Extraction and Bleaching of Acid- and Pepsin-Soluble Collagens from Shark Skin and Muscle (상어 껍질과 육으로부터 산 및 Pepsin 가용성 콜라겐의 추출과 탈색조건)

  • Extraction and bleaching of citric acid- and pepsin-soluble collagens (ASC and PSC, respectively) from shark (Isurus oxyrinchus) skin and muscle were investigated. The optimal sodium hydroxide concentration for extraction was 0.3 M and the optimal treatment time for removal of foreign material was 9 h. The optimal sodium hypochlorite level for bleaching of shark skin was 0.48% (w/v), and sodium hypochlorite was a better bleaching agent than acetone, hydrogen peroxide (10%, v/v), sodium sulfite (0.48%, w/v), sodium thiosulfate (0.48%, w/v), or sodium metabisulfite (0.48%, w/v). Optimal citric acid concentration and extraction time for ASC were 0.3 M and 72 h, respectively, whereas optimal conditions for extraction of PSC were treatment with 0.1 M citric acid containing 0.1% (w/v) pepsin for 24 h. Protein contents in ASSC (acid-soluble shark skin collagen), ASMC (acid-soluble shark meat collagen), PSSC (pepsin-soluble shark skin collagen), and PSMC (pepsin-soluble shark meat collagen) were 88.66%, 83.09%, 90.33%, and 84.81% (on a dry weight basis), respectively, similar to that of commercial marine collagen (88.86%). Net collagen contents of ASSC, ASMC, PSSC, and PSMC, calculated from hydroxyproline levels, were 70.31%, 25.70%, 83.09%, and 32.94%, respectively. The yields of freeze-dried ASSC, ASMC, PSSC,and PSMC were 57.22%, 53.85%, 23.28%, and 20.61%.

Supplemented Effect of Salicornia herbacea Extract Powder on Preparation and Quality Characteristics of Fermented Milk Product. (유산발효유 제조 및 품질특성에 미치는 함초 추출물 첨가의 영향)

  • Fermented milk was prepared from skim milk supplemented with Salicornia herbacea extract powder (SHEP) at the levels of 0∼0.4% (wi v) and was fermented with Lactobacillus acidophilus, Streptococcus thermophillus and Bifidobacterium longum. Quality characteristics of prepared fermented milk were evaluated for acid production, visible cell numbers, viscosity and sensory property during fermentation at 37$^{\circ}C$ for 6 hr. Supplementation of 0.1% SHEP stimulated the growth of lactic acid bacteria which showed the highest number of viable cell counts (9.23 log CFU/ml), and also enhanced the acid production which was pH 4.23 and titratable acidity 0.64%, and increased the viscosity (1,365 cps) after 6 hr incubation. The sensory scores of fermented milk supplemented with 0.1% SHEP were higher than other supplemented contents in taste, texture, flavor, aftertaste and overall acceptability. When the storage abilities of fermented milk supplemented SHEP at 6$^{\circ}C$ for 12 days were evaluated, its quality-keeping properties were relatively good in the fermented milk supplemented with 0.1% SHEP.

Optimum Fermentation Conditions and Fermentation Characteristics of Mulberry (Morus alba) Wine (오디(Morus alba) 와인의 최적 발효조건 및 발효 특성)

  • For the development of mulberry wine, we investigated its optimum fermentation conditions as well as quality changes during fermentation. The physicochemical characteristics of the mulberry fruit used in the study were pH 4.56, 0.50% titratable acidity, and 13.0 $^{circ}Brix$ soluble solids. The mulberry wine fermented with Saccharomyces cerevisiae KCCM 12224 (Sc-24) at 24 $^{circ}Brix$ soluble solids and $26^{circ}C$ showed excellent characteristics in terms of ethanol production, titratable acidity, and redness. The sucrose, fructose, and glucose contents of the mulberry wine drastically decreased with fermentation time. The citric acid content was maintained during the fermentation period, and malic acid decreased, but lactic and succinic acids increased. The cyanidin-3-glucoside content, a major anthocyanin pigment, of the mulberry wine drastically decreased from 195.5 mg% at the initial stage of fermentation to 15.37 mg% at 2 days of fermentation. However, cyanidin-3-rutinoside decreased gradually. In summary, a mulberry wine of high quality was made by fermentation for 8 days at $26^{\circ}C$ using mashed mulberry fruit containing $24^{\circ}Brix$ soluble solids, after adding 200 ppm $K_2S_2O_5$ and inoculating with 3%(v/v) Sc-24.

