A fMRI study on the cerebral activity induced by Electro-acupuncture on Sp9(Yinlingquan) (음릉천(陰陵泉)(Sp9)의 전침자극(電鍼刺戟)이 functional MRI상 뇌활성(腦活性) 변화(變化)에 미치는 영향(影響))

  • Objective : The meridian theory in oriental medicine explains that Sp9(Yinlingquan) has been considered very important for gynecological disorders, spleen and stomach disorders, and water metabolic disorders. Functional MRI is the best method of showing the evidences of the effect of the Electro-acupuncture treatment through scientific methods. On this supposition, Electro-acupuncture stimulation on Sp9(Yinlingquan) was carried out on six healthy volunteers, using the gradient echo sequence with the 3.0T whole-body MRI system(ISOL). Methods : Electro-acupuncture stimulation on Sp9(Yinlingquan), the experiment was carried out on six healthy volunteers, using the gradient echo sequence with the 3.0T whole-body MRI system(ISOL). After the needle insertion on right Sp9(Yinlingquan), 2Hz of electric stimulation was given for 30 seconds, repeated five times, with 30 seconds' intervals. The Image analysis including motion correction, talairach transformation, and smoothing was done using SPM99. Results : The electro-acupuncture stimulation on Sp9(Yinlingquan) activates Brodmann Area 13, 19, 22, 39, 40, 47 which may be the central pathways of the electro-acupuncture stimulation on Sp9(Yinlingquan). We also hypothesize that those brain areas may be directly or indirectly related with the mechanism of the effect of electro-acupuncture stimulation on Sp9(Yinlingquan).

  • Objectve : Recent stuides suggested that there is a strong correlation between acupuncture stimulation and its cortical activation. Another study showed that either positive or negative BOLD effects could be observed depending on anatomical structure in acupuncture. Methods : 1) Subjects and paradigms: Two separate stimulation paradigms were performed in this study. To see the effects of electro-acupuncture stimulation on K7(Fuliu), the experiment was carried out on six healthy volunteers, using the gradient echo sequence with the 3.0T whole-body MRI system(ISOL). After the needle insertion on right K7(Fuliu), 2 Hz of electric stimulation was given for 30 seconds, repeated five times, with 30 seconds' intervals. During the intervals while there was no electro-stimulation, the subjects performed motor task as a reference. The image analysis including motion correction, talairach transformation, and smoothing was done using SPM99. Results: The electro-acupuncture stimulation on K7(Fuliu) activated Brodmann's Areas of 9, 19, 23, 24, 31, 32, 39 which may be the central pathways of the electro-acupuncture stimulation on K7(Fuliu) and those brain areas may be related with the function of the electro-acupuncture stimulation on K7(Fuliu).

Effect of electro acupuncture at Kugmun $(P_4)$ and Naegwan $(P_6)$ on EKG of hypertension patients (극문$(P_4)$, 내관(內關)$(P_6)$ 전침자극(電鍼刺戟)이 고혈압 환자의 EKG에 미치는 영향(影響))

  • Objectives : The purpose of this study was to investigate the Effect of Electro acupuncture at Kugmun $(P_4)$ and Naegwan $(P_6)$ on EKG of hypertension patients. Methods Subjects were ten patients who showed abnormal findings on EKG. They were divided into two Groups, Experiment Group and Control Group. Experiment Croup received electro acupuncture at Kugmun $(P_4)$ and Naegwan $(P_6)$ and Control Group at non acupoint. EKG was measured before and after electro acupuncture stimulation. Results : Five out of ten improved EKG findings by Kugmun $(P_4)$ and Naegwan $(P_6)$ and nine out of ten changed the pulse by Kugmun $(P_4)$ and Naegwan $(P_6)$. It showed significant change compared with Control Group that received electro acupuncture at non acupoint. In fact Kugmun $(P_4)$ and Naegwan $(P_6)$ is able to cure chest pain, hypertension, chest discomfort, palpitation, insomnia, headache, and so on. It proved that it has ability of controling heart function. Conclusions : In conclusion, electro acupuncture at Kugmun $(P_4)$ and Naegwan $(P_6)$ changed EKG findings and Pulse. Especially electro acupuncture at Kugmun $(P_4)$ and Naegwan $(P_6)$ decreased the pulse for tachycardia and increased the pulse for bradycardia. It is thought that it makes to consist vital homeostasis.

