Inhibitory Effects of Xanthine Oxidase by Boiled Water Extract and Tannin from Persimmon Leaves (감잎 열탕 추출물 및 감잎 탄닌의 Xanthine Oxidase 저해 효과)

  • The influence of hot water extracts and tannin obtained from persimmon leaves on xanthine oxidase were investigated. Above two samples had higher inhibitory effect against xanthine oxidase by hot water extracts and tannin obtained from persimmon leaves was 92.4% and 92.1% by addition of 2.0 mg/$m\ell$ of the hot water extracts and the tannin, respectively. The inhibitions by the hot water extracts and the tannin were of competitive mode with respect to xanthine as a substrate.

  • The effects of copper on the anaerobic degradation of propionate were studied using anaerobic batch reactors. The apparent inhibitory effects of copper on the anaerobic degradation of propionate could be observed from behaviors of intermediates, ultimate methane yield(UMY) and specific methanogenic activity(SMA) There was little inhibition at the concentration of $2.5mg\;Cu^{2+}/L$. Beyond this concentration, the inhibitory effects increased with increasing dose of coppers. The 50% inhibition of UMY and SMA occurred at copper dosage of 33.8 and $24.1mg\;Cu^{2+}/gVSS$, respectively. The inhibitory effect based on the UMY was gradually reduced with the operation time dueprobably to the acclimation of microorganisms and/or binding of the added copper by ligands(and possibly ion exchange sites)contained on the cell membrane and extracellular polymer matrix whereas it based on the SMA might exclude the this phenomena. Therefore, the methodology for interpretation of inhibition data based on the SMA was more accurated than the UMY. There was no inhibitory effect in batch reactors supplemented with sulfate due to an antagonistic action of the sulfate reducing bacteria. Propionate degradation was initially retarded for copper inhibited samples but it gradually degraded afterward. Based on the mass removal considering take into account the propionate to acetate conversion, propionate degradation may appeal more affected than acetate. This result revealed that the hydrogenotrophic methanogens were the most affected by copper.

Inhibition Effect of Against Angiotensin Converting Enzyme of Flavan-3-ols isolated Korean Green Tea (한국산 녹차로부터 분리한 Flavan-3-ol 화합물의 Angiotensin Converting Enzyme 저해 효과)

  • For the Purpose of utilizing tannins in the functional foods and crude drugs, the enzyme inhibition of tannins isolated from Korean green tea were determined. Acetone extract from Korean green tea showed inhibition effect against the angiotensin converting enzyme. The galloyl tannins showed higher inhibition activity against angioteosin converting enzyme than the nongalloyl tannins. In terms of stereo isomers, (-)-epicatechins had higher inhibition activity than the (+ )-catechins. The synergistic activity was also observed. Tannins isolated from Korean green tea appeared to be incompetitive inhibitor against the angiotensin converting enzyme.

Effect of Rheum undulatum Extract on Antioxidant Activity and Activity of Matrix Metalloproteinase-1 in Human Skin Fibroblasts (대황 추출물의 항산화 활성 및 MMP-1 저해 활성)

  • Rheum undulatum L. has been commonly used as a cure for hematemesis, dropsy, and haematuria in the Oriental medicine for a long time. The main constituents of R. undulatum are chrysophanol and emodin, which are an antioxidative substance that has an anthraquinone structure. In the present study, to develop a new anti-aging agent, we examined the antioxidant activity and the inhibitory effect of the R. undulatum extract on the synthesis of MMP-1 in UVA-irradiated human dermal fibroblasts and MMP-1 activity. The R. undulatum extract was found to scavenge 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals and superoxide radicals in the xanthine/ xanthine oxidase system by a dose-dependent manner, respectively. UVA-induced MMP-1 expression was reduced about 79.5% by 1 ${\mu}g$/ml of the R. undulatum extract and also inhibited MMP-1 activity in a dose-dependent manner. In conclusion, it was observed that the R. undulatum extract has the antioxidant activity, regulation of UVA-induced MMP-1 production, and inhibition of MMP-1 activity. Therefore, these results suggest that the R. undulatum extract can be developed as a new anti-aging component of cosmetics.

