Marginal fit of three-unit zirconia anterior fixed dental prostheses fabricated using CAD/CAM and MAD/MAM system (CAD/CAM과 MAD/MAM 시스템으로 제작된 3-unit 지르코니아 고정성 국소의치의 변연 적합도에 관한 연구)

  • Purpose: The purpose of this study was to compare the marginal fit of three-unit zirconia fixed dental prostheses (FDPs) fabricated using CAD/CAM and MAD/MAM system. Materials and methods: Dentiform maxillary central and lateral incisor were prepared for 3-unit FDP and fixed in yellow stone. This model was duplicated to epoxy resin die. On the resin die, fifteen 3-unit FDPs were fabricated. Metal-ceramic group was three-unit metal-ceramic FDPs, $Everest^{(R)}$ group was zirconia three-unit FDPs fabricated using the $Everest^{(R)}$ system (Kavo Dental GmbH, Biberach, Germany) and $Rainbow^{TM}$ group was zirconia three-unit FDPs fabricated using the $Rainbow^{TM}$ system (Dentium Co. Inc., Seoul, South Korea). They were cemented to resin dies with adhesive resin cement. After removing pontics, each retainers were separated and observed under measuring machine (Presize 440C) and analyzed through one-way ANOVA and Duncan test (${\alpha}$ = .05). Results: Mean values and standard deviations of marginal gap dimensions in each group for three-unit FDPs were $78.5{\pm}11.05\;{\mu}m$ for the metal-ceramic group, $59.30{\pm}11.63\;{\mu}m$ for the $Everest^{(R)}$ group and $70.34{\pm}13.98\;{\mu}m$ for the $Rainbow^{TM}$ group. Conclusion: 1. The $Everest^{(R)}$ group in comparison with metal-ceramic group showed better marginal fit, which had significant differences P<.05. 2. The mean marginal gap values between $Everest^{(R)}$ and $Rainbow^{TM}$ group did not showed significant differences (P>.05). 3. The mean marginal gap values between $Rainbow^{TM}$ group and metal-ceramic group did not showed significant differences (P>.05). 4. The mean marginal gaps of each group were within clinically acceptable range ($120\;{\mu}m$).

  • It is the analyses of replies showed n the questionnaire consisted of four kinds of matters to see level of knowledge among SELIS (SEoul Women's University Library and Information System) OPAC users of keyword/boolean search. The result of this analyses is : in SELIS search, users who prefer keyword search than any other, who satisfy work of retrieval by means of boolean operator, and who think it easier, show lusher level of knowledge than those who deny it in the questionnaire. Knowledges Presented in the survey are ; characteristics of keyword search, single or double keys, using boolean operator in keyword, knowledge of index, knowledge of stop list, uncontrolled term. keyword search technique, right truncation, correct application of boolean logic operator, and selecting major subject in keyword browsing. The above mentioned knowledges will work as important factors n keyword/boolean search, OPAC. For successful search it requires conceptional knowledge of information retrieval processing, or inquiry word transformation how to search required information, and semantic ability to get result questioned In the given system, when and how to apply the characteristics of the system, and scientific record for user's inquiry, or fundamental computer technology and syntax knowledge to make search word in detail. But so far now important knowledge considered as user's online index search, has been emphasized on knowledge of scientific record, and has been lag of semantic and conceptional knowledge. So, it is recommendable for online index user to train to concentrate semantic knowledge, syntax ability, and conceptional knowledge, rather than scientific technique too much.

MICROLEAKAGE OF 2-STEP ADHESIVE SYSTEMS IN DIAMOND-PREPARED CAVITY (다이아몬드 기구로 삭제한 와동에서 2단계 접착제의 미세누출 비교)

  • The purpose of this study was to compare the marginal microleakage of different 2-step adhesive systems in Class V cavities prepared with different diamond points. Forty Class V cavities were prepared with two different (coarse or fine) diamond points on cervical third of extracted molars. The occlusal and gingival margin of cavities was located in enamel and dentin, respectively. They were divided into one of four equal groups (n = 10) and ; Group 1-prepared with coarse diamond point (EX-41), restored with Single Bond and Z 250, Group 2-prepared with fine diamond piont (TF-21F), restored with Sing)e Bond and Z 250, Group 3-prepared with coarse diamond point (EX-41), restored with Clearfil SE Bond and Clearfil AP-X, Group 4-prepared with fine diamond point (TF-21F), restored with Clearfil SE Bond and Clearfil AP-X. Specimens were thermocycled, immersed in a 2% methylene blue solution for 24 hours, and bisected longitudinally. They were observed leakages at enamel and dentinal margins. Data were analyzed using Mann-Whitney and Wilcoxon signed ranked test. In this study, marginal microleakage or Single Bond was not affected by type of diamond points. But Clearfil SE Bond showed higher marginal microleakage at both enamel and dentinal margin when Class V cavity was prepared with coarse diamond point.