Effects of Electroacupuncture of GB30 on Transient Forebrain Ischemia Injury in Rats (일과성(一過性) 전뇌(前腦) 허혈(虛血) 손상(損傷)에 대한 환도(環跳) (GB30) 전침자극(電鍼刺戟)의 효과(效果))

  • Acupuncture and herbal medicine have been used to prevent and treat the cerebrovascular accident, such as a stroke, and many studies of acupuncture and moxibustion concerning to the stroke have been undertaken in the human and various animals. However, it was not published the protective effect of the electroacupuncture (EA) of Huan-do (GB30) on the transient forebrain ischemia injury. The neuroprotective effects of EA (2 ms, 10 Hz, and 1 - 2 mA) of GB30 on the transient forebrain ischemia injury was investigated by western blot of nNOS and hematoxylin and eosin stain in Sprague-Dawley rats. The transient forebrain ischemia injury resulted in increased expression of nNOS in the brain for 6 hrs after ischemia, and EA decreased significantly expression of nNOS protein in brain increased by transient forebrain ischemia injury. The survived neuronal cell stained with hematoxylin and eosin (H&E) decreased in the hippocampus by the 7 days after ischemia comparing with the normal. Comparing with the normal, the survived neurons seriously decreased cell in the hippocampus after the injury. However, the proportion of survived neurons was increased in EA treatment. These results suggest that EA of GB30 have neuroprotective effects on transient forebrain ischemia injury.

  • Objective: To investigate the analgesic effect and its serotonergic mechanism, especially related with 5-HTI and 5-HT2 receptor, of electroacupuncture(EA) in the rat model of collagen-induced arthritis(CIA). Methods : Immunization of male Sprague-Dawley rats with bovine type II (C II) collagen emulsified in Freund's incomplete adjuvant, followed by a booster injection 14 days later, leads to development of arthritis in more than 70% of rats by 21 days postinjection. After three weeks of first immunization, EA stimulation(2 Hz, 0.07 mA, 0.3 ms) was delivered into Jogsamni($ST_{36}$) for 30 minutes. Analgesic effect was evaluated by tail flick latency(TFL). We compared the analgesic effect of EA with TFLs between pretreatment of normal saline and pretreatment of spiroxatrine (5-HT1 receptor antagonist, 1mg/kg, intraperitoneal) and spiperone (5-HT2 receptor antagonist, 1 mg/kg, intraperitoneal) in CIA. Results : 1. TFLs were gradually decreased in CIA as increasing severity of arthritis. 2. Jogsamni($ST_{36}$) EA stimulation in CIA increased TFLs and the effect lasted for 60 minutes. 3. Increased TFLs with Jogsamni($ST_{36}$) EA stimulation were inhibited by pretreatment of spiroxatrine and spiperone in CIA. Conclusions : Jogsamni($ST_{36}$) EA showed analgesic effects in CIA The analgesic effects of Jogsamni($ST_{36}$) EA were inhibited by spiroxatrine and spiperone pretreatment. These observations suggest that 5-HT1 and 5-HT2 serotonergic receptor, which involve the release of serotonin neurotransmitter, play an important roles in analgesic mechanism of EA stimulation.

Heterotopic electroacupuncture modulates formalin-induced pain via descending inhibition in the rat (백서(白鼠)의 formalin 유발(誘發) 통증(痛症)에 대한 전침자극(電鍼刺戟)과 하행성 진통기전)

  • Objectives : The present study was investigated the effect and pathway of heterotopic electroacupuncture (EA) on pain induced by formalin in rats. Methods : Acupoints in the right forepaws, $HT_7$ and $PC_7$, were stimulated with 3 mA, 2 ms, and 10 Hz before subcutaneously formalin injection (5%, $50{\mu}l$) to the left hind paw. Moreover, it was investigated whether the dorsolateral funiculus (DLF), as known to the descending inhibition, mediates analgesia of the heterotopic EA, and an administration of naltrexone blocks the effect of EA. Results : In the immunohistochemistry of cFos-like protein (cFL), there were inhibitory effects of EA on the increased expression of cFL in the lumbar spinal dorsal horn neurons following formalin injection. Especially, EA inhibited the expression of cFL on the superficial laminae than that on the deep laminae at 1 hr after, but that on the deep laminae than that on the superficial laminae at 2 hr after. Also, EA suppressed the increased expression of nitric oxide (NO) and neuronal nitric oxide synthase (nNOS) in the lumbosacral spinal cord after formalin injection, but not Sham-EA. Suppressed expressions of cFL, NO and nNOS in the spinal cord were eliminated after transection of the ipsilateral DLF at $T_{10}{\sim}T_{11}$ levels. However, pretreatment of naltrexone could not prevent the suppressive expressions of cFL, NO and nNOS at the spinal cord. Conclusions : These results suggest that the analgesia of heterotopic EA may be modulated through the DLF constituting the descending inhibition.

