Studies on the Mechanism of Varietal Resistance of Rice to the Brown Planthopper(Nilaparvata lugens $ST{\AA}L$ (벼멸구에 대한 수도품종(水稻品種)의 저항성기작(抵抗性機作)에 관(關)한 연구(硏究))

  • This paper was performed to study the nature of varietal resistance of some Korean-new rice cultivars to the brown planthopper (BPH), Nilaparvata lugens ($ST{\AA}L$). The rice cultivars tested were Cheongcheong, Gaya, Hangangchal, Samgang, Nampoong and Yeongpoong which have been reported as having resistant genes for the BPH. The check varieties were Jinheung, Sangpoong and Chucheong for susceptible and IR-36 for resistant. The factors studied were referred to the seedling responses, preference in feeding and oviposition of BPH, antibiosis (nymphal development, adult emergence and sex ratio, adult body weight, population build-up, feeding amount and amylase activity), and chemical composition (inorganic components, chlorophyll contents, cell wall components, amino acids and esterase isozymes) of leaf- sheath and/or roots of rice plants. In conclusion, the natures of varietal resistance of rice cultivars to the BPH were not only correlated with the resistant reaction of rice plant, but also they were related with the non preference in feeding and oviposition and those resistant cultivars had the antibiotic effects to the insects. Their antibiotic effects of rice cultivars to the BPH would be related with some of the chemical components of rice plants, such as the contents of magnesium oxide (MgO), and chlorophyll and the different esterase isozymes.

Resistance of Soybean Cultivars to Root-Knot Nematode Species (Meloidogyne incognita and M. hapla) in Korea (뿌리혹선충(Meloidogyne incognita와 M. hapla)에 대한 우리나라 콩 장려품종의 저항성 검정)

  • Two species of root-knot nematode, Meloidogyne incognita and M. hapla, were used in tests of soybean cultivars for resistance. Sixteen soybean cultivars were planted in the 20-cm clay pot and inoculated with each of M. incognita and M. hapla. Four soybean cultivars, Eundaedu, Baegcheon, Dongbugtae and Danyeobkong were resistant; Kwangdu, Hwanggeumkong, and Buseog were moderate, and the rest nine cultivars were susceptible to M. incognita. Jangyeobkong only was resistant, Danyeobkong, Hill, Dongbutae, Hamandaerip and Chungbugbaeg were moderate, and the rest ten cultivars were susceptible to M. hapla. There were no cultivars resistant to both nematodes although Dongbugtae and Danyeobkong were resistant to M. incognita and moderately resistant to M. hapla.

Resistance of Chili Pepper Cultivars to Isolates of Phytophthora capsici (Phytophthora capsici 균주들에 대한 고추 품종들의 저항성)

  • Resistance of one hundred commercialized cultivars of chili pepper to four isolates of Phytophthora capsici was evaluated under controlled environmental conditions. The cultivars are commercialized as resistant (59%) and susceptible (41%) to Phytophthora blight in Korea. Mean disease severities of the cultivars on P. capsici MY-1, KPC-1, JHAI1-7, and KPC-7 isolates were 37, 55, 60, and 74%, respectively. In addition, 38 for MY-1, 48 for KPC-1, 56 for JHAI1-7, and 76 cultivars for KPC-7 showed susceptibility. To P. capsici MY-1, the weakest pathogenicity isolate among them, 59 cultivars represented high resistance. By contrast, only six cultivars showed high resistance to P. capsici KPC-7, the strongest isolate. Furthermore, resistance of most cultivars except for three cultivars was negatively correlated with the virulence of P. capsici isolates. And isolate-specific resistance of the chili pepper cultivars could not be found. Among them, six cultivars showing resistance to all the tested isolates were selected for further study. The development of Phytophthora blight on the six cultivars according to inoculum density ($5{\times}10^4$ to $1.5{\times}10^6$ sporangia/pot) and incubation temperature (25 to $30^{\circ}C$) after inoculation of P. capsici was tested. Resistance of the cultivars to P. capsici KPC-1 and JHAI1-7, moderately pathogenic isolates, was hardly affected. But to KPC-7 isolate, the highly resistant cultivars showed susceptiblility or moderate resistance when the seedlings were inoculated with inoculum density of $1.5{\times}10^6$ sporangia/pot and incubated at 28 to $30^{\circ}C$. From these results, it is likely that resistance of chili pepper cultivars to Phytophthora blight is affected by the virulence of P. capsici isolate.