Antimicrobial Activity of the Monoterpenes of Pinus Plants on Escherichia coli and Aspergillus nidulans (소나무과 식물이 지닌 Monoterpenes가 Escherichia coli와 Aspergillus nidulans의 성장저해에 미치는 영향)

  • We examined the effect of 12 main monoterpenes in Pinus plants on growth inhibition of Escherichia coli and Aspergillus nidulans. We tested four concentrations of each compound by comparing the clear zone with controls. (R)-(-)carvone, (S)(+)carvone, (1R)(-)fenchone, (-)menthone, α-pinene, (1S) (-)verbenone and (+)β - pinene had a inhibition effect on E. coli. (R)-(-)carvone, (S)(+)carvone, (+) β-pinene, geranyl-acetate, α-pinene, and (1S)(-)verbenone had inhibitory effects on the growth of A. nidulans. Geranyl-acetate inhibit growth of A. nidulans, however not to E. coli. And (1R)(-)fenchone and (-)menthone inhibit growth of E. coli, but not to A. nidulans. Myrcene, sabinene, bornyl acetate, and limonene had no inhibitory effects on E. coli and A. nidulans, eventhough at the highest concentration. All these results suggested that some selected monoterpenes had antifungal activities depend on the species of microorganism.

Inhibitive Effects of Meju Extracts Made with a Single Inoculum of the Fungi Isolated from the Traditional Meju on the Human Leukemia Cell Line (전통 메주에서 분리된 단독균으로 제조한 메주추출물의 혈액암세포에 대한 저해효과)

  • In order to study the antitumoral effect of meju extracts, which was made with a single inoculum of the microorganism, the cytotoxicity effects on several human leukemia cells such as promyelocytic leukemia cell (HL60), histiocytic lymphoma cell (U937) and acute T-cell leukemia Jurkat cell, and lymphocyte were analyzed by MTT assay. Twenty one microbes, mainly fungal genera, were isolated from Korean traditional mejus of different regions. From those collected isolates, meju was manufactured and extracted with 80% methanol, respectively. Meju methanol extracts exhibited low activites in cytotoxicity tests on HL60 cell, but high antitumoral effects of meju methanol extracts were shown on U937 and Jurkat cells. Meju methanol extracts made with a genera of Mucor, Absidia and Aspergillus showed prominant cytotoxic activities, especially. However all these extracts had no inhibitory effects on the cell growth of lymphocyte under the same conditions.

Antibrowning Effect of Licorice (Glycyrrhiza glabra) Extracts on Chopped Galic (감초추출물을 이용한 다진 마늘의 갈변 저해 효과)

  • Chemical methods using browning inhibitors have been used to control the browning of chopped garlic. However, consumer demand for natural products is increasing, and we therefore investigated the antibrowning effect of natural licorice root extract on chopped garlic. We used a combination of licorice extract and citric acid. Chopped garlic was exposed to citric acid and licorice extract at various levels and L values were measured during storage at $10^{\circ}C$. Licorice extract was effective as an initial antibrowning, and a mixture of licorice extract and citric acid was effective in inhibition of browning during storage. The effective mixing ratio of licorice extract and citric acid was 5:5 and 2:8. This indicates that licorice extract and citric acid synergistically act to inhibit browning of chopped garlic. Licorice extracts with higher levels of glycyrrhizin were more effective as antibrowning agents for chopped garlic.