COMPARISON OF BOND STRENGTH OF A FIBER POST CEMENTED WITH VARIOUS RESIN CEMENTS (다양한 레진시멘트로 합착한 섬유포스트의 결합강도 비교)

  • The purpose of this study was to compare the push-out strength of a fiber post cemented with various resin cements. Newly extracted 36 human mandibular premolars which had single root canal were selected and their crown portions were removed. The root canal was instrumented using $PROTAPER^{TM}$ system and obturated using continuous wave technique. In each root, a 9-mm deep post space was prepared. #2 translucent fiber post (DT Light post, Bisco Inc., Schaumburg, IL, U.S.A.) was cemented using injection technique with Uni-dose needle tip (Bisco) and six different resin cements. The tested resin cements were Duo-Link (Bisco Inc., Schaumburg, IL, U.S.A.), Variolink II (Ivoclar-Vivadent AG, Schann, Liechtenstein), Panavia F (Kuraray Medical Inc., Okayama, Japan), Multilink Automix (Ivoclar-Vivadent AG, Schann, Liechtenstein), RelyX Unicem (3M ESPE Dental Products, St. Paul, MN, U.S.A.), and Maxcem (Kerr Co., CA, U.S.A.). After storage in distilled water for 24 hours, each root was transversally sectioned into approximately 1-mm thick sections. This procedure resulted in 6 serial sections per root. Push-out test wasperformed using a universal testing machine (EZ Test, Shimadzu Co.) with a crosshead speed of 1 mm/min. The data were analyzed with one-way ANOVA and Tukey HSD (p=0.05). The push-out strength of the groups which cemented fiber post with Panavia F and Multilink Automix were lower than those of the other groups. But, there were no statistically significant difference among groups at a probability level of 0.05.

Variation of Harbor Oscillations in Yeongil Bay (영일만 항만에서의 부진동 변화에 관한 연구)

  • Today the harbor oscillation problems are the most significant factor to design harbors serving the very large ships. Large vessels moored in the elastic hawsers at the coastal harbors are often displaced due to the resonance between the long period waves and mooring system. The cargo handling may be interrupted and the hawsers may be broken, especially when the amplification becomes remarkable. The most significant harbor which is confronted with harbor oscillation problem in Korea is the Pohang New Port. Many cases of problems are being notified by the pilot association and local office of MOMAF. However, it is difficult to prevent the arrival of long waves musing oscillation within this harbor. Moreover, government already started new port plan at the mouth of Yeongil Bay without treating problems occurred in the Pohang New Port. This study deals with the variation of harbor oscillation due to the construction of 4.1km breakwater at the bay mouth and new port plan. Numerical method used are fairly standard form from the extended mild slope equation The obtained numerical results were compared with the field measurement from the previous study and this will bring a certain level of discussion and consideration of variation in the future port development.

In vitro propagation of oil palm (Elaeis guineensis Jacq.) clones through somatic embryogenesis and analysis of somaclonal variation by RAPD (체세포배발생을 통한 오일팜나무(Elaeis guineensis Jacq.) 클론의 기내증식 및 RAPD를 이용한 체세포변이의 검정)

  • This study was carried out to develop reliable systems for somatic embryogenesis in oil palm tree (Elaeis guineensis Jacq.), and to verify the somaclonal variants by RAPD analysis. Embryogenic callus was induced successfully on modified half-strength MS medium containing $NaH_2PO_4{\cdot}2H_2O$ and casein. Embryogenic callus was further developed to somatic embryo mass (SEM), which is very hard and bonded tightly each other. Plantlets were proliferated when SEM was cultured on modified MS medium containing half strength $NH_4NO_3$, casein and L-ascorbic acid. Plantlets were transplanted into pots containing artificial soils. When RAPD analysis was conducted using randomly selected 95 in vitro plantlets and 19 random primers, somaclonal variation was detected using BNR35 primer. There was missing band around 1 kb in #22, #28, #35, and #77 plantlets. In addition, bands obtained from #28, #35, and #77 was much stronger than other normal bands. The blast results at NCBI revealed that somaclonal variation observed in this study was related to chloroplast genome of oil palm. The results also revealed that oil palm reproduction system through somatic embryogenesis is quite reliable and early detection of somaclonal variants seem to be possible at in vitro stage by RAPD analysis.