Screening of Rice Cultivars for Adult-Plant Resistance to Pyricularia oryzae (도열병에 대해 성체식물 저항성을 지닌 벼 품종의 선발)

  • Thirty two rice cultivars which have been cultivated or used as breeding materials in Korea were tested for screening rice cultivars resistant to leaf blast at adult-plant stages in the blast nursery hill plots. When compared on the basis of disease severities in individual leaves at different growth stages of rice plants under natural field infection, the 16 indica-japonica hybrids tested were highly resistant but the 16 japonica cultivars tested showed various degrees of resistance to leaf blast. With aging of rice plants, the quantitative levels of resistance to leaf blast increased in all the cultivars, although the levels of resistance to leaf blast varied according to rice genotypes. The leaf position of rice plants in which changed from susceptible to resistant reactions varied also with rice genotypes. The susceptible reactions of the rice cultivars to Pyricularia oryzae were distinctly changed to a resistant reaction on upper leaves of rice plants. The rice cultivars, in which the quantitative level of resistance to leaf blast was higher, were resistant on the lower leaves of rice plants. The cultivars Akibare, Palkeum, Jinheung, Olchal, Dobong and Ginga which drastically decreased blast infection at late growth stage were evaluated as adult-plant-resistant to leaf blast in the field. The adult-plant-resistant cultivars became resistant to leaf blast, both qualitatively and quantitatively, as rice plants matured.

Survey of Barley (Hordeum vulgare L.) Cultivars Resistance to Mosaic Viruses and Areas Infected with Soil-borne Barley Mosaic Viruses (토양전염성 맥류바이러스 발생지역 및 맥류품종의 저항성 조사)

  • Soil-borne barley viruses were investigated in 42 regions during 1999-2003, in Korea. BaYMV, BaMMV and SbWMV symptoms were found in 37 regions. Crops resistance to mosaic viruses were investigated by growing them on 10 regions infected with soil-borne viruses. It was found that 10 unhulled barley, 6-beer bar]ey, 16 rye and 2 wheat species showed resistance to virus. Most of 15 Japanese cultivars showed infection symptoms but many of them showed relatively higher resistance in Ikasan, Youngkwang and Yesan areas region. Mixed virus infection was investigated and it was found that inter-regional species except in 10 areas, were mix infected with BaYMV and BaMMV. But in Youngkwang area all of crop species except Secheon-6 were infected only with BaYMV. Japanese cultivars were mixed infected with BaMMV and BaYMV except at Yesan that were infected only with BaYMV.

Selection of Desirable Cultivar for Organic Cultivation of Carrot (당근 유기재배를 위한 병해충 저항성 품종 선발)

  • This research was carried out to selection of desirable cultivar for organic cultivation of carrot in Korea. The assay of disease resistance, insect tolerance and rate of bolting were investigated on 32 cultivars of genetic resources of carrot. This experiment was carried out at experimental field and greenhouse of Gangneung-Wonju National University. 'PI 223360' was resistant against powdery mildew. Ten cultivars including 'Oxheart Carrot Heirloom' were moderately resistant, 22 cultivars including 'Long Impeator #58' were susceptible to powdery mildew. Although there was no resistant cultivar against black rot and leaf blight, our results showed that several cultivars have moderate resistance. Thirteen cultivars including 'Oxheart Carrot Heirloom' were having insect (Erythroplusia pyropia) tolerance and 'SA 102' and 'Scarlet Keeper Carrot Rare' were susceptible. According to the investigation of bolting, 6 cultivars including 'Hongsim Ouchon Carrot' were early bolting and 4 cultivars including 'Japanese Imperial Long Carrot' were moderate bolting. Other cultivars were identified as late bolting. From above results, we confirmed that 'Oxheart Carrot Heirloom' was suitable cultivar for organic cultivation among 32 genetic resources of carrot.

Blast Reaction of Korean Rice Cultivars against Korean and Philippines Isolates (국내 주요 벼 품종의 한국과 필리핀 도열병 균주에 대한 저항성 평가)

  • Blast resistance of one hundred and thirty-one rice cultivars bred in Korea was tested with thirty Korean isolates and twenty-two Philippines isolates using three screening methods. In the blast nursery conducted in Korea and in the Philippines, average disease index of rice cultivars were 4.6 and 2.2, respectively. Seventy-nine cultivars showed different resistance reaction in Korea and in the Philippines, and 19 cultivars showed the same resistant reaction in two locations. In the seedling test, Korean blast isolates displayed different levels of virulence. 93-093, a Korean isolate, was compatible with 90 cultivars whereas 97-057 showed a compatible reaction with 13 cultivars. Twenty-three cultivars showed high level of resistance against Korean and Philippines isolates but Chucheongbyeo, Heugnambyeo, and Manmibyeo showed susceptible reaction to all blast isolates. Through the sequential planting test in Korea and in the Philippines, Palgongbyeo and Seomjinbyeo displayed durable resistance, and Nagdongbyeo and Gihobyeo showed high level of disease infection over the planting time. These results indicate that blast isolates collected in two countries have different genetic background and number of compatible isolates should be considered in definition the durability of rice cultivar to rice blast.