Evaluation of Antioxidant, Tyrosinase and Collagenase Inhibitory of Grateloupia elliptica Extracts after Aureobasidium pullulans Fermentation (흑효모를 이용한 참도박 발효 추출물의 항산화 효과와 티로시나제 및 콜라게나제 저해효과)

  • In this experiment, the Grateloupia elliptica (G. elliptica) was fermented using the fungus Aureobasidium pullulans (A. pullulans) and its extract was obtained from hot water and 70% ethanol solution. The extract was studied for their biological activities such as antioxidant effect, Collagenase and tyrosinase inhibition in comparison to the nonfermented exatract in same solvents. Antioxidative activity test using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) showed that ethanol extract had higher antioxidant activity than water extract. Among all of the samples, the antioxidant activity of G. elliptica fermented ethanol extracts (GEFEE) was highest. Tyrosinase inhibitory activity of GEFEE was highest with 9.8% at the 1,000 ㎍/mL concentration. No inhibition of collagenase from G. elliptica water extract (GEWE) and G. elliptica fermented water extract (GEFWE) was observed, but G. elliptica ethanol extracts (GEEE) and GEFEE showed increased collagenase inhibition activity with increasing concentrations of them. Collagenase inhibitory activity of GEFEE was highest with 50.3% at the 1,000 ㎍/mL concentration. MTS cell proliferation assay was conducted with the GEWE, GEEE, GEFWE, GEFEE and cell viability was over 90% at the 10 ㎍/mL ~ 1000 ㎍/mL concentrations for all of the samples, which suggested that the extracts were noncytotoxic. In conclusion, fermented extracts of G. elliptica could be developed to bioactive functional material for cosmetics with antioxidant and wrinkle improvement effects.

  • 제품간장에 산막효모가 번식하여 제품의 품질을 제하시키는것을 방지하기 위하여 기존의 합성보존료를 대체할 수 있는 천연물질로서 allyl isothiocyanat (AITC)와 가수분해 되었을때 AITC를 생성하는 sinigrin울 주요 성분으로 함유하고 있는 고추냉이 분말의 효과를 실험하였다.AITC와 고추냉이 분말을 가열처리와 조미를 하지 않은 발표직후의 제균 생간장에 첨가하여 3$0^{\circ}C$에서 30일간 배양하면서 산막효모의 생육저해효과를 실험한 결과 AITC 20ppm과 고추냉이 분말(몇%)를 첨가한 시험구에서 산막효모의 생 이 저해되어 시험기간동안 막이 나타나지 않았다. 고추냉이 분말을 그대로 첨가하거나 간장을 반응액으로 사용한 시험군은ㄹ 고추냉이 분말을 물에 분산시켜 37$^{\circ}C$에서 반응시킨 뒤 간장에 첨가한 때보다 산막효모 저해 효과가 낮았다. 고추냉이를 물과 함께 활성화시켰을때에 비해 훨씬 많은 양의 AITC가 생성되었다. 산막 생성 저해효과 면에서 보았을때, 물로 활성화시켰을때는 간장액에서 활성화시켰을때보다 3배이상의 AITC가 생성된것으로 나타났다.로 나타났다.

Inhibitory Effect of Zizyphi fructus on ${\beta}-Glucuronidase$ and Tryptophanase of Human Intestinal Bacteria (대추의 장내세균 유해효소 ${\beta}-Glucuronidase$와 Tryptophanase 저해효과)

  • In human intestine, more than 100 species of bacteria reside and dietary factors may alter the bacterial flora which produce bacterial enzymatic activities. Especially ${\beta}-glucuronidase$ and tryptophanase activities in colon are closely associated with occurrence of colon cancer. Therefore, the inhibitory effect of traditional herbal food extracts on these intestinal bacterial enzymes are measured. The results of this study showed that Zizyphi fructus and Glycyrrhiziae radix decreased not only ${\beta}-glucuronidase$ and tryptophanase productions of human intestinal bacteria but also inhibited potently ${\beta}-glucuronidase$ and tryptophanase. Among solvent-extracted fraction of tested herbal foods, ether fraction of Glycyrrhiziae radix and ethylacetate fraction of Zizyphi fructus inhibited potently ${\beta}-glucuronidase$ and tryptophanse. Thus, ethylacetate fraction of Zizyphi fructus separated six components by silica gel column chromatography. The component having Rf=0.34 and Rf=0.43 $(developing\;solvent,\;CHCl_3/MeOH\;(3:1))$ shwed the highest inhibitory effect of ${\beta}-glucuronidase$ and tryptophanase among them.