Analysis of Isoflavone Contents of Soybean By-products with Acid Hydrolysis Method (산 가수분해시 가열방법과 시간 및 추출조건에 따른 대두가공 부산물의 이소플라본 함량 변화)

  • To establish a rapid and effective method for the analysis of soy isoflavone which is known to have lots of variation in derivatives of glucoside, conversion rate from isoflavone conjugates to its aglycones, and decomposition of conversed aglycones were investigated with various acid hydrolysis conditions. A number of heating conditions for acid hydrolysis including heating at convection oven $(105^{\circ}C)$, water bath $(95^{\circ}C)$, heating block $(120^{\circ}C)$, and hot plate $(120^{\circ}C)$ were applied. Acid hydrolysis in heating block with reflux was chosen as the best heating condition. From the stability test of isoflavone aglycone during acid hydrolysis, genistein, daidzein, and glycitein did not show any significant changes in their contents for 60 min of hydrolysis. Ten to thirty milligram of sample per 1 mL HCl was the best ratio of sample to acid. As conclusion, acid hydrolysis for 60 min after addition of 15 mL HCl solution to 0.5 g soybean, and then fill up to 50 mL with methanol, followed by HPLC analysis was set as a final analysis method. From this method, isoflavone contents expressed as total aglycone of feed meal was the highest with content of $1288.5{\mu}g/g$ followed by those of dehulled meal.

Quality characteristics of high-acidity vinegar prepared with grape juice (포도과즙을 이용하여 제조한 고산도 식초의 품질특성)

  • This study was conducted to develop a high-acidity vinegar production (over TA 10%) technology using grape juice to reduce the importation of high-acidity vinegar. The manufacturing condition's effect on the quality of high-acidity vinegar using pure grape juice without the addition of other nutrients for fermentation was investigated. Twelve percent acidity in vinegar was obtained from grape wine with 6% alcohol content. The acetic-acid yield from grape wine decreased when the wine's initial alcohol content was high, which extended the induction time. The pH value was similar in all the treatment groups. The sugar content of the 1st-stage fermentation (1st AAF) was proportional to the initial alcohol content whereas in the 2nd-stage fermentation (2nd AAF), the sugar content was highest in the 6%-alcohol treatment. The major organic acids of the high-acidity grape vinegar included tartaric acid, malic acid, and citric acid. The acid content of the high-acidity initial alcohol group was higher than that of the low-acidity initial alcohol group due to the alcohol content added by the fed-batch and acetic-acid yield difference. The ethyl alcohol content was 364~6,091 ppm (the main alcohol while the others had only traces in all the groups). In conclusion, it was possible to manufacture 12% high-acidity vinegar without the addition of an external nutrient source to grape wine containing 6% initial alcohol content. Finally, a complementary study will be required to shorten the fermentation period through the fed-batch-style addition of alcohol for the purpose of industrialization.

Substitution effects of enzymatically saccharified Korean rice wine lees powder on skim milk in yogurt fermentation (요구르트 발효에서 효소로 당화시킨 주박 분해물의 탈지분유 대체 효과)

  • Yogurt was prepared with different substitution ratio [10, 20, 30, and 50% (w/w)] of skim milk with enzymatically saccharified Korean rice wine lees powder (eKRWLP) and fermented with commercially available mixed lactic acid bacteria (Bifidobacterium longum, Lactobacillus acidophilus, Streptococcus thermophilus) at $40^{\circ}C$ for 18 h. Fermentation characteristics were evaluated in terms of acid production (pH and titratable acidity) and viable cell counts of lactic acid bacteria. The pH of yogurts decreased with increasing eKRWLP substitution ratio of skim milk. After 12 h fermentation, titratable acidities of eKRWLP substitution and control (yogurt made without eKRWLP) were 0.84~1.04% and 0.93%, respectively. The titratable acidities of yogurts prepared with 10 and 20% substitution ratio increased than that of the control yogurt, but titratable acidities of yogurts of 30 and 50% substitution ratio decreased. After 9 h fermentation, the number of viable lactic acid bacterial cell were increased to 8.18~8.24 log CFU/g in all yogurts. In sensory evaluation, there were similar preference for eKRWLP yogurts prepared with 10 and 20% substitution ratio and the control. When eKRWLP substitution and control yogurts fermented for 9 h were incubated at $4^{\circ}C$, their pHs and titratable acidities were slightly changed but the number of viable lactic acid bacteria were well maintained above $10^7CFU/g$ for 11 days in yogurts prepared with 10 and 20% substitution ratio among eKRWLP substitution yogurts. These results suggest that eKRWLP can be used as substituent of skim milk and the optimum substitution ratio is around 10~20%.