Evaluation of Pepper Genetic Sources (Capsicum spp.) for Disease Resistance Breeding (병저항성 육종을 위한 고추 유전자원의 저항성 평가)

  • For initiation of resistance breeding program of the red pepper, 21 PR ($Phytophthora$ resistance) cultivars, 36 cultivars collected from USA and 'Supermanitta' which is a susceptible cultivar against phytophthora blight were assayed against phytophthora blight, powdery mildew, and anthracnose. For seedling assay of phytophthora blight, three different mating type strains of $Phytophthora$ $capsici$ were used (A1, A2, Sterile). The result showed that most of the pepper of PR cultivars were resistance or moderately resistance at each mating type. 'Yeokganghongjanggun' was resistant to all three $P.$ $capsici$ strains and 'PR-Datta' and 'PR-Manitta' were resistant or moderately resistant at each type. In case of the collected cultivars, 'NuMex J.E.Parker', 'Omni Color', and 'SCM334' were resistant to all the three types and some cultivars including 'Sweet Banana' and 'Tabasco' were moderately resistant to each type fungi. 'Orange Habanero' and 'Black Cuban' were resistant to powdery mildew and 'Supermanitta' and 'PR Keumdong' were moderately resistant, while 'Santa Fe Grande', 'NuMex Pinata' and 'Puya' were very susceptible. In the case of anthracnose, 'Aji Limon' and 'Capsicum baccatum var. pendulum 3-4' were resistant and 'Pobalno', 'Omni Color', 'Negro', 'Mesilla', 'Mulato', 'Bhut Jolokia', 'Big Dipper', 'Black Cuban', 'NuMex Pinata', and 'NuMex Big Jim' were moderately resistant. The most PR cultivars except 'Taesan' were susceptible or very susceptible. These resistant individuals identified through this experiment can be used as sources of resistance to pepper pathogens in the future breeding programs.

Varietal reaction of potato late blight in Korea (감자역병에 대한 품종간의 저항성차이에 관한 시험)

  • Fifty three varieties of potatoes were planted at Alpine Potato Research station at Takwalyong Kangwando Province since in 1962. The varieties planted were introduced front Japan, the United States and Germany. The varietal reaction was divided into five classes, and the results were following. Immune : Kennebec, Isola, Cherokee, Merrimack, Yoraku, Lisili. Cosima. Highly Resistant : Antze, Ragis Isola, Hessenkrone, Plymouth, Sebago, Pungo, Sieglinde, Anco, Essex. Resistant : Tawa, B-605-10, Sequoia, Grata, Concordia, Onaway, Holing 1. Nisego. Susceptible: Nordak, Pontiac, Benimaru, Early Gem, Chippewa, Chitose, Ohjiro, Red Bake, Norland, Katahdin. Highly Susceptible: Russet Burbunk, May Queen, Earlaine, Irish Cobbler, Feldeslohn, Red Warba, Chisago, Osseo, Paunee. Warba, Norgleam, Red Beauty, Red Burt, Danshaku, Namchak (Native variety), In general the verieties introduced from the United States and Germany were more resistant to late blight than those introduced from Japan. Especially the German varieties were immune or highly resistant to the disease except Feldeslohn. Early maturing varieties were more susceptible to late blight than the late maturing varieties in southern Korea.

Resistance Evaluation of Tomato Germplasm against Bacterial Wilt by Ralstonia solanacearum (토마토 유전자원의 Ralstonia solanacearum에 의한 풋마름병 저항성 평가)

  • This study was conducted to evaluate tomato plant resistance against bacterial wilt by Ralstonia solanacearum using tomato cultivars or tomato breeding lines maintained in RDA-Genebank of Rural Development Administration and to select resistant tomato lines for breeding purpose. We evaluated the disease responses of a total of 13 cultivars and 39 breeding lines from RDA-Genebank using R. solanacearum SL341 strain, which is a representative strain in Korea. Tomato cultivar Hawaii 7996 and Moneymaker were used as a resistant control plant and a susceptible control plant, respectively. A total of 32 cultivars were susceptible and 10 cultivars showed various disease response suggesting resistant phenotype segregation in the lines. Five commercial cultivars and 5 breeding lines exhibited strong resistance to bacterial wilt by the SL341 strain. These 5 breeding lines might be used for further study of plant defense response against bacterial wilt and cloning of the resistance gene from tomato plants. Ultimately, the selected lines could be used for tomato breeding to generate bacterial wilt resistant tomato plants.