The Effects of electroacupuncture stimuli to the Doublecortin and PSA-NCAM positive cells in the Dentate Gyrus of spontaneously hypertensive rats (전침(電鍼)자극이 SHR 흰쥐의 Dentate Gyrus에서의 Doublecortin과 PSA-NCAM 양성 신경세포에 미치는 영향)

  • Objective : This experimental in vitro study investigates the effects of electro acupuncture stimuli to the DCX and PSA-NCAM positive cells in the dentate gyrus of spontaneously hypertensive rats. Methods : We used immunohistochemical methods to observe the change of neuron cells, and then counts the immunoactive cells under the $100{\times}$ visual field optical microscope. Results : 1. The counts of DCX-positive cells in the dentate gyrus significantly increased in the 2Hz group versus control group. Otherwise 100Hz group did not have a significant differance. 2. The counts of PSA-NCAM-positive cells in the dentate gyrus rather decreased in the both 2Hz group and 100Hz group versus control group. Conclusions : We find out the electro acupuncture stimuli have some effects upon cerebral neuron cell in the dentate gyrus spontaneously hypertensive rats.

Comparative prospective short-term study on Effect of Electro-Acupuncture on thigh circumference (저주파 전침 자극이 허벅지둘레 감소에 미치는 영향에 관한 단기 연구)

  • Objectives : To compare the reduction of thigh circumference between electro acupuncture treatment group and no treatment controlled group. Method : Trial was carried out 24 volunteers for 2 weeks. Volunteers were divided into 2 groups ; electro acupuncture and very low calorie diet group(n=8) or only very low calorie diet group(n=9). The main outcome of measure was the reduction of thigh circumference(cm) per weight loss(kg). Result: The main outcome measure showed no significant difference(p=0.14). There was no significant difference in weight loss(p=0.67) either.

  • Objectives & Methods : This study was to investigate effect of low frequency electroacupuncture on NADPH-d positive neurons in the brain cortex of rat with adjuvant induced rheumatoid arthritis. Experimental groups were divided into 6 groups ; Normal, Control, $ST_{36}$, $SP_9$, $ST_{36}+SP_9$ and Non-Acupoint. Normal group, non-arthritic group, was injected normal saline, and the other groups were injected FCA. Each acupoint groups were treated by 2Hz electroacupuncture at each acupoints and NA group was treated by 2Hz electroacupuncture at non-acupoint. Each groups were evaluated by the number of NADPH-d positive neurons in primary somatosensory area(S1), secondary somatosensory area(S2), motor area and caudate putamen by using an image analyzer and a microscope. Results : 1. In S1, the number of NADPH-d positive neuron cells in the $ST_{36}$ group were significantly(p<0.05) increased compared with the control group. 2. In S2, the number of NADPH-d positive neuron cells in all electroacupuncture groups were not significantly changed compared with the control group. 3. In motor area, the number of NADPH-d positive neuron cells in $ST_{36}$ group, $SP_9$ group, NA group were significantly(p<0.05) increased compared with the control group. 4. In Caudate putamen, the number NADPH-d positive neuron cells in all electroacupuncture groups were significantly(p<0.05) decreased compared with the control group. Conclusions : Our result demonstrated that low frequency electroacupuncture on $ST_{36}$ & $SP_9$ normalized expression of NADPH-d positive neurons in the brain cortex of the rheumatoid arthritis model in rats.

Hepatoprotective effect of electroacupuncture at GB34 (Yangreungcheon) in $CCl_4$-intoxicated rats (양릉천 저주파 전침자극이 $CCl_4$로 유발된 흰쥐의 간 손상에 미치는 영향)

  • Objectives : To investigate the effect of electro-acupuncture (EA) at GB34 on hepatotoxicity in $CCl_4$-intoxicated rats. Methods : Rats were injected with $CCl_4$ and treated with acupuncture or 2 Hz electro-acupuncture (EA) at left GB34 three times a week for 10 weeks. A non-acupoint in left gluteal area was selected as a sham point. To estimate the effects of EA on hepatotoxicity in rats, body weight, liver weight and liver index were measured, and biochemical assays for serum ALT, AST, ALP and total cholesterol, and hematological analysis for RBC, WBC, PLT, hemoglobin, lymphocytes, neutrophils and monocytes, and histology analysis of liver tissue were performed. Results : 1. Lymphocyte level in blood was significantly decreased by $CCl_4$-intoxication and significantly increased by acupuncture and 2 Hz EA at left GB34. 2. Neutrophill and monocyte level in blood was increased by $CCl_4$-intoxication and significantly reduced by acupuncture and 2 Hz EA at left GB34. 3. Acupuncture and 2 Hz EA at left GB34 significantly reduced serum ALT and AST which were increased by $CCl_4$-intoxication. 4. EA at GB34 significantly reduced serum ALT and AST as compared with EA at sham point in $CCl_4$-intoxicated rat. 5. No significant difference was found between the effects of acupuncture and that of 2 Hz EA on $CCl_4$-induced liver damage in rats. Conclusions : 2 Hz EA at GB34 has hepatoprotective effects on $CCl_4$-induced liver damage in rats and the point-specificity of GB34 may be involved in these effects.
