament and secondary filament (lamellae). The following cells are identified and described: pavement cell, pillar cell, blood cell, mucose cell and chloride cell etc. Simple epithelial layer consists of squamous epithelium contained large nucleus, intracellular organelles etc. and the surface is covered with some of microridges. The lamella pillar structures are characterized by the axial microtubules and lateral membrane interdigitations. The mucous cells were globular in shape, and had almost the mucous granules of same size with various electron density. Chloride cells contain a lot of mitochondria and specifically developed tubular systems.

The Alleviating Effects of Green Tea on N-Nitrosodimethylamine Toxicity to Mucins in the Rat Lingual Salivary Glands (흰쥐 혀침샘의 점액질에 미치는 N-Nitrosodimethylamine 독성에 대한 녹차의 완화 효과)

  • This study was performed to determine alleviating effects of green tea (GT) on the N-nitrosodimethylamine (NDMA) toxicity to mucins in the rat lingual salivary glands. Sprague-Dawley rats were divided into five groups: untreated (Control), one week-NDMA administrated (NDMA), one week-GT administrated after NDMA for one week (NDMAGT), one week-NDMA administrated with GT (GTNDMA), and one-week NDMA administrated with GT after GT for one week (GTGTNDMA). The mucin properties were elucidated by periodic acid Schiff (PAS) reaction, alcian blue (AB) pH 2.5, AB pH 2.5-PAS, aldehyde fuchsin (AF) pH 1.7-AB pH 2.5, and high iron diamine (HID)-AB pH 2.5 staining. The lingual von Ebner's glands of NDMA group showed some changes such as contraction and destruction of the serous acini, decrease and disappearance of the cytoplasmic granules, vacuolation of cytoplasm, and the mucigenous duct cells. Also, in the lingual mucous glands, enlarged lumens, fused acini, disappearance of granules, vacuolation of cytoplasm, and mucigenous duct cells were observed. All the GT administrated groups had a tendency to recover. GTGTNDMA group recovered almost up to the state of the control group. The lingual von Ebner's glands of NDMA group showed a decrease of neutral mucin and the lingual mucous glands showed a decrease of both neutral and acidic mucins decreased in comparison with control group, although mucous acini secreting strong sulfomucin decreased while those secreting non-sulfomucin (sialomucin) increased. All the GT administrated groups had a tendency to recover. The degree of recovery in the GTNDMA group was stronger than in NDMAGT group and that of the GTGTNDMA group was almost up to the state of the control group. Plus, lingual von Ebner's glands of GTGTNDMA group contained much more neutral mucin than in the control group. In conclusion, it is suggested that NDMA exhibit the toxicity which affects on the mucin properties and which green tea alleviates.

Effects of Sulfur Dioxide Exposure in Histological Structure and Mucosubstances of the Nasal Respiratory Mucosa of Rat ($SO_2$ 흰쥐 비강 호흡부 점막의 조직학적 구조 및 점액질에 미치는 영향에 관한 연구)

  • This experiment was performed to investigate the effects of sulfur dioxide on the histological changes, properties of mucosubstances and glycoconjugates of the nasal respiratory mucosa in the rat. Sprague-Dawley male rats weighing about 200~250g were divided into a control group and SO$_2$ exposed groups. Again SO$_2$ exposed groups were divided into 10 ppm, 25 ppm, 50 ppm, 100 ppm, and 200 ppm subgroups, according to concentrations of SO$_2$ and each SO$_2$exposed groups were divided into 1, 3 and 6 hours groups. For the histological changes, hematoxylin-eosin(H-E) and periodic acid Schiff's(PAS) stainings were used, and for the properties of mucosubstances, PAS, alcian blue (AB) pH 2.5, pH 2.5-PAS, AB pH 1.0 and aldehyde fuchsin (AF) pH1.7-AB pH 2.5 were used. In all the SO$_2$ exposed groups, loss of cilia and detachment of epithelial cells, vacuolation of goblet cells were observed in the respiratory epithelium while epithelial squamous metaplasia and intraepthelial mucous cells were observed in the higher concentration of SO$_2$ and the degree of the loss cilia was higher according as concentration was higher and exposed time was longer. The intraepitheial mucous cells appeared most remarkable in the 50 ppm SO$_2$ exposed group. The numbers of goblet cells and acini of nasal septal gland were varied according to concentration of SO$_2$ and exposed time, but the numbers in the 25 ppm and 50 ppm, SO$_2$ exposed increased remarkably. However, the numbers in the 100 ppm and 200 ppm SO$_2$ exposed group had a tendency to decrease noticeably, or disappeared.

Mucous Cells and Their Structure on the Epidermis of Five Appendages in the Korean Flat- headed Goby, Luciogobius guttatus (Pisces; Perciformes) (한국산 미끈망둑 Luciogobius guffafus(농어목) 5개 부속지의 표피구조 및 점액세포)

  • The flat-headed goby, Luciogobius guttatus, inhabits tidepools and river mouths, and stays under stones on the dried bottom for the duration of the low tide. To know the relationship of its respiration and habit in this fish, the epidermis of five appendages was observed. The epidermis has three layers: the outermost layer, middle layer and stratum germinativum. The outermost layer is composed of polygonal cells or rather flattened cells, and mucous cells. The unicellular mucous cells showing acid mucopolysaccharides are 11.1 to $16.1{\mu}m$ in mean height and in one or two rows. The middle layer consists mainly of large epidermal cells that are swollen by adjacent epidermal cells and arranged in a web-shaped structure. The swollen cells are 12.3 to $15.2{\mu}m$ in mean height and arranged in one to 11 layers. Since the swollen cells occupy the entire height of the epidermis, the epidermis is thick. A large number of blood capillaries are present just below the stratum germinativum. Taste buds are distributed at intervals on the surface of the epidermis. Based on these epidermal strucures, it is likely that L. guttatus utilizes cutaneous respiration in a dual respiratory systems.

Isolation of Dispersed Mutants from Wild Myxobacteria. (분산 돌연변이 점액세균의 분리)

  • Clumping of cells is one of the major obstacles to culture wild myxobacterial strains in liquid media. In an effort to solve this problem, we tried a method isolating spontaneous mutants that grow dispersed in liquid media from a wild myxobacterial strain. Myxococcus stipitatus KYC1001, a newly isolated strain from Gyearyongsan National Park in Korea, clumps and sticks to the surface of culture vessels as other wild myxobacteria behave in liquid media. Taking an advantage of the characteristics that dispersed mutant cells would grow dispersed while most other wild type cells would clump and stick to the surface of culture vessels, spontaneous dispersed mutants were enriched by repeated subculturing of culture supernatant. A resultant mutant, KYC2001, did not form any clumps nor stick to the surface of culture flasks, but grew completely dispersed in liquid. Meanwhile, three other spontaneous mutants, KYC2002, KYC2003, and KYC2004, shelved partially dispersed phenotype. A major portion of the cells grew dispersed in liquid but they still formed some clumps.