Biosynthesis of recombinant human prominiinsulin in E. coli and plant systems (대장균과 식물시스템에서 재조합 인간 prominiinsulin 생합성 분석)

  • Recently, the number of people with diabetes is rapidly increasing, coupled with the fact that the insulin market is remarkably increasing. Therefore, molecular farming for plant-derived pharmaceutical protein production is reported as becoming more attractive than ever. In this study, we carried out experiments step by step for development of recombinant insulin constructs, which were transformed into E. coli system, in vitro transcription and translation system, and tobacco cells. At first, recombinant proinsulin protein was successfully produced in in vitro transcription and translation system with wheat germ extract. After which, recombinant construct of prominiinsulin encoded a fusion protein of 7.8 kDa with trypsin cleavage sites at N terminus and C terminus of minimized C-peptide was tried to in vitro expression using E.coli culture. After purification with His-tag column, the resulting recombinant prominiinsulin protein was processed with trypsin, and then checked insulin biosynthesis by SDS-PAGE and western blot analysis with anti-insulin monoclonal antibody. The immunoreactive product of trypsin-treated miniinsulin was identical to the predicted insulin hexamer. The construct of 35S promoter-driven preprominiinsulin recombinant gene with signal peptide region for ER-targeting and red fluorescence protein gene [N terminus ${\rightarrow}$ tobacco E2 signal peptide ${\rightarrow}$ B-peptide (1-29 AA) ${\rightarrow}$ AAK ${\rightarrow}$ A-peptide (1-21 AA) ${\rightarrow}$ RR ${\rightarrow}$ His6 ${\rightarrow}$ KDEL ${\rightarrow}$ C terminus] was transformed into BY-2 tobacco cells. A polypeptide corresponding to the 38-kDa molecular mass predicted for fusion protein was detected in total protein profiles from transgenic BY-2 cells by western analysis. Therefore, this recombinant preprominiinsulin construct can be used for generation of transgenic tobacco plants producing therapeutic recombinant insulin.

High-level Expression of Human Procaspase-9 in Escherichia coli and Purification of its GST-tagged Recombinant Protein (대장균을 이용한 세포사멸 유도 단백질 caspase-9의 발현에 관한 연구)

  • Human caspase-9, an essential apoptosis initiator protease, was excessively degraded when expressed in Escherichia coli under the conventional induction condition. To optimize the conditions for induction and develop a rapid purification method for obtaining significant amounts of wild-type procaspase-9, we expressed procaspase-9 as GST fusion in E. coli. The addition of 0.01 mM IPTG as an inducer to the bacterial culture and decreasing the culture temperature to 25oC improved the production of procasapse-9 protein by circumventing proteolytic degradation in E. coli. The wild-type procaspae-9 was purified to approximately 70% purity with relatively high yields using the method developed in this study. In addition, we found that GST-caspase-9 is autocatalytically cleaved after aspartic acid 315, which is the same site for processing in mammalian cells, during expression in E. coli.

The Expression Patterns of Human Parkin in E. codi and Mammalian Cells (대장균과 포유류 세포 내에서 parkin의 발현 양상에 관한 연구)

  • Parkin, known as an E3 ubiquitin ligase, has essential role in protein quality control, and its severe dysfunction leads to neurodegenerative disorders. Human Parkin was excessively degraded when expressed in Escherichia coli under the conventional induction condition ($37^{\circ}C$ culture condition with 0.5 mM IPTG). To optimize the induction and culture conditions for recombinant human Parkin and develop a rapid method for the Parkin purification, we expressed Parkin by using PCEX system at the different culture temperatures and IPTC concentrations. The intact Parkin protein was purified to approximately $90\%$ purity with suitable amounts of protein under the optimal culture condition ($25^{\circ}C$E with 0.01 mM IPTG). Additionally, we constructed various parkin plasmids with different tagging systems and investigated their expression patterns in HEK293 cells. We found that the proteolytically sensitive site is localized within a ubiquitin-like domain of Parkin. This study developes a method for generating useful reagents to investigate biochemical properties of Parkin.