  • A possibility of evaluation of the major internal quality factors-Brix, moisture contents, firmness and acid content in the Korean domestic 'Fuji'apple fruits by near-infrared reflectance spectroscopic (NIRS) methods were investigated. A multiple linear regression(MLR) analysis between the data obtained by physico- chemical analysis method using refractometer, freeze drier, texture analyzer and titrater and NIR spectral data was carried out to make a calibration. The standard error of prediction(SEP) of Brix, moisture, firmness and acid content were $0.50^{\circ}Brix,\;0.64%,\;0.14kg/cm^2$ and 0.07%. It is concluded that NIRS methods can be used to evaluate Brix and moisture contents of in a apple non-destructive and rapid way but the accuracy for determination of firmness and acid content was slightly low.

Quality characteristics of the bread with sprouted brown rice flour (발아현미분을 첨가한 식빵의 품질 특성)

  • Breads were prepared with the addition of sprouted brown rice flour(SBRF) as a funtional ingredient up to 50%, and their quality characteristics were evaluated through analyses of proximate composition amino acid by HPLC, fatty acid by GC, and sensory evaluation. The optimum amount of SBRF for bread preparation was 30%(w/w). The more SBRF was added, the greater the proximate components such as ash, crude protein, crude fat, fiber and amino acid content were, although the baking quality such as volume decreased. The results of sensory evaluation for bread showed that the more SBRF was added, the lower the scores of color, appearance. texture and the feelings in the mouth were, but the higher the score of taste.

A Study on the Dissolution of Aluminum Hydroxide with Mineral and Organic Acid (Aluminum Hydroxide의 유무기산(有無機酸)에 의한 용해특성(溶解特性) 연구(硏究))

  • The dissolution of domestic aluminum hydroxide of 99.7% purity has been performed with mineral and organic acid prior to the synthesis of aluminum compounds from aluminum solution. Mean particle size of aluminum hydroxide used in the work was $14.4{\mu}m$, $22.9{\mu}m$ and $62.3{\mu}m$, respectively and the effect of reaction temperature, concentration of acid and reaction time on the dissolution of aluminum hydroxide has been examined. As a result, the dissolution of aluminum hydroxide was increased with the concentration of HCl and more than 70% dissolution was obtained with 5 mole/l HCl at $70^{\circ}C$ for reaction time of 4 hr. As far as the dissolution of aluminum hydroxide with sulfuric acid was concerned, it was found that the optimum concentration of sulfuric acid was about 6 mole/l for the effective dissolution of aluminum hydroxide. When oxalic acid was used for the dissolution of aluminum hydroxide, nearly complete dissolution could be obtained by the dissolution for 16 hr with 1.0 mole/l oxalic acid at $90^{\circ}C$.

Effect of Storage Conditions on the Chemical Characteristics of Traditional Kochujang (저장조건에 따른 전통고추장의 성분 특성 변화)

  • Effect of anti-microbial agents, such as alcohol, garlic, chitosan, K-sorbate, mustard, or pasteurization on the quality of traditional kochujang was investigated during storage at $30^{\circ}C$ for 24 weeks. The pH of kochujang decreased during storage and titratable acidity changed little in the group of mustard and garlic addition. Reducing sugar increased during storage, with higher values for K-sorbate of alcohol added groups than the other groups. Alcohol contents increased during storage with higher values in control or chitosan added group. Amino nitrogen contents of kochujang decreased gradually during storage and were lower in control or chitosan added group. Ammonia nitrogen contents were lower in alcohol, garlic, K-sorbate or mustard added kochujang. The content of total free amino acid increased slightly during storage and were higher in pasteurized, garlic or alcohol added kochujang. After 24 weeks of storage, the results of sensory evaluation showed K-sorbate, garlic or alcohol added kochujang were more acceptable than control in the sense of taste.
